Xanthostemon chrysanthus (F.Muell.)
Benth. is
a species from the Myrtaceae family with limited
phytochemical and pharmacological studies. In this study, ten compounds
( 1 – 10 ) were isolated and purified
from X. chrysanthus , among which compound 2 , identified as des-O-methyllasiodiplodin (DML), exhibited the most
potent anti-inflammatory activity. This is the first report of DML
being isolated from this species. DML significantly inhibited lipopolysaccharide
(LPS)-induced inflammation in RAW264.7 macrophages by reducing the
expression of TNF-α, IL-1β, and IL-6, as well as nitric
oxide (NO) and reactive oxygen species (ROS) production. Moreover,
DML promoted autophagy by increasing LC3 II/LC3 I conversion and decreasing
p62 expression, accompanied by suppression of the PI3K/AKT/mTOR signaling
pathway. The autophagy inhibitor chloroquine (CQ) reversed these effects,
confirming that DML exerts its anti-inflammatory action through autophagy
activation. In a mouse ear edema model, DML effectively reduced inflammation,
showing even greater efficacy than indomethacin. Collectively, this
study provides the first evidence that DML isolated from X. chrysanthus exerts strong anti-inflammatory effects by promoting
autophagy and modulation of the PI3K/AKT/mTOR pathway. These findings
enrich the chemical and pharmacological understanding of X. chrysanthus and highlight DML as a promising natural candidate
for the development of novel anti-inflammatory therapeutics.

Efficient separation of cellulose from lignocellulosic biomass is essential for the development of sustainable cellulose-based materials. In this study, a two-step deep eutectic solvent (DES) pretreatment strategy was applied to selectively remove hemicellulose and lignin, thereby obtaining cellulose with high purity and structural integrity. The DES-treated residue exhibited an increased crystallinity index (55.72% → 74.36%) and specific surface area (0.5931 → 2.3452 m 2 /g), confirming the disruption of the dense biomass structure and the degradation of polysaccharides in non-crystalline regions. The recovered lignin showed lower molecular weight (Mw = 2972 g/mol, Mn = 1401 g/mol) and better homogeneity (PDI = 2.12). The purified cellulose was subsequently employed to fabricate cellulose-based films using [Emim][OAc]/DMSO (1:4) as solvent. The films exhibited excellent properties, including tensile strength of 25.6 ± 1.5 MPa, elongation at break of 40.5%, high transparency (90.73%), water content of 67.47%, and weight loss rate of 63.94% after 30 days. Structural analysis revealed compact crystalline regions, good compatibility between solvent and cellulose, and stable thermal and interfacial properties. These results demonstrate that dual DES pretreatment enables high-purity cellulose isolation and facilitates the preparation of biodegradable films with promising potential in agricultural applications.

A DES-guided self-assembled polypeptide system (DAC-SAP/BGP) enables synergistic neuromuscular signaling inhibition. • MD simulations reveal dual permeation mechanisms: nanostructure reduces resistance; DES enhances lipid mobility. • DAC-SAP/BGP achieves 42% transdermal penetration at 48 h, 4.53-fold higher than CTX alone. • Inhibits Nav1.4 (94.1%) and Cav1.1 (60%), reduces ACh; 28-day trial improves skin elasticity by 10.86%. Skin aging involves complex mechanisms such as overactivation of neuromuscular signals and oxidative stress. Herein, we developed a synergistic transdermal delivery system combining self-assembled tripeptide nanoparticles (DAC-SAPs) and a ternary deep eutectic solvent (DES, betaine-glycerol-propylene glycol, BGP) for anti-aging. DAC-SAP integrates dipeptide diaminobutyryl benzamide diacetate (DDB), argireline (ARG), and μ-conotoxin (CTX) into a single nanostructure through hydrophobic interactions, hydrogen bonds, and electrostatically driven molecular self-assembly. This spatial co-localization ensures synchronous delivery and spatiotemporal synergy, effectively inhibiting neuromuscular signaling. BGP DES optimizes polarity regulation, destroys stratum corneum lipids, and significantly promotes transdermal penetration. Molecular dynamics (MD) simulations reveal a dual permeation-promoting mechanism, with an appropriate nanostructure size reducing transmembrane resistance, whereas DES promotes lipid mobility and weakens tight junctions in the stratum corneum (SC). In vitro, in vivo, and exploratory clinical assessments demonstrate that DAC-SAP/BGP significantly inhibits sodium and calcium channel activity, reduces acetylcholine (ACh) secretion, and improves skin elasticity and wrinkle parameters. This multicomponent system achieves a synergistic anti-aging effect by blocking and enhancing the transmission of neuromuscular signals, thus providing a new strategy for anti-aging skin care. To enable the efficient extraction of ursolic acid, a non-polar bioactive compound, the applicability of alcohol-based deep eutectic solvents (DESs) was investigated. A series of DESs were prepared by combining a fixed hydrogen bond acceptor, choline chloride (ChCl), with various hydrogen bond donors (HBDs) such as glycerol, ethylene glycol, 1,3-propanediol, 1,2-propylene glycol, and 1,4-butanediol at different molar ratios. To evaluate their potential as green solvents, the cytotoxicity of the alcohol-based DESs was assessed, and the 1,2-propylene glycol-based DESs exhibited the lowest cytotoxicity. Due to the high viscosity of DESs, direct solubility measurements of ursolic acid were limited; thus, the COSMO-SAC model was employed to predict solubility and guide the screening of suitable DESs. Computational results suggested that DESs containing 1,2-propylene glycol and 1,4-butanediol provided superior solubility for ursolic acid. Based on both cytotoxicity and predicted solubility, the DES composed of ChCl and 1,2-propylene glycol at a 1:4 molar ratio was identified as the most biocompatible solvent system. Using this DES, ursolic acid was successfully extracted from rosemary and apple peel. Experimental validation confirmed that PG-based DES (ChCl:1,2-propylene glycol = 1:4) exhibited a solubility 70% lower for ursolic acid than pure ethanol. This study supports a strategic shift toward sustainable extraction technologies using DESs and provides a foundation for future research on their industrial applicability.

To achieve efficient lignin removal while preserving cellulose, ternary deep eutectic solvents (DESs) incorporating Lewis acids were prepared for the microwave-assisted pretreatment of corn stover. The effects of Lewis acid types and pretreatment conditions on delignification were investigated, and the structural characteristics of corn stover samples before and after pretreatment were characterized. The results indicated that the ternary DES composed of choline chloride (ChCl), FeCl 3 , and ethylene glycol (EG), denoted as ChCl-FeCl 3 -EG, exhibited optimal performance. Under the conditions of a pretreatment time of 41 min, a temperature of 134 °C, a liquid-solid ratio of 13.80, and a Lewis acid-to-ChCl molar ratio of 0.086, the removal rates of lignin and hemicellulose reached 92.24% and 87.99%, respectively, while the cellulose retention rate reached 94.03%. Compared with traditional oil bath heating, microwave assistance not only enhanced lignin removal efficiency but also significantly shortened the processing time. Furthermore, the mechanism of the ChCl-FeCl 3 -EG pretreatment process for corn stover was analyzed. The DES system achieved efficient separation of corn stover components through the synergistic effects of “acid catalysis, hydrogen bond disruption, and selective dissolution,” providing a green pretreatment pathway for biorefining.

Phytoremediation is an in situ environmentally friendly and economically feasible remediation technology to treat heavy metal contaminated soil. However, the obtained heavy metal enriched biomass is treated as hazardous waste and disposed of traditionally via incineration, posing risks of secondary pollution. In this work, we developed a novel DES-based pretreatment and metal extraction process to enable the valorization of phytoremediation biomass such as genetically modified poplar as the potential feedstock for biomass refining. The results showed that the DES (choline chloride/lactic acid) could remove around 98.3% Cd accumulated in poplar as well as a notable amount of lignin, resulting in a cellulose-rich pulp with trace heavy metals and Cu-containing lignin nanoparticles with a high phenolic hydroxyl content. Moreover, the electrochemical properties of this DES were analyzed. The excellent electrochemical stability and high conductivity of this DES enable its cleaning up via electrochemical deposition and recycling. Furthermore, this DES-based treatment technology has been demonstrated successfully for the hyperaccumulator Sedum alfredii, providing evidence for its wide applicability and potential for scaling-up.

Deep eutectic solvents (DESs) have emerged as promising green media for lignocellulosic biomass fractionation. However, lignin condensation during processing remains a major bottleneck, hindering efficient separation and downstream valorization. Here, a novel ternary DES composed of p -toluenesulfonic (TsOH), choline chloride (ChCl), and 4-methylguaiacol (4-MGA, a lignin-derived phenolic monomer) was developed to address this challenge. The incorporation of 4-MGA provided dual functionality: it not only promotes lignin dissolution in DES by forming hydrogen bonds and π-π interactions, but also inhibits the condensation of lignin macromolecules via an arylation strategy. Under mild conditions (90 °C, 1 h), the DES system achieved 90.1% delignification and 89.7% hemicellulose removal from Eucalyptus wood, while preserving 90.8% of the cellulose. Furthermore, the directional arylation of lignin mediated by 4-MGA offers a strategy with the potential to circumvent isomer interference in the catalytic preparation of bisphenols. This work demonstrates a rational DES design strategy for enhancing lignin extraction efficiency and opens new avenues for high-value lignin utilization in biorefinery applications.

In this study, a green catalytic strategy based on a deep eutectic solvents (DES) was developed for the efficient synthesis of urea diacetic acid. The effect of the formation of DES with different metal salts (ZnCl 2 , FeCl 3 , CuCl 2 , ZnBr 2 ) was systematically evaluated by combining experimental analysis and molecular thermodynamic simulation. The results showed that the DES (ΔG = −315.06 kJ/mol) formed in a 1:2 mol ratio between urea and ZnCl 2 showed the optimal catalytic activity at 353.15 K, with the product yield as high as 91.50 %, which was significantly better than the traditional non-catalytic system (35.59 %). IR and 1 H NMR spectroscopy confirmed the characteristic peaks of the secondary amine and the carboxylic acid group in urea diacetic acid, and the structural integrity of the product was further verified by the Hinsberg reaction. Mechanistic studies show that DES reduces the reaction activation energy through a dynamic hydrogen bonding network, while the electronic effect of metal salts regulates intermediate release. The urea-ZnCl 2 DES system has high catalytic efficiency and environmental friendliness under mild conditions (80℃), and can be recycled, which provides a new idea for the green industrial production of polyurea materials. This study provides the theoretical basis and practical reference for the multifunctional catalytic application and green chemical process design of DES in organic synthesis.

To expand the field of high-value utilization of lignin. The degraded deep eutectic solvent lignin-grafted poly (N-Vinyl caprolactam) (DES-lignin-g-PNVCL) was synthesized by modified DES-lignin and NVCL via the combination of activators regenerated by electron transfer-atom transfer radical polymerization (ARGET-ATRP). Fourier transform infrared spectroscopy (FT-IR), 1 H NMR, X-ray electron spectroscopy (XPS), dynamic light scattering (DLS), differential scanning calorimeter (DSC) were used to characterize the structure and performance of DES-lignin-g-PNVCL. The results indicated that the PNVCL and DES-lignin-g-PNVCL were successfully prepared by ARGET-ATRP. The lowest critical solution temperature (LCST) of PNVCL was 35.75 °C. Due to different strength of hydrogen bond , different energies were required, so the LCST of the polymer can be regulated. When the molar ratio of phenolic hydroxyl group in degraded DES-lignin to 2-bromoisobutyryl bromide (BiBB) was increased from 1:1 to 1:7, the grafting rate of DES-lignin-Br was increased from 32.87% to 60.84%, and the LCST of DES-lignin-g-PNVCL was decreased from 47.98 °C to 27.88 °C. The LCST of DES-lignin-g-PNVCL was increased from 30.98 °C to 44.64 °C when the addition amount of DES-lignin-Br was increased from 20 mg to 200 mg. The LCST of DES-lignin-g-PNVCL was increased from 27.20 °C to 39.86 °C when the ratio of DMF/water was increased from 1:4 to 4:1. The LCST of DES-lignin-g-PNVCL was decreased from 52.10 °C to 31.02 °C when the concentration of DES-lignin-g-PNVCL was increased from 0.5 mg/mL to 2.5 mg/mL. The equation represented the relationship between LCST and influencing factors was obtained, the good predictability provided a tactics for preparing desired LCST thermo-responsible polymer.

Phenolic wastewater, a type of organic refractory wastewater containing chemical poisons such as phenol, have made a great contribution to human health and the social environment. Therefore, it urgently desires to develop efficient treatment approach for phenolic wastewater. The deep eutectic solvents (DES) provide an attractive option for extracting phenolic compounds. In this work, a series of ammonium-based DES were synthesized and were characterized by in situ IR, FT-IR, and TG-DTG. The extraction conditions of phenolic wastewater were optimized including the kind of DES, temperature, and wastewater concentration. The ability to extract phenolic compounds (phenol and p -chlorophenol) from wastewater by using DES was evaluated. The best experimental conditions for TBAC/Gly DES are temperature of 30 °C, stirring time of 30 min, concentration of 350 mg·L −1 , pH between 2 and 6, and volume ratio V W : V DESs of 2. The mechanism of extract phenolic compounds using TBAC/Gly DES was investigated.

Chiral separation of D- and L-Serine is of great value in the fields of biopharmaceuticals. For this purpose, a D-Serine imprinted polymer was prepared by precipitation polymerization with the aid of deep eutectic solvent (DES) and crowding agent (polymethyl methacrylate, PMMA). The preparation conditions were optimized by a L 9 (3 4 ) orthogonal design. Structure characterization was performed by the FTIR, SEM, AFM and XPS techniques and adsorption selectivity and separation efficiency for this imprinted material investigated. The results showed that under the conditions of pH 5.0 and 303 K, the polymer exhibited maximum adsorption capacity, reaching 121.4 mg g -1 , with a selectivity factor as high as 11.87. When this D-serine-imprinted material was used as a chromatographic stationary phase to separate a mixture of D- and L-serine, the resolution achieved was 1.765, demonstrating high chiral separation efficiency. The high adsorption selectivity for the novel imprinted material might result from the synergistic effect of deep eutectic solvent with macromolecular crowding reagent during the preparation of the imprinted materials. One hand, deep eutectic solvent can stabilize the zwitterionic form of D-Serine ( + H 3 N-CH(CH 2 OH)-COO - ) and hence enhance the ionic interactions between the template and functional monomers and the stability of template-functional monomer complexes; on the other hand, crowding agents significantly improved the binding efficiency between the functional monomer and the template molecules during the pre-assembly stage through the "excluded volume effect". This synergy promotes to form higher density and more precise specific recognition sites during the synthesis of the imprinted polymer and to improve the adsorption selectivity of the material obtained.
