High-performance lateral flow immunoassay using aggregation-induced-emission nanospheres for sensitive detection of lambda-cyhalothrin residue in food

Keyang Lai,Lele Ma,Gan Zhang,Esther De Rycke,Erik Beij,Zongyou Chen,Wenjun Gui,Shuying Li,Weihua Lai,Yirong Guo

The fluorescence performance of AIEgens was improved after molecular modification. • The new fluorescence LFIA exhibited broad linear range and high sensitivity for LCT detection. • The proposed LFIA is applicable to detect LCT in various food samples. The extensive and widespread use of lambda-cyhalothrin (LCT) has raised concerns over its adverse effects on the environment and human health, highlighting the urgent need for rapid and sensitive detection of LCT residues in food commodities. In this study, a highly sensitive lateral flow immunoassay (LFIA) was developed using novel aggregation-induced-emission (AIE) fluorescent nanospheres for the determination of LCT. Starting from tetraphenylethylene (TPE), a typical AIE luminogen, three derivatives were synthesized via chemical reaction. Among them, tetraldehyde tetraphenylethylene-based fluorescent nanospheres (TAPEFN) exhibited superior optical performance. Following conjugation of TAPEFN with anti-LCT monoclonal antibodies, the resulting TAPEFN-mAb served as an efficient fluorescent probe within the LFIA platform. The established TAPEFN-LFIA exhibited a linear detection range of 3.9 to 500 ng/mL, with a limit of detection of 3.8 ng/mL, which was 4.2-fold lower compared to TPEFN-LFIA (15.85 ng/mL). The reliability of the established method was demonstrated by the detection of spiked food samples, with acceptable recoveries (72.82%–119.34%) and low coefficients of variation (<16.97%). These findings demonstrate that rational molecular engineering can significantly enhance the fluorescence properties of AIE materials, and that the resulting nanospheres provide a powerful signal-reporting strategy for improving LFIA sensitivity. Multi-omics reveals coordinated intracellular–extracellular metabolic remodeling underlying lambda-cyhalothrin transformation and tolerance in Chlorella sp. XDA-R55

Liping Yang,Nuerbiye Yisimayi,Wanyi Zhang,Mingyang Sun,Xinyue Tang,Lingna Chen,Yongkun Chen,Ai Shanjiang

A highly tolerant Chlorella sp. achieved efficient lambda-cyhalothrin transformation. • Multi-omics revealed coordinated detoxification and metabolic remodeling. • Intracellular–extracellular remodeling drove adaptive LC responses. • XDA-R55 treatment reduced lambda-cyhalothrin-induced oxidative stress and toxicity. • Mechanisms underlying microalgal pesticide adaptation were elucidated. Lambda-cyhalothrin (LC), a widely used pyrethroid insecticide, poses persistent ecological risks due to its toxicity and environmental persistence. In this study, a microalga strain, Chlorella sp. XDA-R55, was investigated as a potential biological platform for LC-contaminated water remediation. XDA-R55 maintained active growth under 20 mg/L LC exposure and promoted 96.6% LC removal within 6 days without external carbon supplementation. Physiological analyses revealed that XDA-R55 alleviated LC-induced oxidative stress by enhancing antioxidant defense systems and maintaining cellular redox homeostasis. Integrated transcriptomic and metabolomic analyses indicated that XDA-R55 facilitates the detoxification and transformation of LC by upregulating genes associated with glutathione metabolism, biosynthesis of secondary metabolites, and ABC transporters. Several putative transformation products, such as 4-hydroxybenzaldehyde, were identified and exhibited transient accumulation followed by decline. Ecological safety assessments demonstrated that water treated with XDA-R55 significantly alleviated oxidative stress, metabolic disturbances, and immune suppression in zebrafish, with most biomarkers recovering to levels comparable to those in the control group. Moreover, as a photoautotrophic organism, XDA-R55 facilitated LC removal and transformation without external carbon supplementation, providing a sustainable and environmentally compatible strategy for pesticide-contaminated water remediation. These findings provide mechanistic insights into microalgal adaptation and LC transformation under pesticide stress, and highlight the potential of Chlorella sp. XDA-R55 for mitigating the ecological risks associated with LC-contaminated water. Download: Download high-res image (189KB) Download: Download full-size image

Detection and classification of lambda-cyhalothrin and iprodione residues in tobacco leaves by SERS combined with supervised machine learning

Weifan Pan,Peng Zhang,Qi Wang,Liangguang Du,Xue Yang,Xiaowei Guo,Jiamin Yu

Surface-enhanced Raman spectroscopy (SERS) is a promising technique for the rapid detection of trace pesticide residues in agricultural produce. Currently, research is predominantly centered on utilizing SERS to discern residues characterized by non-overlapping spectral peaks. Yet, the technology encounters challenges in accurately identifying substances when their Raman spectral peaks overlap. In this paper, a silver nanoparticles (Ag Nps) modified SiO 2 nanoparticles (SiO 2 Nps) arrays composite SERS substrate (SiO 2 @Ag Nps-SERS substrate) was prepared for detecting lambda-cyhalothrin (LCT) and iprodione (IPR) residues in tobacco leaves, with detection limits of 0.1 ppm and 1 ppm respectively. And then, supervised machine learning models were used to identify and classify LCT and IPR residues with overlapped Raman characteristic peaks at 1000 cm −1 . Support Vector Machine (SVM), Partial Least Squares Discriminant Analysis (PLS-DA) and Decision Tree (DT) models could all accurately classify the spectral data of LCT and IPR residues, and the classification accuracy of SVM model was the highest, reaching 99.74 %. This capability is a testament to the innovative strength of our approach, highlighting its potential for universal applicability in spectral data analysis.

Susceptibility of fall armyworm, Spodoptera frugiperda (J.E. Smith), to eight insecticides in China, with special reference to lambda-cyhalothrin

Yun-Xia Zhao,Jing-Mei Huang,Huan Ni,Di Guo,Feng-Xia Yang,Xin Wang,Shun-Fan Wu,Cong-Fen Gao

Fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is the main destructive insect pest of grain crops that occurs in all maize growing regions of the Americas. It has rapidly invaded the Southern China since January 2019. However, the current status of insecticide resistance in S. frugiperda has not been reported in China. In this study, we determined the susceptibility of eight populations of FAW to eight insecticides by an artificial diet incorporation method. The results showed that among eight insecticides, emamectin benzoate, spinetoram, chlorantraniliprole, chlorfenapyr, and lufenuron showed higher toxicity to this pest, while lambda-cyhalothrin and azadirachtin exhibited lower toxicity. Susceptibility of S. frugiperda to indoxacarb was significantly different (10.0-fold for LC 50 ) across the various geographic populations. To investigate the biochemical mechanism of FAW to lambda-cyhalothrin, we performed the synergism tests and the results showed that piperonyl butoxide (PBO) and triphenyl phosphate (TPP) produced a high synergism of lambda-cyhalothrin effects in the two field populations. Sequencing of the gene encoding the acetylcholinesterase (AChE) gene in the two field populations identified two amino acid mutations, all of which have been shown previously to confer resistance to organophosphates (OPs) in several arthropod species. The results of this study provided valuable information for choosing alternative insecticides and for insecticide resistance management of S. frugiperda .

Molecular Cloning and Expression of CYP9A61: A Chlorpyrifos-Ethyl and Lambda-Cyhalothrin-Inducible Cytochrome P450 cDNA from Cydia pomonella

Xueqing Yang,Xianchun Li,Yalin Zhang

Cytochrome P450 monooxygenases (CYPs or P450s) play paramount roles in detoxification of insecticides in a number of insect pests. However, little is known about the roles of P450s and their responses to insecticide exposure in the codling mothCydia pomonella(L.), an economically important fruit pest. Here we report the characterization and expression analysis of the first P450 gene, designated asCYP9A61, from this pest. The full-length cDNA sequence ofCYP9A61is 2071 bp long and its open reading frame (ORF) encodes 538 amino acids. Sequence analysis shows that CYP9A61 shares 51%–60% identity with other known CYP9s and contains the highly conserved substrate recognition site SRS1, SRS4 and SRS5. Quantitative real-time PCR showed thatCYP9A61were 67-fold higher in the fifth instar larvae than in the first instar, and more abundant in the silk gland and fat body than other tissues. Exposure of the 3rd instar larvae to 12.5 mg L−1of chlorpyrifos-ethyl for 60 h and 0.19 mg L−1of lambda-cyhalothrin for 36 h resulted in 2.20-and 3.47-fold induction ofCYP9A61, respectively. Exposure of the 3rd instar larvae to these two insecticides also significantly enhanced the total P450 activity. The results suggested thatCYP9A61is an insecticide-detoxifying P450.Keywords:Cydia pomonella;cytochrome P450;induction;response;insecticides

Identification of 14 glutathione S-transferase genes from Lasioderma serricorne and characterization of LsGSTe1 involved in lambda-cyhalothrin detoxification

Xi-Ya Liu,Hui-Mei Sun,Yi-Hong Luo,Mao-Ye Li,Hai-Bing Liu,Su Liu

In insects, glutathione S -transferases (GSTs) play a pivotal role in the detoxification of a wide range of pesticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest insect of stored products. Recently, pyrethroid insecticides have been used to control this pest. However, little is known concerning the responses and functions of GSTs in L. serricorne under pyrethroid exposure. In this study, transcriptome sequencing was performed on L. serricorne , and a total of 14 GSTs were identified by retrieving the unigene dataset. Of these, 13 predicted GSTs fell into six cytosolic classes, namely, delta, epsilon, omega, sigma, theta, and zeta, and one was assigned to an “unclassified” group. The GST genes were differentially expressed in various larval tissues and at different developmental stages. Exposure to the pyrethroid insecticide lambda-cyhalothrin (LCT) caused oxidative stress in L. serricorne larvae and led to significantly elevated expression levels of six genes, among which LsGSTe1 was the most upregulated. Recombinant LsGSTE1 protein displayed LCT-metabolizing activity. Furthermore, LsGSTE1 protects cells against oxidative stress. Moreover, knockdown of LsGSTe1 by RNA interference dramatically increased the susceptibility of L. serricorne larvae to LCT treatment. The results from this study provide sequence resources and expression data for GST genes in L. serricorne . Our findings indicate that LsGSTE1 plays a dual role in LCT detoxification by metabolizing the pesticide and by preventing LCT-induced oxidative stress. Thus, the LsGSTe1 gene could be used as a potential target for sustainable management of the cigarette beetle.

Exposure to lambda-cyhalothrin and abamectin drives sublethal and transgenerational effects on the development and reproduction of Cydia pomonella

Di Ju,Yu-Xi Liu,Xue Liu,Youssef Dewer,David Mota-Sanchez,Xue-Qing Yang

The codling moth Cydia pomonella (Lepidoptera: Tortricidae) is a major invasive pest of pome fruits and walnuts worldwide. Lambda-cyhalothrin (LCT) and abamectin (AM) have been frequently used in C. pomonella control, but control of this pest is very difficult because shortly after hatching, larvae of this insect bore tunnels and hide inside host plant fruit. In this study, a simulated field spray bioassay method was developed against neonate larvae of C. pomonella and concentration-response bioassays were conducted to evaluate the susceptibility of the neonate larvae to LCT and AM. Exposure of neonate larvae to sublethal concentration (LC 30 ) of LCT or AM significantly reduced the survival rate of larvae (4th and 5th instars), lowered the mean weight of larvae and pupae, and decreased the daily maximal number of eggs laid and the total number of eggs laid (fecundity) per female. The sublethal effects, including reduced body mass, mean fecundity and net reproductive rate, extended mean generation time, and shortened oviposition period, were also found in transgenerational offspring. Furthermore, the transgenerational maternal effects were more obvious for AM than LCT, in comparison to the control. Additionally, the estimated population size was decreased by exposure to LC 30 of LCT and AM, and the observed reduction of fecundity and population size within and across generations was likely the result of the downregulation of the reproduction-related vitellogenin gene ( CpVg ) after exposure to LC 30 of LCT and AM. These results provide a better understanding of the overall effects of LCT and AM on C. pomonella and the transgenerational effects which should be taken into consideration when using insecticides in order to control C. pomonella .

A heat shock protein protects against oxidative stress induced by lambda-cyhalothrin in the green peach aphid Myzus persicae

Bao Dong,Xi-Ya Liu,Bin Li,Mao-Ye Li,Shi-Guang Li,Su Liu

Lambda-cyhalothrin (LCT) is a pyrethroid insecticide widely used to control insect pests . Insect exposure to LCT may cause abnormal accumulation of reactive oxygen species (ROS) and result in oxidative damage. Heat shock proteins (HSPs) may help protect against oxidative stress . However, little is known about the role of HSPs in response to LCT in the green peach aphid , Myzus persicae . This insect is an important agricultural pest causing severe yield losses in crops. In this study, we characterized a cDNA sequence ( MpHsp70 ) encoding a member of the HSP70 family in M. persicae . MpHsp70 encoded a 623 amino acid protein putatively localized in the cytosol. The highest expression level of MpHsp70 occurred in fourth-instar nymphs. Treatment of M. persicae with LCT resulted in oxidative stress and significantly increased H 2 O 2 and malondialdehyde levels. This led to an elevated transcription level of MpHsp70 . Injection of H 2 O 2 into M. persicae also upregulated the MpHsp70 expression level, suggesting that MpHsp70 is responsive to ROS, particularly H 2 O 2 , induced by LCT. Recombinant MpHSP70 protein was expressed in Escherichia coli . E. coli cells overexpressing MpHSP70 exhibited significant tolerance to H 2 O 2 and the ROS generators, cumene hydroperoxide and paraquat . This indicated that MpHSP70 protects against oxidative stress. Furthermore, knockdown of MpHsp70 by RNA interference resulted in increased susceptibility in apterous adults of M. persicae to LCT. These findings indicate that MpHsp70 plays an important role in defense against LCT-induced oxidative stress and insecticide susceptibility in M. persicae .

Rapid determination of lambda-cyhalothrin using a fluorescent probe based on ionic-liquid-sensitized carbon dots coated with molecularly imprinted polymers

Zhang Dianwei,Tang Jiaqi,Liu Huilin

A highly selective and sensitive fluorescent probe for optosensing lambda-cyhalothrin (LC) was prepared. The probe was based on sulfur-doped carbon dots (CDs) coated with molecularly imprinted polymers (MIPs). Doping the CDs with sulfur and modifying the surfaces of the CDs with an ionic liquid enhanced the performance of the fluorescent probe. The selectivity of the probe was improved through the application of molecular imprinting technology utilizing acrylamide and 1-vinyl-3-butylimidazolium tetrafluoroborate [VBIm][BF 4 ] as functional monomers. The resulting probe was used to detect LC, which is a pesticide residue, in vegetables and tea. Under optimal detection conditions, the linear range of the probe was found to be 1–150 μg kg −1 and the limit of detection to be 0.5 μg kg −1 by analyzing excitation/emission maxima at 350/450 nm. The developed method was successfully used to determine LC in vegetables and tea, yielding recoveries of 98.90–116.93%. These results suggest that this fluorescent probe based on MIP-coated, room-temperature ionic-liquid-sensitized, sulfur-doped carbon dots has great potential to be utilized for the precise detection of LC in complex samples. The voltage-gated sodium channel mutation and detoxification enzymes associated with the resistance of Grapholita molesta (Busck) (Lepidoptera: Tortricidae) to lambda-cyhalothrin

Xiaohe Zhang,Sha Su,Caiyun Qiu,Bufei Ma,Zheng Zhao,Xiong Peng,Jaime C. Piñero,Maohua Chen

The oriental fruit moth , Grapholita molesta (Busck) (Lepidoptera: Tortricidae), a major fruit pest, is primarily controlled with pyrethroid insecticides such as lambda-cyhalothrin and resistance has emerged as a serious threat to management. In this study, we compared a lambda-cyhalothrin-resistant strain (RS) with a susceptible strain (SS) of G. molesta . Sequencing of the voltage-gated sodium channel (VGSC) gene revealed the L1014F mutation exclusively in RS individuals, strongly associated with knockdown resistance. Bioassays showed that the RS exhibited a 13.1-fold cross-resistance to deltamethrin and a 6.0-fold cross-resistance to bifenthrin, as well as moderate cross-resistance to chlorantraniliprole, abamectin, and emamectin benzoate. While showing only low or negligible cross-resistance to other insecticides tested. Synergist assays indicated that piperonyl butoxide (PBO) increased lambda-cyhalothrin toxicity by 1.72-fold in RS, implicating cytochrome P450 as key contributors. Biochemical assays confirmed significantly elevated activities of glutathione S-transferase (GST) and P450 in RS relative to SS. Transcriptomic analysis identified 758 differentially expressed genes, including 13 P450s, 5 GSTs, and 2 carboxylesterases (CarEs), with qRT-PCR validating strong up-regulation of CYP333B121 (14-fold), CYP6B230 (15-fold), CYP6AB117 (13-fold), and GSTs2 (5-fold). These findings suggest that the target-site resistance conferred by the L1014F kdr mutation, together with metabolic resistance mediated by overexpressed P450 and GST genes, jointly contribute to the resistance of G. molesta to lambda-cyhalothrin.

Characterization and Functional Analysis of Three Cytochrome P450 Monooxygenases in Lasioderma serricorne: Assessing Their Contribution to Lambda-Cyhalothrin Susceptibility Through RNA Interference

Bin Li,Dao-Shuang Zhu,Yong-Hui Zhang,Chuan-Tao Xu,De-Xin Chen,Mao-Ye Li,Su Liu

In insects, cytochrome P450 monooxygenases (CYPs) play a pivotal role in the detoxification of various insecticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest of stored products, for which pyrethroid insecticides are increasingly used. Nevertheless, the responses and functions of CYPs in L. serricorne under pyrethroid exposure remain poorly understood. In this study, three full-length CYP cDNAs were identified from L. serricorne and designated as CYP6BK29 , CYP6SY1 , and CYP6TE1 . Phylogenetic analysis revealed that these CYPs belong to the CYP3 clan and the CYP6 family. Reverse transcription–quantitative PCR showed that CYP6BK29 and CYP6SY1 were predominantly expressed in the larval midgut, whereas CYP6TE1 transcripts were enriched in the fat body. The expression of CYP6BK29 peaked in the third- and fourth-instar larvae, while CYP6SY1 was specifically highest in the fourth-instar. In contrast, CYP6TE1 showed the highest mRNA levels in adults. Exposure to a sublethal dose (LD 30 ) of lambda-cyhalothrin (LCT) significantly upregulated the mRNA levels of CYP6BK29 and CYP6TE1 , but not that of CYP6SY1 . RNAi-mediated knockdown of CYP6BK29 significantly increased the susceptibility of larvae to LCT. In contrast, silencing CYP6SY1 or CYP6TE1 had no significant effect on larval survival under LCT exposure. These results provide evidence that CYP6BK29 contributes to LCT detoxification in L. serricorne , highlighting its potential as a molecular target for novel pest management strategies. Summary Characterization of three novel CYP genes ( CYP6BK29 , CYP6SY1 , and CYP6TE1 ) in Lasioderma serricorne . Successful transcriptional knockdown of all three genes via RNAi. CYP6BK29 knockdown enhances larval susceptibility to lambda-cyhalothrin, implicating its role in detoxification.
