Widely targeted metabolome profiling of different plateau raspberries and berry parts provides innovative insight into their antioxidant activities

Ren Xiaoli,Wang Shulin,Wang Jinying,Xu Dan,Ye Ying,Song Yangbo

Raspberries are highly nutritious and have powerful antioxidant properties, making them functional berries with positive effects on physiological functioning. However, there is limited information available on the diversity and variability of metabolites in raspberry and its parts, especially in plateau raspberries. To address this, commercial raspberries and their pulp and seeds from two plateaus in China were subjected to LC-MS/MS-based metabolomics analysis and evaluated for antioxidant activity using four assays. A metabolite-metabolite correlation network was established based on antioxidant activity and correlation analysis. The results showed that 1661 metabolites were identified and classified into 12 categories, with significant variations in composition between the whole berry and its parts from different plateaus. Flavonoids, amino acids and their derivatives, and phenolic acids were found to be up-regulated in Qinghai’s raspberry compared to Yunnan’s raspberry. The main differently regulated pathways were related to flavonoid, amino acid, and anthocyanin biosynthesis. The antioxidant activity of Qinghai’s raspberry was stronger than Yunnan’s raspberry, and the order of antioxidant capacity was seed > pulp > berry. The highest FRAP (420.31 µM TE/g DW) values was found in the seed of Qinghai’s raspberry. Overall, these findings suggest that the environment in which the berries grow can affect their chemical composition, and comprehensive exploitation and cultivation of whole raspberry and its parts from different plateaus can lead to new opportunities for phytochemical compositions and antioxidant activity.

Phytochemical profiles, antioxidant and antimicrobial activities of three Potentilla species

Wang Shan-Shan,Wang Dong-Mei,Pu Wen-Jun,Li Deng-Wu

Background Extracts from Potentilla species have been applied in traditional medicine and exhibit antioxidant, hypoglycemic, anti-inflammatory, antitumor and anti-ulcerogenic properties, but little has been known about the diversity of phytochemistry and pharmacology on this genus. This study investigated and compared the phytochemical profiles, antioxidant and antimicrobial activities of leaf extracts from three Potentilla species (Potentilla fruticosa, Potentilla glabra and Potentilla parvifolia) in order to discover new resources for lead structures and pharmaceutical products. Methods Chemical composition and content of six phenolic compounds were evaluated and determined by RP-HPLC; Total phenolic and total flavonoid content were determined using Folin-Ciocalteau colourimetric method and sodium borohydride/chloranil-based method (SBC); Antioxidant activities were determined using DPPH, ABTS and FRAP assays; Antimicrobial properties were investigated by agar dilution and mycelial growth rate method. Results The results showed hyperoside was the predominant phenolic compound in three Potentilla species by RP-HPLC assay, with the content of 8.86 (P. fruticosa), 2.56 (P. glabra) and 2.68 mg/g (P. parvifolia), respectively. The highest content of total identified phenolic compounds (hyperoside, (+)-catechin, caffeic acid, ferulic acid, rutin and ellagic acid) was observed in P. parvifolia (14.17 mg/g), follow by P. fruticosa (10.01 mg/g) and P. glabra (7.01 mg/g). P. fruticosa possessed the highest content of total phenolic (84.93 ± 0.50 mmol gallic acid equivalent/100 g) and total flavonoid (84.14 ± 0.03 mmol quercetin equivalent/100 g), which were in good correlation with its significant DPPHIC50 (16.87 μg/mL), ABTS (2763.48 μmol Trolox equivalent/g) and FRAP (1398.70 μmol Trolox equivalent/g) capacities. Furthermore, the effective methodology to distinguish the different species of Potentilla was also established by chromatographic fingerprint analysis for the first time. The results of antimicrobial activities showed P. fruticosa exhibited the strongest inhibition aganist Gram-positive bacteria, Pseudomonas aeruginosa and Candida albicans with MIC values of 0.78–6.25 mg/mL. P. parvifolia possessed antibacterial and antifungal activities against all the microorganisms tested, with EC50 and MIC values of 20.52–47.02 mg/mL and 0.78–50 mg/mL, respectively. Conclusion sThese results indicated that leaf extracts from three Potentilla species could become useful supplement for pharmaceutical products as a new antioxidant and antimicrobial agents.

Guangkai Wu,Zihan Zhao,Jun Hu,Yue Li,Jianxia Sun,Weibin Bai

The chemical synthesis of anthocyanins, especially delphinidin-3-O-glucoside and petunidin-3-O-glucoside, is preferable due to the challenges associated with their extraction and purification. However, the reported methods for the synthesis are scarce and intricate. Our research focused on exploring a one-step ester-to-ketone process and optimizing the ring formation reaction, simplifying and improving the overall synthesis strategy. Through these attempts, we were able to achieve higher production yields of delphinidin-3-O-glucoside and petunidin-3-O-glucoside. According to the results of DPPH, ABTS, and FRAP, the antioxidant activity of anthocyanins was increased with the number of B ring hydroxyl substituent. Additionally, both delphinidin-3-O-glucoside and petunidin-3-O-glucoside exhibited no cytotoxicity effects, highlighting their potential for safe application in various fields.

Comparative study on optimization of NADES extraction process by dual models and antioxidant activity of optimum extraction from Chuanxiong-Honghua

Rong-Shuai Hu,Li Yu,Sai-Ya Zhou,Hui-Fen Zhou,Hai-Tong Wan,Jie-Hong Yang

The natural deep eutectic solvent (NADES) was used as the extraction solvent to extract the four ingredients from Chuanxiong-Honghua. NADES-7 composed of L-proline and lactic acid (1:2) has a better extraction effect compared with traditional solvents. The extraction process was optimized using response surface methodology (RSM) and ant colony algorithm-enabled backpropagation neural network (ACO-BPNN), which were compared with each other. ACO-BPNN has a higher R 2 and predictive value than RSM. The optimal process parameters predicted by the ACO-BPNN were determined to be as follows: solvent-to-material ratio, 8 mL/g; extraction time, 67 min; water concentration, 48%; and extraction temperature, 50 °C. The best predicted comprehensive evaluation value was 4.2634, and the mean value obtained from the experiment was 3.8678. Compared with traditional solvents, the antioxidant activity of NADES-7 extract was better, and the mean value of DPPH, FRAP (ferric reducing antioxidant power), and ABTS assays were 0.1555, 0.1132, and 0.0894 mmol Trolox/g dry weight, respectively. Experimental results show that ACO-BPNN can better optimize the extraction process of many different polar compounds in plant combinations.

Rapid discovery of natural antioxidants in Hypericum japonicum: Dual roles of the liquid phase mobile phase as extraction and separation solvent

Zheng-ming Qian,Meng-qi Wu,Jing Chen,Qi Huang,Deng-yun Fan,De-qiang Li

Hypericum japonicum is a traditional folk medicine with various bioactivities such as hepatoprotective, antioxidant, and anti-tumorous. The antioxidant effect of H. japonicum is one of the most prominent effects due to its responsibility for many of its activities. To clarify active natural substance, the antioxidant properties of H. japonicum were preliminarily assessed by ferric reducing–antioxidant power (FRAP), 2,2-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) and Oxygen radical absorbance capacity (ORAC), as well as superoxide dismutase (SOD). Then, a straightforward and effective method named online liquid extraction-high performance liquid chromatography combined with ABTS antioxidant assay and mass spectrometry (OLE-HPLC-ABTS/Q-TOF-MS) was developed to swiftly and directly discover the antioxidants in H. japonicum . Using mobile phase as extraction and separation reagent, coupled with online activity analysis and compounds identification by high-resolution MS, the online system enables rapid screening of natural antioxidant bioactives from complex mixture. By using it, a total of 9 compounds including flavonoids and phenolic acids characterized by retention time, precise mass, and fragmentation ions in MS/MS spectra showed antioxidant action. Finally, the antioxidant and SOD activity of main found active compounds were validated by in vitro experiment assay and molecular docking. In summary, these results suggested that H. japonicum could be considered as a potential source of natural antioxidants, and the online integrated system might become a promising candidate for the natural antioxidants discovery in the future.

Yin Xing-shuo,Zhang Xue-qin,Li De-qiang

In this study, the ultrasound-assisted extraction (UAE) of flavonoids with maximal antioxidant and tyrosinase inhibitory activities and identification of the principal individual flavonoids compounds of Semen Oroxyli by UHPLC-DAD-QTOF-MS/MS was proposed. The method was optimized by a three-factor-three-level Box–Behnken design (BBD) and Derringer’s desirability function using methanol concentration, extraction time, and liquid/solid ratio as factors, while, the total flavonoids content (TFC), antioxidant activities (DPPH, ABTS, FRAP, superoxide anion and hydroxyl radicals) and tyrosinase inhibitory activity as responses. The experimental results showed that the optimal extraction conditions were as followed: a solvent of 69.90% methanol, an extraction time of 30.64 min and a liquid/solid ratio of 31.30 mL/g, which give individual response values of 19.79 mg RE/g, 78.66%, 99.92%, 19.05 mM FE/g, 20.49%, and 13.37% for TFC, DPPH, ABTS, FRAP, superoxide anion and hydroxyl radicals, respectively. In addition, the tyrosinase inhibition assay showed that the extract of Semen Oroxyli exhibited strong inhibitory activity against the tyrosinase. There were no significant differences between the results of the optimization experiments and those predicted by the models. By UHPLC-DAD-QTOF-MS/MS analysis, the main flavonoids identified in the optimized UAE extracts of Semen Oroxyli were oroxin B, oroxin A, chrysin-7- O -β- d -glucuronid, baicalein and chrysin.

Qianqian Chen,Li Yu,Huifen Zhou,Haofang Wan,Yangyang Zhang,Haitong Wan,Jiehong Yang

The Box-Behnken design-response surface methodology (BBD-RSM) and Back-propagation neural network prediction model optimized by Simulated annealing (SA-BPNN) were used to optimize the extraction process of Astragali radix - Paeoniae radix alba by the green natural deep eutectic solvent (NADES), and the antioxidant activity of them was discussed. The best NADES for extracting Astragali radix - Paeoniae radix alba need to be screened out. The comprehensive evaluation value of the extraction yield was used as the index. The BBD-RSM and SA-BPNN were established for prediction and optimization. The antioxidant activity of Astragali radix - Paeoniae radix alba in vitro and in vivo was studied by detecting 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate and ferric reducing antioxidant power (FRAP) and the expression levels of SOD, MDA and GSH-Px in rat plasma. The results showed that the best NADES formula screened was Betaine-Urea (Bet-Ur). Compared with the BBD-RSM, the SA-BPNN had a higher degree of fitting, the comprehensive evaluation value was higher, and the error between tested and predicted value was smaller, which was more suitable as a prediction model for the extraction yield of Astragali radix - Paeoniae radix alba . Through the determination of antioxidant kits and oxidative factors, the results showed that Astragali radix - Paeoniae radix alba had antioxidant activity in vitro and in vivo . The study on the extraction process optimization and antioxidant activity of Astragali radix - Paeoniae radix alba were expected to provide a theoretical basis for its effective utilization.

Ultrasound-assisted extraction of polyphenols from Phyllanthi Fructus: Comprehensive insights from extraction optimization and antioxidant activity

Haixia Che,Rui Zhang,Xianyao Wang,Hong Yu,Xuepeng Shi,Jie Yi,Jiawen Li,Qi Qi,Ruizhi Dong,Qian Li

Phyllanthi Fructus (PF) is a valuable botanical resource with a long history of traditional use, known for potent antioxidant and anti-inflammatory effects attributed to its rich contents of bioactive compounds, particularly polyphenols. However, current extraction techniques limit the utilization of polyphenols from PF. This study aimed to achieve the maximum polyphenol yield and improve the antioxidant activity of PF extracts to promise PF’s prospects for modern healthcare. Firstly, ultrasonic-assisted extraction (UAE) was employed to extract the polyphenols in PF and a combination of Plackett-Burman designs (PBD) and response surface methodology (RSM) was applied to optimize UAE’s conditions. Next, cellular superoxide dismutase (SOD) and malondialdehyde (MDA) were used to assess the antioxidant activity of extracted polyphenols. Ultra-Performance Liquid Chromatography coupled with Quadrupole Time-of-Flight Mass Spectrometry (UPLC-Q-TOF MS) was utilized to characterize polyphenol components in the PF extracts. Finally, network pharmacology and molecular docking analysis were performed to screen the potential target proteins of polyphenols from PF. As a result, the optimized polyphenol yield was 213.49 mg/g, and the antioxidant activities, measured by ability of DPPH scavenging, ABTS+ scavenging, and FRAP were 76.95 %, 2.24 mmol/g, 2.34 mmol/g, respectively. PF extracts also showed good antioxidant capabilities at cellular level. 26 polyphenol components were identified in the PF extracts. Among these, ellagic acid, myricetin, and eriodictyol may exert antioxidant effects by interacting with AKT serine/threonine kinase 1 (AKT1). In conclusion, our study provides valuable insights into the optimizing PF extraction and underscores its potential applications in enhancing natural polyphenols extraction using UAE with a combination of PBD and RSM. These findings offer a promising avenue for the development and utilization of PF, and could serve as a reference for similar extraction processes in the future.
