Low-dose H2O2 enhances taste and odor compound removal and DBP control in UV/chlorine process

Mahmoud Radwan,Zhimin Qiang,Yu Lei,Huiyu Dong,Gholamreza Moussavi,Mengkai Li

Ultraviolet/chlorine (UV/Cl 2 ) process is promising for degrading persistent micropollutants, but suffers from toxic disinfection by-products (DBPs) formation. Here, we developed a new UV/Cl 2 /H 2 O 2 process by combining UV/Cl 2 with a low dosage of H 2 O 2 , which not only enhanced micropollutant degradation but also reduced DBP formation. Taking a recalcitrant taste and odor (T&O) compound, 2,4,6-tribromoanisole (TBA) as an example. UV/Cl 2 /H 2 O 2 increased 2,4,6-TBA degradation rate constants by 90 % and 60 % than those of UV/H 2 O 2 and UV/Cl 2 , respectively, primarily due to increased HO • production. The combined contribution of UV and HO • to the degradation was substantial, accounting for 82−96 % of the overall degradation. Regarding simultaneous DBP formation from 2,4,6-TBA, UV/Cl 2 /H 2 O 2 decreased trihalomethanes (THMs) levels by 79 % compared to UV/Cl 2 . Furthermore, during post-chlorination, UV/Cl 2 /H 2 O 2 reduced the DBP formation potential by 18 % compared to UV/Cl 2 . In UV/Cl 2 /H 2 O 2 , THMs formation was reduced by 52.5 %, haloacetamides (HAMs) formation by 48.5 %, and haloacetic acids (HAAs) formation by 16.7 % compared to UV/Cl 2 using real water samples. The reduced DBP formation may result from the competitive reaction of HO • with halogenation agents and the destruction of halogenated sites. This work establishes a practical AOP that simultaneously enhances micropollutant removal and suppresses DBP formation.

The nuclear receptor REV-ERBα regulates coumarin hepatotoxicity via DBP-dependent repression of CYP2A5 in mice

Yongbin Tong,Qi Zhang,Yutong Chen,Zhiqiang Zhao,Jingqi Li,Jinyi Wang,Haishan Xia,Danyi Lu,Yuyu Liu

Introduction Coumarin exhibit various biological activities, including anti-inflammatory, antioxidant and anxiolytic effects. However, its clinical application is restricted due to significant hepatotoxicity observed in multiple animal models. CYP2A5 is the primary enzyme responsible for metabolizing coumarin in the mouse liver, homologous to human CYP2A6.Methods Acute toxicity and pharmacokinetic investigations were conducted on Rev-erbα−/− and wild-type mice following coumarin administration. The regulatory effects of REV-ERBα on CYP2A5 expression were assessed both in vivo and in vitro. Western blotting and qPCR were used to measure the protein and mRNA levels, respectively. Results Firstly, coumarin-induced hepatotoxicity was mitigated in Rev-erbα−/− mice. Secondly, the systemic exposure of coumarin was lowered in Rev-erbα−/− mice compared with wild-type mice. Thirdly, deletion of Rev-erbα resulted in significant upregulation of CYP2A5 mRNA and protein expression in mice. REV-ERBα negatively regulated CYP2A5/2A6 expression in Hepa-1c1c7 and HepG2 cells. Furthermore, luciferase reporter and chromatin immunoprecipitation (ChIP) assays indicated that REV-ERBα modulated Cyp2a5 transcription and expression through the repression of DBP.Conclusions The knockout of Rev-erbα significantly reduces the hepatotoxicity of coumarin by modulating CYP2A5. This research underscores the potential of targeting the REV-ERBα/CYP2A5 axis as a novel strategy for reducing coumarin-induced hepatotoxicity.

Comparative study of degradation of ketoprofen and paracetamol by ultrasonic irradiation: Mechanism, toxicity and DBP formation

Yu-qiong Gao,Jin-qiang Zhou,Yan-yan Rao,Han Ning,Jia Zhang,Jun Shi,Nai-yun Gao

The present study comparatively investigated the ultrasonic degradation of ketoprofen (KET) and paracetamol (PCT) in water. Ultrasonic irradiation at 555 kHz achieved rapid degradation of KET and PCT in water, the removal efficiencies of KET (2.5–80 μM) and PCT (2.5–80 μM) reached 87.7%-100% and 50.6%-86.9%, respectively, after 10 min of reaction under an ultrasonic power of 60 W. The degradation behaviors of both KET and PCT followed the Langmuir-Hinshelwood model. KET was eliminated faster than PCT because of its higher hydrophobicity. Acidic media favored ultrasonic degradation of KET and PCT. Organic compounds in water matrices exerted a great negative effect on the ultrasonic degradation rates of KET and PCT major by competing with target compounds with the generated radicals at the bubble/water interfacial region. The effects of anions were species dependent. The introduction of ClO 4 − and Cl − enhanced KET and PCT degradation to different extents, while the introduction of HCO 3 − posed a negative effect on both KET and PCT. KET and PCT degradation are accompanied by the generation of several transform intermediates, as identified via LC/MS/MS analysis, and corresponding reaction pathways have been proposed. A human umbilical vein endothelial cell (HUVEC) toxicity evaluation indicated that ultrasonic treatment was capable of controlling the toxicity of KET or PCT degradation. Of note, the enhanced formation of disinfection byproducts (DBPs), i.e., trichloromethane (TCM) and trichloronitromethane (TCNM), was found due to chlorination after ultrasonic treatment for both KET and PCT.

Effect of UV/peroxymonosulfate pretreatment on disinfection byproduct (DBP) formation during post-chlorination of humic acid

Dong Lulu,Yao Zhenxing,Sun Shaohua,Wang Mingquan,Jia Ruibao

UV/peroxymonosulfate (UV/PMS) is a promising advanced oxidation technology in water treatment. This study aimed to investigate the impact of UV/PMS on humic acid (HA) and the influence of PMS dosage, pretreatment time, pH pretreatment, nitrate, nitrite, ammonium, and bicarbonate influencing factors on disinfection byproduct (DBP) formation during post-chlorination. With increased PMS dosage or pretreatment time, the UV/PMS treatment significantly reduced ultraviolet absorbance and increased mineralization. It altered the fractional constituent as humic substances were gradually transformed into building blocks and low-molecular-weight acids. However, most DBP formation increased initially and then decreased after subsequent chlorination. Rising nitrate or nitrite concentrations markedly promoted halonitromethane (HNM) formation. The presence of ammonia had a more significant impact on dichloroacetonitrile (DCAN) formation. Bicarbonate in UV/PMS pretreatment increased carbonated disinfection byproduct (C-DBP) formation, whereas it had a negligible impact on nitrogenous disinfection byproduct (N-DBP) formation. The present study revealed the impact of a series of influencing factors on DBP formation in UV/PMS reaction systems, providing comprehensive insights on applying UV/PMS in actual practice.

Efficient Degradation of Bezafibrate Using the Fe(II)/Sulfite Process: Kinetics, Mechanism and Influence on DBP Formation

Gao Yu-qiong,Ma Jia-xin,Rao Yan-yan,Chen Ju-xiang,Gao Nai-yun

Bezafibrate (BZF), an extensively used lipid-regulating agent, has been frequently detected in aqueous environments. In this work, we systematically investigated the Fe(II)/sulfite process for degrading BZF and its impact on disinfection byproducts (DBPs) during postchlorination. Degradation conditions were optimized by adjusting the pH, sulfite concentration, Fe(II), and BZF concentration. Under the conditions of pH = 4, [BZF] 0  = 5 μM, [Fe(II)] 0  = 25 μM, and [sulfite] 0  = 250 μM, the BZF removal efficiency reaches 97.9% in 15 min. Sulfate radicals (SO 4 ●– ) and singlet oxygen ( 1 O 2 ) are recognized as the main reactive agents, with Fe(IV) also contributing to the removal of BZF. Common anions (Cl − and HCO 3 − ) and humic acid generally impede the degradation process, except that trace amounts of Cl − can slightly accelerate BZF degradation. A total of ten products are recognized by ultra high performance liquid chromatography and quadrupole time-of-flight mass spectrometry, and four major degradation pathways are proposed: hydroxylation, cleavage of amino bonds, removal of fibrate chains, and dechlorination. Meanwhile, the toxicity assessment shows that the majority of products exhibit lower biological toxicity and less bioaccumulation potential than BZF itself. The Fe(II)/sulfite pretreatment alters the DBP formation potential, especially when Br − is present. The formation of trichloromethane (TCM) is diminished following pretreatment with the Fe(II)/sulfite process, whereas a noticeable increase in the formation of dichloroacetonitrile (DCAN) is found. Moreover, Fe(II)/sulfite pretreatment enhances the formation of brominated DBPs. Therefore, special consideration should be given to DBP formation when a Fe(II)/sulfite system is employed as a pretreatment for the removal of BZF in water. Graphical abstract

Sporoderm-broken spores of Ganoderma lucidum alleviates liver injury induced by DBP and BaP co-exposure in rat

Jing Chen,Xiu He,Yawen Song,Ying Tu,Wenyan Chen,Guanghong Yang

Dibutyl phthalate (DBP) and Benzo(a)pyrene (BaP) are ubiquitous contaminants in environment and foodstuffs, which increase the chance of their combined exposure to humans in daily life. However, the combined effects of DBP and BaP on liver and the underlying mechanisms are still unclear. In this study, we explored the combined effects of DBP and BaP on liver and the potential mechanisms in a rat model. We found that DBP and BaP co-exposure activated the MyD88/NF-κB pathway through increasing TLR4 acetylation (TLR4ac) level, leading to the imbalance of pro-inflammatory factors (CXCL-13, IL-6 and TNF-α) and anti-inflammatory factors (IL-10), ultimately resulting in liver tissue damage and functional changes. Sporoderm-broken spores of Ganoderma lucidum (SSGL) had strong alleviating effects on liver injury induced by DBP and BaP co-exposure. Our study found that SSGL suppressed TLR4ac-regulated MyD88/NF-κB signaling to reduce the release of pro-inflammatory factors, and promote the secretion of IL-10, thus alleviating liver injury caused by DBP and BaP co-exposure. In conclusion, SSGL contributed to liver protection against DBP and BaP-induced liver injury in rats via suppressing the TLR4ac-regulated MyD88/NF-κB signaling.

Insights into CoFe2O4/Peracetic Acid Catalytic Oxidation Process for Iopamidol Degradation: Performance, Mechanisms, and I-DBP Formation Control

Haiwei Wu,Jiaming Zhang,Fangbo Zhao,Wei Fan,Song Yang,Jun Ma

In chlorination disinfection treatment, residual iodinated X-ray contrast media (ICMs) are the precursors to iodinated disinfection by-products (I-DBPs). This study employed CoFe2O4nanoparticle catalytic peracetic acid oxidation (CoFe2O4/PAA) to remove iopamidol (IPM) and control I-DBP formation. The experimental results demonstrated that over 90% of the IPM degradation was achieved in 40 min. The metastable intermediate (≡Co(II)-OO(O)CCH3), rather than the alkoxyl radicals, was identified as the dominant oxidation species (ROS). The electron transfer pathways between the metastable intermediate and IPM were oxygen-atom transfer and single-electron transfer. The monoiodoacetic acid formation potential (MIAAFP) was investigated. In ultraviolet-activated ClO−(UV/chlorine), a portion of I−generated through IPM dehalogenation underwent conversion to reactive iodine species (RIS), consequently elevating the MIAAFP. In CoFe2O4/PAA, the MIAAFP was less than 43% of that in UV/chlorine, which can be attributed to the complete conversion of I−into iodate IO3−without generating RIS. CoFe2O4/PAA is thus a promising treatment for removing ICMs and controlling I-DBP formation due to the efficient degradation of ICMs while avoiding the generation of RIS.

Novel insights into DBP-induced zebrafish liver inflammatory damage: Ferroptosis activating the HMGB1-TLR4-NF-κB signaling pathway

Zixu Li,Yacan Gong,Emmanuel Sunday Okeke,Dan Li,Yao Chen,Weiwei Feng,Ting Zhao,Liuqing Yang,Guanghua Mao,Xiangyang Wu

Typical plasticizer dibutyl phthalate (DBP) has been demonstrated to induce hepatotoxicity in zebrafish, but the underlying molecular mechanisms remain incompletely elucidated. Numerous studies have shown that ferroptosis is involved in the pathophysiological progression of hepatic disease. However, it remains unclear whether ferroptosis is involved in the DBP-induced hepatotoxicity in zebrafish. Initially, histopathological analyses have preliminarily confirmed that DBP can activate inflammatory responses in the zebrafish liver. Further investigation revealed that DBP induces ferroptosis in the zebrafish liver, characterized by iron overload, lipid peroxidation, and aberrant activation of ferroptosis pathways. Furthermore, DBP exposure induced ferroptosis, disrupting cell membranes and subsequent release of HMGB1, which are sensed by immunocytes TLR4/NF-κB signaling pathways, thereby activating the innate immune response in a context-dependent manner. Moreover, the ferroptosis inhibitor Fer-1 effectively rescues the activation of HMGB1-TLR4/NF-κB-mediated immune processes. Overall, this work enriches the molecular mechanism of DBP-induced zebrafish liver inflammatory damage and provides a reliable biomarker for future environmental risk assessment of DBP.

From Toxic Pollutant to Catalyst: One-Pot Synthesis of Fe3O4-Ni-DBP/NF for Efficient Overall Water Splitting

Qingyuan Song,Jinshou Yao,Liujun Jin,Peiyang Gu,Ye Wang,Ping Liu,Congping Chen,Guohong Dai

Dibutyl phthalate (DBP), a ubiquitous plasticizer pollutant, poses severe risks to environmental and human health, yet its high chemical stability makes valorization highly challenging. Herein, we report a one-pot, mild, and green strategy that directly upcycles DBP as an organic ligand to construct a ferroferric oxide functional Ni-based MOF framework nanocomposite (Fe3O4-Ni-MOF(DBP)) on nickel foam (NF), enabling the simultaneous detoxification of DBP and fabrication of a high-performance electrocatalyst. The in-situ integration of Fe3O4 nanoparticles with the Ni-MOF(DBP) endows the hybrid catalyst with accelerated charge-transfer kinetics, enhanced electrical conductivity, and abundant accessible active sites, leading to remarkable bifunctional activity for overall water splitting. In an alkaline electrolyte, Fe3O4-Ni-MOF(DBP)/NF exhibits outstanding bifunctional electrocatalytic performance, delivering low overpotentials of 233 mV for the OER and 42 mV for the HER at 10 mA cm -2 , along with excellent durability over continuous operation exceeding 40 h. In-situ Raman analysis reveals that the superior catalytic performance of Fe3O4-Ni-MOF(DBP)/NF is attributed to the dynamic reconstruction of surface metal sites into active (oxy)hydroxide intermediates. This work presents an innovative strategy for the valorization of toxic plastic pollutants, thus advancing the cross integration of environmental remediation and clean energy technologies.

Impacts of permanganate/bisulfite pre-oxidation on DBP formation during the post chlorine disinfection of ciprofloxacin-contaminated waters

Guiqiao Wang,Weiye Shi,Defang Ma,Baoyu Gao

Bisulfite-activated permanganate (PM/BS) oxidation process can oxidize ciprofloxacin in complex water matrices rapidly. However, effects of PM/BS pre-oxidation on the formation of disinfection byproducts (DBPs) during post-chlorination of ciprofloxacin-contaminated waters need to be addressed. This study investigated the formation of trihalomethanes (THMs), haloacetonitriles (HANs), haloketones and trichloronitromethane during chlorination of ciprofloxacin-contaminated humic acid (HA), bovine serum albumin (BSA) and alginate solutions, and revealed the effects of PM/BS pre-oxidation on ciprofloxacin degradation and DBP formation during post-chlorination, considering the presence of Br − . Only THMs and HANs were quantifiable. THMs were the most abundant. Ciprofloxacin-contaminated HA exhibited the highest formation potential of DBPs and integrated toxic risk value (ITRV). In the absence of Br − , PM/BS pre-oxidation reduced or hardly affected the toxicity risks derived from DBPs formed from the post-chlorination. However, the presence of Br − greatly reduced the degradation of ciprofloxacin (30–50%) in various waters. In the ciprofloxacin-contaminated waters containing Br − , the total ITRVs of DBPs formed from post-chlorination increased by 60%–800% with PM/BS pre-oxidation, attributing to the enhanced formation of DBPs especially bromochloroacetonitrile and dibromoacetonitrile. Overall, PM/BS is a potential pre-oxidation technology for the treatment of ciprofloxacin-contaminated waters without bromide.

Insight into PPCP degradation by UV/NH2Cl and comparison with UV/NaClO: Kinetics, reaction mechanism, and DBP formation

Simiao Li,Xiuwei Ao,Chen Li,Zedong Lu,Wenfeng Cao,Fangfang Wu,Shuming Liu,Wenjun Sun

The UV/NH 2 Cl process is an emerging advanced oxidation process (AOP) that is greatly effective in degrading pharmaceuticals and personal care products (PPCPs). However, detailed information regarding the process is lacking. The degradation of ibuprofen (IBP, an electron-withdrawing PPCP) and naproxen (NPX, an electron-donating PPCP) in UV/NH 2 Cl and UV/NaClO processes was performed to investigate the applicability and security of the UV/NH 2 Cl process and compare with those of UV/NaClO. UV/NH 2 Cl was effective in degrading both IBP and NPX and the degradation followed pseudo-first order kinetics (k IBP  = 0.0037 cm 2 /mJ and k NPX  = 0.0044 cm 2 /mJ). This indicated the broad applicability of UV/NH 2 Cl to different kinds of PPCPs. Ranges of values of UV intensity (0.3–1.0 mW/cm 2 ) and pH (6.0–8.0) showed little effect on the degradation of PPCPs by UV/NH 2 Cl based on UV Dose but HCO 3 − (2–8 mM), natural organic matter (NOM, 2–8 mg/L), and the natural water matrixes were inhibitory. Increasing the dosage of NH 2 Cl from 0.15 mM to 0.75 mM, resulted in an even increase of k IBP ; however, k NPX increased slowly after 0.3 mM NH 2 Cl. Mechanism experiments involving nitrobenzene showed that •OH was the major radical involved in degrading IBP and NPX via UV/NH 2 Cl. The electron spin resonance spectroscopy and kinetic modeling results also indicated the larger amount of •OH and weaker reactive chlorine species (mainly ClO• and ClO 2 •) in UV/NH 2 Cl compared with UV/NaClO. Compared to UV/NaClO in synthetic and natural water, UV/NH 2 Cl was a more stable degrader with little pH- and substrate-dependence, while UV/NaClO preferred degrading the electron-donating PPCP and at low pH. The UV/NH 2 Cl produced less halogenated disinfection byproducts (DBPs) (even nitrogenous DBPs) and was less cytotoxic theoretically than UV/NaClO based on the DBPs included in this study. Thus UV/NH 2 Cl process may be an effective AOP for water treatment .

Endophytic Phthalate-degrading Bacillus subtilis N-1-gfp colonizing in soil-crop system shifted indigenous bacterial community to remove di-n-butyl phthalate

Li-Hui Liu,Jia-Yan Zhang,Guang-Xuan Tang,Yu-Hong Huang,Xiang-Qing Xie,Jun Geng,Hui-Xiong Lü,Hui Li,Yan-Wen Li,Ce-Hui Mo,Hai-Ming Zhao,Quan-Ying Cai

Endophytic bacteria can degrade toxic phthalate (PAEs). Nevertheless, the colonization and function of endophytic PAE-degrader in soil-crop system and their association mechanism with indigenous bacteria in PAE removal remain unknown. Here, endophytic PAE-degrader Bacillus subtilis N-1 was marked with green fluorescent protein gene. Inoculated strain N-1- gfp could well colonize in soil and rice plant exposed to di-n-butyl phthalate (DBP) as directly confirmed by confocal laser scanning microscopy and realtime PCR. Illumina high-throughput sequencing demonstrated that inoculated N-1- gfp shifted indigenous bacterial community in rhizosphere and endosphere of rice plants with significant increasing relative abundance of its affiliating genus Bacillus than non-inoculation. Strain N-1- gfp exhibited efficient DBP degradation with 99.7% removal in culture solutions, and significantly promoted DBP removal in soil-plant system. Strain N-1- gfp colonization help plant enrich specific functional bacteria (e.g., pollutant-degrading bacteria) with significant higher relative abundances and stimulated bacterial activities (e.g., pollutant degradation) compared with non-inoculation. Furthermore, strain N-1- gfp displayed strong interaction with indigenous bacteria for accelerating DBP degradation in soil, decreasing DBP accumulation in plants and promoting plant growth. This is the first report on well colonization of endophytic DBP-degrader Bacillus subtilis in soil-plant system and its bioaugmentation with indigenous bacteria for promoting DBP removal.
