Deep eutectic solvent-based ultrasonic-assisted extraction of phenolic compounds from Moringa oleifera L. leaves: Optimization, comparison and antioxidant activity

Lingfeng Wu,Lei Li,Shengjie Chen,Lu Wang,Xue Lin

In this study, an ultrasonic-assisted extraction (UAE) procedure with selected deep eutectic solvents (DES) as solvent was first designed to simultaneously optimize the total phenolic/flavonoid content (TPC/TFC) and antioxidant activities of Moringa oleifera L. leaves (MOLs) by using response surface methodology (RSM). The key factors for RSM were selected based on the design of the experimental results along with a three-factors-five-level, central composite design (CCD), including 20 experimental runs. The analysis of variance (ANOVA) results revealed that the water content in DES had a significant influence on all responses, while the ultrasonic time and the ratio of liquid to solid had no statistically significant effects on the total phenolic content. The optimal conditions of the combination of TPC/TFC and antioxidant activities were obtained as follows: 37% water content in DES, 144 W ultrasonic power, and 40 °C ultrasonic temperature. The measured parameters corresponded with the predicted results. Moreover, a comparative study confirmed that the optimized DES-based UAE yielded further higher TPC, TFC, and antioxidant activities than other extraction methods. The results of HPLC analysis in optimized conditions verified that the MOLs extracts with DES-based UAE included 14 phenolic compounds with high concentrations of vicenin-2 (17.6 mg/g) and orientin (23.6 mg/g). The present study supplied a green and high-efficient method for extracting high levels of anti-oxidative phenolic compounds from MOLs.

Amelioration of hypoglycemic peptide MoHpP-2 from Moringa oleifera seeds in C2C12 / IR cells and type 2 diabetes mellitus mice

Yaozhu Fan,Xuefeng Wang,Yiyan Li,Lingfei Li,Yang Tian

The hypoglycemic peptide MoHpP-2 from Moringa oleifera seeds exhibits excellent α-glucosidase inhibitory activity, but its amelioration in insulin resistant (IR) cells and type 2 diabetes mellitus (T2DM) mice is still unclear. Therefore, the present study aimed to investigate the hypoglycemic effect of MoHpP-2 on C2C12/IR cells, and gut microbiota and metabolite changes of T2DM mice using 16S rRNA high-throughput sequencing and metabolomics. MoHpP-2 within the concentration range could effectively enhance the glucose consumption levels and antioxidant capacity in C2C12/IR cells. MoHpP-2 intervention improves the blood sugar levels, liver function, and organ indices, reverses oxidative damage, and enhances the antioxidant capacity of T2DM mice. Furthermore, it increases the diversity of cecum microorganisms, decreases the ratio of Firmicutes to Bacteroides and some pathogenic bacteria , and reshapes the gut microbial composition of T2DM mice. Moreover, the metabolomic analysis results showed that MoHpP-2 intervention might promote unsaturated fatty acid biosynthesis, inhibit bile acid formation, and regulate linoleic acid metabolism and amino acid metabolism related to oxidative stress, thereby improving insulin resistance and sensitivity in T2DM mice. Overall, the experimental results provide insights into the hypoglycemic effect of MoHpP-2 in vitro and in vivo and its application in hypoglycemic functional food products.

In vitro antioxidant activity of Moringa oleifera lam. Leaf after Monascus purpureus fermentation and chemical component changes by untargeted metabolomics

Zilin Wang,Ying Feng,Yang Tian,Min Yang,Yanju He,Yujia He,Yue Wang,Chongying Shi,Liang Tao

This study aims to explore the enhancement effect of semi-solid fermentation of Monascus purpureus on the antioxidant properties of Moringa oleifera Lam. leaf (MOLL), as well as to analyze the potential antioxidant substances and their production mechanisms. In this study, the antioxidant activity of MOLL fermented with Monascus purpureus was evaluated in vitro . To gain further insight, we employed untargeted metabolomics to analyze the differentially abundant metabolites, and 30 key differentially expressed metabolites were identified. Notably, 15 of these metabolites were positively correlated with free radical scavenging activity. By utilizing a combination of KEGG topological and enrichment analyses, tryptophan metabolism was identified as a crucial pathway. Interestingly, Monascus purpureus fermentation significantly enhanced the conversion of tryptophan into metabolites with antioxidant properties, such as melatonin, kynurenine, and 3-hydroxyanthranilic acid, suggesting that fermentation with Monascus purpureus is a promising approach for enhancing the antioxidant properties of MOLL.

Effects of UV/H2O2 degradation on Moringa oleifera Lam. leaves polysaccharides: Composition, in vitro fermentation and prebiotic properties on gut microorganisms

Min Yang,Liang Tao,Zi-Lin Wang,Ling-Fei Li,Cun-Chao Zhao,Chong-Ying Shi,Jun Sheng,Yang Tian

Moringa oleifera Lam. leaves are a new raw food material rich in polysaccharides. These polysaccharides exhibit various biological properties, including antioxidant, hypoglycemic and immunoregulatory effects. However, the use of Moringa oleifera Lam. leaves polysaccharides (MOLP) may be limited by their large molecular weight ( M W ) and presence of numerous impurities, such as pigments. Research has indicated that degraded polysaccharides usually exhibit high biological activity because of changes in physical structure and chemical properties. In this study, we focused on the extraction of a degraded-modified fraction from MOLP using the Ultraviolet/ Hydrogen peroxide (UV/H 2 O 2 ) method. Specifically, the physicochemical properties and glycosidic bond composition of a particular fraction (UV/H 2 O 2 degraded Moringa oleifera Lam. leaves polysaccharides in 3 h called DMOLP-3) were investigated. In addition, in vitro simulated digestion experiments showed that DMOLP-3 was only partially degraded during gastrointestinal digestion, indicating that DMOLP-3 can be utilised by gut microorganisms. Furthermore, the prebiotic properties of MOLP and DMOLP-3 was studied using an in vitro faecal fermentation model. The results indicated that compared with MOLP, DMOLP-3 led to a decrease in both the colour and M W of the polysaccharides. In addition, this model exhibited enhanced solubility and antioxidant capabilities while also influencing the surface morphology. Moreover, DMOLP-3 can facilitate the proliferation of advantageous microorganisms and enhance the synthesis of short-chain fatty acids (SCFAs). These results provide valuable insights into the utilization of bioactive components in Moringa oleifera Lam. leaves for the intestinal health.

Exploring the protection on islet β-cells and hypoglycemic effect of Moringa oleifera stem based on network pharmacology and experimental validation

Xilin Jiang,Jian Tang,Zhiqiang Gao,Caixia Chen,Wenjun Li,Lu Peng,Hongwei Li,Hongying Zhao,Yun Zhao

Moringa oleifera Lam. ( M. oleifera ) has been used as a remedy for diabetes clinically. To discover the underlying mechanism, we investigated its hypoglycemic compounds and shed light on its regulatory targets and pathways using network pharmacology. A total of 13 active compounds of stem were shot, and they may relieve diabetes through 122 core intersecting targets, mainly regulating the biological processes of inflammatory response and apoptosis. Molecular docking showed binding advantages of the active compounds with proteins in apoptosis process. In experiments conducted on MIN6 cells, an islet β-cell line, the extract of M. oleifera stem was validated to protect cells from apoptosis through reducing cellular ROS and endoplasmic reticulum stress. And consistently with its protective effect on islet β-cells, M. oleifera stem alleviated hyperglycemia in Type 2 diabetes mellitus (T2DM) mice. These results offer novel evidence for the development of functional foods to treat T2DM with M. oleifera stem.

Exploration of anti-osteoporotic peptides from Moringa oleifera leaf proteins by network pharmacology, molecular docking, molecular dynamics and cellular assay analyses

Deying Men,Jiahe Dai,Yuxue Yuan,Haifen Jiang,Xinyu Wang,Yifan Wang,Liang Tao,Jun Sheng,Yang Tian

Moringa oleifera   leaf peptide, which is a plant-derived bioactive peptide, exhibits several advantages, including safety, high efficiency and nontoxic side effects. The goal of this study was to identify representative antiosteoporotic peptides from   Moringa oleifera   leaf proteins and determine their regulatory mechanisms using network pharmacology, molecular docking and molecular dynamics. The following core targets of Moringa oleifera leaf antiosteoporotic peptides were screened by network pharmacology: SRC, MAPK1, JUN and STAT3. Through molecular docking, DPYLGK was identified as a representative active peptide from the Moringa oleifera leaf protein, and SRC was determined to be a potential primary target. Molecular dynamics revealed the intermolecular mechanism of DPYLGK and SRC. Cellular assays showed that DPYLGK promotes bone formation and inhibits bone resorption. These findings demonstrated that DPYLGK is an anti-osteoporotic peptide with dual regulatory effects and will aid in the development of functional foods to prevent osteoporosis.

Preconcentration and Determination of Cadmium, Lead, and Cobalt in Moringa Oleifera (Moringaceae) Using Magnetic Solid-Phase Extraction and Flame Atomic Absorption Spectrometry

Shuangqiao Ju,Mousheng Liu,Yaling Yang

Magnetic solid-phase extraction was used for the preconcentration of cadmium, lead, and cobalt in Moringa Oleifera with determination by flame atomic absorption spectrometry. Fe3O4 nanoparticles were modified by oleic acid to form the sorbent for magnetic solid-phase extraction. The modified sorbent was characterized by transmission electron microscopy, infrared spectroscopy, and X-ray diffraction. The preconcentration was optimized for the mass of nanoparticles, pH, adsorption time, the presence of coexisting ions, solution volume, eluent, and reuse. The optimized limits of detection were 0.09, 0.26, and 0.54 µg/L for Cd(II), Pb(II), and Co(II) with relative standard deviations below 4.3%. The method was employed for the analysis of Moringa oleifera and recoveries between 90.0 and 102.0% were obtained.

Unveiling the formation mechanism of Moringa oleifera leaf flavonoids aglycones during Monascus anka fermentation by using integrated widely targeted metabolomics and proteomics analysis

Youjing Wei,Ganqi Zhang,Tao Fei,Xue Lin,Lu Wang

This study revealed the formation mechanism of flavonoid aglycones in Moringa oleifera leaf (MOL) during Monascus anka fermentation via widely targeted metabolomics and data-independent acquisition-based proteomics. Bioactivity assays demonstrated that M. anka fermentation significantly enhanced MOL's bioactivity; total flavonoid and phenolic content, ABTS and DPPH radical scavenging capacities were increased by 226.8%, 45.59%, 1.28-, and 1.25-fold compared with the unfermented control, respectively. Widely targeted metabolomics identified 172 phenolic metabolites, of which flavonoids accounted for 70.9%. Notably, flavonoid aglycones (quercetin and kaempferol) were significantly accumulated during fermentation, being 5.05-, 2.65-, and 5.54-fold higher than the unfermented control. Proteomics analysis identified key carbohydrate-active enzymes associated with cell wall degradation. Importantly, β -glucosidases (GH1, GH3) and α -L-rhamnosidases (GH78) hydrolyzed flavonoid glycosidic linkages, facilitating free aglycone accumulation. Collectively, these findings indicated that M. anka -secreted enzymes exerted catalytic roles in cell wall degradation and flavonoid deglycosylation, providing a theoretical basis for targeted enhancement of MOL bioactivity.

Efficient polyphenol extraction from Moringa oleifera Lam. leaves using natural deep eutectic solvents: COSMO-RS screening, ANN-GA optimization and antioxidant activity evaluation

Weilong Peng,Lijun Wang,Xiaoguang Wang,Yaya Wang,Weimei Wang,Junjie Huang,Ruigang Zhou,Chaoyang Chen,Ruonan Bo,Mingjiang Liu,Jingui Li

A novel method for extracting polyphenols from Moringa oleifera Lam. ( M. oleifera ) using natural deep eutectic solvents (NADES) has been developed, with extraction conditions optimized via an artificial neural network-genetic algorithm (ANN-GA) to maximize efficiency. Traditional extraction methods for polyphenols from M. oleifera leaves, such as ethanol extraction, are often associated with low efficiency, environmental concerns, and high operational costs. Furthermore, the empirical selection of solvents and extraction conditions is time-consuming and lacks precision. In this study, 28 NADES were employed to predict the solubility of polyphenol molecules through a conductor-like screening model for realistic solvents (COSMO-RS) model. Based on the results, eight highly effective NADES combinations were selected and experimentally verified. The betaine-maleic acid (Bet-MA) combination with the ultrasonic-assisted extraction showed the highest extraction rate of M. oleifera leaves polyphenol (MOLP), which was 1.56 times that of ultrasonic-assisted ethanol solvent. Simplex centroid mixture design (SCMD) and two-level factor experiments were conducted to optimize the ratio of betaine, maleic acid, and water and identify the most influential extraction factors on MOLP yield. The ANN-GA model was further applied to refine the extraction conditions and analyze their interactions on MOLP content. Additionally, antioxidant assays demonstrated that MOLP extracted with the Bet-MA solvent displayed significant antioxidant activity. In conclusion, this study demonstrates that COSMO-RS and ANN-GA may serve as powerful tools for solvent selection and extraction process optimization, offering a green and efficient approach for polyphenol extraction from M. oleifera leaves. The findings highlight the potential of NADES-based extraction methods for broader applications in the extraction of various bioactive substances, contributing to the development of sustainable and environmentally friendly extraction technologies.

In vitro/in vivo hepatoprotective properties of 1-O-(4-hydroxymethylphenyl)-α-L-rhamnopyranoside from Moringa oleifera seeds against carbon tetrachloride-induced hepatic injury

Congyong Sun,Wenjing Li,Yingkun Liu,Wenwen Deng,Michael Adu-Frimpong,Huiyun Zhang,Qilong Wang,Jiangnan Yu,Ximing Xu

1-O-(4-hydroxymethylphenyl)-α-L-rhamnopyranoside (MPG) is a phenolic glycoside that exists in Moringa oleifera seeds with various health benefits, whereas its hepatoprotective effect is lacking clarification. Herein, MPG was isolated from Moringa oleifera seeds, and its hepatoprotection against CCl 4 -induced hepatotoxicity in L02 cells and ICR mice was investigated. Toxicity studies showed that MPG did not induce significant changes in organ coefficients and histological analysis, as well as exhibited no cytotoxicity. In vitro studies indicated that MPG substantially increased cell viability and intracellular SOD activities, and significantly inhibited LDH leakage in CCl 4 -treated cells. In vivo studies demonstrated that MPG significantly alleviated CCl 4 -induced hepatotoxicity in mice, as indicated by diagnostic indicators of hepatic injury, as well as the histopathological analysis. Moreover, MPG reduced the lipid peroxidation levels and regulated the inflammatory cytokines. Notably, MPG substantially suppressed the significant elevation of ROS production in hepatocytes of mice intoxicated with CCl 4 . Moreover, TUNEL assay demonstrated that MPG obviously inhibited hepatic apoptosis induced by CCl 4 . Altogether, these results suggested that MPG has excellent liver-protecting effects against hepatocytotoxicity induced by CCl 4 in mice and L02 cells, which can be further developed as a valuable functional food additive or drug for the treatment of hepatic injury.

Temperature-responsive biphasic deep eutectic solvent system enables integrated extraction and self-separation of polyphenols from Moringa oleifera Lam. leaves: a machine learning-optimized biorefinery approach

Weilong Peng,Lijun Wang,Lichao Song,Yan Liu,Yu Ren,Yaya Wang,Weimei Wang,Jun Chen,Ruonan Bo,Mingjiang Liu,Jingui Li

A novel amine-free temperature-responsive biphasic DES (BLevA-OA) was developed. • Reversible phase transition integrated polyphenol extraction and separation. • GSWOA-SVR machine learning accurately optimized the extraction process. • MD simulations revealed molecular mechanisms of extraction and phase switching. • TRDES system outperformed traditional ethanol in extraction efficiency. The sustainable recovery of bioactive phytochemicals is frequently hindered by the toxicity and separation bottlenecks of conventional volatile organic solvents. Herein, a novel amine-free temperature-responsive biphasic deep eutectic solvent (TRDES) system, composed of betaine, levulinic acid, and octanoic acid (BLevA-OA), was constructed for the integrated extraction and separation of polyphenols from Moringa oleifera Lam. ( M. oleifera ) leaves. This system exhibits reversible thermomorphic behavior, transitioning from a homogeneous single phase at 60 °C to maximize mass transfer, to a biphasic state at 25 °C for spontaneous phase-selective separation. To address the nonlinear complexity of the extraction process, a hybrid Gaussian Whale Optimization Algorithm-Support Vector Regression (GSWOA-SVR) model was developed, which precisely optimized the parameters to achieve a maximum total polyphenol content of 39.8 mg GAE/g DW, which is 1.7 times that of conventional ethanol extraction. Furthermore, multiscale computational analyses combining COSMO-RS and Molecular Dynamics (MD) simulations elucidated the microscopic mechanism: the superior extraction efficiency stems from a cooperative network of specific hydrogen bonding and hydrophobic interactions, while the temperature-triggered phase transition is driven by the kinetic modulation of component miscibility and diffusion. This work establishes a sustainable, closed loop strategy for the high efficiency recovery of polyphenols from M. oleifera leaves, offering a theoretical and practical foundation for the industrial application of smart green solvents in the circular bio-economy. Effects of impregnate temperature on extraction of caffeoylquinic acid derivatives from Moringa oleifera leaves and evaluation of inhibitory activity on digestive enzyme, antioxidant, anti-proliferative and antibacterial activities of the extract

Yahya Saud Hamed,Mohamed Abdin,Guijie Chen,Hafiz Muhammad Saleem Akhtar,Xiaoxiong Zeng

Summary In an ultrasonic bath and for 30 min in the presence of water as extraction solvent in a ratio of 1:10 (g/mL), effect of extraction temperature on extraction of caffeoylquinic acids (CQAs) from Moringa oleifera leaves (MOL) was investigated. Gradually increasing in extraction temperature caused a growing in mono-CQA and declining in 3,5-diCQA. The extract obtained at 80 °C showed the highest antioxidant capacity, and IC 50 values for scavenging activities on DPPH radicals and ABTS radicals were 56.9 and 53.6 µg/mL, respectively. Compared with α-glucosidase and α-amylase, the extract showed the highest inhibition on pancreatic lipase (IC 50 of 0.073 mg/mL). Increasing the extraction temperature improved cytotoxicity of the extract on cancer cell lines of HeLa and HepG2. Compared with gram-negative Escherichia coli and Salmonella typhimurium , all the extracts showed higher inhibition on the growth of gram-positive Bacillus cereus and Staphylococcus aureus . Our results suggested that use of higher extraction temperature (80 °C) improved the biological activity of MOL extract.
