Background Larval zebrafish ( Danio rerio ) is not only an ideal vertebrate applied in Fish Embryos Toxicity (FET) test but also a well-accepted model in behavioral neurotoxicity research. By applying the commercial standard behavioral tracking system (Zebrabox), the locomotion profiles (neurobehavioral-phenomics) of larval zebrafish can be comprehensively monitored and systematically analyzed to probe ecotoxicological neurotoxicity of nano-pollutants at environmental relevant concentration level. Results Herein, the potential toxicity of at environment relevant concentration level on embryonic zebrafish was evaluated by FET and neurobehavioral-phenomics (NBP). The embryos were exposed to the environmental relevant concentration (0.05, 0.1,1, 5, 10, 100 μg/L). The FET criteria were utilized to evaluate the ecotoxicological effect induced by silica NPs. Subsequently, behavioral neurotoxicity of silica NPs was further quantified via locomotion response (LMR). Specifically, the alteration of Light/Dark challenge (LDC) evoked by light/dark stimulation was detected and analyzed by commercially standard behavioral protocols using zebrabox. We revealed that the exposures of silica NPs at environmental relevant concentration (0.05, 0.1, 1, 5, 10,100 μg/L) significantly disturbed locomotion profiles of larval zebrafish. Additionally, it was obviously noted that low, environmentally relevant silica concentrations might result in altering the total behavioral profiles in developing zebrafish. Conclusions In sum, neurobehavior phenomics profiling based on LMR and LDC is a potent methodology for the evaluation of sub-lethal or sub-teratogenic toxicity. Compared with the FET tests characterized by the detection of embryonic teratogenicity, the neurobehavior phenomics based method can be more sensitive to determine sub-teratogenic toxicity of silica NPs at environmental concentrations. With the combination of multivariate data analysis, this approach would offer effective technical reference for environmental nano-toxicology research.

Two-dimensional transition-metal dichalcogenides (TMDs) are of interest for beyond-silicon electronics 1 , 2 . It has been suggested that bilayer TMDs, which combine good electrostatic control, smaller bandgap and higher mobility than monolayers, could potentially provide improvements in the energy-delay product of transistors 3 , 4 , 5 . However, despite advances in the growth of monolayer TMDs 6 , 7 , 8 , 9 , 10 , 11 , 12 , 13 , 14 , the controlled epitaxial growth of multilayers remains a challenge 15 . Here we report the uniform nucleation (>99%) of bilayer molybdenum disulfide (MoS 2 ) on c -plane sapphire. In particular, we engineer the atomic terrace height on c -plane sapphire to enable an edge-nucleation mechanism and the coalescence of MoS 2 domains into continuous, centimetre-scale films. Fabricated field-effect transistor (FET) devices based on bilayer MoS 2 channels show substantial improvements in mobility (up to 122.6 cm 2 V −1 s −1 ) and variation compared with FETs based on monolayer films. Furthermore, short-channel FETs exhibit an on-state current of 1.27 mA μm −1 , which exceeds the 2028 roadmap target for high-performance FETs 16 .

Herein, a novel and simple electrospray (ES) printing technique was developed for the fabrication of ultrathin graphene layers with precisely controlled nanometer-scale thickness, where graphene oxide (GO) was electrosprayed on wafers and subsequently chemically reduced into reduced GO (rGO). Utilizing that technique, we prepared ultrathin rGO in-plane graphene field-effect transistor (GFET)-based biosensors coupled with a portable prototype measuring system for point-of-care detection of pathogens. We illustrate the use of such prepared GFETs to detect COVID-19, using the SARS-CoV-2 nucleocapsid protein antigen (N-protein) and genomic viral RNA as detection targets. The electrosprayed and chemically reduced rGO films enhance the molecular detection in GFET sensors through significant local gating effects. The device detects the N-protein from the SARS-CoV-2 Omicron variant in a culture medium with an LOD of 1.44 PFU/mL and in clinical oropharyngeal samples with an LOD of 45 genome copies/mL in 5 min. It also successfully detects viral RNA in oropharyngeal swabs within 10 min. The GFET sensor responses were further analyzed using our proprietary wireless, miniaturized, and portable FET analyzer, coupled with a smartphone detecting app. Altogether, we present low-cost and mass-producible GFETs with high-quality graphene channels, enabling a portable, efficient, and accurate solution for point-of-care pathogen detection and in clinical testing. This technology has the potential to become a crucial tool in preventing future global epidemic outbreaks.

Field effect transistor (FET) has attracted high attention in biomolecules detection. However, the sensitivity of FET-based biosensors is often limited by the charge screening effect. In addition, the facile and ultra-sensitive detection for small biomolecules, which possess weak charge or electroneutral, remains to be studied. Here, the self-assembled monolayer AuNPs/three-dimensional (3D) crumpled graphene FET is structured for a biosensor by shrinking flexible polystyrene (PS) films through heat treatment method and realized label-free and ultra-sensitive detection of dopamine (DA) using DA aptamer as probes immobilized on the AuNPs/3D crumpled graphene surface. The nanoscale deformation caused by thermal expansion effect of flexible graphene/PS can effectively reduce charge screening and the synergistic application of crumpled graphene and AuNPs can promote electron transfer and the graphene carrier concentration, leading to conductivity increase and hydrophilicity enhancement of 3D graphene. In addition, the binding of DA aptamer to DA will cause conformational changes of the aptamer molecule, which affects the charge transport properties of the sensor, thus improving its selectivity and stability. The biosensor can also easily distinguish interfering substances for DA detection in complex components (PBS, human urine, and fetal calf serum) with the detection limits as low as 60, 240 and 316 zM (10 −19 –10 −11 M), respectively. DA released from exocytosis induced by K + stimulation was selectively detected. The results show that the biosensor can be used as an excellent tool for ultrasensitive molecular recognition and detecting the effects of exogenous reagents on living cells, which is promising for clinical diagnosis and early disease prevention.
