Decellularized extracellular matrix (dECM) has been widely used in tissue engineering and regenerative medicine due to its excellent biocompatibility and biological activities. In this work, the dECM was obtained by treating porcine Achilles tendon tissue via a combination of chemical, physical, and biological approaches. The obtained dECM was interpenetrated with genipin crosslinked quaternary ammonium chitosan (QCS) network to fabricate the composite genipin/QCS/dECM (QGD) hydrogels. The prepared QGD hydrogels exhibited an interconnected microstructure and a suitable water-absorption capacity. Furthermore, the QGD hydrogels exhibited strong adhesion to the surfaces of different matrices and various organs, including lung, liver, trachea, heart, and spleen. The remodelling of dECM gave the QGD hydrogels adjustable self-healing ability and mechanical properties. In addition, the QGD bio-adhesives exhibit effective antibacterial activity against the typical Gram-positive S. aureus and Gram-negative E. coli . 2,2′-Azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) radical scavenging and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging tests confirmed the antioxidant abilities of QGD hydrogels, and the cellular reactive oxygen species (ROS) scavenging potential was also investigated using RAW264.7 cells. Furthermore, the QGD hydrogels possessed excellent hemocompatibility, cytocompatibility, and cell pro-migration abilities. The results confirmed that the proposed QGD hydrogels could serve as potential bioadhesives for biomedical applications.

Microsphere systems have been used to deliver drugs, proteins, and cells. However, dispersed microspheres can induce harmful effects after they are introduced into the body. To agglomerate these dispersed microspheres, an in situ forming system was developed to fabricate microsphere/hydrogel composites. Polycaprolactone microspheres with a porous surface and hollow core were physically incorporated into a genipin-crosslinked gelatin hydrogel. The incorporation of microspheres reduced the swelling capacities and weakened the unexpected volume expansion, which is not favorable for in vivo implants. Additionally, the reacted genipin ratio increased with increasing gelatin concentration. The degradation of the composite was also determined, and it was proposed that the degradation mechanism of the composite was bulk collapse, whereas that for the pure hydrogel was surface erosion. The obtained microsphere/hydrogel composite might have a great potential application as an injectable system for tissue regeneration. © 2012 Wiley Periodicals, Inc. J. Appl. Polym. Sci., 2013

Nanocarriers play an important role in improving the photo- and thermal-stability of photosensitizers to gain better pharmacokinetics behavior in tumor photothermal therapy. Herein, PEGylated chitosan (CG-PEG; PEG: polyethylene glycol) nanoparticles with ultrasmall size (∼5 nm) were prepared through a water-in-oil reverse microemulsion method using genipin as a cross-linker. Particle size and zeta-potential can be tuned by varying the molar ratio between chitosan amino groups and genipin. CG-PEG-ICG (ICG: indocyanine green) nanoparticles were fabricated by adding ICG to CG-PEG aqueous solution through a self-assembly method via electrostatic interaction. The resultant CG-PEG-ICG nanoparticles exhibited improved photo- and thermal-stability, good biocompatibility, and low toxicity. When irradiated with a laser, the cells incubated with CG-PEG-ICG nanoparticles showed very low cell viability (15%), indicating the CG-PEG-ICG nanoparticles possess high in vitro photothermal toxicity. Moreover, the CG-PEG nanocarriers can significantly alter the biodistribution and prolong the retention time of ICG in the mice body after intravenous injection. In vivo photothermal study of tumors injected with CG-PEG-ICG nanoparticles containing ICG at a concentration greater than 100 μg·mL–1 (100 μL) induced irreversible tissue damage. The growth of U87 tumors was dramatically inhibited by CG-PEG-ICG nanoparticles, demonstrating that the CG-PEG nanoparticles may act as potential ICG nanocarriers for effective in vivo tumor photothermal therapy.

Enzyme immobilization is an effective strategy to improve catalyst recovery and operational stability, but conventional approaches are often limited by diffusion resistance, steric hindrance, and insufficient control over the enzyme microenvironment. Hydrogel-based materials are attractive supports because their hydrated, tunable, and porous networks help preserve enzyme conformation while improving substrate diffusion and mass transfer. Here, a gas-shearing microfluidic strategy was developed to fabricate alginate/chitosan hydrogel microreactors for dual-mode lipase immobilization through entrapment and genipin-mediated covalent binding. By tuning gas-liquid shear, microspheres with controllable and narrow size distribution were generated without surfactants or UV-initiated polymerization. Importantly, genipin was introduced as a mild and biocompatible cross-linker to reinforce the alginate/chitosan network and create a stable interfacial microenvironment for enzyme immobilization. Compared with conventional cross-linkers, the milder and more controllable reactivity of genipin is advantageous in preserving enzyme conformation and catalytic performance. The resulting microreactors exhibited enhanced catalytic activity, improved storage stability, and good reusability. After five consecutive cycles, the entrapped and covalently immobilized systems retained 70% and 81% of their initial activity, respectively. These results demonstrate that gas-shearing microfluidics combined with genipin-mediated network reinforcement provides a robust and biocompatible platform for constructing hydrogel-based enzymatic microreactors with improved catalytic performance and operational durability.

Export Rat nerve growth factor and total flavonoids from hawthorn leaf contribute to the recovery of neurological function after spinal cord injury, including traumatic, non-traumatic spinal cord injuries. However, it remains challenging to efficiently deliver nerve growth factor and total flavonoids from hawthorn leaf to spinal cord injury sites, ensure their sustained release, and minimize further damage. In the present study, we chose a biocompatible and biodegradable gelatin as the substrate, which was crosslinked with the natural biological crosslinker genipin to form a gelatin–genipin hydrogel carrier for the slow release of nerve growth factor and total flavonoids from hawthorn leaf in spinal cord injury sites. The prepared gelatin–genipin hydrogel had good injectable properties and photothermal effects. Furthermore, when the hydrogel with 2% genipin, 200 ng/mL nerve growth factor, and 320 μg/mL total flavonoids from hawthorn leaf was combined with near infrared irradiation, there was a slow release of total flavonoids from hawthorn leaf and nerve growth factor, reduced oxidative stress, an improved inflammatory microenvironment, and accelerated angiogenesis and axonal regeneration via inhibition of the nuclear factor kappa-B signaling pathway, thereby promoting recovery from spinal cord injury. Collectively, our results indicate that this new hydrogel may improve the prognosis of spinal cord injury, and may represent a new strategy for treating spinal cord injury.

As awareness of health concerns continues to rise, the development of new generations of sustainable food packaging materials has emerged as a focal point of significant attention. This study presents the development of a fully bio-based grease resistant composite coating composed of chitosan (CS), genipin (GP), and microfibrillated cellulose (MFC) for sustainable paper food packaging. The synergistic effects of crosslinking between CS and GP, combined with electrostatic interactions between CS and MFC, enabled the formation of a dense, interconnected coating layer on paper substrates. Systematic optimization of coating formulations revealed that a 2 wt% CS, 0.1 wt% GP, and 15 wt% MFC composition achieved maximum grease resistance (kit rating: 12/12) at a remarkably low coating weight of 4 g/m 2 , outperforming existing chitosan-based coatings requiring higher application weights for medium performance. The composite coating significantly enhanced mechanical properties, with two kinds of food wrapping paper after coating exhibiting tensile strength improvements of 25 % and 13 %, and burst strength increases of 25 % and 41 %, respectively. Additionally, water vapor transmission rates decreased by 22 % and 28 %, demonstrating enhanced moisture barrier functionality. Practical evaluations have confirmed the coating's robust resistance to various organic solvents, as well as its stability in hot oil. Simulated food contact tests using fried chicken and sausages revealed no detectable grease penetration or structural compromise in coated papers. This study establishes the CS/GP/MFC coating as a high-performance, environmentally sustainable alternative to fluorinated and non-biodegradable materials, with broad applicability in grease resistant food packaging systems.

The development of genipin modification methods has attracted much attention to improve the functional properties of proteins in the fields of food science . In this work, two types of pH-responsive genipin-crosslinked bovine bone gelatin conjugates with high surface hydrophobicity were developed for emulsion stabilization. One was genipin molecule-crosslinked gelatin conjugates prepared at acidic pH and the other was genipin macromer-crosslinked gelatin conjugates prepared at neutral and basic pH. The degrees of crosslinking increased with the increase of preparation pH. They increased with the increase of genipin-to-gelatin mass ratios at acidic pH, whereas decreased with the increase of genipin-to-gelatin mass ratios at neutral and basic pH. The surface hydrophobicity values, molecular weights, and emulsifying properties (creaming stability, emulsion activity index, and emulsion stability index) of the obtained conjugates increased with the increase of both preparation pH and genipin-to-gelatin mass ratios. This work provided useful knowledge to understand how the preparation parameters (preparation pH and genipin-to-gelatin mass ratio) and genipin structures (molecule and macromer) affect a protein's degrees of crosslinking, physicochemical properties, and emulsifying properties.

Design and fabrication of multifunctional nano-carrier is a promising tool in the field of nanomedicine for glioma theranostics. In this study, blue color nanoparticles with targeted tumor cell staining and drug delivery capabilities were simply fabricated by crosslinking lysine (Lys) and lysine-folic acid (Lys-FA) mixture with genipin as the crosslinking agent through a reversed microemulsion method. Doxorubicin (DOX) as the model drug was loaded through electrostatic interaction. The nanoparticles before (Lys-FA NPs) and after (Lys-FA/DOX NPs) loading of DOX were characterized by ultraviolet–visible (UV–Vis) spectroscopy, Fourier transform infrared spectroscopy (FTIR), dynamic laser light scattering (DLS), transmission electron microscopy (TEM), and fluorescence spectroscopy. Ultimately, the glioma cell staining ability of Lys-FA NPs and cytotoxicity of Lys-FA/DOX NPs were tested in vitro . The results showed that the particle size of Lys-FA NPs can be tuned by adjusting the ratio of Lys to Lys-FA simply. The Lys-FA NPs exhibited dark blue and the blue color is stable under different pH values and temperatures. The feature makes the staining glioma cell pellets visible to naked-eyes. The model drug DOX was loaded on Lys-FA NPs indicated by the increase of size and zeta potential , and change of optical properties. The DOX loaded on the Lys-FA NPs can be sustainedly released, other than burst release. Encouragingly, the Lys-FA NPs can deliver DOX into cells and dramatically reduce cell viability, as proved by U87 cell cytotoxicity analysis and confocal fluorescence imaging. In addition, the Lys-FA NPs (≤2.5 mg mL −1 ) are nontoxic to CHO cells. In a short, the blue color Lys-FA NPs can visibly stain glioma cell pellets and deliver DOX into glioma cells, which can potentially be applied as a multifunctional nanocarrier for tumor imaging and drug delivery.

The driving performance of current biomimetic artificial muscles has not yet met the requirements for engineering applications, limiting their practical prospects. This paper presents a test scheme for the driving characteristics of biomimetic artificial muscles, with chitosan and graphene as the main materials. Key evaluation indicators are proposed, including output force, output displacement, and response speed. The study focuses on analyzing the effects of multi-walled carbon nanotubes (MCNT) and Genipin on the driving characteristics of the artificial muscles. MCNT crosslinked with graphene enhances the conductivity of the electrode layer, while Genipin crosslinked with chitosan improves the structural stability and flexibility of the driving layer. The combined addition of MCNT and Genipin significantly enhances the driving performance of the artificial muscles, especially in terms of output displacement, output force, and response speed. The experiment summarizes the optimal additive ratio, providing important guidance and experimental support for further optimization of the design and performance of biomimetic artificial muscles.

High-speed shearing induced the unfolding of soybean lipophilic protein (SLP). • Primary amino groups of SLP served as reactive sites for genipin crosslinking. • Genipin facilitated intermolecular crosslinking of SLP after shearing. • Shearing and genipin treatment enhanced SLP adsorption at the oil-water interface. • Modified SLP improved emulsion stability and enhanced its viscoelasticity. Native soybean lipophilic protein (SLP) readily forms compact aggregates through strong hydrophobic interactions, impairing its water solubility and interfacial adsorption capacity. This study investigated the effects of high-speed shearing followed by genipin crosslinking on the structural and functional properties of SLP. Spectroscopic analyses revealed enhanced exposure of tryptophan and tyrosine residues, indicating that shearing induced conformational unfolding of SLP. Subsequent genipin crosslinking resulted in a clear blue shift of the amide II band, confirming the formation of covalent crosslinks. The treatment consisting of shearing at 12,000 rpm for 4 min and 0.004% genipin yielded the highest SLP solubility of 76.29%. Under these conditions, the emulsifying activity index and emulsion stability index of modified SLP increased by 14.33 m 2 /g and 135.88 min compared with native SLP. In summary, this study provides an important reference for addressing the stability issues of plant protein emulsions in the food industry. Multifunctional hydrogel adhesives have emerged as promising candidates for advanced biomedical applications, particularly in surgical suture alternatives, hemostatic management, and regenerative wound care. This study developed an interpenetrating polymer network (IPN) hydrogel adhesive system through the synergistic integration of dopamine-functionalized hyaluronic acid (HA-DA) and genipin-crosslinked quaternary ammonium chitosan (QCS). The successful preparation of HA-DA and QCS was confirmed via 1 H nuclear magnetic resonance ( 1 H NMR) and Fourier transform infrared (FT-IR) analysis. The fabricated hydrogel adhesives exhibited the interconnected microstructure, suitable mechanical strength, and elastic solid properties. Additionally, the hydrogel adhesives exhibited expected self-healing abilities and injectability. The hydrogels displayed strong adhesion on different matrix and tissues. The quaternary ammonium group pendants in the hydrogel network result in the excellent antibacterial activity of hydrogels against Escherichia coli and Staphylococcus aureus . Furthermore, hemolysis test, fluorescence images, CCK-8 assay, and wound scratch assay demonstrated that the hydrogel adhesives possessed good hemocompatibility, biocompatibility, and cell migration ability. These multifunctional characteristics, combining structural integrity, rapid self-repair, surgical-grade adhesion, antimicrobial protection, hemocompatibility, and cytocompatibility, establish this IPN hydrogel as a promising candidate for biomedical adhesives.

Soy hull polysaccharide (SHP), an anionic pectin-like polysaccharide derived from soybean processing by-products, exhibits moderate emulsifying capacity but limited interfacial strength due to its reliance on weak physical interactions. To enhance its interfacial performance, this study systematically investigated how genipin (GP) induced covalent crosslinking regulates the adsorption kinetics, conformational evolution, and self-assembly behavior of SHP at the oil-water interface. Dynamic interfacial tension and dilatational rheology analyses revealed a non-monotonic dependence of interfacial properties on crosslinking density. At a moderate SHP:GP mass ratio of 10:5, the system formed a continuous covalent network dominated by Schiff base linkages, achieving the highest interfacial dilatational modulus (120 mN/m) and the fastest molecular rearrangement rate. Correspondingly, high internal phase emulsions (HIPEs) prepared under this condition exhibited the smallest droplet size (460 nm), the highest absolute ζ-potential (−69.69 mV), and enhanced rheological and thermal stability. In contrast, excessive crosslinking (10:8) induced structural over-constraint, interfacial embrittlement, and aggregate formation, leading to reduced adsorption efficiency and compromised emulsion uniformity. Spectroscopic analyses confirmed progressive covalent bond formation and conformational restriction with increasing GP concentration. At high GP levels, the interfacial network transitioned from a homogeneous structure to a heterogeneous phase-separated system, accompanied by GP self-aggregation and SHP conformational collapse These findings demonstrate that crosslinking density critically governs the balance between network connectivity and chain mobility during interfacial self-assembly. This work provides mechanistic insight into the controlled design of polysaccharide-based HIPEs through precise covalent modification and supports the high-value utilization of soybean by-products in advanced food hydrocolloid applications.
