Effects of HMW-GSs at Glu-B1 locus on starch–protein interaction and starch digestibility during thermomechanical processing of wheat dough

Pai Peng,Xiaolong Wang,Mei Liao,Xiaoyang Zou,Qianying Ma,Xiaoke Zhang,Xinzhong Hu

Background The composition of glutenin protein significantly affects protein–starch interactions and starch digestion characteristics in wheat dough matrices. To elucidate the effects of high molecular weight glutenin subunits at the Glu-B1 locus on dough processing quality, the detailed structural changes of protein, starch, and their complexes were compared in Mixolab dough samples of two near isogenic lines 7 + 8 and 7 + 9. Results The results showed that the degree of protein aggregation increased continuously during dough processing, as did the destruction and rearrangement of the gluten network. Compared to 7 + 8, the stronger and more stable protein network formed in 7 + 9 dough induced intensive interactions between protein and starch, primarily through hydrogen bonds and isomeric glycosidic bonds. In 7 + 9 dough, the more compact and extensive protein–starch network significantly inhibited starch gelatinization during dough pasting, while during the dough cooling stage [from C4 (82.8 °C) to C5 (52.8 °C)], more protein–starch complexes composed of monomeric proteins and short-chain starch were generated, which remarkably inhibited starch retrogradation. All protein–starch interactions in the 7 + 9 dough improved the starch digestion resistance, as reflected by the high content of resistant starch. Conclusion The more extensive and intensive protein–starch interactions in the 7 + 9 dough inhibited the gelatinization and enzymatic hydrolysis of starch, thereby producing more slowly digestible starch and resistant starch. These findings demonstrate the feasibility of optimizing the texture and digestibility of wheat-based food products by regulating the behavior and interactions of proteins and starch during dough processing. © 2022 Society of Chemical Industry.

Neuro-stimulating effect of Citri Reticulata Pericarpium Viride essential oil through regulating Glu/NMDAR on olfactory bulb to improve anxiety-like behavior

Jingbin Chen,Yali Liu,Yonggui Song,Huihui Liang,Genhua Zhu,Bike Zhang,Liangliang Liao,Jian Luo,Ming Yang,Dan Su

Ethnopharmacological relevance Citri Reticulata Pericarpium Viride (also known Qing-Pi or QP) is a plant in the Rutaceae family, QP is a traditional Qi-regulating medicine in Chinese medicine that is compatible with other Chinese medicine components and has extensive clinical practice in treating anxiety and depression. Reports on the pharmacological effects of QP have demonstrated its neuroprotective effects and antioxidant capacities. Numerous pharmacological benefits of QP are attributed to its antioxidant abilities. Anxiety disorders are a broadly defined category of mental illnesses. Oxidative stress and an imbalance in the antioxidant defense system are typical pathological features of these disorders. Aim of the study The aim of this study was to evaluate the effects of QP essential oil on anxiety using animal models and investigate the underlying neurobiological mechanisms. Materials and methods This study aimed to develop an animal model of anxiety using chronic restraint stress and investigate the effects of inhalation of Citri Reticulata Pericarpium Viride essential oil on anxiety-like behavior, olfactory function, and olfactory bulb neurogenesis in mice with anxiety. Results The results showed that long-term chronic restraint stimulation caused a decrease in olfactory function, significant anxiety-like behavior, and a notable reduction in the number of neurons in the olfactory bulb. However, inhalation of Citri Reticulata Pericarpium Viride essential oil reversed these effects, improving the olfactory function, neuro-stimulating effect, alleviating anxiety-like behavior, and regulating theta (4-12Hz) oscillation in the hippocampus DG area. These effects were associated with changes in the expression levels of glutamate receptor NMDAR and NeuN in olfactory bulb. Conclusions The study revealed that mice with anxiety induced by chronic restraint stress exhibited significant olfactory dysfunction, providing strong evidence for the causal relationship between anxiety disorders and olfactory dysfunction. Moreover, QP essential oil has the potential to be developed as a therapeutic drug for anxiety disorders, in addition to its role as a complementary anxiolytic.

Antidepressant effects of cherry leaf decoction on a chronic unpredictable mild stress rat model based on the Glu/GABA-Gln metabolic loop

Jiang Chuan,Wang Hua,Qi Jiaying,Li Jinghan,He Qianqian,Wang Chaonan,Gao Yonggang

Cherry leaves ( Prunus pseudocerasus Lindl. [Rosaceae] ), a traditional Chinese herbal medicine, can regulate the factors closely related to depression including inflammatory cytokines, oxidative stress and blood glucose level. However, the antidepressant effects of cherry leaves and underlying neuromodulatory mechanisms remain relatively have not been elucidated explicitly. The present study investigated the antidepressant effects of cherry leaf decoction (CLD). The underlying neuromodulatory mechanism was explored by examining the glutamate (Glu)/γ-aminobutyric acid (GABA)-glutamine (Gln) metabolic loop. The chronic unpredictable mild stress (CUMS) rodent model was used in this study. The main flavonoids components of CLD were identified using high-performance liquid chromatography (HPLC). The antidepressant effects of CLD were assessed throughout behavioural tests including the bodyweight, sucrose preference test (SPT), forced swimming test (FPT) and tail suspension test (TST). Moreover, The baseline levels of serum adrenocorticotropic hormone (ACTH) and corticosterone (CORT) were quantified. The expression of proteins integrally involved in the Glu/GABA-Gln metabolic loop were observed and quantified by Western blotting, reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. This study found that CLD ameliorated depressive-like behaviours induced by CUMS. The increase of serum ACTH and CORT baseline levels induced by CUMS was also reversed after CLD intervention. Furthermore, CUMS reduced the expression of GAD65, GAD67, GLT-1, GS and GABA A and increased NMDAR1 levels in the rat hippocampus, which was normalized by CLD treatment. The findings demonstrated that CLD could ameliorate the depression-like behaviours induced by CUMS, potentially through the inhibition of hypothalamic–pituitary–adrenal (HPA) axis hyperactivity and the regulation of Glu/GABA-Gln metabolic loop. Enzyme-integrated metal-organic framework platform for cascade detection of α-amylase

Nana Li,Liangqiang Chen,Wanqiu Huang,Mengdi Hao,Huabin Tu,Hao Shen,Fan Yang,Shaoning Yu

As one of the most important industrial enzymes, α-amylase is widely used in food processing, such as starch sugar and fermentation, bringing high added value to industry of more than a trillion dollars. We developed a multi-enzyme system (Glu&Gox@Cu-MOF-74) prepared by embedding α-glucosidase (Glu) and glucose oxidase (Gox) into the biomimetic metal-organic framework Cu-MOF-74 using in situ encapsulation within 15 min at room temperature for efficient and sensitive detection of α-amylase activity. Benefitting from the remarkable peroxidase-mimicking property and rigid skeleton of Cu-MOF-74, the biocatalytic platform exhibited excellent cascade activity and tolerance in various extremely harsh environments compared to natural enzymes. On this basis, a cascade biocatalytic platform was constructed for the detection of α-amylase activity with wide linear range (5–100 U/L) and low limit of detection (1.45 U/L). The colorimetric cascade scheme is important for the sensitive and selective determination of α-amylase in complex fermentation samples, and the detection time is short (∼0.5 h). This work provides new ideas for the detection of α-amylase based on the cascade amplification method.

Platinum Glutamate Acid Complex as a Peroxidase Mimic: High Activity, Controllable Chemical Modification, and Application in Biosensor

Yuanyuan Zhang,Lexian wu,Jing Yang,Guoming Li,Keqin Deng,Haowen Huang

Recent strides in nanotechnology have given rise to nanozymes, nanomaterials designed to emulate enzymatic functions. Despite their promise, challenges such as batch-to-batch variability and limited atomic utilization persist. This study introduces Pt(Glu)2, a platinum glutamic acid complex, as a versatile small-molecule peroxidase mimic. Synthesized through a straightforward method, Pt(Glu)2 exhibits robust catalytic activity and stability. Steady-state kinetics reveal a lower Km value compared to natural enzymes, signifying strong substrate affinity. Pt(Glu)2 was explored for controllable chemical modification and integration into cascade reactions with natural enzymes, surpassing other nanomaterials. Its facile synthesis and seamless integration enhance cascade reactions beyond the capabilities of nanozymes. In biosensing applications, Pt(Glu)2 enabled simultaneous detection of cholesterol and alkaline phosphatase in human serum with high selectivity and sensitivity. These findings illuminate the potential of small molecule mimetics in catalysis and biosensing, paving the way for their broader applications.

Facile fluorescent sensor for rapid and selective detection of phytic acid in food based on phosphatase-like activity of Zr(IV)-Glu complex

Xing Cheng,Yue Tang,Xiaohuan Huang,Linghui Huang,Han Tao,Yuangen Wu

Artificial enzymes have attracted much attention in food analysis because of their simple preparation, low cost, and strong stability. However, the most used artificial enzymes are based on oxidoreductases such as oxidase, peroxidase, catalase, and laccase. There are few reports on hydrolases like phosphatase for food analysis. Herein, a novel green zirconium-glutamic acid (Zr(IV)-Glu) complex was prepared using Zr ions and glutamic acid. The Zr(IV)-Glu complex was first found to possess excellent phosphatase-like activity, which was used to establish a fluorescent sensor for detecting phytic acid (PA). The Zr(IV)-Glu complex can catalyze the hydrolysis of 4-methylumbellione phosphate disodium salt (4-MUP) to produce blue fluorescent products with fluorescence emission peaks at 448 nm. PA with six phosphate groups can attach on the surface of Zr(IV)-Glu to form Zr-O-P bond and inhibit the catalytic activity of Zr(IV)-Glu, leading to the decrease of fluorescence in the system. The change of fluorescent intensity at 448 nm was positively correlated with the PA content, so that a fluorescent sensor using the phosphatase-like activity of Zr(IV)-Glu was proposed to detect PA. The established sensor has high sensitivity, low detection limit (LOD) of PA to 58 μg·L -1 , strong anti-interference ability, and excellent selectivity. This work can provide a new method for PA detection in food and further expand the application of phosphatase mimics.

Glu-Co-Assisted Iron-Based Metal–Organic Framework-Derived FeCo/N Co-Doped Carbon Material as Efficient Bifunctional Oxygen Electrocatalysts for Zn–Air Batteries

Pengfei Sha,Xiao Yong,Di Chen,Xing Chen,Fengying Yan,Beili Pang,Hongzhou Dong,Jianhua Yu,Liyan Yu,Lifeng Dong

A Zn–air battery serves as an energy storage solution to address fossil energy and environmental concerns. However, sluggish kinetics in oxygen reduction reactions (ORRs) and oxygen evolution reactions (OERs) demand innovative, cost-effective, and stable bifunctional catalysts to replace precious metal catalysts. In this study, an FeCo-CNTs/KB catalyst was synthesized by pyrolyzing NH2-MIL-101(Fe) coated with glu-Co and conductive carbon (KB). This hierarchical structure comprises carbon nanotubes (CNTs) grafted onto a carbon matrix, housing abundant FeCo nanoparticles within the nanotubes or matrix. KB introduction enhances FeCo nanoparticle dispersion and fosters uniform CNT formation with smaller diameters, thus exposing active sites. Consequently, the FeCo-CNTs/KB catalyst exhibits remarkable bifunctional electrocatalytic activity: an ORR half-wave potential of 0.84 V and an OER overpotential of 0.45 V (10 mA cm−2). Furthermore, the FeCo-CNTs/KB catalyst in a secondary Zn–air battery showcases enduring charge–discharge performance (≥300 h).

A fluorescence-activatable tumor-reporting probe for precise photodynamic therapy

Jian Li,Tingting Wang,Feng Jiang,Zhangyong Hong,Xinhui Su,Shuang Li,Shoufa Han

Approaches that could enable precise photodynamic therapy (PDT) are of therapeutic potential. We herein report a trifunctional probe (Glu-RdEB) that could be activated to generate fluorescent rhodamine species to pinpoint tumor foci. The probe contains a γ-glutaminyl moiety cleavable to γ-glutamyl transpeptidase (GGT) overexpressed in multiple tumors, an entity of an ENBS photosensitizer for PDT, and an entity of rhodamine fluorescently quenched by ENBS. Upon activation by tumor-associated GGT, the probe releases highly fluorescent rhodamine that is selectively confined in tumors whereby light irradiation leads to effective tumor regression in mice. These results indicate the feasibility of a fluorescently quenched dye–photosensitizer pair to yield tumor-activatable fluorescence to direct PDT.

Coating TiO2 with MIL-101(Fe) to decarboxylate D-Glu and D-MeAsp of microcystin-LR from HA-rich water: Diminishing the inhibitory effect of humic acids

Yarui Wang,Chengzhuo Yu,Sen Wang,Jin Xu,Fanglan Geng,Silong Wang,Jitao Lv,Lixia Zhao,Yawei Wang

Humic acids (HA) released during algal bloom poses a key challenge for efficient photocatalytic degradation of Microcystin-LR (MC-LR) due to their scavenging function on active species. Herein, MIL-101(Fe)/TiO 2 -P25 (M/P) was constructed to diminish the inhibitory effect of HA on the photocatalytic degradation of MC-LR. Results showed that MC-LR could be efficiently removed assigning to the photogenerated holes (h + ) and superoxide radical (O 2 •− ) oxidation under visible light irradiation, where the carboxy groups of D -glutamic acid (Glu) and methyl- D -aspartic acid (MeAsp), and Adda side chain related to MC-LR biotoxicity were selectively destroyed to achieve MC-LR detoxication. More importantly, HA exerted non-inhibitory effect on MC-LR removal by M/P. The counteracting mechanisms were studied. Firstly, the location of TiO 2 in the pore structure of MIL-101(Fe) could occupy the adsorption sites of HA, and decrease the HA adsorption on M/P. Furthermore, XPS analyses suggested the coordination of unsaturated Fe metal sites on the surface of M/P hybrids complexed with –OH and –COOH of HA, which could accelerate the electron transfer and reduce hole consumption. Thus, the inhibitory effect of HA was alleviated during photocatalytic MC-LR treatment. This work offers a possible way to remove MC-LR in an abundant HA matrix water system efficiently.

Modification of a polyethersulfone membrane with a block copolymer brush of poly(2-methacryloyloxyethyl phosphorylcholine-co-glycidyl methacrylate) and a branched polypeptide chain of Arg–Glu–Asp–Val

Xiaoqin Niu,Dan Li,Yuhong Chen,Fen Ran

Polyethersulfone (PES) has good thermal stability, superior pH, chlorine tolerance, and excellent chemical resistance; however, the hydrophilicity and biocompatibility of PES need to be improved for its real applications. In this study, we report a surface modification method for the preparation of a functional PES membrane with hydrophilic polymer chains (MPC and GMA) via surface-initiated electrochemically-mediated atom-transfer radical polymerization (SI-eATRP) technology, and the Arg–Glu–Asp–Val polypeptide groups (REDV) were immobilized onto the modified membrane by a ring-opening reaction. XPS and SEM were used to analyze the chemical composition and morphology of the modified membrane surfaces, confirming that the hydrophilic polymer chains MPC and GMA and the polypeptide group REDV were successfully grafted onto the PES membrane surface. The static water contact angle decreased from 89° to 50–65°, and the hydrophilic property of the modified membrane was enhanced. The water flux increased from 4.29 L m−2 h−1 for the pristine PES membrane to 25 L m−2 h−1 for the modified membrane with PGMA chains grafted on it and REDV functional groups immobilized on it; note that the antifouling tests showed that all the modified membranes had the higher flux recovery ratio values (FRR) of above 80% than the pristine PES membrane (about 60%), and the APTT for the modified membrane increased from 46 s to 93 s, indicating that these modified membranes could be applied in the separation and blood purification fields.

A novel phosvitin phosphopeptide (Glu-Asp-Asp-pSer-pSer) as a potential food ingredient to intervene in osteoporosis: A potential mechanism study on promoting the osteogenic differentiation of MC3T3-E1 cells

Tianrui Xue,Tingting Gong,Wei Zhao,Mingran Zhang,Binfei Lv,Huiping Liu,Qi Xu,Xiaowei Zhang

Food-derived peptides with strong calcium-binding and osteogenic activity are promising functional ingredients for osteoporosis intervention. This study aims to explore whether a novel phosvitin phosphopeptide (Glu-Asp-Asp-pSer-pSer, EDDpSpS) with excellent calcium binding ability can promote the osteogenic differentiation of MC3T3-E1 cells and its potential mechanism. Network pharmacology analysis of EDDpSpS regulating osteoblast differentiation identified that the focal adhesion was a key signaling pathway, which is closely related to cell differentiation. Molecular simulations demonstrated that EDDpSpS was strongly bound to the target proteins within the focal adhesion signaling pathway and maintains a stable state. In addition, EDDpSpS had the potential to activate the key target protein Src tyrosine kinase (Src). Results of cell experiments showed that EDDpSpS (> 100 μg/mL) significantly promoted the differentiation and mineralization of MC3T3-E1 cells. Transcriptome analysis verified that the expression of paxillin (PXN), Src, actin beta/gamma 1 (ACTG1), and other osteoblast genes were regulated by EDDpSpS, leading to an up-regulation of the focal adhesion. In this study, the effect and the potential mechanism of EDDpSpS on osteoblast differentiation were clarified. EDDpSpS may be used as an effective dual-effect food ingredient for developing functional foods targeting osteoporosis.

Circulating metabolites are associated with persistent elevations of ALT in patients with chronic hepatitis B with complete viral suppression

Dekai Zheng,Changhao Cheng,Yanhua Tang,Zhixin Fang,Xuelian Gao,Yuchuan Chen,Qiuhong You,Kaifeng Wang,Heqi Zhou,Zhixian Lan,Jian Sun

Hepatitis B virus (HBV) can be completely suppressed after antiviral treatment; however, some patients with chronic hepatitis B (CHB) exhibit elevated alanine aminotransferase (ALT) levels and sustained disease progression. This study provides novel insights into the mechanism and potential predictive biomarkers of persistently elevated ALT (PeALT) in patients with CHB after complete viral inhibition. Patients having CHB with undetectable HBV DNA at least 12 months after antiviral treatment were enrolled from a prospective, observational cohort. Patients with PeALT and persistently normal ALT (PnALT) were matched 1:1 using propensity score matching. Correlations between plasma metabolites and the risk of elevated ALT were examined using multivariate logistic regression. A mouse model of carbon tetrachloride-induced liver injury was established to validate the effect of key differential metabolites on liver injury. Of the 1238 patients with CHB who achieved complete viral suppression, 40 (3.23%) had PeALT levels during follow-up (median follow-up: 2.42 years). Additionally, 40 patients with PnALT levels were matched as controls. Ser-Phe-Ala, Lys-Ala-Leu-Glu, 3-methylhippuric acid, 3-methylxanthine, and 7-methylxanthine were identified as critical differential metabolites between the two groups and independently associated with PeALT risk. Ser-Phe-Ala and Lys-Ala-Leu-Glu levels could be used to discriminate patients with PeALT from those with PnALT. Furthermore, N -acetyl- l -methionine (NALM) demonstrated the strongest negative correlation with ALT levels. NALM supplementation alleviated liver injury and hepatic necrosis induced by carbon tetrachloride in mice. Changes in circulating metabolites may contribute to PeALT levels in patients with CHB who have achieved complete viral suppression after antiviral treatment.
