The repair of bone defects from various causes remains a significant challenge in clinical settings. Piezoelectric materials have been demonstrated to promote osteogenic differentiation, among which barium titanate (BT) has been widely reported. Magnesium oxide (MgO) dopants not only enhance the piezoelectric properties but also stimulate the osteogenic and angiogenic properties in stem cells. In this study, a MgO-modified BT/hydroxyapatite (MgO@BT/HA) piezoelectric ceramic scaffold was constructed using three-dimensional (3D)-printed technology. The results indicated that the addition of MgO improved the electrical properties of BT ( d 33 = 68.26 pC/N), and facilitated an increase in the proportion of HA (30%) while maintaining the piezoelectric coefficient ( d 33 = 2.04 pC/N) of the ceramic system. Furthermore, under the induction of low-intensity pulsed ultrasound (LIPUS) stimulation, in vitro biosafety experiments confirmed that MgO@BT/HA exhibits excellent biosafety and promotes the osteogenic differentiation of dental pulp stem cells (DPSCs).

Fe/Mn (hydr)oxides are metallic compounds that exhibit significant redox activity in environmental media and play a pivotal role in geochemical processes, thereby influencing the fate of metals in porous media. The morphology of Fe/Mn (hydr)oxides in natural environments and their interactions with trace metals are significantly influenced by the presence of natural organic matter (NOM). However, there is limited understanding regarding the formation, transport, and stability of Fe/Mn (hydr)oxides in the environment. The present study employed humic acid (HA) as a representative NOM material to investigate the positive influence of HA on the formation of Fe/Mn colloids. However, there remains limited comprehension regarding the formation, transport, and stability of Fe/Mn (hydr)oxides in the natural environment. In this study, we investigated the positive effect of natural organic matter (NOM) on the formation of Fe/Mn colloids using humic acid (HA) as a representative NOM material. We comprehensively characterized the chemical and physical properties of HA–Fe/Mn colloids formed under various environmentally relevant conditions and quantitatively analyzed their subsequent aggregation and stability behaviors. The findings suggest that the molar ratios of C to Fe/Mn (hydr)oxide play a pivotal role in influencing the properties of HA–Fe/Mn colloids. The formation and stability of HA–Fe/Mn colloids exhibit an upward trend with increasing initial molar ratios of C to Fe/Mn. Redox and metal–carboxylic acid complexation reactions between HA and hydrated iron/manganese oxides play a pivotal role in forming colloidal HA–Fe/Mn complexes. Subsequent investigations simulating porous media environments have demonstrated that the colloidal structure resulting from the interaction between HA and Fe/Mn facilitates their migration within surrounding porous media while also enhancing their retention in the surface layers of these media. This study offers novel insights into the formation and stabilization mechanisms of HA–Fe/Mn colloids, which are pivotal for comprehending the behavior of Fe/Mn colloids and the involvement of Fe/Mn (hydr)oxides in geochemical cycling processes within porous media.

Nanomicelles, by virtue of their prominent biocompatibility, degradability, and ability to solubilize hydrophobic drugs, have been widely used as the most effective delivery platform for anticancer drugs. However, undesirable drug-loading capacity, unfeasible modification, poor in vivo stability, and intratumoral penetration remain to be addressed. Herein, we introduce a novel core/shell PEGylated poly(glycerol sebacate) (PEGS)/hydroxyapatite (HA) hybrid nanomicelle based on a unique triblock PEGS substrate with functional carboxyls in terminals and free hydroxyls as pendant groups. The hydrophobic doxorubicin (DOX) can be controllably encapsulated in the core of nanomicelles via hydrogen bonding, and ensuing in situ mineralization of HA occurs as a shell layer with the electrostatic effect between the carboxylate radical (COO–) and calcium ion (Ca2+). Through optimizing the coordination of PEGS nanomicelles and HA mineralization, 20–30 nm spherical nanoparticles can be formed with considerable drug loading (0.38 mg DOX/1 mg nanoparticles) and a sensitive pH-responsive release (about 50% release amount at pH 5.6 while <5% release amount at pH 7.4 in 24 h). In further in vitro studies, this PEGS/HA hybrid nanoparticle system exhibits excellent selective tumor inhibitory efficacy, while in in vivo studies, high efficacy of tumor suppression and low incidence of toxicity can be evidenced in a DOX-loaded PEGS/HA group (71.7% decrease in average tumor volume compared to a control group after 15 day hypodermic treatment). The core/shell PEGS/HA nanoparticle coordinated with PEGS nanomicelles and in situ HA mineralization represents high drug-loading capacity, multifunctional possibility, and tumor-selective and responsive release profiles and could offer a highly promising platform for tumor therapy in clinical application.

Tumor microenvironment (TME)-responsive size-changeable and biodegradable nanoplatforms for multimodal therapy possess huge advantages in anti-tumor therapy. Hence, we developed a hyaluronic acid (HA) modified CuS/MnO 2 nanosheets (HCMNs) as a multifunctional nanoplatform for synergistic chemodynamic therapy (CDT)/photothermal therapy (PTT)/photodynamic therapy (PDT). The prepared HCMNs exhibited significant NIR light absorption and photothermal conversion efficiency because of the densely deposited ultra-small sized CuS nanoparticles on the surface of MnO 2 nanosheet. They could precisely target the tumor cells and rapidly decomposed into small sized nanostructures in the TME, and then efficiently promote intracellular ROS generation through a series of cascade reactions. Moreover, the local temperature elevation induced by photothermal effect also promote the PDT based on CuS nanoparticles and the Fenton-like reaction of Mn 2+ , thereby enhancing the therapeutic efficiency. Furthermore, the T 1 -weighted magnetic resonance (MR) imaging was significantly enhanced by the abundant Mn 2+ ions from the decomposition process of HCMNs. In addition, the CDT/PTT/PDT synergistic therapy using a single NIR light source exhibited considerable anti-tumor effect via in vitro cell test. Therefore, the developed HCMNs will provide great potential for MR imaging and multimodal synergistic cancer therapy.

The colloid of ferrous sulfide modified by humic acid (HA-n-FeS) shows good reduction and immobilization efficiency for variable-valence heavy metals in wastewater. The removal efficiency of HA-n-FeS for halogenated organic pollutants, however, remains unclear, especially in the absence and presence of oxygen. This study addressed this issue by exploring the effect and mechanism of dissolved oxygen on the degradation of tetrabromobisphenol A (TBBPA) by the HA-n-FeS colloid in water. The results showed that the removal efficiency of different concentrations of TBBPA (5,10, and 20 μm) by the HA-n-FeS colloid was 33.16%, 20.48%, and 22.37% in the absence of oxygen, respectively. When TBBPA reacted with the HA-n-FeS colloid, the concentration of Fe(II) and S(-II) remained stable. The adsorption of HA-n-FeS was the main mechanism of removing TBBPA in the absence of oxygen. In the presence of oxygen, the removal efficiency of TBBPA by the HA-n-FeS colloid was 82.37%, 56.80%, and 43.78% (for the above-mentioned TBBPA concentrations), respectively. In addition, the removal capacity of TBBPA by HA-n-FeS was 39.63, 52.21, and 89.75 mg/g, respectively. The concentration of Fe(II) and S(-II) decreased rapidly in time. Among them, the HA-n-FeS colloid removed part of the TBBPA through chemical adsorption . The main way of chemical adsorption was pore adsorption and functional group (olefin C C, phenolic hydroxyl group O–H, alcohol group C–O) combination. Besides, the HA-n-FeS colloid degraded part of the TBBPA into BPA through reduction, in which 17.72% of TBBPA was removed by the reduction of HA-n-FeS colloid. Fe(II) was the main contributor to the reductive degradation of TBBPA. Furthermore, active species ( 1 O 2 and •O 2 − ) played a minor role in the removal of TBBPA by the HA-n-FeS colloids with oxygen, where 13% of TBBPA was removed by 1 O 2 and •O 2 − . Therefore, in practical applications, the aeration method can be used to significantly improve the removal efficiency of TBBPA by HA-n-FeS colloids in water.

Due to biological safety and negligible toxicity, nitric oxide (NO) therapy has gained increasing interest in the field of cancer therapy during the past few years. However, individual NO cancer therapy normally exhibited limited therapeutic efficiency. In order to acquire satisfactory therapeutic outcomes, the NO therapy is usually combined with other therapeutic treatments, mostly chemotherapy. Herein, we constructed HA-TPP@NORM nanoparticles based on the co-assembly of an NO donor (NORM) and tetraphenylporphyrin (TPP)-modified hyaluronic acid, which can efficiently generate a highly biocidal molecule of peroxynitrite (ONOO−) via the synergistic interactions of the nitric oxide (NO) therapy and photodynamic therapy (PDT) to enhance the cancer therapeutic efficiency. In addition, MTT results exhibited that without light irradiation, the HA-TPP@NORM nanoparticles have favourable biocompatibility with the cell viability above 95% at the maximum TPP concentration (20 μg mL−1). Under simultaneous irradiation with 365 nm and 650 nm light, ONOO− can be efficiently produced in cancer cells via the direct coupling reaction of the generated NO and superoxide anion radical (O2˙−), which significantly enhanced the anticancer effect, when compared with individual NO therapy or PDT therapy. Therefore, the HA-TPP@NORM nanoparticles may provide a new insight into the design of efficiently NO-related cancer therapeutic systems.

To engineer bone tissue, an ideal biodegradable implant should be biocompatible, biodegradable, osteoinductive and osteoconductive. However, the lack of bioactivity has seriously restricted the development of biodegradable implants in bone tissue engineering. In this study, we have developed a three-dimensional porous poly(L-lactic-co-glycolic acid)/hydroxyapatite (PLGA/HA) scaffold based on the dual delivery of bone morphogenic protein 2 (BMP-2) and insulin-like growth factor 1 (IGF-1) via polydopamine (PDA) coating. The aim of this study was to improve the osteoinductivity of biodegradable implants for clinical bone repair via a PDA-assisted BMP-2 and IGF-1 surface modification strategy. Our study demonstrated that PDA layer surface modification could more efficiently immobilize BMP-2 and IGF-1 on the scaffold surfaces than physical adsorption, and the immobilized growth factor was released slowly and steadily from the scaffold in a sustained manner. MC3T3-E1 cell attachment and proliferation on IGF-1 and BMP-2-immobilized porous scaffolds were much higher than other groups. According to an in vitro osteogenesis assay, alkaline phosphatase (ALP) activity, the expression of osteogenesis-related genes and the mineralization capacity of MC3T3-E1 cells were increased by the incorporation of BMP-2 and IGF-1. In vivo, digital radiograph evaluation demonstrated that there was the most rapid healing in the defects treated with the IGF-1 and BMP-2 immobilized porous scaffold compared with the other groups eight weeks after implantation. In vitro and in vivo results of this study revealed that the PDA-assisted surface modification method can be a useful tool to graft biomolecules onto biodegradable implants, and the dual release of BMP-2 and IGF-1 could promote cell proliferation and osteogenesis differentiation. Based on the results, the surface modification strategy with growth factors has great potential for the enhancement of osteointegration of biodegradable bone implants.

As biodegradable orthopedic implant materials, magnesium alloys have been attracted enough attentions recently. However, too fast degradation in vivo , limited biocompatibilities, and insufficient antibacterial properties are three main challenges at present. In order to solve these problems, a multifunctional composite coating of Chi(Zn/BMP2)/HA was constructed on AZ31B magnesium alloy surface, successively by the alkali heating treatment, self-assembly of 16-phosphonyl-hexadecanoic acid, in situ immobilization of Chi(Zn/BMP2) (chitosan, zinc ions, and bone morphogenetic protein 2), and the deposition of HA (hydroxyapatite). The results of ATR-FTIR (attenuated total reflection Fourier transform infrared spectrum) spectra and elemental compositions confirmed that 16-phosphonyl-hexadecanoic acid, Chi(Zn/BMP2), and HA were successfully immobilized on the surface. Compared with Mg, Mg-OH, Mg-16, and Mg-Chi(Zn/BMP2), Mg-Chi(Zn/BMP2)/HA with the concave–convex structure surface significantly enhanced the hydrophilicity and corrosion resistance. On the other hand, Mg-Chi(Zn/BMP2)/HA coating also showed excellent biocompatibilities, which not only significantly promoted the osteoblast adhesion and proliferation, but also upregulated ALP and OCN expression of osteoblasts. Furthermore, due to the synergistic antibacterial effect of zinc ions and chitosan, Mg-Chi(Zn/BMP2)/HA showed a good antibacterial property against Escherichia coli ( E. coli ). Therefore, it can be said that the method used in this work has a good application prospect in improving the corrosion resistance, biocompatibility of magnesium alloys, and inhibiting infections against E. coli .

This study explored the applicability, cellular efficacy, and osteogenic activities of porous nano-hydroxyapatite/Poly (glycerol sebacate)-grafted maleic anhydride (n-HA/PGS-g-M) composite scaffolds. Nuclear magnetic resonance (NMR) analyses indicated that approximately 43% of the hydroxide radicals in PGS were displaced by maleic anhydride. Resonance bands at 1036 cm −1 occurred in scaffolds containing nHA powders, and peak areas increased when n-HA weight increased in PGS-M-n-HA-0.4, PGS-M-n-HA-0.5, and PGS-M-n-HA-0.6 scaffolds. The n-HA/PGS-g-M composite scaffolds exhibited porous microstructure with average pore size of 150–300 µm in scanning electron microscopy (SEM) analysis. Differential scanning calorimetry (DSC) identified the glass transition temperature (Tg) as −25–30 °C, indicative of quality resilience. The modulus of compressibility increased when n-HA content increased. Interestingly, viability of human adipose-derived stem cells (hADSCs) in vitro and expression of the osteogenic related genes RUNX2 , OCN , and COL1A1 was enhanced in the n-HA/PGS-g-M composite scaffolds compared to those factors observed in PGS-g-M scaffolds. Finally, simulated body fluid (SBF) tests indicated more apatite deposits on the surface of n-HA/PGS-g-M scaffolds compared to PGS-g-M scaffolds. Overall, porous n-HA/PGS-g-M composite scaffolds possessed acceptable biocompatibility and mechanical properties, and they stimulated hADSC cell proliferation and differentiation. Given these qualities, the composite scaffolds have potential applications in bone tissue engineering.

Silicone hydrogels have emerged as promising candidates for ocular drug delivery due to their exceptional oxygen permeability. However, their inherent hydrophobicity often results in suboptimal drug-loading capacity and patient discomfort. To address these limitations, this study introduces an innovative dual-network silicone/β-CD-HA hydrogel. In this design, the primary silicone network preserves high oxygen permeability, while the secondary β-CD-HA network enhances hydrophilicity and introduces additional drug-loading sites via host-guest interactions. Levofloxacin (LVF), a broad-spectrum antibiotic, was incorporated into the hydrogel through non-covalent binding with β-CD, and the system’s drug-loading capacity, sustained release profile, in vitro bactericidal activity, and post-loading properties were systematically evaluated. The results demonstrated a total LVF loading of 959.90 ± 36.60 μg/lens in the silicone/β-CD-HA hydrogel, with sustained release extending up to 8 h. Bactericidal assays revealed markedly reduced E. coli survival rates in treated groups: 14.46 ± 0.76 % (p < 0.0001) for LVF-loaded silicone hydrogel and 10.42 ± 1.66 % (p < 0.0001) for the β-CD-HA/silicone hydrogel, underscoring the enhanced antimicrobial efficacy of the dual-network system. The integration of β-CD-HA significantly improved hydrogel hydrophilicity, increasing equilibrium water content (EWC) from 27.69 ± 0.18 % to 45.60 ± 0.19 % and reducing the surface contact angle from 99.41 ± 0.24° to 70.36 ± 0.14°. Crucially, the modified hydrogel retained functionality-critical properties, exhibiting 93.45 ± 0.42 % light transmittance and 110.45 ± 0.32 barrer oxygen permeability values well within practical application standards. Post-drug-loading characterization showed minimal changes in oxygen permeability (105.47 ± 0.18 barrer), transmittance, or mechanical integrity, confirming structural stability. These findings collectively highlight the dual-network silicone/β-CD-HA hydrogel as a robust platform for extended ocular drug delivery, effectively balancing enhanced drug loading, sustained release kinetics, without compromising essential material properties.

Improving the drug loading and delivery efficiency of biodegradable nanomaterials used for targeting prostate cancer (PCa) remains a challenging task. To accomplish this task, herein, a new surface molecularly imprinted polymer (ZIF-8/DOX-HA@MIP) was designed and constructed with a hyaluronic acid (HA)-modified zeolitic imidazolate framework-8 (ZIF-8) metal–organic framework loaded with doxorubicin (DOX) as a substrate and a responsive molecularly imprinted polymer film as a shell. Owing to the large surface area of ZIF-8, DOX was successfully loaded into the ZIF-8/DOX-HA@MIP with a high drug loading efficiency (more than 88%). In vitro cell experiments have shown that the strengthened targeting ability of ZIF-8/DOX-HA@MIP to PCa cells was realized through the synergistic effect of HA and the molecularly imprinted membrane. Under the condition of simulated tumor microenvironment solution, Zn species were released and the particle size of ZIF-8/DOX-HA@MIP decreased gradually by the synergistic effect of hyaluronidase, pH, and glutathione, showing excellent biodegradability. In vivo antitumor research indicated the excellent antitumor activity and biocompatibility of ZIF-8/DOX-HA@MIP. The multifunctional ZIF-8/DOX-HA@MIP constructed herein provides a novel impetus for the development of targeted drug delivery in PCa treatment and a new strategy for treating other tumors.

Following dental implantation, the characteristic bacterial milieu of the oral cavity may lead to peri-implant inflammation, which can negatively impact osseointegration and cause implant failure. To improve soft tissue sealing around the implant, enhance osseointegration, and improve implant success rates, this paper proposes a composite multifunctional coating (PHG) prepared using gelatin and polydopamine/hydroxyapatite nanoparticles, investigates the effects of this novel coating on cell adhesion, proliferation, antibacterial activity, osteogenic differentiation, and evaluates its immune-related properties. The PHG coating was proved to have satisfactory hydrophilicity and wettability for cell attachment. Furthermore, it improved the expression of adhesion-related genes and proteins in human gingival fibroblasts, indicating its adhesion-promoting effect. Additionally, bone marrow mesenchymal stem cells exhibited strong osteogenic differentiation potential and mineralization on PHG-coated surfaces. Notably, the PHG coating exhibited antibacterial activity against Streptococcus mutans , as well as anti-inflammatory effects, potentially via the regulation of macrophages. Therefore, the proposed PHG coating may promote soft tissue sealing and bone bonding, providing a potential strategy for the surface modification of dental implants.
