Effects of gallic acid alkyl esters and their combinations with other antioxidants on oxidative stability of DHA algae oil

Yan Shen,Chao Guo,Ting Lu,Xu-Yang Ding,Man-Tong Zhao,Min Zhang,Hui-Lin Liu,Liang Song,Da-Yong Zhou

The most effective composite antioxidants for DHA algae oil were optimized by combining the selected gallic acid (GA) alkyl ester with other commonly used antioxidants. Results of Rancimat induction time, peroxide value, thiobarbituric acid-reactive substances, and free radical generation indicated that octyl gallate (OG) was the best one in DHA algae oil among GA alkyl esters with various chain lengths. Therefore, OG was used to combine other antioxidants (antioxidant of bamboo leaves, rosemary extract, tea polyphenols, tea polyphenol palmitate (TPP), ascorbyl palmitate, vitamin E, phytic acid and phospholipid) for further improving the oxidative stability of DHA algae oil. The combination of OG + TPP showed the best antioxidant effect among the composite antioxidants of two and three components. Through optimization of mixture ratio, the combination of 53.20 mg/kg OG + 360 mg/kg TPP demonstrated the best antioxidant capacity, which prolonged the shelf life of DHA algae oil by 4.24 folds.

Integrative metabolomics and gut microbiota analyses reveal the protective effects of DHA-enriched phosphatidylserine on bisphenol A-induced intestinal damage

Qiaoling Zhao,Fei Yang,Qiuyan Pu,Rui Zhao,Su Jiang,Yunping Tang

The protective effects of docosahexaenoic acid-enriched phosphatidylserine (DHA-PS, 50 or 100 mg/kg) on bisphenol A (BPA)-induced intestinal damage were assessed. The biochemical indices, pathological examination, non-targeted metabolomics integrated with gut microbiota analysis, and immunofluorescence analysis were used to investigate the protective effects of DHA-PS and its underlying regulatory mechanism. The DHA-PS treatment improved the pathology of intestinal tract and increased the expression levels of Claudin-1, Occludin, and ZO-1. In addition, the DHA-PS treatment also notably decreased the levels of interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α, and increased the antioxidant enzymes activities. Moreover, DHA-PS treatment increased the relative abundances of Akkermansia, Alistipes, Butyricicoccus, Coriobacteriaceae_ UCG-002 , Enterorhabdus , and Lachnospiraceae_ UCG-006. DHA-PS treatment alleviated BPA-induced intestinal damage by regulating the kynurenine pathway of tryptophan, lipid, and arachidonic acid metabolisms. Overall, this study suggested that DHA-PS could alleviate BPA-induced intestinal damage by enhancing the intestinal barrier integrity, improving gut microbial composition and metabolites, and inhibiting the TLR4/NF-κB pathways.

Transesterification of phosphatidylcholine with DHA-rich algal oil using immobilized Candida antarctica lipase B to produce DHA-phosphatidylcholine

Liwen Shu,Xin Zheng,Shuhua Qi,Shuizhi Lin,Yinghua Lu,Chuanyi Yao,Xueping Ling

Docosahexaenoic acid (DHA) enriched with phospholipids (PLs) (DHA-PLs) is a type of structured PL with good physicochemical and nutritional properties. Compared to PLs and DHA, DHA-PLs has higher bioavailability and structural stability and many nutritional benefits. To improve the enzymatic synthesis of DHA-PLs, this study investigated the preparation of phosphatidylcholine (PC) enriched with DHA (DHA-PC) via enzymatic transesterification of algal oil, which is rich in DHA-triglycerides, using immobilized Candida antarctica lipase B (CALB). The optimized reaction system incorporated 31.2% DHA into the acyl chain of PC and converted 43.6% PC to DHA-PC within 72 h at 50 °C, 1:8 PC: algal oil mass ratio, 25% enzyme load (based on total substrate mass), and 0.02 g/mL molecular sieve concentration. Consequently, the side reactions of PC hydrolysis were effectively suppressed and products with high PC content (74.8%) were produced. Molecular structure analysis showed that exogenous DHA was specifically incorporated into the sn-1 site of the PC by immobilized CALB. Furthermore, the evaluation of reusability with eight cycles showed that the immobilized CALB had good operational stability in the present reaction system. Collectively, this study demonstrated the applicability of immobilized CALB as a biocatalyst for synthesizing DHA-PC and provided an improved enzyme-catalyzed method for future DHA-PL synthesis.

Glycerol-Regulated Water Content in DHA Algal Oil Emulsions: Effects on Emulsion Stability and Oxidative Stability of DHA Algal Oil

Li Ma,Dian Liu,Xiaoqing Li,Liqiang Zou,Qin Wang,Wei Liu

This study constructed DHA algae oil emulsions using sodium caseinate (NaCS) as an emulsifier and by controlling the mass fraction of glycerol in the aqueous phase (0–90 wt%). The differences of physicochemical properties, oxidative stability, and bioaccessibility were systematically investigated. The findings indicated that the incorporation of glycerol significantly enhanced the emulsion performance. The emulsion with high glycerol content (90 wt%) exhibited the smallest droplet size (185.6 ± 4.05 nm), the highest apparent viscosity and shear modulus, demonstrating weak gel characteristics, and maintained excellent stability within the pH range of 3–8 and under centrifugal conditions. By inhibiting the dissolved oxygen in the aqueous phase and delaying oxygen diffusion, the glycerol emulsion significantly slowed down the oxidation process of DHA algae oil, with lower primary oxidation products (POV) and secondary oxidation products (TBARS) compared to the conventional emulsion (0 wt% glycerol). Additionally, it inhibited the oxidation of tryptophan and the formation of Schiff bases in NaCS, effectively suppressing protein oxidation. In vitro simulated digestion experiments showed that the DHA bioaccessibility of all emulsions was above 85%, which was three times higher than that of pure algae oil (27.8%), and the glycerol content had no significant effect on it. This study confirms that the low-water glycerol–NaCS emulsion system can synergistically enhance the oxidative stability and bioavailability of DHA algae oil, providing a new strategy for developing long-lasting and stable functional lipid delivery systems.

DHA emulsion-assisted gel encapsulation enhances probiotic stress resistance

Ruixin Lai,Yue Wu,Mengyuan Zhang,Yucheng Lv,Jiahui Zhao,Yichen Li,Jianguo Zhu,Yanan Li

Innovatively proposed the idea of using long-chain fatty acids beneficial to the human body, such as docosahexaenoic acid (DHA), to fill the three-dimensional network structure of hydrogels. • DHA oil-in-water emulsions can enhance the viability of probiotics under simulated industrial processing conditions. • DHA oil-in-water emulsions can effectively fill the pores in the hydrogel network. • Modified hydrogels prepared using DHA emulsions demonstrate significantly superior encapsulation performance for embedded probiotics compared to unmodified traditional hydrogels. • Modified hydrogels provide significantly better protection for embedded probiotics in simulated gastric and intestinal fluids than traditional hydrogels. Although gel encapsulation is widely used in the delivery of probiotics, its three-dimensional porous structure has inherent drawbacks, probiotics may leak out through the pores, and gastrointestinal stress factors may also penetrate into the gel through the pores. In this study, we prepared a docosahexaenoic acid (DHA) oil-in-water emulsion (DHA-ME) and evaluated its effects on probiotics, showing that the 0.0075% (w/v) DHA-ME improved the survival rate of probiotics against the heat and freeze-drying stress. We then incorporated this into a gelatin-gellan gum gel to prepare a modified gel (GDG). Taking Bifidobacterium longum subsp. longum BL21 as an example, the addition of DHA-ME yielded a retention rate of nearly 100% in simulated intestinal fluid after 24 h. Moreover, following incubation in simulated intestinal and gastric fluids, BL21 activity increased by 25.3% and 657.5%, respectively. This strategy offers a new approach to the delivery of probiotics via gels.

A comprehensive study of lipid profiles of round scad (Decapterus maruadsi) based on lipidomic with UPLC-Q-Exactive Orbitrap-MS

Chen He,Zexin Sun,Xingchen Qu,Jun Cao,Xuanri Shen,Chuan Li

Round scad ( Decapterus maruadsi ) is rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). However, there is no comprehensive information covering its lipidomic profile. In this study, lipid profiles including fatty acid composition, distribution and detailed structure information were explored using gas chromatography-flame ionization detector (GC-FID) and ultra-high-performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry (UPLC-Q-Exactive Orbitrap-MS). The results showed that triacylglycerol (TG) was the dominant class, and the EPA and DHA were concentrated as glycerophospholipid (GP). The saturated fatty acids and monounsaturated fatty acids were mainly esterified in positions 1 and 3 of TG and sn -1-position of GP, while the polyunsaturated fatty acids (PUFA) were mainly esterified in sn -2-position of TG and GP. A total of 1282 species from six classes (21 subclasses) including glycerolipids (GL), sphingolipids (SP), GP, fatty acyls (FA), saccharolipids (SL) and prenol lipids (PR) were identified. Several molecular species were characterized with PUFA, especially EPA and DHA in GP. Considering the superior fatty acids composition and distribution of round scad, it is deserved for further exploitation of its marine lipid source on account of the healthy and nutritional functions.

DHA-enriched salmon–konjac emulsion gel improves cognitive function in mice

Shen Yemeng,Fei Siyuan,Bai Yihua,El-Aty A. M. Abd,Tan Mingqian

Cognitive impairment is considered as a neurological deficit, which affects millions of people worldwide. Docosahexaenoic acid (DHA), an essential part of brain cell membranes, is crucial for supporting neural communication and preventing cognitive decline. To address this concern, emulsion fillers comprising salmon myofibrillar protein (SMP), Konjac glucomannan (KGM), and octenyl succinic anhydride (OSA)-starch were used. An DHA-loaded emulsion gel stabilized by OSA-satrch through the modulation of the oil volume fraction was developed to strengthen the structure of SMP gels. The stabilization by the SMP-KGM gel and OSA-starch SMP–KGM (OSA-SMPG) promoted the formation of a compact three-dimensional network structure in the emulsion gel, which resulted in an increased storage modulus, enhanced viscoelastic properties, and good thixotropic behavior. DHA-loaded OSA-SMPG emulsion gel alleviated organ atrophy in mice and reduced escape latency by 48% while improving learning and memory abilities. The total cholesterol level in mouse serum decreased by 32%. In addition, OSA-SMPG alleviated the oxidative stress induced by D-galactose in mice, increased the body’s antioxidant defenses, and increased superoxide dismutase levels in brain tissue by 43%. These findings suggest that promising prospects and potential for nutritional intervention in individuals with cognitive impairment are offered by OSA-SMPG.

High sn-2 DHA in Dietary Triglyceride Enhances Cognitive Performance of Mice

Qi Fang,Xinyao Liu,Wei Huang,Kai Qian,Yanjianxiong Yang,Jing Li,Zeyuan Deng

Docosahexaenoic acid (DHA), one of the most critical polyunsaturated fatty acids, is vital for the neurological growth and cognitive function of infants and children. Approximately 98% of DHA in breast milk exists as triglycerides, with 60% esterified at the sn-2 position. To demonstrate the necessity of mimicking the form of DHA present in breast milk in nutritional food for young children, this study administered diets with varying sn-2 DHA contents (10%, 30%, and 50%) to four groups of mice and analyzed their behavioral performance, brain DHA concentration, expression of brain fatty acid transport proteins, histopathology, and expression of synaptic-related proteins in the hippocampus after 4 weeks. The results showed that compared with the control group, mice in the 50% sn-2 DHA group exhibited superior learning and memory capabilities in behavioral tests, with the most pronounced behavioral improvements in mice, which correlated with higher brain DHA accumulation (from 0.870 ± 0.055 mg/g brain to 1.809 ± 0.132 mg/g brain, p < 0.05), increased levels of MFSD2A (1.40-fold, p > 0.05), FABP5 (2.36-fold, p < 0.05), FATP1 (1.47-fold, p < 0.05), and ACSL6 (1.48-fold, p < 0.05), improved hippocampal neuron morphology, and enhanced the level of BDNF (1.55-fold, p < 0.05), SYN (1.45-fold, p < 0.05), and PSD-95 (1.57-fold, p < 0.05). These findings establish a foundation for developing DHA nutritional supplements.

Antioxidant Activity of Docosahexaenoic Acid (DHA) and Its Regulatory Roles in Mitochondria

Guiling Li,Yuanyuan Li,Baoping Xiao,Dongyue Cui,Yanqi Lin,Jun Zeng,Jian Li,Min-Jie Cao,Jingwen Liu

Reactive oxygen species (ROS) are single-electron-bearing oxidation–reduction products that are mainly produced in mitochondria. Excessive ROS accumulation may lead to oxidative damage. Docosahexaenoic acid (DHA) is an essential component of brain phospholipids and is mainly derived from the diet. Its antioxidant activities have been extensively studied. However, its regulatory roles in mitochondria and the underlying mechanism remain to be elucidated. In this study, the DHA’s effect on cellular antioxidant capacity and mitochondrial functions was examined in HepG2 cells. The results showed that 100 μM DHA decreased cellular and mitochondrial ROS levels to 75.2 ± 9.4% (P < 0.05) and 55.1 ± 1.4% (P < 0.01), respectively. It also increased the total antioxidant capacity by 55.6 ± 0.1 and 49.2 ± 1.1% (P < 0.05), based on ABTS and FRAP assay results, respectively. Consistently, it increased the activities and gene expression of major antioxidant enzymes by at least 35 and 40% (P < 0.05), respectively. Furthermore, DHA promoted mitochondrial functions and biogenesis. These data suggested that DHA’s antioxidant activity can be attributed to its enhancement of mitochondrial functions and biogenesis. This study may shed light on the molecular mechanisms underlying DHA’s function in improving resistance to and relieving the symptoms of chronic disease.

Bio-imprinted lipase-catalyzed production of medium- and long-chain structured lipids rich in n-3 polyunsaturated fatty acids by acidolysis

Xiaoqiang Zou,Mudassar Hussain,Imad Khan,Yanxi Wang,Bangzhi Jiang,Lei Zheng,Yuechao Pan,Jijie Hu,Azqa Ashraf

Medium- and long-chain structured lipids (MLSLs) rich in docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were synthesized by acidolysis of fish oil (Omax 1812, a mixture of sardine and anchovy oils) with caprylic acid (CA) using a lipase bio-imprinted with fatty acids as a catalyst. A bio-imprinted lipase for the acidolysis reaction was first prepared under optimal conditions, and its activity was 1.43 times higher than that of the non-bioimprinted. The bio-imprinted lipase was used as a catalyst to produce such MLSLs, and the reaction conditions were optimized. Under the optimal conditions, equilibrium was achieved in 8 h, and the total and the sn -1, 3 CA contents in the obtained product were 28.72% and 42.26%. Additionally, the total and the sn -2 DHA contents were 14.23% and 21.10%, respectively, and the total and the sn -2 EPA contents were 16.43% and 10.14%, respectively. These obtained MLSLs, rich in sn -1, 3 CA, and sn -2 DHA, EPA can deliver significant health benefits.

Novel Room-Temperature-Prepared Covalent Organic Framework as Adsorbent for Rapid Extraction of Aromatic Esters in Baijiu

Yongqing Zhang,Zhen Fan,Xiaolong Shang,Ling Ao,Miao Liu,Feng Lin,Baoguo Sun,Wei Dong,Xiaotao Sun,Yanfei Xiong,Bo Deng

Developing a rapid, efficient, and environmentally friendly analytical methodology for the determination of aromatic esters (AREs) in Baijiu presents a significant challenge. In this study, we developed a room-temperature method to synthesize core-shell Fe 3 O 4 @TPB-Dha in 1 h using pre-prepared Fe 3 O 4 nanoparticles, enabling rapid and sensitive detection of AREs. The Fe 3 O 4 @TPB-Dha demonstrated exceptional adsorption capabilities (52.63–58.82 mg/g) with a brief adsorption duration (4 min) for four AREs as magnetic solid phase extraction (MSPE) adsorbent. Moreover, density functional theory (DFT) calculations revealed the key adsorption mechanisms to be hydrogen bonds, van der Waals forces, and π–π interactions. Ultimately, the method enabled the rapid and accurate quantification of the four AREs in real Baijiu samples, with concentrations spanning from 0.17 ± 6 × 10 −4 to 2.40 ± 0.03 mg/L, and exhibited relative standard deviations below 6.57% ( n = 3). These findings suggest that Fe 3 O 4 @TPB-Dha holds promise as an effective adsorbent for efficient extraction and precise quantification of aromatic esters in Baijiu.
