Collective influence of substrate chemistry with physiological fluid shear stress on human umbilical vein endothelial cells

Yan Li,Zhongjie Qin,Lin Zhou,Khawar Ali Shahzad,Delin Xia

In the treatment of cardiovascular diseases, vascular scaffold materials play an extremely important role. The appropriate substrate chemistries and 15 dynes/cm 2 physiological fluid shear stress (FSS) are both required to ensure normal physiological activity of human umbilical vein endothelial cells (HUVECs). The present study reported the collective influence of substrate chemistries and FSS on HUVECs in the sense of its biological functions. The CH 3 , NH 2 , and OH functional groups were adopted to offer a variety of substrate chemistries on glass slides by the technology of self-assembled monolayers, whereas FSS was generated by a parallel-plate fluid flow system. Substrate chemistries on its own by no means had noticeable effects on eNOS, ATP, NO, and PGI 2 expressions, while FSS stimuli enhanced their production. While substrate chemistries, as well as FSS, were both exerted, the releases of ATP, NO, and PGI 2 were dependent on substrate chemistries. Study of F-actin organization and focal adhesions (FAs) formation of HUVECs before FSS exposure proves that F-action organization and FAs formation followed similar chemistry-dependence. Hereby proposed a feasible mechanism, that is, the F-actin organization and FAs formation of HUVECs are controlled by substrate chemistries, further advancing the modulation of FSS-triggered responses of HUVECs.

Microencapsulation Facilitates Low-Cryoprotectant Vitrification of Human Umbilical Vein Endothelial Cells

Yufang Li,Kashan Memon,Yuanyuan Zheng,Yue Cheng,Momoh Karmah Mbogba,Peitao Wang,Xilin Ouyang,Gang Zhao

Vitrification has become one of the promising cryopreservation methods for biosamples including cells and tissues because the vitreous state reduces the damage of ice crystals to cells. However, besides extremely high cooling rates, routine vitrification protocols require a high concentration of penetrating cryoprotectants (pCPAs, ∼6–8 M), which is toxic for cells and brings trouble when removing pCPAs. Therefore, reducing the concentration of toxic pCPAs in vitrification remains a challenge, and advanced strategies are urgently needed. Hydrogel encapsulation has become one effective method to achieve low-cryoprotectant (CPA) concentration preservation of stem cells with rapid cooling, but there are very few related studies about endothelial cells (ECs). In this study, we achieved pCPA concentration (up to 3 M) vitrification by encapsulating human umbilical vein endothelial cells (HUVECs) into core–shell alginate hydrogel microcapsules. Alginate encapsulation increased HUVEC cryosurvival up to 80%, which is 60% improvement compared to control without encapsulation. Furthermore, two different sizes of capsules (diameter: ∼900 and 400 μm) were produced to explore the effects of microcapsule volume on the cell preservation results, and it was found that larger capsules (∼900 μm) have no significant effect on cell survival while improving encapsulation efficiency. This encapsulation method provides a new strategy for EC preservation and serves as an improvement to optimize the preservation of biosamples.

Enhanced hydrophilicity of one-step electrosprayed red blood cell-like PLGA microparticles by block polymer PLGA-PEG-PLGA with excellent magnetic-luminescent bifunction and affinity to HUVECs

Junwei Xu,Yi Cui,Meili Liu,Zitong An,Kun Li,Xuenan Gu,Ping Li,Yubo Fan

The incidence of cancer and other diseases continues to increase, posing a serious threat to human health. Drug delivery systems using bionic red blood cell (RBC)-like microparticles (MPs) as drug carriers have been extensively studied, as they are considered ideal carriers due to the unique shape. However, most methods of preparing multifunctional RBC-like MPs are complicated, and the cell affinity and therapeutic effect of MPs prepared using simple electrospray are unsatisfactory. In this context,   hydrophilicity-enhanced and magnetic-luminescent bifunctional RBCs-like Janus MPs [Fe 3 O 4 @poly(lactic-co-glycolic acid) (PLGA)-poly(ethylene glycol) (PEG)-PLGA/PLGA]//[NaYF 4 :Eu 3+ @PLGA-PEG-PLGA/PLGA] were successfully prepared by one-step double-needle electrospray method. The physical and chemical properties, such as morphology, particle size, structure, and thermal stability of the MPs, were studied. The addition of block polymer PLGA-PEG-PLGA was found to enhance the hydrophilicity of the MPs, promote cell spreading and adhesion proteins (focal adhesion kinase and Paxillin) mRNA expressions of human umbilical vein endothelial cells, significantly improving the cell affinity. The results also demonstrated that RBC-like MPs exhibited excellent magnetic and enhanced luminescent properties, making them a promising candidate for various biomedical applications, including drug delivery systems, magnetic targeting, bio-imaging, and fluorescent tracing.

Alleviation of endothelial dysfunction of Pheretima guillemi (Michaelsen)-derived protein DPf3 in ponatinib-induced thrombotic zebrafish and mechanisms explored through ox-LDL-induced HUVECs and TMT-based proteomics

Yali Wu,Yunnan Ma,Wanling Zhong,Huijuan Shen,Jinhong Ye,Shouying Du,Pengyue Li

Ethnopharmacological relevance Thrombus generation is one of the leading causes of death in human, and vascular endothelial dysfunction is a major contributor to thrombosis. Pheretima guillemi (Michaelsen), a traditional medicinal animal known as “Dilong”, has been utilized to cure thrombotic disorders for many years. DPf3, a group of functional proteins extracted from P. guillemi , has been characterized and identified to possess antithrombotic bioactivity via in vitro and ex vivo experiments. Aim of the study This study is aimed to investigate the vascular-protection activity and related mechanism of antithrombotic protein DPf3 purified from Pheretima guillelmi systematically. Materials and methods The antithrombotic activity and vascular endothelium protection effect of DPf3 was explored in vivo using ponatinib-induced vascular endothelial injury zebrafish thrombus model. Then, (hi) ox-LDL-induced HUVECs was applied to investigate the protection mechanism of DPf3 against the injury of vascular endothelium. In addition, TMT-based proteomics analysis was used to study the biomarkers, biological processes and signal pathways involved in the antithrombotic and vascular protective effects of DPf3 holistically. Results DPf3 exerted robust in vivo antithrombosis and vascular endothelial protection ability. DPf3 was identified to prevent HUVECs from damage by reducing ROS production, and to reduce monocyte adhesion by decreasing the protein content of adhesion factor VCAM 1. DPf3 was also observed to weaken the migration ability of injured cells and inhibit abnormal angiogenesis. The mechanism of DPf3's antithrombotic and vascular protective activity was mainly related to the regulation of lipid metabolism, energy metabolism, complement and coagulation system, ECM receptor interaction, MAPK signal pathway, etc. Conclusions This study demonstrates that DPf3 has strong antithrombotic and endothelial protective effects. The endothelial protective ability and related mechanisms of DPf3 provide a scientific reference for the traditional use of earthworms in the treatment of thrombosis.

On-Chip Label-Free Sorting of Living and Dead Cells

Guowei Wang,Chengpan Li,Chunguang Miao,Shibo Li,Bensheng Qiu,Weiping Ding

With the emergence of various cutting-edge micromachining technologies, lab on a chip is growing rapidly, but it is always a challenge to realize the on-chip separation of living cells from cell samples without affecting cell activity and function. Herein, we report a novel on-chip label-free method for sorting living and dead cells by integrating the hypertonic stimulus and tilted-angle standing surface acoustic wave (T-SSAW) technologies. On a self-designed microfluidic chip, the hypertonic stimulus is used to distinguish cells by producing volume differences between living and dead cells, while T-SSAW is used to separate living and dead cells according to the cell volume difference. Under the optimized operation conditions, for the sample containing 50% of living human umbilical vein endothelial cells (HUVECs) and 50% of dead HUVECs treated with paraformaldehyde, the purity of living cells after the first separation can reach approximately 80%, while after the second separation, it can be as high as 93%; furthermore, the purity of living cells after separation increases with the initial proportion of living cells. In addition, the chip we designed is safe for cells and can robustly handle cell samples with different cell types or different causes of cell death. This work provides a new design of a microfluidic chip for label-free sorting of living and dead cells, greatly promoting the multi-functionality of lab on a chip.

Chao Han,Dingsong Lu,Suoping Yang,Chong Liu,Feng Guo,Kai Zhang,Peng Li

The design and construction of a new and excellent synthetic graft is of great significance in the field of bone defect repair and reconstruction. In this study, a dopamine modified chitosan hydrogel doped with Cu ions with a mild photothermal effect was designed to provide a better microenvironment to advance the bone repair via promote the angiogenesis and osteogenesis. Characterizations showed the successful synthesis of the material while it also presented excellent biocompatibility and mild photothermal effect under the irradiation of near-infrared light. Further, it could enhance the angiogenesis of HUVECs cells through promoting the ability of migration and tube formation and enhance the osteogenic differentiation of MC3T3-E1 cells via increasing the content of vital osteogenic factors including Runx2, Col-1, OPN, OCN, OSX, etc. The in vivo experiment also testified that it could promote the bone defect repair in rat models. These results indicate the multifunctional hydrogel is an ideal material for the treatment of bone defects and has good clinical application potential.

In Situ Visualization and SERS Monitoring of the Interaction between Tumor and Endothelial Cells Using 3D Microfluidic Networks

Ziting Qian,Jiayuan Fei,Shenfei Zong,Kuo Yang,Lang Li,Ruiyang Liu,Zhuyuan Wang,Yiping Cui

A multifunctional microfluidic platform was demonstrated to monitor the interaction between tumor cells and endothelial cells by integrating a three-dimensional (3D) cell culture unit with a protein detection unit. In such a chip, breast cancer cells MCF7 were seeded into the collagen to form a 3D tumor environment while human umbilical vein endothelial cells (HUVECs) are seeded in the channel next to the collagen matrix. Thus, an in situ growth of angiogenic sprouting can be visualized through fluorescence in the 3D collagen matrix after a coculture of MCF7 and HUVEC after 4 days, which cannot be observed in the 2D culture environment. On the other hand, gold@silver core–shell nanorods were used as surface-enhanced Raman scattering (SERS) immunoprobes for the detection of the secretion of cytokine (vascular endothelial growth factor, VEGF). The limit of detection of the VEGF is 100 pg/mL. Further, as LiCl and bevacizumab can act as a promoter and an inhibitor of VEGF, the dynamic change of the concentration of VEGF under the stimulation of them was monitored by SERS signals. Thus, this integrated SERS microfluidic platform creates opportunity for the fundamental research of interaction between tumors and endothelial cells.

3D bioprintable methacrylated carrageenan/sodium alginate dual network hydrogel for vascular tissue engineering scaffolding

Anduo Zhou,Yasong Hu,Chongshuai Chen,Heyi Mao,Lei Wang,Shanfeng Zhang,Xia Huang

3D printing technology has significant advantages in the processing field of vascular tissue engineering. We have developed KCMA/SA hydrogels based on methacrylated carrageenan (KCMA) and sodium alginate (SA), which have low viscosity and shear thinning properties and can be used for 3D printing. The physicochemical double cross-linked network structure leads to good mechanical properties. The composite hydrogels maintain the liquid crystallinity of KC, which causes the HUVECs to align. The KCMA/SA gel filaments printed by the 3D extrusion printing system formed a connected microstructure. These characteristics all indicate the potential of KCMA/SAs in the field of tubular tissue regeneration.

Kaempferol Protects Blood Vessels From Damage Induced by Oxidative Stress and Inflammation in Association With the Nrf2/HO-1 Signaling Pathway

Yao He,Sun Jingyu,Wei Jie,Zhang Xin,Chen Bing,Lin Yajun

Over recent years, an increasing number of studies have confirmed that the occurrence and development of vascular pathological changes are closely related to oxidative stress and the inflammatory response of the vascular endothelium. Kaempferol is the most common flavonoid compound found in fruits and vegetables. Our present research identified that kaempferol had the capability to protect the vascular endothelium in a mouse model of vascular injury and explored the specific mechanisms underlying these effects by investigating oxidative stress, the extent of cardiovascular injury, and inflammatory markers such as NF-κB, TNF-α, IL-6, and the Nrf2/HO-1 signaling pathway. Analysis showed that kaempferol reduced oxidative stress and inflammation mediated by H2O2 and paraquat, respectively, both in vitro and in vivo. Furthermore, kaempferol suppressed the levels of TNF-α and IL-6, and the activation of NF-κB, in aortic tissues and human umbilical vein endothelial cells (HUVECs). Finally, we observed that kaempferol corrected the levels of antioxidants and elevated the protein levels of Nrf2 and HO-1 in aortic tissues and HUVECs. Collectively, our findings prove that kaempferol protects blood vessels from damage induced by oxidative stress and inflammation and that the Nrf2/HO-1 signaling pathway plays a key role in mediating these effects.

Comparative study of in vitro effects of different nanoparticles at non-cytotoxic concentration on the adherens junction of human vascular endothelial cells

Tao Wen,Aiyun Yang,Lingyu Piao,Suisui Hao,Lifan Du,Jie Meng,Jian Liu,Haiyan Xu

Background Effects of different nanoparticles (NPs) exposure at acutely non-cytotoxic concentrations are particularly worthy to figure out, compare, and elucidate. Objective To investigate and compare the effect of a small library of NPs at non-cytotoxic concentration on the adherens junction of human umbilical vein endothelial cells (HUVECs), obtaining new insights of NPs safety evaluation.Materials and methods The HUVECs layer was exposed to NPs including gold (Au), platinum (Pt), silica (SiO2), titanium dioxide (TiO2), ferric oxide (Fe2O3), oxidized multi-walled carbon nanotubes, with different surface chemistry and size distribution. Cellular uptake of NPs was observed by transmission electron microscopy. and the cytotoxicity was determined by Cell Counting Kit-8 assay. The NP-induced variation of intracellular reactive oxygen species (ROS) and catalase (CAT) activity was measured using the probe of 2ʹ7’-dichlorodihydr fluorescein diacetate and a CAT analysis kit, respectively. The level of VE-cadherin of HUVECs was analyzed by Western blot, and the loss of adherens junction was observed with laser confocal microscopy. Results The acutely non-cytotoxic concentrations of different NPs were determined and applied to HUVECs. The NPs increased the level of intracellular ROS and the activity of CAT to different degrees, depending on the characteristics. At the same time, the HUVECs lost their adherens junction protein VE-cadherin and gaps were formed between the cells. The NP-induced oxidative stress and gap formation could be rescued by the supplementary N-acetylcysteine in the incubation. Conclusion The increase of intracellular ROS and CAT activity was one common effect of NPs, even at the non-cytotoxic concentration, and the degree was dependent on the composition, surface chemistry, and size distribution of the NP. The effect led to the gap formation between the cells, while could be rescued by the antioxidant. Therefore, the variation of adherens junction between endothelial cells was suggested to evaluate for NPs when used as therapeutics and diagnostics.

Mechanisms of action underlying the effect of Tongsaimai on wound healing based on experimental and network pharmacology

Fanxing Xu,Jingxian Wu,Yumei Hu,Chun Chu,Wenjun Liu,Xiang Li,Wen Zheng,Weishuo Yang,Boyan Zhao,Jiangxue Guo,Zhenzhong Wang,Ying Jia,Wei Xiao

Ethnopharmacological relevance Tongsaimai (TSM) is a traditional Chinese medicine that has several therapeutic qualities, including anti-inflammatory, anti-oxidative, and anti-vasculitis effects. However, its impacts and underlying mechanisms on wound healing remain unclear. Aim of the study The aim of our study was to evaluate TSM for its pro-healing effect and the relevant mechanisms using both experimental validation and network pharmacology analysis. Materials and methods The components of TSM were detected by high-performance liquid chromatography combined with diode array detector (HPLC-DAD). Skin wounds with a diameter of 4 mm were created on the backs of mice, after which, topical treatments of 2.5–10% TSM were applied onto the lesions once daily for either 2 or 7 days. Then, the wound tissues were collected to determine the impacts of TSM on collagen deposition, epithelial cell proliferation , oxidative stress, inflammation, and angiogenesis. Moreover, the effects of TSM (0.5–2 mg/mL) on the cell viability of HUVECs and HaCaT cells were evaluated. Results A total of 11 components in TSM were identified by HPLC-DAD. TSM was found to enhance the rate of wound contraction and increase epithelial thickness and collagen deposition during the healing process. In addition, TSM increased SOD activity and downregulated MDA and IL-1β levels in the wound tissues. Immunofluorescence analysis further indicated an increased expression of Ki67, CD31, and VEGF in wound tissues following TSM administration. Results of the network pharmacology analysis revealed that multiple pathways including VEGF, PI3K/Akt, and MAPK pathways were involved in the pharmacological actions of TSM on wound healing. Accordantly, in vitro experiments revealed that TSM promoted the proliferation of HUVECs and HaCaT cells while activating the PI3K/Akt pathway. Conclusions Our results suggest that TSM may serve as a therapeutic medication to improve wound healing by employing multiple regulatory mechanisms that affect proliferation, angiogenesis, collagen deposition, oxidative stress, and inflammation.

Ferulic acid suppresses the inflammation and apoptosis in Kawasaki disease through activating the AMPK/mTOR/NF-κB pathway

Wu Huilan,Wang Yijia,Tan Pingping,Ran Yuqing,Guan Yuting,Qian Songwei,Feng Xing,Jiang Yalan,Peng Yongmiao,Sheng Ke,Xi Haitao,Ji Weiping,Guo Xiaoling

Background: Kawasaki disease (KD) is a self-limiting and acute systemic vasculitis of unknown etiology, mainly affecting children. Ferulic acid (FA), a natural phenolic substance, has multiple pharmacological properties, including anti-inflammatory, anti-apoptosis, and anti-fibrosis, and so on. So far, the protective effects of FA on KD have not been explored. Methods: In this study, we established Candida albicans water soluble fraction (CAWS)-induced mouse coronary artery vasculitis of KD model and the tumor necrosis factor α (TNF-α)-induced human umbilical vein endothelial cells (HUVECs) injury model to investigate the anti-inflammatory and anti-apoptosis effects of FA on KD, and try to elucidate the underlying mechanism. Results: Our in vivo results demonstrated that FA exerted anti-inflammatory effects on KD by inhibiting the infiltration of CD45-positive leukocytes and fibrosis around the coronary artery. Additionally, FA downregulated the levels of inflammatory and chemotactic cytokines, alleviated splenomegaly, and exhibited anti-apoptotic effects on KD by reducing TUNEL-positive cells, downregulating BAX expression, and upregulating BCL-2 expression. In addition, Our in vitro findings showed that FA could effectively inhibit TNF-α-induced HUVEC inflammation like NF-κB inhibitor QNZ by downregulating the expression of pro-inflammatory cytokines as well as attenuated TNF-α-induced HUVEC apoptosis by reducing apoptotic cell numbers and the BAX/BCL-2 ratio, which could be reversed by the AMPK inhibitor compound c (CC). The further mechanistic study demonstrated that FA could restrain vascular endothelial cell inflammation and apoptosis in KD through activating the AMPK/mTOR/NF-κB pathway. However, FA alone is hard to completely restore KD into normal condition. Conclusion: In conclusion, FA has potential protective effects on KD, suggesting its promising role as an adjuvant for KD therapy in the future.
