This study developed and optimized a fully natural W1/O/W2 double emulsion system for oral iron delivery, using diosgenin, a natural triterpenoid saponin, to stabilize the inner water–oil (W1/O) interface and pea protein nanoparticles (PPNs) to stabilize the outer oil–water (O/W2) interface. The optimal primary emulsion, containing 9 % diosgenin in the oil phase and a W1:O ratio of 2:8, exhibited excellent centrifugal and storage stability. External aqueous phases (W2) were prepared from 2 %, 4 %, and 6 % pea protein dispersions, which underwent ultrasound treatment followed by centrifugation to produce corresponding PPNs suspensions designated as PPN-2 %, PPN-4 %, and PPN-6 %, respectively. The primary iron-containing W1/O emulsion was dispersed into these W2 phases at a W1/O: W2 ratio of 2:8, yielding stable W1/O/W2 emulsions with iron encapsulation efficiencies exceeding 99 %. In vitro simulated digestion revealed that the emulsions with PPN-6 % effectively minimized iron release in the gastric phase (<1 %) while promoting controlled release in the intestinal phase, thereby improving bioavailability and potentially reducing gastric irritation. Compared to synthetic PGPR-stabilized emulsions, the diosgenin-PPNs system demonstrated superior structural stability and digestive performance. This multilayered, food-grade emulsion system offers a promising, health-oriented alternative for iron fortification in functional foods.

Acute pancreatitis (AP) is one of the most common causes of hospitalization for gastrointestinal diseases, with high morbidity and mortality. Endoplasmic reticulum stress (ERS) and Gasdermin D (GSDMD) mediate AP, but little is known about their mutual influence on AP. Diosgenin has excellent anti-inflammatory and antioxidant effects. This study investigated whether Diosgenin derivative D (Drug D) inhibits L-arginine-induced acute pancreatitis through meditating GSDMD in the endoplasmic reticulum (ER). Our studies were conducted in a mouse model of L-arginine-induced AP as well as in an in vitro model on mouse pancreatic acinar cells. The GSDMD accumulation in ER was found in this study, which caused ERS of acinar cells. GSDMD inhibitor Disulfiram (DSF) notably decreased the expression of GSDMD in ER and TXNIP/HIF-1α signaling. The molecular docking study indicated that there was a potential interaction between Drug D and GSDMD. Our results showed that Drug D significantly inhibited necrosis of acinar cells dose-dependently, and we also found that Drug D alleviated pancreatic necrosis and systemic inflammation by inhibiting the GSDMD accumulation in the ER of acinar cells via the TXNIP/HIF-1α pathway. Furthermore, the level of p-IRE1α (a marker of ERS) was also down-regulated by Drug D in a dose-dependent manner in AP. We also found that Drug D alleviated TXNIP up-regulation and oxidative stress in AP. Moreover, our results revealed that GSDMD-/-mitigated AP by inhibiting TXNIP/HIF-1α. Therefore, Drug D, which is extracted from Dioscorea zingiberensis, may inhibit L-arginine-induced AP by meditating GSDMD in the ER by the TXNIP /HIF-1α pathway. Keywords:acute pancreatitis;L-arginine;Gasdermin D;endoplasmic reticulum stress;TXNIP

Green manufacture of steroid precursors from diosgenin by microbial replacing multistep chemical synthesis has been elusive. It is currently limited by the lack of strain and degradation mechanisms. Here, we demonstrated the feasibility of this process using a novel strain Mycolicibacterium sp. HK-90 with efficiency in diosgenin degradation. Diosgenin degradation by strain HK-90 involves the selective removal of 5,6-spiroketal structure, followed by the oxygenolytic cleavage of steroid nuclei. Bioinformatic analyses revealed the presence of two complete steroid catabolic gene clusters, SCG-1 and SCG-2, in the genome of strain HK-90. SCG-1 cluster was found to be involved in classic phytosterols or cholesterol catabolic pathway through the deletion of key kstD1 gene, which promoted the mutant m-∆kstD1 converting phytosterols to intermediate 9α-hydroxyandrostenedione (9-OHAD). Most impressively, global transcriptomics and characterization of key genes suggested SCG-2 as a potential gene cluster encoding diosgenin degradation. The gene inactivation of kstD2 in SCG-2 resulted in the conversion of diosgenin to 9-OHAD and 9,16-dihydroxy-pregn-4-ene-3,20-dione (9,16-(OH) 2 -PG) in mutant m-ΔkstD2 . Moreover, the engineered strain mHust-ΔkstD1,2,3 with a triple deletion of kstDs was constructed, which can stably accumulate 9-OHAD by metabolizing phytosterols, and accumulate 9-OHAD and 9,16-(OH) 2 -PG from diosgenin. Diosgenin catabolism in strain mHust-ΔkstD1,2,3 was revealed as a progression through diosgenone, 9,16-(OH) 2 -PG, and 9-OHAD to 9α-hydroxytestosterone (9-OHTS). So far, this work is the first report on genetically engineered strain metabolizing diosgenin to produce 21-carbon and 19-carbon steroids. This study presents a promising biosynthetic platform for the green production of steroid precursors, and provide insights into the complex biochemical mechanism of diosgenin catabolism.

Using natural-based lipids to construct biocompatible, controllable and efficient nanocarriers and elucidating their structure–function relationships, was regarded as an important area for creating sustainable biomaterials. Herein, we utilized two natural steroids: cholesterol and diosgenin (bearing different hydrophobic tails) as the building blocks, to synthesize a series of natural steroid-based cationic random copolymers PMA6Chol-r-PDMAEMA and PMA6Dios-r-PDMAEMA via RAFT polymerization. The results demonstrated that the steroid-r-PDMAEMA copolymers could efficiently bind pDNA (N/P < 3.0) and then form near-spherical shape (142–449 nm) and positively-charged (+11.5 to +19.6 mV) nanoparticles. The in vitro cytotoxicity and gene transfection efficiency greatly depend on the steroid hydrophobic tail structures and steroid/PDMAEMA block ratios. Optimum transfection efficiency of the (Chol-P1/pDNA and Dios-P3/pDNA) nanoplexes could reach to 18.1–31.2% of the PEI-25K/pDNA complex. Moreover, all of the steroid-r-PDMAEMA/Cy3-pDNA nanoplexes have an obvious “lysosome localization” effect, indicating the steroid structures do not remarkably influence the intracellular localization behaviors of these nanoplexes.

Diosgenin is an important intermediate of steroid hormone drugs, green production of diosgenin with high yield is a key research issue. In this study, a convenient, efficient, and eco-friendly approach was developed to produce diosgenin from saponins in D. zingiberensis C. H. Wright (Curcuma longa L) using a magnetic solid acid. Fe 3 O 4 @SiO 2 -Pr-S-SO 3 H is a heterogeneous catalyst and was obtained by loading 3-(mercaptopropyl) trimethoxysilane on the surface of Fe 3 O 4 @SiO 2 , followed by sulfonation with chlorosulfonic acid. The magnetic solid acid catalyst was fully characterized by Fourier Transform-Infrared (FT-IR) spectra, thermo gravimetric analysis (TGA), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), and X-ray photoelectron spectroscopy (XPS), scanning electron microscope(SEM), transmission electron microscope (TEM), NH 3 -TPD, elemental mapping analysis. The effects of single factors, such as the solid acid mass, alcoholysis temperature, extraction time, and solvent volume on the diosgenin yield were analyzed. The extraction parameters for diosgenin were optimized by response surface methodology (RSM). The optimized values for the solid acid mass, solvent volume, extraction time, and temperature were 0.22 g, 7.8 mL, 7.4 h, and 100 °C, respectively. Under the optimized conditions, the maximum yield of diosgenin was 19.15 %. The solid acid reported in this work exhibited higher catalytic activity at a smaller dosage than traditional sulfuric acid and other solid acids. The alcoholysis products were identified by high-performance liquid chromatography-mass spectrometry (HPLC-MS). Alcoholysis catalysed by an as-prepared magnetic solid acid is clearly superior in terms of catalytic activity, dosage, and reusability. It is a promising strategy for the green production of diosgenin.

To obtaining diosgenin from Dioscorea nipponica Makino ( D. nipponica ) was usually subjected to the multiple processes such as extraction, hydrolysis, transformation and separation etc. In this study, a novel method for rapid production and recovery of diosgenin from D. nipponica was developed by integrating microwave-assisted extraction (MAE), microwave-assisted hydrolysis (MAH) and two-phase extraction (TPE) in one-step procedure, that named MAEH-TPE. Using a two-phase system of ethanol aqueous solution-sulfuric acid-petroleum ether, the MAEH-TPE process was optimized by single-factor experiments and response surface methodology (RSM). Under optimum conditions of ethanol aqueous solution of 60 % (v/v), sulfuric acid concentration of 1.4 mol/L, liquid-to-material ratio of 20 mL/g, extraction temperature of 90 ℃, and extraction time of 35 min, the yield of diosgenin reached 17.09 ± 0.32 mg/g which conformed very closely to the predicted value of 17.185 mg/g, and validation experiments confirmed effectiveness and reliability of the RSM model. Compared with the conventional methods, MAEH-TPE exhibited higher extraction yield, shorter extraction time and simpler operation. The mechanism in MAEH-TPE process was explored in detail by chromatographic analysis and multi-phase equilibria. Enzyme inhibition assay demonstrated that the inhibition rate of the extracted diosgenin on porcine pancreatic lipase (PPL) reached 56.18 ± 1.38 % with IC 50 of 191.42 µg/mL, further proving that diosgenin as the main parent structure of the saponins in D. nipponica dominantly contributed to PPL inhibitory effect. Also, the results proved that MAEH-TPE had excellent extraction performance and simplified the complex processes, reducing energy consumption and saving solvents and resources.

In situ discrimination of Paris saponin isomers by DESI-IM-MSI using CCS information. • Distinct tissue-specific distributions of diosgenin- and pennogenin-type saponins were revealed. • Spatial metabolomics and transcriptomics reveal potential pennogenin conversion. Although the spatial distribution of plant metabolites is critical for understanding their biosynthesis, transformation, transport, and accumulation, the tissue-specific organization of saponins in Paris polyphylla Smith var. yunnanensis (PPY) remains poorly understood. In this study, saponins, flavonoids, and ecdysteroids in the rhizomes, fibrous roots, stems, leaves, and calyces of PPY were analyzed using UPLC-IM-MS. Based on these results, desorption electrospray ionization–ion mobility–mass spectrometry imaging (DESI-IM-MSI) was employed to map their spatial distributions, with collision cross section (CCS) information enabling the in situ discrimination of isomeric saponins. Integration with transcriptomic analysis of key cytochrome P450 hydroxylase genes further provided insights into the biosynthesis and transformation of tissue-specific saponins. Diosgenin-type saponins were predominantly enriched in the rhizome cortex and pith and exhibited pronounced spatial redistribution during rhizome development, whereas pennogenin-type saponins were consistently localized in the epidermis and fibrous root scar regions. Transcriptomic evidence further suggested that the rhizome served as a major site of saponin biosynthesis, while their occurrence in aerial tissues likely reflected subsequent transport and local metabolic modification. Overall, this study establishes a DESI-IM-MSI-based spatial metabolomics workflow and reveals a spatial metabolic trajectory of saponins involving rhizome biosynthesis, peripheral conversion of diosgenin-type to pennogenin-type saponins, and subsequent transport and modification in other tissues.
