Secondary formation of PM2.5-bound PFAS in the Pearl River Delta, China: Field evidence and implications for precursor control

Weiru Wang,Shizhen Zhao,Lele Tian,Shaojun Lv,Jun Li,Zhen Zhao,Duohong Chen,Ping’an Peng,Gan Zhang

Per- and polyfluoroalkyl substances (PFAS) are global pollutants of concern, yet their atmospheric sources and secondary formation mechanisms remain poorly understood. Here, we collected PM 2.5 samples during winter and summer across nine cities in the Pearl River Delta (southern China). Among 44 targeted PFAS in PM 2.5 , 15 were detected with a median concentration of 222 pg/m 3 , with ultra-short-chain PFAS accounting for 42% of the total concentration. Higher Σ 15 PFAS concentrations occurred in winter (261 pg/m³) than in summer (141 pg/m³), with distinct seasonal compositional patterns. Source apportionment analysis indicated that potential secondary formation contributed an average of 16% to Σ 15 PFAS. Significant correlations between selected perfluoroalkyl acids (PFAAs) and atmospheric oxidants, aerosol liquid water content, and succinic acid point to aqueous-phase precursor degradation as a potential pathway for PFAS formation. These results suggest that PM 2.5 may act as a sink for PFAS, especially for ultrashort-chain species during winter. Overall, our study provides field-based evidence for potential secondary PFAS formation in ambient aerosols and underscores the importance of controlling precursor emissions to mitigate atmospheric PFAS pollution.

Identification and Functional Analysis of a Delta Class Glutathione S-Transferase Gene Associated with Insecticide Detoxification in Bradysia odoriphaga

Bowen Tang,Wu Dai,Lijun Qi,Qi Zhang,Chunni Zhang

A delta class glutathione S-transferase gene (BoGSTd2) is identified from Bradysia odoriphaga for the first time. Developmental expression analysis showed that expression of BoGSTd2 is significantly higher in the fourth instar larval stage and the adult stage. Tissue-specific expression analysis found that BoGSTd2 was expressed predominantly in the midgut and Malpighian tubules in the fourth instar larvae and the abdomen of adults. Expression of BoGSTd2 was significantly upregulated following exposure to chlorpyrifos and clothianidin. In vitro inhibition and metabolic assays indicated that recombinant BoGSTd2 could not directly metabolize chlorpyrifos and clothianidin. Nevertheless, disk diffusion assays indicated that BoGSTd2 plays an important role in protection against oxidative stress. RNAi assays showed that BoGSTd2 participates in the elimination of reactive oxygen species induced by chlorpyrifos and clothianidin. These results strongly suggest that BoGSTd2 plays an important role in chlorpyrifos and clothianidin detoxification in B. odoriphaga by protecting tissues from oxidative stress induced by these insecticides.

Atomically Thin Delta-Doping of Self-Assembled Molecular Monolayers by Flash Lamp Annealing for Si-Based Deep UV Photodiodes

Shannan Chang,Jiajing He,Slawomir Prucnal,Jieyin Zhang,Jianjun Zhang,Shengqiang Zhou,Manfred Helm,Yaping Dan

Delta doping (δ-doping) can find a wide range of applications in advanced metal oxide semiconductor field effect transistors, deep UV photodetectors, quantum devices, and others. In this work, we formed a δ-doping layer in silicon by employing flash lamp annealing to treat the PCl3 monolayers grafted on silicon surfaces. The δ-doping layer is atomically thin (<1 nm). Low-temperature Hall measurements show that the δ-doping layer is in a metallic state and exhibits a weak localization phenomenon, implying that a two-dimensional electron gas is formed. When we form such an n-type δ-doping layer on a highly doped p-type Si substrate, a highly sensitive solar-blind UV photodetector is created, which traditionally was only possible by using wide band gap semiconductors such as gallium nitride (GaN) or silicon carbide (SiC).

Guilongwan Ameliorates Experimental Diabetic Foot Ulcer in Rats via the Inhibition of Delta-Like 4/Notch1 Signaling in M1 Macrophages

Xi-Ting Lv,Wen-Xiu Yang,Xiao Zhang,Bao-Ling Li,Wen-Ying Wang,Guo-En Wang

Guilongwan (GLW), a representative of traditional Chinese Medicine (TCM) has been utilized to treating diabetic foot ulcer (DFU)-related syndrome including an intolerance of cold with cold limbs, blood circulation disorder, and immune dysfunction for decades. However, the chemical and biological mechanisms of GLW remain unclear. This study aims to discover the biological mechanisms of GLW on DFU by using streptozotocin- and skin-puncher-induced DFU rat models, in vitro macrophage models, and in silico analysis. The alterations in pathology, Notch1 signaling, and macrophage polarization are detected. The results indicated that GLW promoted wound healing, cutaneous cell proliferation, and angiogenesis in DFU rats by inhibiting delta-like (DLL) 4/Notch1 signaling. In addition, GLW inhibited M1 polarization and promoted M2 polarization in diabetic wounds. Seventeen chemical compounds in GLW-medicated serum are identified. In silico analysis and in vitro experiments demonstrated that GLW-medicated serum and its main compounds inhibited the expression of DLL4 in matrix metalloproteinase-9-induced M1 macrophages. In conclusion, GLW ameliorated experimental DFU rats via the inhibition of DLL4/Notch1 signaling in M1 macrophages. This study provided a new biologic mechanism for GLW in the treatment of DFU.

Licorice-saponin A3 is a broad-spectrum inhibitor for COVID-19 by targeting viral spike and anti-inflammation

Yang Yi,Wenzhe Li,Kefang Liu,Heng Xue,Rong Yu,Meng Zhang,Yang-Oujie Bao,Xinyuan Lai,Jingjing Fan,Yuxi Huang,Jing Wang,Xiaomeng Shi,Junhua Li,Hongping Wei,Kuanhui Xiang,Linjie Li,Rong Zhang,Xin Zhao,Xue Qiao,Hang Yang,Min Ye

Currently, human health due to corona virus disease 2019 (COVID-19) pandemic has been seriously threatened. The coronavirus severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike (S) protein plays a crucial role in virus transmission and several S-based therapeutic approaches have been approved for the treatment of COVID-19. However, the efficacy is compromised by the SARS-CoV-2 evolvement and mutation. Here we report the SARS-CoV-2 S protein receptor-binding domain (RBD) inhibitor licorice-saponin A3 (A3) could widely inhibit RBD of SARS-CoV-2 variants, including Beta, Delta, and Omicron BA.1, XBB and BQ1.1. Furthermore, A3 could potently inhibit SARS-CoV-2 Omicron virus in Vero E6 cells, with EC 50 of 1.016 μM. The mechanism was related with binding with Y453 of RBD determined by hydrogen-deuterium exchange mass spectrometry (HDX-MS) analysis combined with quantum mechanics/molecular mechanics (QM/MM) simulations. Interestingly, phosphoproteomics analysis and multi fluorescent immunohistochemistry (mIHC) respectively indicated that A3 also inhibits host inflammation by directly modulating the JNK and p38 MAPK pathways and rebalancing the corresponding immune dysregulation. This work supports A3 as a promising broad-spectrum small molecule drug candidate for COVID-19.

Developing a biodegradable tricalcium silicate/glucono-delta-lactone/calcium sulfate dihydrate composite cement with high preliminary mechanical property for bone filling

Zhengwen Ding,Wenjing Xi,Mizhi Ji,Hong Chen,Qiyi Zhang,Yonggang Yan

Bone cements with the feature of easily shaping could ideally match the defect site and prevent the ingrowth of fibrous tissue. In this manuscript, a biodegradable tricalcium silicate (C 3 S)/glucono-delta-lactone (GDL)/calcium sulfate dihydrate (CSD) organic-inorganic composite cement was fabricated with shorter setting time (less than 15 min) and high preliminary mechanical property (5.27 MPa in the first hour). Many methods were applied to study the physicochemical and biological properties of the cement in vitro. The weight loss in PBS can reach 58% after 12 weeks soaking indicating the better biodegradability. The excellent bioactivity in vitro was emerging after the cement was soaked in the simulated body fluid. The cell experiments showed that suitable concentration of the extract liquid of cement was conducive to the proliferation, differentiation and extracellular matrix calcification of the mouse bone marrow stromal cells. Briefly, the C 3 S/GDL/CSD composite cement would have the bright capacity for bone filling.

Unveiling the complex PFAS landscape in firefighting foams applied in the Yangtze River Delta of China: Comprehensive composition analysis and toxicity profiling

Chong Tan,Xin Lyu,Xiaole Weng,Kun Yang,Lifeng Cao

Per- and polyfluoroalkyl substances (PFAS) in firefighting foams represent a persistent source of groundwater contamination due to their extreme environmental stability and toxicological properties. This study provides a comprehensive characterization of PFAS in eleven in-use firefighting foams collected from China's Yangtze River Delta, integrating non-targeted screening with total oxidizable precursor (TOP) assays. A total of 185 distinct PFAS compounds were identified, including 119 anionic, 100 cationic, and 34 zwitterionic species, representing the most diverse PFAS profile reported to date for firefighting foams in the Yangtze River Delta. Parallel analysis detected 139 hydrocarbon surfactants, underscoring the chemical complexity of real-world foam residues. QSAR-ToxPi modeling classified the identified PFAS into three tiers, with 95 compounds (51.4%) falling into the high-hazard category. TOP assays demonstrated that ostensibly PFAS-reduced formulations contained precursor reservoirs capable of generating terminal PFOA at concentrations up to 5198.4 mg/L, nearly 30-fold above initial measurements. These findings demonstrate that expanding water-quality monitoring programs to include PFAS precursors is essential to improve risk assessments for AFFF-impacted groundwater and drinking water sources.

Impact of COVID-19 lockdown on small-scale farming in Northeastern Nile Delta of Egypt and learned lessons for water conservation potentials

Tarek Selim,Mohamed Galal Eltarabily

The coronavirus disease-2019 (COVID-19) pandemic had a great impact on the agricultural sector, especially in developing countries. In particular, it caused exceptional challenges to small and local-scale farmers. Field questionnaires and interviews were used to investigate the effects of COVID-19 on small-scale farmers in the Northern Nile Delta of Egypt. Agricultural farms in Southern Port Said City represent emerging agricultural communities on newly reclaimed land, while those in Damietta are stable agricultural communities (old land of the Nile Delta). The questionnaire was divided into four questions groups to identify and analyze the different reasons that contributed to the disruption of farming systems and the agricultural sector. These groups were farmers' data, the effect of COVID-19 on agriculture inputs, infection rates and precautionary measures against COVID-19, and potential measures and governmental policies for controlling the negative impacts of COVID-19 and achieving agricultural sustainability. Results showed that the effect of lockdown was slightly lower in Damietta as compared to Port Said. Although fertilizers and labors costs, as well as water availability near Port Said, was not considerably affected during the lockdown, the total income of the small-scale farmers' notability decreased. The reluctance of major traders to buy crop production and keep the required balance of cash during the pandemic dramatically affected the crop production selling prices. Results showed that in the absence of agricultural extensions possibilities, there is a lack of awareness toward improving agricultural practices and switching to smart irrigation systems as a way of saving water and increasing crop productivity. The resistance against applying new agricultural practices and switching to smart irrigation systems depends mainly on farmers' financial capability and the nature of agricultural land either old or new. Activating the agricultural extensions roles is considered a keystone for enhancing agricultural sustainability in small-scale farming not only in developing regions of Egypt but also in other similar agricultural communities worldwide.

Eugenol Potentially Targets Oleate Delta-12 Desaturase of Botrytis cinerea and Controls Gray Mold as Part of a Combination of Multiple Fungicidal Components

Xiansu Wang,Chengyan Xia,Miss Dongxue Li,Miss Fenghua Liu,Jun Zhang,Miss Wenjing Xie,Dan Cheng,Libo Zhang,Zhuo Chen

Eugenol exhibits a broad spectrum of antimicrobial activities and fungicide efficacy against crop diseases. However, the antifungal mechanisms of eugenol remain incompletely understood. In this study, we assessed the inhibitory activity of eugenol against several phytopathogenic fungi, finding it to be particularly effective against Botrytis cinerea . The half-maximal effective concentration for inhibiting hyphal growth of B. cinerea was determined to be 72.11 µg/ml. Eugenol exhibited significant inhibition effect of 66.72 and 43.62% against gray mold on tomato fruit and leaves at the 1,500.0 µg/ml dose. Micromorphological analysis revealed that eugenol induced abnormalities in hyphal structures, including disrupted cell membranes and unclear organelle boundaries. Transcriptomic analysis indicated that differentially expressed genes in hyphae treated with eugenol were primarily enriched in processes related to lipid metabolism, transmembrane transport, and electron transfer activity. Molecular docking studies suggested that eugenol may bind to oleate delta-12 desaturase (FAD2) with a low free energy of −9.9 kcal/mol. Assays of cell membrane permeability and hyphal staining confirmed that eugenol disrupts the cell membrane, resulting in leakage of hyphal contents. Quantitative PCR assay showed that eugenol significantly altered the expression of genes involved in lipid metabolism. Compared to the wild type, fad2 mutant displayed slower hyphal growth rates and became less sensitive to the effects of eugenol. This study demonstrates that eugenol target FAD2, disrupt cell membrane integrity, thereby inhibiting the proliferation of B. cinerea . Several formulations of eugenol, including feneptamidoquin or meridianin C, exhibit stronger inhibitory effects, offering promising potential for the control of gray mold.

Baicalein exerts anxiolytic and antinociceptive effects in a mouse model of posttraumatic stress disorder: Involvement of the serotonergic system and spinal delta-opioid receptors

Lina Ruan,Kaiyu Guan,Yue Wang,Ming Gu,Yue Chen,Lisha Cai,Ruixuan Ye,Zhengwei Huang,Anqi Guo,Zhengkang Su,Xi Li,Jianchun Pan

Post-traumatic stress disorder (PTSD) is a serious mental disease featured by a stress dysfunction that occurs after an individual has faced intense mental stress, often accompanied by anxiety and chronic pain. Currently, the mainstream drug for PTSD is serotonin reuptake inhibitors (SSRIs), however, their pain management for patients is limited. Baicalein , a Chinese traditional herbal medicine, has shown promising results in treating anxiety, depression, and pain. In this study, we found that baicalein may alleviate single prolonged stress (SPS)-induced PTSD-like behaviors in mice without altering baseline nociceptive sensitivity or activity. Meanwhile, baicalein increased the noradrenaline (NE) and serotonin (5-HT) content and decreased the ratio of 5-hydroxyindoleacetic acid (5-HIAA)/5-HT by inhibiting the activity of monoamine oxidase A (MAO-A) in SPS-induce mice. The anxiolytic and antinociceptive effects induced by baicalein were totally abolished by 5-HT depleting agents. Moreover, the anxiolytic effects of baicalein could be abolished by the 5-HT 1A receptor antagonist WAY-100635, and the analgesic effects could be abolished by delta-opioid receptor antagonists in the spinal. Taken together, our study provides compelling evidence that baicalein reversed anxiety-like behaviors and neuropathic pain in PTSD through serotonergic system and spinal delta-opioid receptors.

Distribution, homologue pattern, sources, and environmental behavior of short-chain chlorinated paraffins in multi-media environments within the petrochemical industry of the Yangtze River Delta

Jiaying Wang,Xue Wang,Zaihong Zhu,Saixia Ying,Kashif Hayat,Xiaoxia Bai,Shuren Liu,Xuexi Xiao,Chongwei Jin,Weiping Liu

Short-chain chlorinated paraffins (SCCPs) are widely found in the environment. However, limited information exists on the inter-media exchange and migration of SCCPs in the same region. In this study, a comprehensive survey of SCCPs was performed on various environmental substrates (including the surface soil, groundwater, and air) from a large petrochemical park in the Yangtze River Delta, and a total of 24 congener groups were measured. The SCCP concentrations, spatial variations, congener group profiles, and environmental behaviors during the water–land–atmosphere cycle in a typical industrial area were investigated. The ΣSCCP concentrations in the surface soil and groundwater within the petrochemical factory and in the atmosphere in the petrochemical factory perimeter were 108–745 ng g−1, 1133–2994 ng L−1 and 12.1–30.5 ng m−3, respectively. The concentrations and homologue patterns of SCCP significantly varied across different sampling sites, which was attributed to the distances between the sampling sites and workshops, as well as the diverse CP products or byproducts involved in the processing activities. The congener profile revealed that C10Cl8,9 and C13Cl8,9 were the major SCCP homologue groups in the soil within the petrochemical park, whereas C10Cl5–7 and C13Cl5–7 were the predominant congeners in the groundwater, and C12,13Cl7,8 was the dominant SCCP congener group in the atmosphere around the petrochemical park. With respect to the air-soil and soil–water exchange behaviors, the majority of fugacity fraction values for the air–soil and soil–groundwater samples were less than 0.3 and greater than 0.7, respectively, indicating the dominant deposition of SCCP congeners from the air to the soil and permeation into groundwater from the soil within the petrochemical park. To date, the preliminary risk assessments have indicated that SCCPs pose a low ecological risk in the petrochemical zone and pose a low risk to humans through dust ingestion and dermal contact.

Design, synthesis, and biological evaluation of a novel dual peroxisome proliferator-activated receptor alpha/delta agonist for the treatment of diabetic kidney disease through anti-inflammatory mechanisms

Kai Liu,Xing Zhao,Xue Qi,Dong-Liang Hou,Hao-Bin Li,Yu-Hao Gu,Qing-Long Xu

Diabetic kidney disease (DKD) is a major feature of the final stage of nearly all cause types of diabetes mellitus (DM). To date, few safe and effective drugs are available to treat. Peroxisome proliferator-activated receptors (PPARs), comprised of three members: PPAR-α, PPAR-δ and PPAR-γ, play a protective role in the DKD through glycemic control and lipid metabolism, whereas systemic activation of PPAR-γ causes serious side-effects in clinical trials. GFT505 is a dual PPAR-α/δ agonist, and the selectivity against PPAR-γ is still to be improved. Sulfuretin has been shown to suppress the expression of PPAR-γ and improve the pathogenesis of diabetic complications . In this study, by hybridizing the carboxylic acid of GFT505 and the parent nucleus of sulfuretin, we pioneeringly designed and synthetized a series of novel dual PPAR-α/δ agonists, expecting to provide a better benefit/risk ratio for PPARs. Of all the synthesized compounds, compound 12 was identified with highly activity on PPAR-α/δ and higher selectivity against PPAR-γ than that of GFT505 (EC 50 : hPPAR-α: 0.26 μM vs. 0.76 μM; hPPAR-δ: 0.50 μM vs. 0.73 μM; hPPAR-γ: 4.22 μM vs. 2.79 μM). The molecular docking studies also depicted good binding affinity of compound 12 for PPAR-α and PPAR-δ compared to GFT505 . Furthermore, compound 12 exhibited an evidently renoprotective effect on the DKD through inhibiting inflammatory process, which might at least partly via JNK/NF-κB pathways in vivo and in vitro. Overall, compound 12 hold therapeutic promise for DKD.
