Transgenerational Effects of Atrazine on Daphnia magna Based on Life-History Traits and Population Dynamics

Wang Jun,Li Meng,Gu Lei,Sun Yunfei,Zhu Xuexia,Yang Zhou

Atrazine is a predominant herbicide globally, and its residues are commonly found in natural water bodies due to its extensive use. Atrazine is known for its detrimental effects on the reproductive abilities of aquatic plants and animals. Our study explored the impact of maternal exposure to atrazine on the survival and performance of offspring using the water flea Daphnia magna as a model organism. We investigated how the life-history traits and population dynamics of maternal D. magna and their offspring are influenced by atrazine. We hypothesized that exposure to atrazine in mothers would negatively affect the survival and performance of their offspring. The findings revealed that short-term exposure to atrazine adversely affects fitness-related traits, manifesting as decreased survivorship and reduced fecundity. Offspring from atrazine-exposed mothers displayed a limited recovery in fitness; no mortality was observed during the 14-day experiment, yet fecundity remained significantly reduced. At the population level, maternal exposure to atrazine led to a decreased population carrying capacity. The notable maternal effects on offspring reproduction underscore the long-term toxicity of atrazine to D. magna populations, suggesting prolonged consequences even after the cessation of atrazine pollution. This study underscores the transgenerational effects of atrazine, highlighting its persistent threat to D. magna populations.

Driving mechanism of suspended minerals on the multi-level bioavailability of citalopram in Daphnia magna

Haohan Yang,Hanyu Jiang,Hao Xing,Zhankun Chen,Qi Zhang,Zhenhua Yan

Minerals modulated the multi-level bioavailability of CIT in D. magna . • Illite caused severe damages to the digestive system at 100 μg/L-CIT. • Montmorillonite obviously aggravated the oxidative stress of CIT. • Sensitive responses of feeding and swimming behaviors were observed. The impacts of water conservancy projects on the ecological risk of pollutants have raised intensive concerns in recent years. The widespread occurrence of antidepressant citalopram (CIT) could pose potential threats to the aquatic non-organisms and even human health. However, information regarding the bioavailability of CIT in the presence of suspended minerals is still limited. In this study, the driving mechanism of kaolinite, montmorillonite (Mon), and illite on the multi-level bioavailability of CIT was investigated in Daphnia magna . Corresponding to the inhibition of feeding behavior, the CIT-associated minerals in the gastrointestinal tracts caused severe cell swelling, cavitation and microvilli degeneration, especially for CIT+illite group. All co-exposure of minerals and CIT significantly alleviated the cellulase activity, while kaolinite further aggravated the inhibition of CIT on trypsin activity at 100 μg/L. Decreased CIT body burden, swimming activities were observed in the presence of three minerals at 10 μg/L, whereas no remarkable function at 100 μg/L. Sensitive oxidative stress was observed for CIT+Mon group with significant increase of reactive oxygen species, total antioxidant capacity, catalase as well as the inhibition of catalase and glutathione S-transferase genes expression. Illite obviously aggravated the biological effects of CIT at both concentrations, whereas Mon was effective only at low concentrations with the most limited action of kaolinite. The feeding and swimming behaviours of D. magna represented the comprehensive stress response among the correlation of biomarkers. The rapid and extensive release of CIT from minerals in gastrointestinal fluids indicates the critical role of secondary release in modulating its bioavailability. Impact of atrazine on the dynamic response of Daphnia pulex populations to fish predation risk

Qin Shanshan,Xia Tian,Li Gongzheng,Gu Lei,Sun Yunfei,Yang Zhou

Herbicide pollution is persistent, which not only has a negative impact on individual organisms, but also may alter population dynamics and stability of interspecific relationships. Cladocerans, an important part of zooplankton, are often simultaneously exposed to environmental pollutants and predation risk in the aquatic environment. To evaluate the combined effects of atrazine and fish predation risk on the population traits of cladocerans, we exposed Daphnia pulex to different concentrations of atrazine (0, 0.05, 0.10, and 1.0 mg L−1) with or without fish (Rhodeus ocellatus) kairomone, recorded the key population traits, and fitted Gaussian model to population dynamics. Results showed that fish kairomone increased the population density at the end of the experiment and resting eggs production, and tended to decrease the total biomass and the average dry weight per individual of D. pulex. Atrazine reduced the total biomass, the average dry weight per individual, and resting eggs production of D. pulex populations. Atrazine also decreased the population density at the end of the experiment of D. pulex in fish kairomone treatment, and attenuated the promoting effect of fish kairomone on resting eggs production and the reduction of the total biomass. The findings highlighted the importance of considering the combined impact of environmental pollutants and predation risks on zooplankton populations.

Toxicity of microplastics and nanoplastics to Daphnia magna: Current status, knowledge gaps and future directions

Oluwadamilola Pikuda,Eva Roubeau Dumont,Qiqing Chen,Jun-Ray Macairan,Stacey A. Robinson,Dimitrios Berk,Nathalie Tufenkji

We conducted a systematic review of 124 published articles that investigated the toxicity of microplastics and nanoplastics to Daphnia magna . This review summarizes studies assessing acute, chronic, and multigenerational impacts, as well as the effects observed via leached chemicals from plastics and the role of plastics as contaminant vectors. Overall, observed toxicity varies across different polymer types, and shapes. One of the most visible findings is that targeted research synthesis of the acute toxicity tests found more toxicity in smaller-sized particles. Most studies use spherical plastics that are commercially available, especially polystyrene, while the use of irregular-shaped and/or secondary plastics is still emerging. Also, there are still various confounding factors that make the comparison of the observed results difficult. Future studies should focus on irregular-shaped particles, and other polymer types, besides polystyrene. More research efforts are needed to understand the impacts of environmental factors and complex matrices.

Toxicity of Arsenic to Photobacterium phosphoreum, Daphnia magna, and Danio rerio at Different pH Levels

Qun He,Ruijuan Qu,Xinghao Wang,Zhongbo Wei,Ping Sun,Zunyao Wang

Although it has been widely known that arsenic poses great threat to organisms and arsenic species account for the toxic effect of the metalloid, relatively little attention has been given to the arsenic toxicity to different organisms at different pH values. Three kinds of aquatic organisms including Photobacterium phosphoreum , Daphnia magna , and Danio rerio were used to study the effects of pH on the toxicity of arsenic. Toxicity test results were analyzed by calculating EC 50 values and for arsenic species at five different pH values (5.0, 6.0, 7.0, 8.0, and 9.0). As a result, As(III) is more toxic to the tested organisms than As(V), and P. phosphoreum was more sensitive to the toxicity of arsenic than D. magna and D. rerio . The toxicity of As(III) species is different for different organisms: dihydrogen arsenite, H 2 AsO 3 – , is more toxic to P. phosphoreum and D. rerio than arsenous acid, H 3 AsO 3 , while H 3 AsO 3 is more toxic to D. magna . As for As(V), arsenate, HAsO 4 2− , is the most toxic to P. phosphoreum , whereas dihydrogen arsenate, H 2 AsO 4 – , may be responsible for the toxicity to D. magna and D. rerio . As(III) and As(V) are more toxic to P. phosphoreum in an alkaline environment, while for D. magna in an acidic environment and As(III) is more toxic to D. rerio in an alkaline environment and As(V) in an acidic environment.

The detrimental effects and mechanisms of Orlistat in disrupting energy homeostasis and reproduction in Daphnia magna

Zhihua Ni,Xinling Tian,Wenbo Zhao,Wenkai Hu,Jinghua Lv,Xiaoli Sun,Yajie Zhang,Yiwen Zhang,Yuming Zhang,Baoku Li,Fengsong Liu

Orlistat (ORL) has been employed as an anti-obesity pharmaceutical for several decades. Given its low absorption rate, the majority of administered ORL is excreted into the environment with feces. It is crucial to collect scientific information regarding the possible ecological risks associated with ORL. Here, the effects of ORL on Daphnia magna were evaluated using a 21-day chronic test at concentrations of 1, 10, 100, and 1000 μg/L. We found the inhibition of feeding and swimming activities in the 100 and 1000 μg/L ORL exposed D. magna , respectively. Their digestive enzyme activities and metabolites were reduced even at 1 μg/L ORL exposure. It is noteworthy that exposure to 100 μg/L ORL induced a decrease in the reproductive capacity of D. magna , although no discernible genotoxicity was observed. To identify the toxicological mechanisms of ORL, a metabolic analysis was conducted on D. magna exposed to 1000 μg/L ORL. A comprehensive reduction in carbohydrates, lipids, and amino acids was observed, which resulted in a blockage of metabolic flux towards the TCA cycle, as evidenced by mitochondrial dysfunction. These findings substantiate the detrimental impact of ORL on D. magna and provide insights into the underlying molecular mechanisms from a metabolic perspective.

Evaluating the Impact of Individual and Combined Toxicity of Imidacloprid, Cycloxaprid, and Tebuconazole on Daphnia magna

Yanli Man,Tian Sun,Chi Wu,Xingang Liu,Mingyuan He

The risks posed by chemicals in the environment are typically assessed on a substance-by-substance basis, often neglecting the effects of mixtures. This may lead to an underestimation of the actual risk. In our study, we investigated the effects of three commonly used pesticides—imidacloprid (IMI), cycloxaprid (CYC), and tebuconazole (TBZ)—both individually and in combination, using various biomarkers to assess their impact on daphnia. Our findings indicated that the order of toxicity, from highest to lowest, was TBZ, IMI, and CYC, as determined by acute toxicity as well as reproduction. The effects of the ITmix (IMI and TBZ) and CTmix (CYC and TBZ) combinations on immobilization and reproduction were evaluated by MIXTOX, revealing a higher risk of immobilization at low concentrations for ITmix. The effect on reproduction differed depending on the ration of pesticides in the mixture, with synergism observed, which may be caused mainly by IMI. However, CTmix showed antagonism for acute toxicity, with the effect on reproduction depending upon the composition of the mixture. The response surface also exhibited a switch between antagonism and synergism. Additionally, the pesticides extended the body length and inhibited the development period. The activities of superoxide dismutase (SOD) and catalase (CAT) content was also significantly induced at different dosage points in both the single and combination groups, indicating changes in the metabolic capabilities of detoxifying enzymes and target site sensitivity. These findings highlight the need for more attention to be focused on the effects of pesticide mixtures. Keywords:ecotoxicity;pesticide mixture;combined toxicity;MIXTOX;risk assessment

Triazophos exposure on maternal Daphnia magna at environmental-related concentrations revealed toxic effects to its offspring

Shaoquan Liu,Jianlu Zhang,Ruijiao Li,Chunyun Zhang,Lixin Wang,Hao Liang,Guangpeng Feng,Dongmei Xiong

Due to chemical and photochemical stability, triazophos has been frequently detected in rivers and oceans over the years with extensive use for pest control in agriculture , and it has become a worldwide ecological concern to the aquatic environment . Until now, fewer data are available regarding the potential long-term adverse effects of triazophos on aquatic invertebrates , which plays an essential role in aquatic food webs , as a key group for water ecosystems. In this experiment, the F 1 - and F 2 progenies of Daphnia magna were recovered when daphnias (F 0 ) exposure to triazophos at environmental-related concentrations (0.1 and 1.0 μg/L) for 21 d; and the indexes related to phenotypic traits, reproduction and gene expression were measured in tested animals. The results showed that heart rate and total number of neonates in exposed F 0 -daphnias were significantly lower than those of control group, and the detoxification genes ( HR96 and P-gp ) were up-regulated while genes related reproduction ( Vtg ) and molting ( Nvd and Shd ) were significantly down-regulated. The heart rate and individual size of F 1 -daphnias (<24 h) were significantly reduced in the treatment group. After 21-d recovery, the heart rate and expression of HR96 , P-gp, Vtg, Nvd and Shd were declined in F 1 -daphnias. There was no obvious difference of morphological traits and heart rate between treatment and control in F 2 -daphnias (<24 h). In summary, daphnias (F 0 ) exposure to triazophos with environmental dose could raise toxic effects on its offspring (F 1 ), which is mainly manifested by reduced heart rate, the accumulated number and individual size of offspring and decreased expression of genes related to molting and reproduction.

Dual drive acute lethal toxicity of methylene blue to Daphnia magna by polystyrene microplastics and light

Xiangmei Xiang,Juanjuan Zhou,Shaochen Lin,Nan Zhang,Gulizhabaier Abulipizi,Guikui Chen,Zhanjun Li

Microplastics (MPs) can adsorb and influence the toxicity of traditional pollutants significantly. Although the complex toxicity of MPs and molecular pollutants were frequently reported, rare work has been done on the influence of MPs on the phototoxicity of photosensitive pollutants under light illumination condition. Herein, polystyrene microplastics (PS) (~1 μm in diameter, 5.0 mg/L) was used as a model MP to investigate its influence on the phototoxicity of a soluble blue dye, methylene blue (MB) using Daphnia magna as a model organism. The results indicate that PS could adsorb MB effectively and quickly, thus led to concentrated MB on PS/water interface. D. magna ingested MB-adsorbed PS very quickly within tens of minutes. Although MB or PS alone led to negligible lethal phototoxicity to D. magna , PS significantly enhanced the lethal phototoxicity of MB (0.25 mg/L) to D. magna after light illumination (10 h) with the survival rate decreased by 63.3 % compared with the control in the dark. Further, the phototoxicity of MB was found positively consistent with PS concentration from 0.50 mg/L to 7.50 mg/L. The singlet oxygen fluorescence assay indicates that the presence of PS did not increase the total amount of singlet oxygen in the aquatic environment but increased the local concentration in the gut area via non-selective ingestion of D. magna . High level singlet oxygen generated in the gut might possibly be the main reason that led to the massive death of D. magna . Surface adsorption of photosensitive pollutants may transform inert MPs into persistent solid sources of singlet oxygen production and become a new potential lethal threat to aquatic small organisms and ecological equilibrium. This kind of MPs and light dual drive phototoxicity of photosensitive pollutants needs to paid more attention in understanding the uncertain ecological risk of MPs.

Chronic toxicity of diclofenac, carbamazepine and their mixture to Daphnia magna: a comparative two-generational study

Nkoom Matthew,Lu Guanghua,Liu Jianchao

The chronic toxicity of diclofenac (DCF) and carbamazepine (CBZ) as separate substances and in conjunction with their mixture on Daphnia magna was assessed in the parental (F0) and first filial (F1) generations. The second (F1–B2) and fifth (F1–B5) broods of F1 offspring were investigated and compared. Both drugs and their mixture were exposed to each generation of Daphnia magna for 21 days with life history, behavioural and gene expressions as measured endpoints. After the parental exposure, offspring from these two broods were transferred to a clean medium for a 21-day recovery. Exposure to diclofenac, carbamazepine and their mixture significantly inhibited growth, reproduction, swimming activities, heart rate, thoracic limb activities, reproductive and antioxidant-related genes in the parental as well as the first filial generations. These effects were relatively greater in the F1 generation. This indicates that Daphnia magna ’s sensitivity improved while its fitness declined over the two generations, which is an indicator of greater energy requirements for maintenance. Besides, the significant inhibition in the antioxidant-related genes implies that oxidative stress occurred in Daphnia magna under the exposure to these drugs. The significant reduction in the reproductive output, moulting frequency and cyp314 gene expression as a result of exposure to CBZ simultaneously obtained herein may indicate that this drug could act as an endocrine disruptor. Most of these significant effects were not recoverable after the 21-day recovery period. The findings reported herein highlight the necessity to include maternal effects in environmental risk assessment processes, considering that pollutant effects are underestimated during single-generational exposure.

Daphnia magna and mixture toxicity with nanomaterials – Current status and perspectives in data-driven risk prediction

Diego Stéfani T. Martinez,Laura-Jayne A. Ellis,Gabriela H. Da Silva,Romana Petry,Aline M.Z. Medeiros,Hossein Hayat Davoudi,Anastasios G. Papadiamantis,Adalberto Fazzio,Antreas Afantitis,Georgia Melagraki,Iseult Lynch

The aquatic ecosystem is the final destination of most industrial residues and agrochemicals resulting in organisms being exposed to a complex mixture of contaminants. Nanomaterials (NMs) are being increasingly applied in many technologies and industrial sectors, so there is an increasing concern about the negative impacts of NMs in the environment after their interaction with co-contaminants. Consequently, mixture toxicology has been gaining attention in nanotoxicology recently. Usually, mixture toxicity or combined toxicity is estimated from the individual effects of the chemicals using the mathematical models of concentration addition (CA) or independent action (IA), however these models do not account for metabolic interactions between the chemicals, when they act in related metabolic pathways and molecular targets . As NMs unique physico-chemical properties make them highly reactive with a high surface area for adsorption, those models may not realistically estimate the toxicological effects of mixtures containing NMs. The co-exposition of NMs and other environmental contaminants (e.g., organic pollutants and heavy metals) may cause different mixture effects such as addition, synergism, antagonism, or even other complicated responses, including altered toxicokinetics/toxicodynamics, which vary according to the individual components properties, environmental exposure conditions, and the biological system. Therefore, the large number of factors that may influence the toxicity of a NM and contaminant mixture makes NMs mixture risk assessment a complex task. Daphnia magna are one of the most commonly used model species in nanotoxicology, including in mixture studies. It’s advantages include short generation time, small body sizes, ability to produce large populations rapidly, coupled with its completely mapped genome which allows the use of a multitude of omics techniques to understand the stress responses of daphnids to NMs and chemicals. Here, we analyse the toxicological effects of NMs and contaminant mixtures using Daphnia as a model organism, and discuss future perspectives for NMs-mixtures risk assessment focusing on harmonization of methodologies and application of data-driven science in mixture ecotoxicology.

Combined exposure of polystyrene microplastics and carbamazepine induced transgenerational effects on the reproduction of Daphnia magna

He Yide,Zhang Yiqun,Zhou Wei,Freitas Rosa,Zhang Yunhai,Zhang Yongjun

Polystyrene microplastics (PS MPs) and carbamazepine (CBZ) are frequently detected in freshwater ecosystems. However, the transgenerational effects of PS MPs and CBZ on the reproduction of aquatic organisms and the corresponding mechanisms are still unclear. In the present study, Daphnia magna was used to evaluate the reproductive toxicity in two consecutive generations (F0, F1). The molting and reproduction parameters, the expression of reproduction, and the toxic metabolism genes were examined after 21-day exposure. A significantly enhanced toxicity was observed in the presence of 5 μm PS MPs and CBZ. Chronic exposure results showed that the 5 μm PS MPs alone, CBZ alone, and their mixtures exerted significant reproductive toxicity of D. magna . The results of RT-qPCR showed transcripts of genes related to reproduction ( cyp314 , ecr-b , cut , vtg1 , vtg2 , dmrt93b ) and toxic metabolism ( cyp4 , gst ) were altered in both the F0 and F1. In addition, for the F0, gene transcriptional changes of reproduction were not fully translated into physiological performance, probably due to the compensatory responses caused by the low dose of PS MPs alone, CBZ alone, and their mixtures. Whereas for the F1, the trade-off between reproduction and toxic metabolism at gene levels was observed, which translated into a significant reduction in the total neonate number of F1. These findings suggest that long-term exposure to MPs and CBZ can cause serious reproduction damage to aquatic animals, which needs to be given sufficient attention. Graphical Abstract
