Homeostasis and energy and substance metabolism reprogramming shape various tumor microenvironment to sustain cancer stemness, self-plasticity and treatment resistance. Aiming at them, a lipid-based pharmaceutical loaded with CaO 2 and glucose oxidase (GOx) (LipoCaO 2 /GOx, LCG) has been obtained to disrupt calcium homeostasis and interfere with glycometabolism. The loaded GOx can decompose glucose into H 2 O 2 and gluconic acid, thus competing with anaerobic glycolysis to hamper lactic acid (LA) secretion. The obtained gluconic acid further deprives CaO 2 to produce H 2 O 2 and release Ca 2+ , disrupting Ca 2+ homeostasis, which synergizes with GOx-mediated glycometabolism interference to deplete glutathione (GSH) and yield reactive oxygen species (ROS). Systematical experiments reveal that these sequential multifaceted events unlocked by Ca 2+ homeostasis disruption and glycometabolism interference, ROS production and LA inhibition, successfully enhance cancer immunogenic deaths of breast cancer cells, hamper regulatory T cells (Tregs) infiltration and promote CD8 + T recruitment, which receives a considerably-inhibited outcome against breast cancer progression. Collectively, this calcium homeostasis disruption glycometabolism interference strategy effectively combines ion interference therapy with starvation therapy to eventually evoke an effective anti-tumor immune environment, which represents in the field of biomedical research.

GOx@ZIF-90@CuCo-MOF composite was synthesized via synergistic self-assembly. • Bimetallic sites and ZIF-90 shell significantly enhanced nanozyme activity. • The GOx@ZIF-90@CuCo-MOF paper-based nanozyme sensor was fabricated for rapid and noninvasive glucose detection in saliva. • The sensor achieved a 13.5 μM detection limit within a 20-min reaction. Glucose biosensors are pivotal to diabetes management; however, traditional fingerstick blood collection methods are often challenging due to their invasiveness, pain, high cost, and inability to provide continuous monitoring. Here, we constructed a paper-based colorimetric sensing platform using a multifunctional metal–organic framework (MOF) composite system to enable rapid, noninvasive detection of glucose in saliva. This platform utilized a CuCo-MOF framework, loaded with glucose oxidase (GOx) and ZIF-90 via a synergistic self-assembly mechanism, to catalyze the colorimetric reaction of H 2 O 2 and tetramethylbenzidine (TMB). The synergistic effect of the composite significantly enhanced the overall catalytic efficiency, enabling a sensitive response to glucose levels in saliva. Using ImageJ, color images captured with a smartphone were analyzed using RGB color channels to quantitatively determine glucose levels in saliva. The detection time was 20 min, with a detection limit (LOD) of 13.5 μM and a linear range of 50–2000 μM. This platform offered advantages with the absence of blood sample collection, ease of operation, and visual response. It provided new material strategies and design ideas for developing portable, non-invasive blood glucose monitoring technology. In the frontier of drug delivery research, the amalgamation of nanotechnology with 3D printing presents a transformative avenue for patient-centric therapies. This study presents the development of insulin/GOx-loaded Zeolitic Imidazolate Framework-8 (ZIF-8) microneedles, utilizing the unique properties of Metal-Organic Frameworks (MOFs) and advanced 3D printing techniques to enhance transdermal drug delivery systems. A key innovation lies in the use of a Polymerizable Deep Eutectic Solvent (PDES)-based ink for digital light processing (DLP) 3D printing, enabling accelerated photopolymerization and precise fabrication of microneedles. The PDES formulation, consisting of Betaine, Methacrylic Acid (MAA), 2-Hydroxyethyl Methacrylate (HEMA), Polyethylene Glycol Diacrylate (PEGDA), and the photoinitiator trimethylolpropane triacrylate (T-POL), was specifically designed to exploit hydrogen bonding interactions, thereby enhancing polymerization efficiency and structural integrity. The insulin/GOx-loaded ZIF-8 microneedles demonstrated responsive and controlled drug release, with notable sensitivity to glucose and pH changes, making them particularly suitable for diabetic applications. In vitro and in vivo results confirmed the superior drug encapsulation, stability, and release kinetics of the microneedles, showcasing their potential for effective and sustained transdermal delivery of insulin. This work establishes a new paradigm in the synthesis of multifunctional microneedles through the integration of MOFs and PDES for DLP 3D printing. The findings highlight the capability of this approach to address current challenges in drug delivery, offering a scalable, biocompatible, and responsive platform for advanced therapeutic applications.

Salmonella typhimurium ( S. typhimurium ) contamination has become the main source of foodborne diseases and has substantial effects on human well-being. Herein, we present a nucleic acid-responsive colorimetric biosensor for ultrasensitive detection of S. typhimurium , integrating a multifunctional Fe 3 O 4 @Zr-MOF/GOx nanoprobe with exponential amplification reaction (EXPAR). The system integrates glucose oxidase (GOx) encapsulated within a Zr-based metal-organic framework (Zr-MOF) modified with magnetic Fe 3 O 4 nanoparticles. In the presence of S. typhimurium , the EXPAR reaction is triggered, and the controlled release of GOx is achieved by DNA strand displacement. The detection mechanism leverages GOx-catalyzed glucose oxidation to generate H 2 O 2 , which oxidizes 3,3’,5,5’-tetramethylbenzidine (TMB) for colorimetric signal readout. The biosensor demonstrates high specificity for S. typhimurium , with a detection limit of 2.26 CFU mL⁻¹ , and practical applicability in spiked milk samples, achieving recoveries of 89.6–111.5 %. The strategy employing stimuli-responsive DNA/MOF composites demonstrated significant potential in food quality monitoring and analysis.

The intratumoral microbiota plays a crucial role in tumor progression, yet they are often disregarded when formulating cancer treatment strategies. The present study introduces a novel therapeutic approach targeting glucose utilization, and develops an acid-stable adhesive hydrogel co-loaded with glucose oxidase (GOx) and allicin (AG@G). This enables sustained and simultaneous intervention in tumor and bacterial proliferation within tumor. Allicin, a traditional Chinese medicine (TCM) extracted from garlic, exhibits dual efficacy by inhibiting both tumor growth and bacterial proliferation, while GOx catalyzes glucose consumption, depleting cellular energy reserves and initiating NADPH-mediated disulfidptosis. The synergistic combination of allicin and GOx amplifies intracellular disulfide stress and oxidative stress, effectively triggering disulfidptosis and ferroptosis. In vivo studies confirm the potent antitumor activity of AG@G, effectively suppressing the growth of tumor and associated microbiota in breast cancer, even in the drug-resistant lung cancer. Notably, AG@G treatment prevents lung metastasis of breast cancer and the homing of subcutaneous osimertinib-resistant lung cancer. This work suggests promising avenues for integrating traditional Chinese medicine in oncological treatment, and underscores the pivotal role of antibacterial strategies in cancer therapy.

Colorectal cancer (CRC), a leading cause of cancer-related mortality, remains challenging due to drug resistance, insufficient targeting, and systemic toxicity. We engineered a pH/glucose dual-responsive zeolitic imidazolate framework-8 (ZIF-8) nanocarrier for the co-delivery of berberine (BBR) and glucose oxidase (GO X ). BBR and GO X were encapsulated into ZIF-8 nanoparticles via one-pot synthesis and subsequent surface modification with polyethylene glycol–folic acid (PEG–FA), yielding BBR@GO X @ZIF-8–FA (BGZF). At pH 5.4, BBR and GO X release was approximately 74% and 69%, respectively. Cellular uptake and JC-1 assays revealed that the delocalized positive charge of the nanoparticles enhanced mitochondrial targeting, synergizing with GO X -mediated glucose depletion for amplified starvation therapy. Reactive oxygen species (ROS) quantification showed that BBR and GO X synergistically exacerbated oxidative stress, resulting in higher levels of ROS than those observed with monotherapy. Mechanistically, BGZF induced apoptosis through dual pathways: endogenous mitochondrial dysfunction and exogenous ROS overproduction. BGZF’s tumor-specific targeting capabilities were validated in vivo , demonstrating FA-mediated active homing and prolonged biosafety with negligible systemic toxicity. This research pioneers a dual-responsive nanoplatform that integrates mitochondrial targeting and cascade catalytic amplification to achieve synergistic antitumor effects between BBR and GO X , offering a novel paradigm for CRC treatment.

Metal-organic frameworks (MOFs) have attracted extensively interests for enzyme immobilization. However, their rigid structures remain a great challenge in improvement the catalytic performance of the carriers due to no nanoconfinement. Herein, carriers based on rigid UiO-66-NH 2 (U) modified with soft smart polymer were designed by surface engineering. A thermo-responsive polymer, poly(N-isopropylacrylamide) (PNP), was synthesized through an in-situ polymerization strategy, which was employed as a “soft tent” on the surface of rigid U. Glucose oxidase (GOx)-horseradish peroxidase (HRP) was chemically bonded on PNP-U using glutaraldehyde as a linker. Benefiting from the nanoconfinement via variation temperatures, PNP-U@GOx-HRP carriers exhibited 2.01-fold catalytic activity ( K cat value) at 37 °C higher than that at 25 °C. Furthermore, a colorimetric method for analysis of glucose in grape samples was developed. This study offers a strategy for construction of “soft tent” smart polymer-MOFs@enzyme carriers, which provides a pivotal nanoconfinement for boosting their catalytic performance, and broadens their applications in bio-molecules sensing.

Diabetic wound healing is a great clinical challenge due to the microenvironment of hyperglycemia and high pH value, bacterial infection and persistent inflammation. Here, we develop a cascade nanoreactor hydrogel (Arg@Zn-MOF-GOx Gel, AZG-Gel) with arginine (Arg) loaded Zinc metal organic framework (Zn-MOF) and glucose oxidase (GOx) based on chondroitin sulfate (CS) and Pluronic (F127) to accelerate diabetic infected wound healing. GOx in AZG-Gel was triggered by hyperglycemic environment to reduce local glucose and pH, and simultaneously produced hydrogen peroxide (H 2 O 2 ) to enable Arg to release nitric oxide (NO) for inflammation regulation, providing a suitable microenvironment for wound healing. Zinc ions (Zn 2+ ) released from acid-responsive Zn-MOF significantly inhibited the proliferation and biofilm formation of S.aureus and E.coli . AZG-Gel significantly accelerated diabetic infected wound healing by down-regulating pro-inflammatory tumor necrosis factor (TNF)-α and interleukin (IL)-6, up-regulating anti-inflammatory factor IL-4, promoting angiogenesis and collagen deposition in vivo . Collectively, our nanoreactor cascade strategy combining “endogenous improvement (reducing glucose and pH)” with “exogenous resistance (anti-bacterial and anti-inflammatory)” provides a new idea for promoting diabetic infected wound healing by addressing both symptoms and root causes. Statement of Significance A cascade nanoreactor (AZG-Gel) is constructed to solve three key problems in diabetic wound healing, namely, hyperglycemia and high pH microenvironment, bacterial infection and persistent inflammation. Local glucose and pH levels are reduced by GOx to provide a suitable microenvironment for wound healing. The release of Zn 2+ significantly inhibits bacterial proliferation and biofilm formation, and NO reduces wound inflammation and promotes angiogenesis. The pH change when AZG-Gel is applied to wounds is expected to enable the visualization of wound healing to guide the treatment of diabetic wound. Our strategy of “endogenous improvement (reducing glucose and pH)” combined with “exogenous resistance (anti-bacterial and anti-inflammatory)” provides a new way for promoting diabetic wound healing.

A sensitive fluorescent microfluidic sensor based on carbon quantum dots (CQDs), cadmium telluride quantum dots (CdTe QDs) aerogel and glucose oxidase (GOx) for urinal glucose detection was fabricated via a simple method. CQDs, emitting green fluorescence (emission peak at 520 nm) and the CdTe QDs, emitting red fluorescence (emission peak at 620 nm) under 365 nm ultraviolet irradiation, were synthesized. The as-prepared CQDs, CdTe QDs and GOx (C/CdTe QD-GOx) were mixed in a certain ratio and were then introduced into the microfluidic chip to construct a three-dimensional porous aerogel sensor after freeze-drying. A ratiometric fluorescence response was realized through the quenching of the red fluorescence by H2O2 produced from the glucose/GOx reaction, while the green fluorescence intensity stays constant. A small change in the ratio of the two fluorescence intensities led to a clear change in the fluorescent colour of the sensor, which can be easily observed under a UV lamp. The microsensor displayed a serial colour change from red to green with the changes in glucose concentration. The intensity ratio between the red fluorescence and green fluorescence (R/G value) indicates the glucose concentration. In our experiments, the optimal concentration ratio of CdTe QDs and CQDs was 0.5 : 1, and the optimal response time was 5 min. The detection range of glucose concentration was from 0 mM to 13 mM, and the detection limit was 0.223 mM. We also found that this glucose sensor has excellent selectivity, specificity and stability. The as-prepared microsensor can be stably stored for 30 days under −20 °C. Such a C/CdTe QD-GOx aerogel based microfluidic assay sensor provides a new approach towards the optical and quantitative detection of glucose, which enables daily urine glucose monitoring for the diabetic patients in a convenient way.

Glucose (Glu) is an important source of energy and metabolic intermediate for animal and plant cells. In this study, a high-performance biosensor for specific detection of Glu was constructed by combining a light-addressable potentiometric biosensor (LAPS) and a specific sensitive unit. The specific sensitive unit was adopted a silicon-based capacitive structure, and the glucose oxidase/reduced graphene oxide -chitosan-ferrooxidase/gold nanoparticles (GOX/RGO-CS-Fc/AuNPs) was modified onto the silicon-based working electrode to construct an nanosensing membrane. The interaction between the specific sensitive unit and Glu resulted in the formation of a bilayer potential, and the field effect generated by the bilayer potential caused a change in the capacitance of the silicon structure, which resulted in a change in the photocurrent-voltage characteristics of the LAPS. Through the potential shift in the photocurrent-voltage characteristics, the LAPS was capable of detection the concentration of Glu. Under optimal conditions, the potential shift was linearly correlated with the Glu concentration from 0.01 mg/mL to 4.00 mg/mL with R 2 of 0.9821, and the limited detection was 0.001 mg/mL. In addition, the LAPS showed good recovery (97.92 %–105.72 %), admirable specificity and stability in detection Glu in human serum samples.

Starvation therapy has shown promise as a cancer treatment, but its efficacy is often limited when used alone. In this work, a multifunctional nanoscale cascade enzyme system, named CaCO 3 @MnO 2 -NH 2 @GOx@PVP (CMGP), was fabricated for enhanced starvation/chemodynamic combination cancer therapy. CMGP is composed of CaCO 3 nanoparticles wrapped in a MnO 2 shell, with glucose oxidase (GOx) adsorbed and modified with polyvinylpyrrolidone (PVP). MnO 2 decomposes H 2 O 2 in cancer cells into O 2 , which enhances the efficiency of GOx-mediated starvation therapy. CaCO 3 can be decomposed in the acidic cancer cell environment, causing Ca 2+ overload in cancer cells and inhibiting mitochondrial metabolism. This synergizes with GOx to achieve more efficient starvation therapy. Additionally, the H 2 O 2 and gluconic acid produced during glucose consumption by GOx are utilized by MnO 2 with catalase-like activity to enhance O 2 production and Mn 2+ release. This process accelerates glucose consumption, reactive oxygen species (ROS) generation, and CaCO 3 decomposition, promoting the Ca 2+ release. CMGP can alleviate tumor hypoxia by cycling the enzymatic cascade reaction, which increases enzyme activity and combines with Ca 2+ overload to achieve enhanced combined starvation/chemodynamic therapy. In vitro and in vivo studies demonstrate that CMGP has effective anticancer abilities and good biosafety. It represents a new strategy with great potential for combined cancer therapy.
