Topological Control of Lanthanide-Doped Upconversion Nanocrystals for Improved FRET Coupling Efficiency in Second Near-Infrared-Triggered Photodynamic Therapy

Wang Wang,Qiqing Li,Haoran Chen,Xiaoyu Xie,Bin Li,Xianggui Kong,Yulei Chang

The precise control of the topological morphology and hierarchical composition is essential for the specific design of multifunctional lanthanide-doped nanocrystals (NCs). This control also enhances our understanding of the epitaxial growth mechanism in core–shell nanostructures, particularly in the case of NaErF4-based NCs, which exhibit minimal detectable upconversion emission without an epitaxial shell. Herein, we established a precise regulation strategy for the fabrication of disk- and rod-shaped core–shell NCs, catering to the growing interest in anisotropic core–shell NCs and aiming to improve Förster resonance energy transfer (FRET) efficiency for photodynamic therapy (PDT) applications. We synthesized NaErF4@NaREF4 NCs (RE = Y, Lu) with distinct topological morphologies by deliberately controlling the Ostwald ripening process. Furthermore, we observed that sacrifice NCs obtained through varying treatment times had diverse ion and oleate ligand release rates. These differences in the ripening reaction pathways were responsible for the ultimate variation in topological morphologies. Steady-state and dynamic luminescence studies revealed that the ligand-free rod-shaped NCs exhibited more robust upconversion emissions, and higher FRET coupling efficiency (54.0%) with photosensitizers, resulting in more generation efficiency of singlet oxygen. These findings suggest promising applications in second near-infrared-triggered PDT.

A Targeted and FRET-Based Ratiometric Fluorescent Nanoprobe for Imaging Mitochondrial Hydrogen Peroxide in Living Cells

Fangkai Du,Yunhao Min,Fang Zeng,Changming Yu,Shuizhu Wu

Hydrogen peroxide (H 2 O 2 ) is a prominent member of the reactive oxygen species family and plays crucial roles in living organisms, thus detecting H 2 O 2 and elucidating its biological functions has become an important area of biological and biomedical research. Herein, a multifunctional fluorescent nanoprobe is demonstrated for detecting mitochondrial H 2 O 2 . The nanoprobe is prepared by covalently linking a mitochondria-targeting ligand (triphenylphosphonium, TPP) and a H 2 O 2 recognition element (PFl) onto carbon dots (CDs). For this nanoprobe, the CD serves as the carrier and the FRET donor. In the presence of H 2 O 2 , the PFl moieties on a CD undergo structural and spectral conversion, affording the nanoplatform a FRET-based ratiometric probe for H 2 O 2 . The nanoprobe displays excellent water dispersibility, high sensitivity and selectivity, satisfactory cell permeability, and very low cytotoxicity. Following the living cell uptake, this nanoprobe can specifically target and stain the mitochondria; and it can detect the exogenous H 2 O 2 in L929 cells, as well as the endogenously produced mitochondrial H 2 O 2 in Raw 264.7 cells upon stimulation by PMA. This study shows that CDs can serve as promising nano-carriers for fabricating practical multifunctional fluorescent nanosensors.

A ratiometric fluorescent sensor based on g-CNQDs@Zn-MOF for the sensitive detection of riboflavin via FRET

Shasha Feng,Fubin Pei,Yi Wu,Jingjing Lv,Qingli Hao,Tinghai Yang,Zhaoyang Tong,Wu Lei

A novel ratiometric fluorescent sensor based on Förster resonance energy transfer (FRET) platform was designed for riboflavin (RF) detection. The graphitic carbon nitrides quantum dots - Zn-MOF composite (g-CNQDs@Zn-MOF) was used as the fluorescent probe . In the FRET system, g-CNQDs@Zn-MOF and RF acted as donor and acceptor, respectively. The probe exhibited high sensitivity and good selectivity to RF, and had been successfully used for the detection of RF in milk and vitamin B 2 tablets. The detection limit of the sensor was 15 nM. The strategy expanded the application of MOF in sensing filed and provided a new method for the detection of RF.

Dual-FRET platform based on dsDNA-functionalized Ti3C2Tx MXene for highly sensitive detection of bacteria

Langjin Zhang,Yingwen Wang,Yubing Jiao,Min Mao,Weiliang Xiong,Yingjie Zhang,Youning Wang,Songbai Zhang,Wei Zhang,Jinguang Yang

Pathogenic bacteria represent a considerable threat to public health and food safety, necessitating the development of advanced detection methodologies. In this study, a novel fluorescence sensing platform based on fluorescence resonance energy transfer (FRET) was established to detect Pathogenic bacteria with rapid response, exceptional sensitivity, and stable selectivity. Using carboxyl-modified Ti 3 C 2 T x MXene (Ti 3 C 2 T x -COOH) as nanocarriers, double-stranded DNA (dsDNA) were assembled to functionalize the MXene for rapidly efficient quenching of fluorescence dye-labeled aptamers and specific recognition of target bacteria. Upon the incorporation of target bacteria, the aptamer preferentially bound to bacteria, dissociating from the MXene and resulting in a significant and rapid increase in fluorescence intensity. Using E. coli O157:H7 and S. aureus as exemplar biotargets, the sensor demonstrated ultra-sensitive detection of E. coli O157:H7 and S. aureus with limits of detection (LOD, S/N = 3) as low as 0.61 CFU/mL and 1.62 CFU/mL, respectively, within a rapid response time of 15 min, thereby demonstrating superior performance in comparison to the majority of existing FRET-based biosensors. Moreover, this method demonstrates excellent reproducibility and stability, having been successfully applied to spiked milk samples. The recovery rates for S. aureus ranged from 90.2 % to 114.3 %, with acceptable RSDs of 1.4 % to 7.4 %, while those for E. coli O157:H7 ranged from 91.7 % to 93.6 %, with acceptable RSDs of 5.2 % to 8.7 %, confirming its strong potential for practical sample analysis. The present study establishes a foundational framework for subsequent research into biosensors for detecting targets and provides a new pathway for the analysis of other biological factors.

Conjugation of PPV functionalized mesoporous silica nanoparticles with graphene oxide for facile and sensitive fluorescence detection of TNT in water through FRET

Haixia Zhang,Lijuan Feng,Bingxin Liu,Cuiyan Tong,Changli Lü

This paper reported a convenient method to prepare water-soluble graphene oxide (GO) functionalized by amine-modified mesoporous silica nanoparticles (MSNs) containing poly(p-phenylenevinylene) via covalent bonds . The resulted GO–PPV@MSN hybrid materials had a good dispersion and strong green fluorescence in aqueous solution. The structure, morphology and optical properties of the resulting hybrid materials were characterized. The GO-PPV@MSN can be used as a novel and effective fluorescent sensing platform for detection of 2,4,6-trinitrotoluene (TNT) in aqueous solution by fluorescence resonance energy transfer (FRET). Upon the addition of TNT, the amino groups on the surface of PPV@MSN can bind TNT molecules from solution by forming Meisenheimer complex, and this complex can absorb the green part of visible light, and strongly suppress the fluorescence emission of GO-PPV@MSN through FRET. The observed linear fluorescence intensity change allowed the quantitative detection of TNT with a detection limit down to 1.3 × 10 −7  M.

Development of a PCR fluorescence sensor utilizing an upconversion FRET system for rapid and ultra-sensitive determination of Escherichia coli

Han Lu,Song Yaqi,Chen Min,Pan Leiqing,Tu Kang,Su Jing

An upconversion-gold nanoparticle detection system that integrates PCR amplification and fluorescence resonance energy transfer was constructed to enable swift and highly sensitive identification of Escherichia coli . The forward primer used in the PCR amplification is modified with sulfhydryl groups, enabling its connection to gold nanoparticles via Au–S bonds. The complementary strand of the forward primer, which is attached to the upconversion nanomaterials, can hybridize with the free forward primer through base complementary pairing. This interaction induces fluorescence resonance energy transfer, resulting in fluorescence quenching. The concentration of the target bacteria influences the amount of free primer in the system after PCR amplification, which subsequently alters the intensity of the upconversion fluorescence. The fluorescent PCR sensor developed based on the aforementioned principles demonstrated a detection limit of 14 CFU/mL for E. coli , with a quantitative detection range of 18–1.8 × 10 7  CFU/mL. In comparison to the qPCR method, the number of PCR cycles required for the constructed biosensor can be reduced to approximately 22 to achieve the same detection limit, effectively decreasing the detection time by about 24 min. The spiked recoveries in chicken were 91.8–106.0% with the relative standard deviations less than 10%, indicating that the constructed method exhibits good applicability. Graphical abstract

Multiplexed and highly sensitive FRET aptasensor for simultaneous assay of multiple antibiotics via DNAzyme and catalytic strand displacement amplification cascades

Tingting Gong,Huaifeng Yan,Daxiu Li,Bingying Jiang,Yun Xiang,Ruo Yuan

Background The emergence of antibiotic-resistant microorganisms poses significant risks to public health. Therefore, the development of technologies capable of detecting antibiotics with high sensitivity and selectivity is essential for monitoring and controlling the spread of antibiotic resistance. Yet, current major available antibody-based antibiotic detection methods often face limitations in sensitivity, complexity, and cost, and commonly one target antibiotic can be detected in one assay. Results On the basis of a three-way DNA junction (3-WJ) signal construct, we describe a multiplexed fluorescence resonance energy transfer (FRET) aptasensor strategy for highly sensitive simultaneous detection of sarafloxacin (SAR) and enrofloxacin (ENR) through cyclic DNAzyme and catalytic strand displacement reaction (CSDR) signal amplification cascades. Target antibiotics are recognized separately by the aptamers in DNAzyme/apamer duplexes to release active DNAzyme sequences, which cleave the dumbbell substrate hairpins to free ssDNAs to trigger subsequent CSDR between the assistance hairpins and the 3-WJ constructs for formation of many fluorophores 5-carboxyfluorescein (FAM)- and 2',7'-dimethoxy-4', 5'-dichloro-6-carboxyfluorescein (JOE)/6-carboxy-X-rhodamine (ROX)-labeled DNA duplexes. This leads to the pulling of FAM dye donor in proximity to the ROX and JOE dye acceptors, facilitating the yield of considerably amplified FRET signals at 555 nm and 605 nm for the SAR and ENR assays, respectively, with detection limits of 1.95 pM (0.76 ng/L) and 5.01 pM (1.8 ng/L) within 2.5 hours. Additionally, this sensing method can selectively discriminate SAR and ENR against non-target antibiotics and has been validated for the simultaneous detection of SAR and ENR in milk samples. Significance Featured with the advantages of convenient and significant signal amplification capability as well as single excitation for multiplexed detection, the successful demonstration of our method for sensitive and simultaneous detection of two antibiotics therefore shows its promising potential for constructing different multiplexed aptasensors for detecting various low levels of biomolecules.

Molecularly imprinted fluorescent probe based on FRET for selective and sensitive detection of doxorubicin

Zhifeng Xu,Peihong Deng,Junhua Li,Li Xu,Siping Tang

In this work, a new type of fluorescent probe for detection of doxorubicin has been constructed by the combined use of fluorescence resonance energy transfer (FRET) technology and molecular imprinting technique (MIT). Using doxorubicin as the template, the molecularly imprinted polymer thin layer was fabricated on the surfaces of carbon dot (CD) modified silica by sol-gel polymerization. The excitation energy of the fluorescent donor (CDs) could be transferred to the fluorescent acceptor (doxorubicin). The FRET based fluorescent probe demonstrated high sensitivity and selectivity for doxorubicin. The detection limit was 13.8   nM. The fluorescent probe was successfully applied for detecting doxorubicin in doxorubicin-spiked plasmas with a recovery of 96.8–103.8%, a relative standard deviation (RSD) of 1.3–2.8%. The strategy for construction of FRET-based molecularly imprinted materials developed in this work is very promising for analytical applications.

An innovative near-infrared fluorescent probe with FRET effect for the continuous detection of Zn2+ and PPi with high sensitivity and selectivity, and its application in bioimaging

Yixuan Yao,Fudong Zeng,Jinli Ma,Liangqiang Wu,Shuo Xing,Haiqin Yang,Yapeng Li,Jianshi Du,Qingbiao Yang,Yaoxian Li

As the second most abundant transition metal element in the human body, zinc ions play an important role in the normal growth and development of the human body. We have successfully synthesized a near-infrared fluorescent probe with FRET effect for the detection of Zn 2+ . Probe DR6G has good selectivity and anti-interference ability for Zn 2+ . When Zn 2+ is added to the probe DR6G solution, it responds completely within seconds, releasing red fluorescence with a detection limit of 2.02 × 10 −8 M. As the main product of ATP hydrolysis, PPi is indispensable in various metabolic activities in cells and the human body. Due to the strong binding ability of Zn 2+ and PPi, it is easy to form ZnPPi precipitation, so we added PPi to the solution to complete the Zn 2+ detection, and realized the continuous detection of PPi, and the detection limit was 2.06 × 10 −8 M. Since Zn 2+ and PPi play an important role in vivo, it is of great practical significance to design and synthesize a fluorescent probe that can continuously detect Zn 2+ and PPi. Biological experiments have shown that the probe DR6G has low cytotoxicity and can complete the detection of exogenous Zn 2+ and PPi in cells and living mice in vitro. Bacterial experiments have shown that the DR6G probe also has certain research value in the field of environmental monitoring and microbiology. Due to the constant variation of the fluorescence signals of Zn 2+ and PPi during detection, we designed the logic gate program. In practical applications, the probe DR6G can quantitatively detect Zn 2+ in zinc-containing oral liquids and qualitatively detect PPi in toothpaste.

A cascade FRET photosensitizer that enhances photodynamic therapy for ocular melanoma

Jing Ruan,Fang Li,Hao Tian,Jie Yu,Hongpei Deng,Shengfang Ge,Kam W. Leong

Photodynamic therapy (PDT) relies on the generation of cytotoxic singlet oxygen ( 1 O 2 ) from an otherwise nontoxic photosensitizer accumulated in tumor tissue to treat cancer. However, PDT of uveal melanoma, the most common adult intraocular cancer, using potent porphyrin derivatives as photosensitizers has shown limited therapeutic efficacy due to low bioavailability and 1 O 2 yield. Here, considering the uveal melanoma microenvironment of endogenous melanin , which has a high absorption between 300 nm and 500 nm, we design a carbon dot-based photosensitizer that uses multiple different pathways to generate 1 O 2 . We conjugated gadolinium (Gd)-doped carbon dots (Gd@Cdots) with the porphyrin-derivatives chlorin e6 (Ce6) and verteporfin (Ver). The resulting nanoparticle (Gd@Cdots/Ce6/Ver) generates 1 O 2 under UV irradiation mainly in cascade FRET from Gd@Cdots to Ce6 and then to Ver; the photoactivated Ver, in turn, transfers its energy to 3 O 2 to generate 1 O 2 . In human uveal melanoma MUM-2B cells in vitro, Gd@Cdots/Ce6/Ver generated 7.1- and 2.4-fold higher production of hydroxyl radical (·OH) and superoxide anions (O 2 - ·) following UV irradiation than Gd@Cdots, indicating greater induction of tumor cell apoptosis. In an orthotopic mouse model of uveal melanoma, PDT with Gd@Cdots/Ce6/Ver showed the same trend of superior potency in tumor growth inhibition. This photosensitizer design improves the PDT efficacy of uveal melanoma by enhancing the production of 1 O 2 and should be of broad utility to other PDT.

A sandwich FRET biosensor for lysozyme detection based on peptide-functionalized gold nanoparticles and FAM-labeled aptamer

Meiqing Liu,Hongyuan Zhuang,Yun Zhang,Yanwei Jia

Lysozyme (LYZ) plays a crucial role in the body's immune defense system. Monitoring LYZ levels can provide valuable insights into the diagnosis and severity assessment of various diseases. Traditionally, antibody-based sandwich assays are employed for LYZ detection, but they are often time-consuming and operationally complicated. In this research, a novel sandwich FRET biosensor was developed, which enables rapid detection of LYZ based on peptide-functionalized gold nanoparticles (pAuNPs) and FAM-labeled aptamer (Apt-FAM). Initially, a mixture of Apt-FAM and pAuNPs resulted in partial quenching of the Apt-FAM fluorescence emission through an inner filter effect (IFE), with negligible energy transfer because of the electrostatic repulsion between the negatively charged pAuNPs and Apt-FAM. The introduction of LYZ into the mixture drove the specific binding of Apt-FAM and pAuNPs to LYZ, facilitating the formation of a pAuNPs-LYZ-aptamer sandwich structure. The formation of this complex drew the pAuNPs and Apt-FAM into close enough proximity to enable FRET to occur, which in turn effectively quenched the fluorescence emission of FAM. The decrease in FAM fluorescence intensity was correlated with the increasing concentration of LYZ. Thus, a sandwich FRET biosensor was successfully developed for LYZ detection with a linear detection range of 0–1.75 μM and a detection limit of 85 nM. Additionally, the biosensor allowed visual detection of LYZ in a 96-well microplate, with a rapid response time of just 15 s. This study introduces a innovative sandwich FRET biosensor that combines aptamer and peptide recognition elements, offering a fast and antibody-free method for protein detection.

Plasmon-enhanced FRET biosensor based on Tm3+/Er3+ co-doped core-shell upconversion nanoparticles for ultrasensitive virus detection

Xinyue Lao,Yuan Liu,Lihua Li,Menglin Song,Yingjin Ma,Mo Yang,Guanying Chen,Jianhua Hao

Outbreaks of infectious viruses offer a formidable challenge to public healthcare systems and early detection of viruses is essential for preventing virus propagation. In this work, an ultrasensitive plasmon-enhanced fluorescence resonance energy transfer (FRET) biosensor based on core-shell upconversion nanoparticle (csUCNP) and gold nanoparticle (AuNP) for accurate detection of SARS-CoV-2 viral RNA is presented. In this biodetection assay, the Tm 3+ /Er 3+ co-doped csUCNP NaGdF 4 :Yb/Tm@NaYF 4 :Yb/Er acts as an energy donor and AuNP serves as an energy acceptor. The upconversion emission of Tm 3+ and the design of the core-shell structure led to a simultaneous surface plasmon effect of AuNP. The localized surface plasmon resonance (LSPR) arising from collective oscillations of free electrons significantly enhanced FRET efficiency between Er 3+ and AuNP. The as-prepared biosensor obtained a limit of detection (LOD) as low as 750 a m , indicating that the integration of FRET and surface plasmon into one biodetection assay significantly boosted the sensitivity of the biosensor. In addition, samples extracted from clinical samples are also utilized to validate the effectiveness of the biosensor. Therefore, this innovative plasmon-enhanced FRET biosensor based on Tm 3+ /Er 3+ co-doped csUCNP may pave the way for rapid and accurate biodetection applications.
