The extracellular matrix (ECM) is a dynamic, hierarchically structured network essential for mechanical support and regulation of cell behavior. Mimicking its complexity in synthetic materials remains a major challenge. Here, we present a straightforward strategy to fabricate ECM-inspired, dynamically phase-separated hydrogels (DyPS-MAPgel) by methacrylation of mussel adhesive proteins (MAPs), followed by metal ion-induced phase separation and in situ photopolymerization. Leveraging the intrinsic ability of MAPs to undergo biomolecular phase separation and subsequent covalent crosslinking, the resulting hydrogels exhibit tunable mechanical properties, strong tissue adhesion, self-healing, antibacterial, and antioxidant activities. Importantly, DyPS-MAPgels support stem cell proliferation and lineage-specific differentiation both in vitro and in vivo through integrin-dependent mechanotransduction pathways. This work demonstrates the potential of phase separation–mediated assembly as a robust strategy for constructing ECM-mimetic materials and provides insights into ECM-cell interactions. Our findings offer a versatile platform for regenerative medicine applications, including bioadhesion, 3D bioprinting, and tissue engineering.

Photoaging, caused by chronic ultraviolet (UV) exposure, leads to extracellular matrix (ECM) degradation, impaired skin function, and visible signs of aging. While collagen-based implants offer therapeutic potential, most rely on animal-derived sources, posing risks of immunogenicity and pathogen transmission. Recombinant collagens overcome these limitations but often lack the ability to form native-like fibers. Herein, we report the first injectable, self-assembled recombinant type I collagen nanofiber (SARCI) implant for reversing photoaged skin damage. The SARCI implant exhibits excellent injectability, enhanced thermal and enzymatic stability, and low immunogenicity. In vitro, it significantly promotes fibroblast proliferation, migration, and myofibroblast differentiation, while inducing chondrogenic commitment of mesenchymal stem cells. In a UV-induced photoaging mouse model, SARCI restores ECM homeostasis, improves epidermal thickness, dermal density, hydration, and transepidermal water loss, and reduces oxidative stress. Transcriptomic analysis confirms that SARCI activates ECM-receptor interaction, PI3K-Akt, and focal adhesion signaling pathways. These findings establish SARCI as a safe and effective recombinant biomaterial for regenerative dermatology and photoaging intervention.

Clinically, cartilage injuries often involve simultaneous damage to the subchondral bone. However, the extracellular matrix (ECM) microenvironment of cartilage and subchondral bone varies greatly, making it a challenge to design scaffolds that match the osteochondral repair microenvironment. Herein, we proposed a bilayer scaffold with dual-tissue-specific ECM microenvironments for spatially guiding osteochondral regeneration via a multiple biomimetic strategy. Specifically, bone ECM-like chitosan/chitin whisker (CHW) liquid crystal (LC) hydrogel radial microchannels and deferoxamine-loaded polyethylene glycol diacrylate (PEGDA)/CHW LC hydrogel central longitudinal canal were constructed in 3D printed poly( l -lactide) scaffold as the osteogenic layer, while a cartilage ECM-like viscoelastic PEGDA/CHW LC hydrogel encapsulated with baicalin was designed as the chondrogenic layer. Results indicate that bone ECM-like LC state and microchannels, in conjunction with deferoxamine, can efficiently synergistically boost osteogenic differentiation and vascularization. Furthermore, LC state and viscoelastic hydrogel remodels the stem cell microenvironment and promote the chondrogenic differentiation of stem cells with baicalin. This multiple biomimetic scaffold exhibits satisfactory regenerative ability around rabbit osteochondral defect, facilitating simultaneous regeneration of cartilage and vascularized subchondral bone. This multiple biomimetic strategy achieves highly simulated construction of dual-tissue-specific ECM microenvironments, providing new insights for guiding osteochondral regeneration.

Thin endometrium (TE), a major cause of embryo implantation failure, increases the risk of early miscarriage, ectopic pregnancy, and perinatal complications. Owing to the poor utilization and short-term effectiveness of current stem cell therapies, their efficacy in improving reproductive outcomes in patients with TE is limited. Endometrial extracellular matrix (EMECM) modified hemispherical hydrogels (ECMHPs) are developed to deliver menstrual blood-derived stem cells (MenSCs) to promote TE repair. The irregular porous structure on one side of the hemispherical hydrogel microspheres significantly enhances their retention capability on the uterine wall, whereas the bioactive decellularized EMECM promotes MenSCs proliferation and paracrine function. In vitro experiments demonstrate that ECMHPs@MenSCs promote the proliferation of damaged human endometrial stromal cells (HESCs) while inhibiting their fibrosis and apoptosis. In TE rat models, intrauterine transplantation of the ECMHPs delivering MenSCs system (ECMHPs@MenSCs) extends retention time, promotes endometrial thickness by 2.3-fold, increases glandular number by 3.7-fold, inhibits endometrial fibrosis. Additionally, this system improves endometrial regeneration markers and uterine receptivity, and restores fertility, with a mean gestational sac number of 11.0 ± 2.8 compared with only 2.3 ± 2.1 in the untreated group. This study provides an efficient and convenient therapeutic strategy for TE repair and fertility restoration.

Cranial defect repair is frequently hindered by limited intrinsic regenerative capacity, infection risks, and chronic inflammation, whereas conventional grafts and inert implants often suffer from poor host integration. Herein, we report an extracellular matrix–mimicking hydrogel that synchronizes angiogenesis, osteogenesis, and immunomodulation through dynamic thiol–disulfide chemistry and sacrificial prevascularization. Hyaluronic acid functionalized with L-cysteine ethyl ester (HACys) was crosslinked with allicin to yield a viscoadaptive, stress-relaxing network, within which type I collagen was extruded into VEGF-loaded threads to serve as sacrificial templates (HACys-VEGF@Coll-A). Upon enzymatic degradation, these threads generated VEGF-lined microchannels designed to guide rapid vascular ingress. The composite demonstrated excellent cytocompatibility with BMSCs and selectively enhanced HUVEC viability and spreading. Furthermore, VEGF-presenting constructs significantly promoted endothelial tube formation and migration while upregulating VEGF mRNA, confirming preserved bioactivity. In BMSCs, VEGF-containing hydrogels increased alkaline phosphatase activity and mineral deposition, concomitant with the upregulation of osteogenic genes (ALP, COL1, RUNX2, OCN). Notably, macrophages shifted from a CD86high/CD206low toward a CD86low/CD206high phenotype with decreased TNF-α and increased IL-10 secretion, indicating the establishment of a pro-resolution immune microenvironment. Collectively, HACys-VEGF@Coll-A forms a dynamic, remodelable scaffold that integrates preformed vascular conduits while supporting osteogenesis and tempering inflammation, thereby addressing major barriers to cranial defect repair and warranting further in vivo evaluation of release kinetics, channel architecture, and mechanics.

Proper niche environment is the key factor for mesenchymal stem cells (MSCs) involved tendon-lineage differentiation and regeneration. In the present study, soluble tendon derived extracellular matrix (sTECM) was used to modify aligned electrospun fibers with acid neutralizing effect in order to create an optimal niche environment for tendon regeneration, and its efficacy was evaluated using mouse MSC tenogenic differentiation in vitro and rat Achilles tendon regeneration in vivo . The results showed that sTECM modified ultrafine fibers were more cytocompatible and capable of inducing a predominant tenogenic phenotype of mouse MSCs with the enhanced expression of tendon markers including scleraxis , tenomodulin , collagen III , mohawk homeobox , decorin , fibromodulin and biglycan . By contrast, pure sTECM components led to enhanced multi-lineage differentiation of mouse MSCs including tenogenic lineage. This inductive effect was further enhanced when unilateral mechanical loading was applied in vivo. Moreover, the implantation of sTECM modified scaffold created an ideal niche environment including bioactive sTECM components, topological and mechanical signals, upon in vivo implantation for in situ tendon regeneration. As the results, sTECM modified scaffold in comparison with the non-modified one regenerated higher quality tendon tissue with the features of more mature tissue structure, better tissue organization, cell density and alignment as well as better collagen ultrastructure. Quantitative analysis demonstrated significantly bigger collagen fibril diameter, stronger mechanical property and better tissue grading score and higher expression levels of tenogenic markers when compared to those of control scaffold (p < 0.05). This study demonstrated that sTECM modified pH-neutral ultrafine fibers may represent a novel bioactive scaffold for in situ tendon regeneration, which deserves further investigation in large animals.

Aberrant destruction of the articular extracellular matrix (ECM) has been considered to be one of the pathological features of osteoarthritis (OA) which results in chondrocyte changes and articular cartilage degeneration. The MAPK signaling pathway serves a key role by releasing cartilage‑degrading enzymes from OA chondrocytes. However, the use of MAPK inhibitors for OA is hindered by their potential long‑term toxicity. Vicenin 3 is one of the major components of the Jian‑Gu injection which is effective in the clinical treatment of OA. However, its potential impact on OA remain poorly understood. Therefore, the present study aimed to assess the effects of vicenin 3 on interleukin (IL)‑1β‑treated SW1353 chondrocytes, which mimic the microenvironment of OA. These chondrocytes were pretreated with vicenin 3 (0, 5 and 20 µM) for 1 h and subsequently stimulated with IL‑1β (10 ng/ml) for 24 h. Nitric oxide (NO) production was measured using the Griess reaction, whereas the production of prostaglandin E2 (PGE2), matrix metalloproteinases (MMPs), A disintegrin‑like and metalloproteinase with thrombospondin motifs (ADAMTSs), collagen type II and aggrecan were measured using ELISA. The mRNA expression of MMPs and ADAMTSs were measured using reverse transcription‑quantitative PCR. The protein expression levels of MAPK were measured using western blotting. Vicenin 3 was found to significantly inhibit IL‑1β‑induced production of NO and PGE. Increments in the expression levels of MMP‑1, MMP‑3, MMP‑13, ADAMTS‑4 and ADAMTS‑5 induced by IL‑1β, in addition to the IL‑1β‑induced degradation of collagen type II and aggrecan, were all reversed by vicenin 3 treatment. Furthermore, vicenin 3 suppressed IL‑1β‑stimulated MAPK activation, an effect that was similar to that exerted by SB203580, a well‑known p38 MAPK inhibitor. In conclusion, vicenin 3 may confer therapeutic potential similar to that of the p38 MAPK inhibitor for the treatment of OA.

The effective management of oral ulcers is hindered by the constantly moist oral environment and persistently dysregulated inflammatory microenvironment. Inspired by the hierarchical architecture of native mucosa, we engineered a biomimetic bilayer patch to achieve immune modulation and synergistic tissue regeneration. Compositionally mimicking the bioactive lamina propria, the patch features a bioadhesive inner layer enriched with dopamine-modified esophageal mucosa decellularized extracellular matrix (eECM-DA) to specifically recapitulate the native biochemical niche, embedded within hyaluronic acid methacrylate (HAMA) networks loaded with dexamethasone (Dex). Structurally mimicking the protective epithelial barrier, this inner matrix was coupled with a hydrophobic ethylcellulose (EC) backing to shield against saliva erosion. After contact with the moist oral environment, this eECM-DA/HAMA/EC-Dex (EDH/EC-Dex) bilayer patch rapidly contacts the wound and displaces interfacial water to achieve robust wet adhesion. Functionally, the EDH/EC-Dex patch actively reprogrammed the inflammatory milieu in lipopolysaccharide (LPS)-stimulated macrophages by modulating JAK-STAT signaling and antioxidant networks, thereby driving inflammation resolution and promoting orderly regulation of the ulcer microenvironment. In a rat oral ulcer model, EDH/EC-Dex patches demonstrated superior efficacy in accelerating re-epithelialization and functional recovery compared to commercial products. This work introduces an immune-modulatory, ECM-derived platform, offering a promising strategy for oral ulcer management and broader tissue engineering applications. Statement of significance A mucosa-inspired bilayer patch is developed for treating oral ulcers. This ECM-derived patch achieves robust wet adhesion and provides a protective barrier against saliva erosion through its bioadhesive inner layer and hydrophobic backing. Mechanistically, it actively regulates JAK-STAT signaling and antioxidant networks, effectively reprogramming the dysregulated inflammatory microenvironment to promote rapid re-epithelialization and functional recovery. Designing dressings that mimic the composition, structure, and function of native skin is of great significance. Despite commercial decellularized extracellular matrix (dECM)-based wound dressings promoted skin regeneration process, reconstructing the fibrous microstructure of native dermis remains a significant challenge. In this work, a dECM-based composite nanofibrous membranes loaded with stem cell secretomes (SCS/dECMM) have been developed for accelerating wound repair, which comprehensively mimicked the dermal tissue in terms of composition, structure, and function. In detail, dermal dECM was prepared by eco-friendly supercritical carbon dioxide (SC-CO 2 ) technology and then mixed with SCS to obtain SCS/dECMM by using electrospinning technology. The resulting nanofibrous membrane with 555.19 nm diameter that maintained SCS bioactivity while enabling sustained SCS release. SCS/dECMM significantly enhanced cell adhesion, proliferation, migration, and angiogenesis, thereby continuously promoting wound repair. In vivo wound healing results revealed that SCS/dECMM accelerated wound healing by promoting re-epithelialization, collagen deposition, and vascularization. SCS/dECMM offered a novel strategy for accelerating wound healing by replicating the composition, structure, and function of native skin.

Aim: To develop an effective strategy for increasing angiogenesis at diabetic wound sites and thereby accelerating wound healing. Materials & methods: A micropatterned nanofibrous scaffold with bioglass nanoparticles encapsulated inside coaxial fibers was prepared by electrospinning. Results: Si ions could be released in a sustained manner from the scaffolds. The hierarchical micro-/nano-structure of the scaffold was found to act as a temporary extracellular matrix to promote endothelial cell adhesion and growth. The scaffold greatly improved angiogenesis and collagen deposition at the wound site, which shortened the healing period of diabetic wounds. Conclusion: This study provides a promising therapeutic option for chronic diabetic wounds with improved angiogenesis.

Mesenchymal stromal cells (MSCs) hold great promise for tissue regeneration due to their potent paracrine effects. However, the absence of extracellular matrix (ECM) support following transplantation significantly compromises their survival and therapeutic efficacy. To address this, we developed a single-cell encapsulation strategy using an ECM-mimetic supramolecular hydrogel system based on host–guest chemistry. In this approach, cholesterol–polyethylene glycol–adamantane is inserted into the MSC membrane via hydrophobic interactions, enabling the subsequent formation of a uniform hydrogel coating through specific recognition between cyclodextrin- and adamantane-modified hyaluronic acids. This facile and biocompatible strategy achieves high encapsulation efficiency without the need for complex equipment, while preserving cell viability and function. Encapsulated MSCs exhibited enhanced resistance to pathological stress, improved survival, and superior therapeutic efficacy in a rat model of myocardial infarction. These findings highlight the potential of supramolecular single-cell encapsulation to augment MSC-based therapies for tissue repair and regenerative medicine.

Fibrotic diseases, which impair tissue function and contribute to organ failure, remain a major clinical challenge with limited treatment options. Mesenchymal stromal cells (MSCs) offer promise for antifibrotic therapy via paracrine signaling, but their clinical efficacy is hindered by poor survival and limited functional activity after transplantation. Here, we present a cell surface engineering strategy that reprograms the antifibrotic function of MSCs by constructing a pseudofibrotic extracellular matrix (ECM) on their surface. Through in situ self-assembly of peptide-modified hyaluronic acid, we generate a nanofiber-based matrix that mimics the dense, disordered architecture of fibrotic ECM. This matrix activates the Piezo1/PI3K-Akt signaling pathway, inducing up-regulation of Mmp13—a key collagen-degrading matrix metalloproteinase—in engineered MSCs. In a rat model of myocardial infarction–associated fibrosis, engineered MSCs exhibit robust antifibrotic activity compared to unmodified MSCs. These findings establish a bioinspired strategy for MSC reprogramming and offer a path toward more effective cell-based therapies for fibrotic disease.
