Rapid and high-throughput analysis of 5-hydroxymethylfurfural in honey by MALDI-MS with in-situ tetra (4-aminophenyl)-porphyrin derivatization

Sun Zhi,Wang Fangfang,Zhou Peipei,Chen Di,Zuo Lihua

5-Hydroxymethylfurfural (HMF), a product of the Maillard reaction and caramelization in honey, serves as a crucial marker of food quality and thermal processing, underscoring the importance of regulatory surveillance for consumer safety. This study introduces a method for rapid, high throughput and eco-friendly detection of HMF, using derivatization in matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). 5,10,15,20-Tetrakis-(4-aminophenyl)-porphyrin was employed for in-situ derivatization of aldehydes after extraction by ethyl acetate, enhancing detection sensitivity by shifting low-molecular-weight aldehydes to higher regions. The calibration curves for HMF determination exhibited outstanding linearity. The detection limit reached 0.347 mg/kg with good accuracy and precision. Variations in the content of HMF in honey during heating were also assessed, revealing the increased formation of HMF under high temperature and prolonged heating. The method’s proficiency was evidenced through the detection of various aldehydes. This method represents an advance in rapid analysis for food safety, aligning with green chemistry principles.

High-throughput screening and spatial profiling of low-mass pesticides using a novel Ti3C2 MXene nanowire (TMN) as MALDI MS matrix

Chunxia Ma,Xiao Wang,Huamin Zhang,Wei Liu,Daijie Wang,Feng Liu,Heng Lu,Luqi Huang

Pesticides play critical roles in agricultural fields; however, pesticide residues can cause serious damage to human health and the ecological environment; therefore, developing a rapid and sensitive method for pesticide detection is urgently needed. Nanostructure-assisted matrix laser desorption/ionization (MALDI) mass spectrometry (MS) has great potential for the detection of low-mass pesticides. In this study, a novel Ti 3 C 2 MXene nanowire (TMN) was prepared by a facile sol-gel method and served as a matrix to enhance MALDI MS performance in the analysis of pesticides in positive ion mode. The TMN showed superior performance in the high-throughput detection of six kinds of pesticides (organophosphorus, organochlorine , carbamate, neonicotinoids, triazole, and oxadiazines), with ultrahigh sensitivity (detection limits at sub-ppt levels), remarkable repeatability, excellent salt tolerance, and extremely low background compared to traditional organic matrices due to the specific polyaromatic structure and the doping of nitrogen. Furthermore, this matrix was successfully employed for the analysis of residual pesticides in traditional Chinese herbs, and the level of diniconazole was quantified with a linear range of 0–50 ng/mL (R 2  > 0.99). More importantly, the spatial distribution of various endogenous compounds (e.g., amino acids and saccharides, fatty acids, alkaloids, and plant hormones) and xenobiotic pesticides from the intact root of the medicinal plant P. quinquefolium was clearly visualized using the TMN self-assembly film as a matrix for MALDI imaging mass spectrometry (IMS). With superior advantages such as sensitivity, simplicity, rapidness, and minimal sample requirement, TMN as a matrix-assisted MALDI MS shows great promise for various applications.

Fluoro-functionalized ionic covalent organic frameworks (F-iCOFs) for highly selective enrichment and sensitive determination of perfluorinated sulfonates by MALDI-MS

Liu Xiao-Pan,Sun Wen-Qian,Zhao Meng-Ge,Zhang Xing-Jie,Liu Li-Hong,Chen Chang-Po

An easily prepared fluoro-functionalized ionic covalent organic framework (F-iCOF) has been implemented into MALDI-TOF MS, enabling the highly selective enrichment and sensitive determination of perfluorinated sulfonate (PFS) contaminants in a rapid and convenient manner. The good thermal stability and excellent optical absorption properties of F-iCOF makes it a brilliant matrix with no background noise. Moreover, benefitting from the large surface area, appropriate pore size, good water dispersibility, and abundant fluorine atom and cationic characteristic of F-iCOF, it exhibited superior adsorption capacity and enrichment selectivity towards PFSs. Good signal responses for PFSs were obtained in the presence of various interfering compounds such as BSA, HA, or even more than 100-fold excess of glutamic acid and similar in structure sodium alkyl sulfonates, highlighting the specific selectivity of F-iCOF. Calibration curves for potassium perflurobutane sulfonate (PFBSK) in tap water and whole blood were established with good linear correlation in the range 1–500 pg mL −1 . The limits of detection and quantification for PFBSK were as low as 0.04 pg mL −1 and 0.05 pg mL −1 , respectively, which are comparable or better than the existing methods for the determination of PFSs. Combination of nano-material enrichment and dead-end filtration for uniform and rapid sample preparation in matrix-assisted laser desorption/ionization mass spectrometry

Zengnan Wu,Mashooq Khan,Sifeng Mao,Ling Lin,Jin-Ming Lin

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) is a fast analysis tool for the detection of a wide range of analytes . However, heterogeneous distribution of matrix/analyte cocrystal, variation in signal intensity and poor experimental reproducibility at different locations of the same spot means difficulty in quantitative analysis. In this work, carbon nanotubes (CNTs) were employed as adsorbent for analyte cum matrix on a conductive porous membrane as a novel mass target plate. The sample pretreatment step was achieved by enrichment and dead-end filtration and dried by a solid-liquid separation. This approach enables the homogeneous distribution of analyte in the matrix, good shot-to-shot reproducibility in signals and quantitative detection of peptide and protein at different concentrations with correlation coefficient (R 2 ) of 0.9920 and 0.9909, respectively. The simple preparation of sample in a short time, uniform distribution of analyte, easy quantitative detection, and high reproducibility makes this technique useful and may diversify the application of MALDI-MS for quantitative detection of a variety of proteins.

A cheap 3D-printed boron, nitrogen, sulfur-MXene quantum dots-based target plate for multi-dimensional MALDI MS imaging of small-molecule environmental pollutants

Zifang Peng,Yanhao Zhang,Yide Dong,Wuduo Zhao,Jian Mao,Qidong Zhang,Gangfeng Ouyang,Shusheng Zhang,Jianping Xie

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging (MALDI-TOF MSI) generally possess difficulties for analysis of small-molecule environmental pollutants. In the present study, an unreported straightforwardly synthesized ternary-doped boron, nitrogen, sulfur-MXene quantum dots (BNS-MQDs) was incorporated into photopolymer clear resin, and the mixture was applied to manufacture of a novel 3D-printed MALDI target plate (3D-MTP). Due to BNS-MQDs’ nano-size, uniform shape and excellent capabilities of energy absorption/transfer, their doping helped the self-made 3D-MTP successfully achieve multi-dimensional MALDI-TOF MSI of small-molecule environmental contaminants. 3D printing technology also made construction of 3D-MTP cheap ($ 0.1) and time-saving (20 min), considerably reducing experimental cost. Furthermore, MALDI-TOF MSI using BNS-MQDs-based 3D-MTP did not need manual and instrumental matrix spray, which is conducive to obtaining accurate and reproducible imaging results. Besides excellent performance of 2D MSI of environmental contaminants, 3D-MTP was also successfully employed to 3D MALDI-TOF MSI of an emerging polluted antioxidant, N, N’ -di-2-naphthyl-p-phenylenediamine (DNPD), in female zebrafish after exposure. The enrichment of DNPD in female zebrafish was found in eye, gill, liver, intestine and ovary for the first time. Overall, the developed BNS-MQDs-based 3D-MTP showed compelling application prospects in multi-dimensional MALDI-TOF MSI research of small-molecule environmental xenobiotics.

A highly sensitive isotope-coded derivatization method and its application for the mass spectrometric analysis of analytes containing the carboxyl group

Jiapeng Leng,Haoyang Wang,Li Zhang,Jing Zhang,Hang Wang,Yinlong Guo

A novel isotope labeling reagent d 0 -/ d 6 -2, 4-dimethoxy-6-piperazin-1-yl pyrimidine (DMPP) has been developed for derivatization toward the carboxyl group based on carbodiimide chemistry for mass spectrometry (MS) analysis. The strengths of this derivatization strategy involve fast labeling (15   s), low chemical background and general access to most carboxylic analytes. This has been demonstrated using a series of compounds containing carboxylic acids, including peptides and proteins. To enhance the MS response of the derivatized analytes, the design of DMPP has been based on integration of the theoretical consideration of high gas-phase hydrogenation capacity and hydrophobicity. In addition, the high abundance product ions at m / z 225 and m / z 231 from d 0 -/ d 6 -DMPP labeled carboxylic acids indicate high efficiency of the gas-phase cleavage induced by the labeling reagent. Quantitative determination of these ions can also be used in single reaction monitoring to achieve extremely high sensitivity toward the target analytes. This has subsequently been used to determine trace free fatty acids in human urine. Furthermore, the DMPP labeled peptides also provide additional sequence information in MALDI–MS/MS because of the formation of sequence-related isotope fragment ions. This DMPP-oriented labeling technique is expected to be a promising tool for the MS detection of many varieties of compounds containing carboxyl groups.

In-depth profiling of carbohydrate isomers in biological tissues by chemical derivatization-assisted mass spectrometry imaging

Lei Wang,Yuhao Han,Yaqi Zhang,Haoyuan Geng,Zihan Zhu,Panpan Chen,Xiaoqing Cui,Xiao Wang,Chenglong Sun

Carbohydrates play crucial regulatory roles in various physiological and pathological processes. However, the low ionization efficiency and the presence of linkage pattern, monosaccharide composition and anomeric configuration isomers make their in-depth analysis very challenging, especially for heterogeneous biological tissues. In this study, we propose a high-sensitive and isomer-specific imaging approach to visualize the spatial distributions of monosaccharide and disaccharide isomers by integrating chemical derivatization and matrix-assisted laser desorption/ionization tandem mass spectrometry imaging (MALDI-MS 2 I). 2-Pyridinecarbohydrazide (PYD) is developed as a novel derivatization reagent which can not only improves the MS sensitivity of carbohydrates, but also enables the identification and visualization of ketose and aldose monosaccharide isomers, as well as linkage pattern, monosaccharide composition and anomeric configuration disaccharide isomers by mass spectrometry imaging of isomer-specific MS/MS fragment ions. Moreover, we build quantitative MALDI-MS 2 and MALDI-MS 2 I methods for disaccharide isomers based on the diagnostic fragment ions, and good linear relationships could be achieved both in solution and on glass slides. We expect that this study should provide new ideas for in-depth profiling of the spatial signatures of carbohydrates in biological tissues and lay the foundation for a deeper understanding of carbohydrates’ structure.

Molecularly Imprinted Heterostructure-Assisted Laser Desorption Ionization Mass Spectrometry Analysis and Imaging of Quinolones

Ruya Wang,Weidong Zhang,Weiqiang Liang,Xiao Wang,Lili Li,Zhenhua Wang,Miaomiao Li,Jun Li,Chunxia Ma

Quinolone residues resulting from body metabolism and waste discharge pose a significant threat to the ecological environment and to human health. Therefore, it is essential to monitor quinolone residues in the environment. Herein, an efficient and sensitive matrix-assisted laser desorption/ionization mass spectrometry (MALDI/MS) method was devised by using a novel molecularly imprinted heterojunction (MIP-TNs@GCNs) as the matrix. Molecularly imprinted titanium dioxide nanosheets (MIP-TNs) and graphene-like carbon nitrides (GCNs) were associated at the heterojunction interface, allowing for the specific, rapid, and high-throughput ionization of quinolones. The mechanism of MIP-TNs@GCNs was clarified using their adsorption properties and laser desorption/ionization capability. The prepared oxygen-vacancy-rich MIP-TNs@GCNs heterojunction exhibited higher light absorption and ionization efficiencies than TNs and GCNs. The good linearity (in the quinolone concentration range of 0.5–50 pg/μL, R2 > 0.99), low limit of detection (0.1 pg/μL), good reproducibility (n = 8, relative standard deviation [RSD] < 15%), and high salt and protein resistance for quinolones in groundwater samples were achieved using the established MIP-TNs@GCNs–MALDI/MS method. Moreover, the spatial distributions of endogenous compounds (e.g., amino acids, organic acids, and flavonoids) and xenobiotic quinolones from Rhizoma Phragmitis and Rhizoma Nelumbinis were visualized using the MIP-TNs@GCNs film as the MALDI/MS imaging matrix. Because of its superior advantages, the MIP-TNs@GCNs–MALDI/MS method is promising for the analysis and imaging of quinolones and small molecules.

Polydopamine-capped AgNPs as a novel matrix overcoming the ion suppression of phosphatidylcholine for MALDI MS comprehensive imaging of glycerophospholipids and sphingolipids in impact-induced injured brain

Chao Han,Shumu Li,Qingwei Yue,Na Li,Hui Yang,Zhenwen Zhao

Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) is a powerful tool for the characterization and localization of analytes without the need for extraction, purification, separation or labeling of samples. However, in tissue sections the most abundant lipids, phosphatidylcholines (PCs), could suppress the signals of other classes of coexisting lipids. In this work, polydopamine (PDA)-capped AgNPs (AgNPs@PDA) were synthesized as a matrix of MALDI MSI to analyze lipids in both positive and negative ion modes. By adjusting the thickness of the PDA layer, the signal of silver cluster ions of AgNPs@PDA was effectively controlled, and the ability of AgNPs@PDA serving as a matrix was optimized. More interestingly, using AgNPs@PDA as a matrix, the sensitivity of PCs was dramatically decreased, and the PC signals were greatly suppressed, while for other lipids (including PE, HexCer, PS, PI, PIP, and ST), they were just the opposite. The reason, we believe, is related to the positively charged surface of AgNPs@PDA, and the polyhydroxy and amino groups of PDA. Benefitting from the suppression of the signals of PCs and the improvement of detection sensitivity of other lipids, 58 glycerophospholipids and 25 sphingolipids in brain tissue sections could be imaged in one run with AgNPs@PDA as a matrix by MALDI MSI, much better than when using traditional organic matrices 2,5-dihydroxybenzoic acid and 9-aminoacridine.

Machine Learning Assisted MALDI Mass Spectrometry for Rapid Antimicrobial Resistance Prediction in Clinicals

Weibo Gao,Hang Li,Jingxian Yang,Jinming Zhang,Rongxin Fu,Jiaxi Peng,Yechen Hu,Yitong Liu,Yingshi Wang,Shuang Li,Shuailong Zhang

Antimicrobial susceptibility testing (AST) plays a critical role in assessing the resistance of individual microbial isolates and determining appropriate antimicrobial therapeutics in a timely manner. However, conventional AST normally takes up to 72 h for obtaining the results. In healthcare facilities, the global distribution of vancomycin-resistant Enterococcus fecium (VRE) infections underscores the importance of rapidly determining VRE isolates. Here, we developed an integrated antimicrobial resistance (AMR) screening strategy by combining matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) with machine learning to rapidly predict VRE from clinical samples. Over 400 VRE and vancomycin-susceptible E. faecium (VSE) isolates were analyzed using MALDI-MS at different culture times, and a comprehensive dataset comprising 2388 mass spectra was generated. Algorithms including the support vector machine (SVM), SVM with L1-norm, logistic regression, and multilayer perceptron (MLP) were utilized to train the classification model. Validation on a panel of clinical samples (external patients) resulted in a prediction accuracy of 78.07%, 80.26%, 78.95%, and 80.54% for each algorithm, respectively, all with an AUROC above 0.80. Furthermore, a total of 33 mass regions were recognized as influential features and elucidated, contributing to the differences between VRE and VSE through the Shapley value and accuracy, while tandem mass spectrometry was employed to identify the specific peaks among them. Certain ribosomal proteins, such as A0A133N352 and R2Q455, were tentatively identified. Overall, the integration of machine learning with MALDI-MS has enabled the rapid determination of bacterial antibiotic resistance, greatly expediting the usage of appropriate antibiotics.

Multi-wavelength visible-light induced [2+2] cycloaddition for identification of lipid isomers in biological samples

Yuhao Han,Panpan Chen,Zhichao Li,Xiao Wang,Chenglong Sun

Ultraviolet (UV) light-catalyzed Paternò-Büchi (PB) reaction has been developed as an efficient lipid C=C double bond (DB) derivatization strategy, which can accurately assign the position of C=C bond in unsaturated lipids when coupled with tandem mass spectrometry (MS/MS). Inspired by this, here we proposed a novel visible-light induced [2+2] cycloaddition reaction combined with ESI-MS/MS and MALDI-MS/MS to identify lipid C=C position isomers. Benz[g]isoquinoline-5,10-dione (BIQD) and 6,9-difluorobenzo[g]isoquinoline-5,10-dione (DF-BIQD) were developed as a new type of [2+2] cycloaddition reagent, which can not only react with C=C bond under 254 nm UV light irradiation, but also quickly combine with lipid C=C bond under the irradiation of 405 nm visible-light and > 400 nm compact fluorescent lamp visible-light. High cycloaddition reaction conversion efficiency can be achieved by irradiating under compact fluorescent lamp light for 2 min. Moreover, we discovered that 437 nm, 489 nm, 545 nm, 581 nm, and 613 nm monochromatic light appearing in compact fluorescent lamp can individually induce the [2 + 2] cycloaddition reaction between DF-BIQD and unsaturated lipids. Using this method, we found that the expressions of lipid DB-positional isomers in rat heart, brain, lung, spleen, thymus, kidney, liver and plasma vary greatly. The relative content of FA-18:1 (Δ9) in rat heart is only 1.49 times that of FA-18:1 (Δ11), while the relative content of FA-18:1 (Δ9) in rat plasma is 5.20 times that of FA-18:1 (Δ11). The above results offer new insight into the development of photocatalytic reagent for visible-light induced [2+2] cycloaddition and structural lipidomic studies.
