Ni-metal-organic-framework (Ni-MOF) membranes from multiply stacked nanosheets (MSNs) for efficient molecular sieve separation in aqueous and organic solvent

Guiliang Li,Yanmin Qi,Haibo Lin,Na Lu,Jiaping Chen,Jianqiang Wang,Qiu Han,Fu Liu

Lamellar membranes are attracting extensive attention for high throughput and precise molecules/ions sieve separation. However, the energy-consuming exfoliation and low yield of single-layer nanosheets restrict the massive production of membranes. Herein, we facilely fabricated 2D Ni-mediated metal-organic framework (Ni-MOF) membranes directly from multiply stacked nanosheets (MSNs) for both aqueous and organic molecular sieve separation. Multiply layered nanosheets were obtained from a high concentration dispersion via simple ultrasonic treatment. The morphology and chemistry of synthesized Ni-MOF nanosheets were characterized via scanning electron microscopy (SEM), N 2 adsorption-desorption, Fourier transform infrared spectra (FTIR), and X-ray photoelectron spectra (XPS). The thickness of Ni-MOF membranes could be well regulated by the volume of dispersion. The membrane exhibited high molecular rejection to 8GX (97.29%), EB (96.28%), CR (99.69%), and CBB (93.31%) in water, respectively and a stable perm-selectivity in continuous 24 h filtration. The lamellar membrane showed excellent pressure and pH (2–10) resistance. The membrane achieved rational permeance of various organic solvents mainly depending on viscosity and good rejection to MB (91.37%), BY (92.01%), and EB (96.90%) in isopropanol.

Nanocellulose composites with enhanced mechanical and flame-retardant properties based on grafting of inorganic organic/multilayer core-shell matter — MSNs-TMSB/DA/TOCNF

Qingshuo Wang,Lanfeng Hui,Yiyi Zhang,Wenwen Guo,Chengfeng Zhu,Mingyue Zhao,Qian Yang,Yu Qu,Zitong Zhang

Flame retardant materials are essential for safety, yet their development is often hindered by trade-offs between efficiency, aging resistance, and mechanical properties. Traditional organic flame retardants are inefficient and degrade over time, while inorganic alternatives, despite their effectiveness, are difficult to integrate into composites. Here we showed the synthesis of a novel inorganic silica-based flame retardant, PDA@MSNs-TMSB, which chemically modified nanocellulose fibrils, enhancing flame retardancy, aging resistance, and toughness without compromising integrity. The results showed that, compared to pure TOCNF, the modified nanocellulose materials (TOCNF-PDA@MSNs-TMSB) exhibited a higher limiting oxygen index (46.5 %), reaching the UL-94 V-0 level (GB) rating with self-extinguishing behavior and no flame propagation. In contrast, pure TOCNF had a limiting oxygen index of only 22 % and burned rapidly upon ignition which did not achieve the UL-94 V-0 level rating. The toughness of the modified TOCNF-PDA@MSNs-TMSB was superior to that of pure TOCNF, representing a 37.5 % increase. Combining powerful tenacity, high flame retardancy, and better aging resistance, the flame retardant nanocellulose material from renewable resource shows great potential for flame retardant applications and emits no toxic byproducts post-combustion.

A Hierarchical Porous Bowl-like PLA@MSNs-COOH Composite for pH-Dominated Long-Term Controlled Release of Doxorubicin and Integrated Nanoparticle for Potential Second Treatment

Jianming Pan,Runrun Wu,Xiaohui Dai,Yijie Yin,Guoqing Pan,Minjia Meng,Weidong Shi,Yongsheng Yan

We chemically integrated mesoporous silica nanoparticles (MSNs) and macroporous bowl-like polylactic acid (pBPLA) matrix, for noninvasive electrostatic loading and long-term controlled doxorubicin (DOX) release, to prepare a hierarchical porous bowl-like pBPLA@MSNs-COOH composite with a nonspherical and hierarchical porous structure. Strong electrostatic interaction with DOX rendered excellent encapsulation efficiency (up to 90.14%) to the composite. DOX release showed pH-dominated drug release kinetics; thus, maintaining a weak acidic pH (e.g., 5.0) triggered sustained release, suggesting the composite’s great potential for long-term therapeutic approaches. In-vitro cell viability assays further confirmed that the composite was biocompatible and that the loaded drugs were pharmacologically active, exhibiting dosage-dependent cytotoxicity. Additionally, a wound-healing assay revealed the composite’s intrinsic ability to inhibit cell migration. Moreover, pH- and time-dependent leaching of the integrated MSNs due to pBPLA matrix degradation allow us to infer that the leached (and drug loaded) MSNs may be engulfed by cancer cells contributing to a second wave of DOX-mediated cytotoxicity following pH-triggered DOX release.

A simple mesoporous silica nanoparticle-based fluorescence aptasensor for the detection of zearalenone in grain and cereal products

Tan Hongxia,Guo Ting,Zhou Hongyuan,Dai Hongjie,Yu Yong,Zhu Hankun,Wang Hongxia,Fu Yu,Zhang Yuhao,Ma Liang

Zearalenone (ZEN) is a type of estrogenic mycotoxin commonly occurring in cereals. The aim of this study was to design a simple, rapid, inexpensive and ultrasensitive fluorescence assay for the determination of ZEN. Here, amino-modified mesoporous silica nanoparticles (MSNs-NH 2 ) were synthesized to be the positive charge-rich reactor. A 6-carboxy-fluorescein-labeled aptamer (aptamer-FAM) was designed as the signal probe, ZEN-capture probe and negative charge reactor. In the absence of ZEN, the negatively charged aptamer-FAM combined with the positively charged MSNs-NH 2 in an electrostatic manner. In the presence of ZEN, the fluorescence intensity in the supernatant increased significantly because the aptamer-FAM could bind to ZEN instead of MSNs-NH 2 . Under the optimal experimental conditions, this assay exhibited excellent specificity, repeatability and a wide linearity range of 0.005–150 ng/mL, with a detection limit of 0.012 ng/mL. Additionally, it showed high recovery (83.3–101.5%) for the spiked samples. There was no statistically significant difference in the ZEN concentrations detected by the proposed assay and HPLC in naturally contaminated samples. Overall, this design provides a new strategy for the rapid, inexpensive and sensitive detection of ZEN, and it could be applied to develop fluorometric assays for different targets by the selection of appropriate aptamers. NAC-loaded electrospun scaffolding system with dual compartments for the osteogenesis of rBMSCs in vitro

Yuanjing Zhu,Fangfang Song,Yanyun Ju,Liyuan Huang,Lu Zhang,Chuliang Tang,Hongye Yang,Cui Huang

Purpose In this study, we aimed to develop a unique N-acetyl cysteine (NAC)-loaded polylactic-co-glycolic acid (PLGA) electrospun system with separate compartments for the promotion of osteogenesis.Materials and methods We first prepared solutions of NAC-loaded mesoporous silica nanoparticles (MSNs), PLGA, and NAC in N, N-dimethylformamide and tetrahydrofuran for the construction of the electrospun system. We then fed solutions to a specific injector for electrospinning. The physical and chemical properties of the scaffold were characterized through scanning electron microscopy, transmission electron microscopy, and Fourier transform infrared spectroscopy. The release of NAC and Si from different PLGA scaffolds was estimated. The cell viability, cell growth, and osteogenic potential of rat bone marrow-derived stroma cell (rBMSCs) on different PLGA scaffolds were evaluated through MTT assay, live/dead staining, phalloidin staining, and Alizarin red staining. The expression levels of osteogenic-related markers were analyzed through real-time PCR (qRT-PCR).Results NAC was successfully loaded into MSNs. The addition of MSNs and NAC decreased the diameters of the electrospun fibers, increased the hydrophilicity and mechanical property of the PLGA scaffold. The release kinetic curve indicated that NAC was released from (PLGA + NAC)/(NAC@MSN) in a biphasic pattern, that featured an initial burst release stage and a later sustained release stage. This release pattern of NAC encapsulated on the (PLGA + NAC)/(NAC@MSN) scaffolds enabled to prolong the high concentrations of release of NAC, thus drastically affecting the osteogenic differentiation of rBMSCs.Conclusion A PLGA electrospun scaffold was developed, and MSNs were used as separate nanocarriers for recharging NAC concentration, demonstrating the promising use of (PLGA + NAC)/(NAC@MSN) for bone tissue engineering.

Characterization and evaluation of an oral vaccine via nano-carrier for surface immunogenic protein (Sip) delivery against Streptococcus agalactiae infection

Chunhua Zhu,Na Zhang,Dingding Jing,Xiaodong Liu,Zhanzhuang Zeng,Jingjing Wang,Fangnan Xiao,Hong Zhang,Hongshu Chi,Chunhe Wan,Pingdong Lin,Hui Gong,Yunkun Wu

Streptococcus agalactiae causes systemic disease in a variety of wild and farmed fish, resulting in high levels of morbidity and mortality, as well as serious economic losses to the Nile tilapia aquaculture industry. The development of economic and applicable oral vaccines is therefore urgently needed for the sustainable development of Nile tilapia aquaculture. In this study, mesoporous silica nanoparticles (MSNs) were fabricated using sol–gel synthesis technology, and the antigens of surface immunogenic protein (Sip) was loaded into MSNs to develop a nanovaccine MSNs-Sip@HP55. The results showed that the prepared nanovaccine exhibited pH-controlled release, which could survive in the simulated gastric environment (pH 1.5), and release antigens in the simulated intestinal environment at pH 7.4. The nanovaccine could induce innate and adaptive immune responses in Nile tilapia. When the challenge doses were 1.5 × 10 6 , 1.18 × 10 6 , and 0.88 × 10 6  CFU/mL, the relative protection rates in immunized Nile tilapia were 63.33 %, 64.23 %, and 76.31 %, respectively. Taken together, the nanovaccine exhibited a high antigen utilization rate and was easily administered orally via feeding, which could protect Nile tilapia against challenge with S. agalactiae in large-scale farms. Oral vaccine based on MSNs carriers is a potentially promising strategy for the development of fish vaccines.

Dendrimer-like mesoporous silica nanospheres with suitable surface functionality to combat the multidrug resistance

Weixia Xu,Xiaofeng Gao,Pengjin Ge,Fuquan Jiang,Xiaokun Zhang,Jingjing Xie

Multidrug resistance (MDR), as a major obstacle in cancer therapy, has resulted in over 90% of cancer chemotherapeutic failure. Mesoporous silica nanospheres (MSNs) have been demonstrated to be tuned with large pore sizes, mediating the MDR-reversal effects. However, the study that surface functionality of the large pore sized-MSNs affects the MDR-overcoming effects hasn’t been extensively studied. In this study, we developed a new dendrimer-like MSNs delivery system based on a rational synthesis strategy and further modified MSNs with various surface functionalities to evaluate their roles in overcoming cancer MDR. Our results showed that the small particle sized-MSNs could be fabricated with dendrimer-like internal structure, resulting in the large pore size of 9 nm. Surface functionality of MSNs, especially hydroxylation and carboxylation, largely improved the intra-nuclear delivery and therapeutic efficiency of DOX for MCF7/ADR cells, which was not up to inhibiting P-gp expression but significantly increasing the intracellular drug accumulation of over 90% even under the strong drug efflux. This study indicates that surface functionality design strategy may display the potential of the large pore sized-MSNs as the efficient chemotherapeutic carriers to combat MDR.

Effects of Skeleton Structure of Mesoporous Silica Nanoadjuvants on Cancer Immunotherapy

Jia Tan,Binbin Ding,Hao Chen,Qi Meng,Jing Li,Chunzheng Yang,Wenying Zhang,Xinyang Li,Di Han,Pan Zheng,Ping'an Ma,Jun Lin

Mesoporous silica nanoparticles (MSNs) have been widely praised as nanoadjuvants in vaccine/tumor immunotherapy thanks to their excellent biocompatibility, easy-to-modify surface, adjustable particle size, and remarkable immuno-enhancing activity. However, the application of MSNs is still greatly limited by some severe challenges including the unclear and complicated relationships of structure and immune effect. Herein, three commonly used MSNs with different skeletons including MSN with tetrasulfide bonds (TMSN), MSN containing ethoxy framework (EMSN), and pure −Si−O−Si− framework of MSN (MSN) are comprehensively compared to study the impact of chemical construction on immune effect. The results fully demonstrate that the three MSNs have great promise in improving cellular immunity for tumor immunotherapy. Moreover, the TMSN performs better than the other two MSNs in antigen loading, cellular uptake, reactive oxygen species (ROS) generation, lymph node targeting, immune activation, and therapeutic efficiency. The findings provide a new paradigm for revealing the structure-function relationship of mesoporous silica nanoadjuvants, paving the way for their future clinical application.

NIR-responsive magnesium phosphate cement loaded with simvastatin-nanoparticles with biocompatibility and osteogenesis ability

Bin Wang,Yanbin Zhao,Yangyang Li,Junyan Yao,Shunjie Wu,Guoping Miu,Chenglin Chu

The insufficient osteogenesis of magnesium phosphate cement (MPC) limits its biomedical application. It is of great significance to develop a bioactive MPC with osteogenic performance. In this study, an injectable MPC was reinforced by the incorporation of a near infrared (NIR)-responsive nanocontainer, which was based on simvastatin (SIM)-loaded mesoporous silica nanoparticles (MSNs) modified with a polydopamine (PDA) bilayer, named SMP. In addition, chitosan (CHI) was introduced into MPC (K-struvite) to enhance its mechanical properties and cytocompatibility. The results showed that nanocontainer-incorporated MPC possessed a prolonged setting time, almost neutral pH, excellent injectability, and enhanced compressive strength. Immersion tests indicated that SMP-CHI MPC could suppress rapid degradation. Based on its physicochemical features, the SMP-CHI MPC had good biocompatibility and osteogenesis properties, as shown via in vitro and in vivo experiments. These findings can provide a simple way to produce a multifunctional MPC with improved osteogenesis for further orthopedic applications.

Enhanced Gut-to-Liver Oral Drug Delivery via Ligand-Modified Nanoparticles by Attenuating Protein Corona Adsorption

Jie Wang,Zilong Zhang,Zhuan Zhang,Zhiwen Zou,Yan Zhuo,Chang Liu,Di Nie,Yong Gan,Miaorong Yu

The development of effective oral drug delivery systems for targeted gut-to-liver transport remains a significant challenge due to the multiple biological barriers including the harsh gastrointestinal tract (GIT) environment and the complex protein corona (PC) formation. In this study, we developed ligand-modified nanoparticles (NPs) that enable gut-to-liver drug delivery by crossing the GIT and attenuating PC formation. Specifically, mesoporous silica nanoparticles (MSNs) were functionalized with peptides targeting the neonatal Fc receptor (FcRn), capitalizing on FcRn expression in the small intestine and liver for targeted drug delivery. We showed that MSNs decorated with a small cyclic FcRn binding peptide (MSNs-FcBP) obtained enhanced diffusion in intestinal mucus and superior transportation across the intestine compared to unmodified MSNs and MSNs decorated with a large IgG Fc fragment (MSNs-Fc), which correlated with diminished protein adsorption and weaker interaction with mucin. After entering the blood circulation, reduced serum PC formation by MSNs-FcBP reduces the proteolytic and phagocytic propensity of the reticuloendothelial system, ultimately ameliorating accumulation in hepatocytes. Pharmacokinetic and pharmacodynamic studies in diabetic mice revealed that MSNs-FcBP effectively transported the therapeutic agent exenatide across the intestinal epithelium, leading to a significant hypoglycemic response and improved glucose tolerance. This study underscores the critical role of ligand selection in limiting protein corona formation, thereby significantly enhancing gut-to-liver drug delivery by increasing mucus permeation and minimizing serum–protein interactions. The effective delivery of exenatide in diabetic mice illustrates the potential of this strategy to optimize oral drug bioavailability and therapeutic efficacy.

Engineered Biomimetic Nanoparticles-Mediated Targeting Delivery of Allicin Against Myocardial Ischemia-Reperfusion Injury by Inhibiting Ferroptosis

Minghui Li,Jiabi Wu,Tao Yang,Yuhang Zhao,Ping Ren,Lingling Chang,Pilong Shi,Jing Yang,Yuhang Liu,Xiaolei Li,Peng Wang,Yonggang Cao

Background Cardiac microvascular damage is substantially related with the onset of myocardial ischaemia-reperfusion (IR) injury. Reportedly, allicin (AL) effectively protects the cardiac microvascular system from IR injury. However, the unsatisfactory therapeutic efficacy of current drugs and insufficient drug delivery to the damaged heart are major concerns. Here, inspired by the natural interaction between neutrophils and inflamed cardiac microvascular endothelial cells (CMECs), a neutrophil membrane-camouflaged nanoparticle for non-invasive active-targeting therapy for IR injury by improving drug delivery to the injured heart is constructed. Methods In this study, we engineered mesoporous silica nanoparticles (MSNs) coated with a neutrophil membrane to act as a drug delivery system, encapsulating AL. The potential of the nanoparticles (named AL@MSNs@NM) for specific targeting of infarcted myocardium was assessed using small animal vivo imaging system. The cardiac function of AL@MSNs@NM after treatment was evaluated by Animal Ultrasound Imaging system, HE staining, and Laser Speckle Imaging System. The therapeutic mechanism was analyzed by ELISA kits, immunofluorescence, and PCR.Results We discovered that AL@MSNs@NM significantly improves cardiac function index, reduced infarct size and fibrosis, increased vascular perfusion in ischemic areas, and also promoted the function of CMECs, including migration, tube formation, shear stress adaptation, and nitric oxide production. Further research revealed that AL@MSNs@NM have cardio-protective functions in IR rats by inhibiting CMEC ferroptosis and increasing platelet endothelial cell adhesion molecule-1 (PECAM-1) expression. Conclusion Our results indicated that AL@MSNs@NM significantly reversed CMEC ferroptosis and increased PECAM-1 expression, enhanced cardiac function, and reduced myocardial infarction size. Therefore, this strategy demonstrates that engineered biomimetic nanotechnology effectively delivers AL for targeted therapy of myocardial infarction.

Sub-femtomolar vertical graphene field effect immunosensor for detection of lung tumor markers

Ruifang Liu,Hang Song,Hao Wu,Zhihao Yang,Surong Fang,Jing Zhu,Long Ba

Lung cancer is the main cancer that endangers human life worldwide, with the highest mortality rate. The detection of lung tumor markers is of great significance for the early diagnosis and subsequent treatment of lung cancer. In this study, a vertical graphene field effect transistor (VGFET) immunosensor based on graphene/C 60 heterojunction was created to offer quantitative detections for the lung tumor markers carcinoembryonic antigen (CEA), cytokeratin 19 fragment (Cyfra21-1), and neuron-specific enolase (NSE). The experimental results showed that the sensitive range for standard antigen is between 1 pg/ml to 100 ng/ml, with a limit of detection (LOD) of 5.6 amol/ml for CEA, 33.3 amol/ml for Cyfra 21–1 and 12.8 amol/ml for NSE (1 pg/ml for all). The detection accuracy for these tumor markers was compared with the clinically used method for clinical patients on serum samples. Results are highly consistent with clinically used immunoassay in its efficient diagnosis concentration range. Subsequently, the mesoporous silica nanospheres (MSNs) with an average size of 90 nm were surface modified with glutaraldehyde, and a second antibody was assembled on MSNs, which fixes nanospheres on the antigen and amplified the field effect. The LODs for three markers are 100 fg/ml (0.56 amol/ml for CEA) under optimal circumstances of detection. This result indicates that specific binding to MSNs enhances local field effects and can achieve higher sensing efficiency for tumor marker detection at extremely low concentrations, providing effective assistance for the early diagnosis of lung cancer.
