This study aimed to investigate the effect of naringenin (Nar) on cadmium (Cd)-induced testicular toxicity. Twenty-four male Sprague–Dawley (SD) rats aged 5 weeks were used. Rats were administered with 0.9% NaCl (control group), CdCl 2 (2 mg/kg b.w. intraperitoneally), Nar (50 mg/kg b.w, orally), and CdCl 2 + Nar (2 mg/kg b.w intraperitoneally and 50 mg/kg b.w. orally, respectively) for 4 weeks. Results showed that body weight, relative testis weights , and sperm quality decreased in the Cd-treated group, and Cd accumulated in serum and testes. Pathological examination showed that Cd can cause testicular damage. Cd decreased the serum concentrations of gonadotropin-releasing hormone (GnRH), follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone. It also decreased the activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx). Moreover, exposure to Cd resulted in decreased content of reduced glutathione (GSH) and total antioxidant capacity (T-AOC) concentrations, as well as increased malondialdehyde (MDA) and hydrogen peroxide (H 2 O 2 ) contents. Cd also provoked testis autophagy by upregulating the expression of the autophagy-related proteins P62 and LC3 II. However, the combined administration of Nar and Cd significantly attenuated the Cd-induced negative effects by increasing the body weight, relative testis weights, and sperm quality and by decreasing testicular damage. Simultaneous supplementation of Nar and Cd markedly restored the decreased levels of GnRH, FSH, LH, testosterone, GSH, and T-AOC and the activities of SOD, CAT, and GPx caused by Cd treatment. Nar further suppressed MDA and H 2 O 2 production and protected the testes from Cd-induced autophagy by downregulating P62 and LC3 II expression. Therefore, Nar protected the testes from Cd-induced toxicity.

Naringenin, a flavonoid compound found in pummelo, is a key biological active compound in some traditional Chinese medicines, including Citri reticulatae pericarpium , Citri reticulatae pericarpium viride , Aurantii fructus immaturus , and Aurantii fructus . These Chinese medicinal preparations are the peels or immature fruits of certain citrus species. Aiming at detecting naringenin in complex matrices such as pummelo and traditional Chinese medicines, we put forward a sensitive and practical indirect competitive enzyme-linked immunosorbent assay (icELISA) based on anti-naringenin monoclonal antibodies (anti-Nar-mAbs). The median inhibitory concentration (IC 50 ) was 4.43 ng/mL, and the working range was 1.15–15.81 ng/mL. The findings of the icELISA for the analysis of naringenin in pummelo and herb samples had a good correlation with the ultra performance liquid chromatography (UPLC) methodology and showed good accuracy and reproducibility. These data demonstrated that the developed icELISA is reliable, accurate, and suitable for detecting naringenin in pummelo and traditional Chinese medicines.

Cocrystallization between a BCS II class drug, carbamazepine , and polyphenolic compounds such as naringenin and naringin , is performed in the present study. New carbamazepine-naringenin (CBZ-NRG) cocrystal with 1: 1 molar ratio was obtained and characterized by using the methods of X-ray diffraction, nuclear magnetic resonance (solid state 13 C-NMRand liquid 1 H NMR), thermal and spectral analyses (FT-IR and solid state fluorescence). CBZ-NRG crystallizes in the P 2 1 /c space group, which belongs to monoclinic system. The molecular interactions were elaborated in the packing diagrams and confirmed by solid state 13 C NMR and FT-IR methods. It is scarce that CBZ-NRG cocrystal presents higher endotherm ( T onset = 272.7 °C) than both starting materials ( T onset = 188.6, 248.6 °C) using DSC analysis. The fluorescence values of CBZ-NRG cocrystal (179 A.U.) decreases significantly in comparison with CBZ (6121 A.U.). The in vitro/vivo assessment of NRG effect on CBZ was conducted in the present study. Compared with CBZ (97.2 ± 1.5 μg/mL) in water the solubility of CBZ-NRG cocrystal decreases to 7.2 ± 0.6 μg/mL, the same trends in pH = 1.2 and 6.8 buffer solution. The IDR values of CBZ and CBZ-NRG cocrystal are 0.042 and 0.016 mg/cm 2 /min respectively. Interestingly, CBZ has one absorption peak while the CBZ-NRG cocrystal has double absorption peaks in plasma concentration-time curves. The peak concentration ( C max = 491.3 ± 97.6 ng/mL) of CBZ-NRG cocrystal decreases with longer elimination half life ( t 1/2 = 8.5 ± 1.0 h) in comparison with CBZ ( C max = 5258.1 ± 904.2 ng/mL, t 1/2 = 0.7 ± 0.2 h). The obtained results suggest that CBZ-NRG cocrystal presents different in vitro/vivo performance in comparison with CBZ.

Thermophysical properties including densities and viscosities of naringenin in aqueous solutions of ethanol/1-propanol with alcohol concentrations of (24.00 to 40.00) mol·kg–1 have been measured as a function of concentration of naringenin at temperatures from (293.15 to 323.15) K and atmospheric pressure. Also, refractive indices for studied systems have been determined at 298.15 K. The measured data were used to calculate the apparent molar volume (Vϕ), standard partial molar volume (Vϕ0), partial molar volume (V̅), viscosity B-coefficients, and molar refractions (Rm). The viscosity and refractive index data have been analyzed by Jones–Dole and Lorentz–Lorenz equations, respectively. The trends of variation of experimental and calculated parameters have been discussed according to the interactions between solvents and solute. The obtained results imply that naringenin acts as a structure maker in the studied system.

Taking into consideration the importance of biofilms in food deterioration and the potential risks of antiseptic compounds, antimicrobial agents that naturally occurring are a more acceptable choice for preventing biofilm formation and in attempts to improve antibacterial effects and efficacy. Citrus flavonoids possess a variety of biological activities, including antimicrobial properties. Therefore, the anti-biofilm formation properties of the citrus flavonoid naringenin on theStaphylococcus aureusATCC 6538 (S. aureus) were investigated using subminimum inhibitory concentrations (sub-MICs) of 5~60 mg/L. The results were confirmed using laser and scanning electron microscopy techniques, which revealed that the thick coating ofS. aureusbiofilms became thinner and finally separated into individual colonies when exposed to naringenin. The decreased biofilm formation ofS. aureuscells may be due to a decrease in cell surface hydrophobicity and exopolysaccharide production, which is involved in the adherence or maturation of biofilms. Moreover, transcriptional results show that there was a downregulation in the expression of biofilm-related genes and alternative sigma factorsigBinduced by naringenin. This work provides insight into the anti-biofilm mechanism of naringenin inS. aureusand suggests the possibility of naringenin being used in the industrial food industry for the prevention of biofilm formation. Keywords:naringenin;biofilm formation;cell surface hydrophobicity;confocal laser scanning microscopy;biofilm-related genes

Naringenin (NRG) is a natural compound with several biological activities; however, its bioavailability is limited owing to poor aqueous solubility. In this study, NRG nanoparticles (NPs) were prepared using the wet media milling method. To obtain NRG NPs with a small particle size and high drug-loading content, the preparation conditions, including stirring time, temperature, stirring speed, and milling media amount, were optimized. The NRG (30 mg) and D-α-tocopherol polyethylene glycol succinate (10 mg) were wet-milled in deionized water (2 mL) with 10 g of zirconia beads via stirring at 50 °C for 2 h at a stirring speed of 300 rpm. As a result, the NRG NPs, with sheet-like morphology and a diameter of approximately 182.2 nm, were successfully prepared. The NRG NPs were stable in the gastrointestinal system and were released effectively after entering the blood circulation. In vivo experiments indicated that the NRG NPs have good antitussive effects. The cough inhibition rate after the administration of the NRG NPs was 66.7%, cough frequency was three times lower, and the potential period was 1.8 times longer than that in the blank model group. In addition, the enzyme biomarkers and histological analysis results revealed that the NRG NPs can effectively regulate the inflammatory and oxidative stress response. In conclusion, the NRG NPs exhibited good oral bioavailability and promoted antitussive and anti-inflammatory effects. Keywords:naringenin;nanoparticle;antitussive effect;antioxidant effect;anti-inflammatory effect

Acute liver injury is a common clinical disease, which easily leads to liver failure and endangers life, seriously threatening human health. Naringenin is a natural flavonoid that holds therapeutic potential against various liver injuries; however it has poor water solubility and bioavailability. In this study, we aimed to develop naringenin-loaded bovine serum albumin nanoparticles (NGNPs) and to evaluate their hepatoprotective effect and underlying mechanisms against acetaminophen overdose toxicity. In vitro data indicated that NGNPs significantly increased the drug solubility and also more effectively protected the hepatocyte cells from oxidative damage during hydrogen peroxide exposure or lipopolysaccharide (LPS) stimulation. In vivo results confirmed that NGNPs showed an enhanced accumulation in the liver tissue. In the murine model of acetaminophen-induced hepatotoxicity, NGNPs could effectively alleviate the progression of acute liver injury by reducing drug overdose-induced levels of oxidative stress, inflammation and apoptosis in hepatocytes. In conclusion, NGNPs has strong hepatoprotective effects against acetaminophen induced acute liver injury.

The aim of this study was to explore the lipid-lowering effect of naringenin and the underlying mechanism in high-fat-diet-fed SD rats and 3T3-L1 cells. In this study, SD rats were divided into the normal chow diet group (NCD), high fat diet group (HFD), three treatment groups feeding high-fat diet with naringenin (100, 200, 400 mg/kg) for 12 weeks. Results indicated that naringenin treatment decreased total cholesterol (TC), triglyceride (TG) and the non-high-density lipoprotein cholesterol (non-HDL-C) levels in serum. Naringenin also alleviated hepatic steatosis and reduced the adipocyte size in the epididymis in high-fat-diet-induced SD rats. In addition, naringenin (25−75 µg/mL) decrease TG and TC levels in 3T3 mature adipocytes. The molecular mechanism of naringenin in the treatment of obesity were predicted by using network pharmacology. Real-time PCR analysis results showed that naringenin regulated the expression of lipid metabolism genes . Meanwhile, naringenin increased the AMPK (AMP-activated protein kinase) activity and the expression of AMPK phosphorylated protein in 3T3 mature adipocytes. And the inhibitory effect of naringenin on lipid accumulation in 3T3 adipocytes was abolished by Compound C. Molecular docking results indicated that naringenin could bind to AMPK protein. These results indicated naringenin reduced lipid accumulation through AMPK pathway.

Naringenin (NAR), a natural flavonoid, possesses potent antibacterial and antioxidant properties. However, its therapeutic potential is severely limited by poor water solubility. Therefore, there is an urgent need to explore efficient drug delivery formulations to improve the physicochemical properties of NAR. In the present work, NAR was successfully encapsulated into methyl- β -cyclodextrin (M β CD) to form inclusion complex, which were further processed into nanofibers (NAR/M β CD-IC-NF) via electrospinning technology. SEM results indicated that the nanofibers were uniform and bead-free, with the NAR/M β CD-IC-NF displaying a remarkably large surface area per unit mass together with a highly developed porous framework. Multiple complementary characterization techniques including Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), proton nuclear magnetic resonance ( 1 H NMR), carbon nuclear magnetic resonance ( 13 C NMR), and X-ray diffraction (XRD) were adopted to verify the successful formation of NAR/M β CD inclusion complex within nanofibers. Molecular docking simulation was further performed to illuminate the host–guest interaction and binding conformation between NAR and M β CD at the molecular level. Additionally, there was a marked increase in the water solubility of NAR, with NAR/M β CD-IC-NF capable of complete dissolution in water within 3 s. Free radical scavenging and antibacterial assays confirmed that NAR/M β CD-IC-NF significantly enhanced the biological activity of NAR. In summary, NAR/M β CD-IC-NF served as an effective carrier system to significantly improve the solubility of NAR, thereby expanding its application prospects and bioavailability in biopharmaceuticals. Intensive farming and global warming exacerbate the impact of heat stress on the broiler industry, often causing significant heart damage to poultry and thus reducing production performance. Naringenin has garnered significant attention due to its potent antioxidant properties and cardioprotective characteristics. This article aims to explore the role of ferroptosis and ferritinophagy in heat stress-induced heart injury and evaluate the preventive effect of naringenin. The chickens were randomly allocated to four groups for the experiment. The experimental results indicate that heat stress can disrupt the homeostasis of heart antioxidant function and lead to the accumulation of free radicals in chickens. The accumulated reactive oxygen species result in heart damage. Moreover, heat stress upregulates the gene transcription and protein expression levels of ferroptosis-related indicators (GPX4, NRF2, ACSL4, TFR1) and ferritinophagy-related indicators (NCOA4, FTH1, P62, LC3B, BECN1). Meanwhile, naringenin intervention can effectively enhance heart antioxidant function and inhibit ferroptosis and ferritinophagy. It significantly reduces heat stress levels and alleviates heat-stress-induced damage to chicken hearts. Thus, it offers a new way to explore the prevention of heat stress in chickens.

EnterotoxigenicEscherichia coli(ETEC) leads to severe diarrhea in piglets. Naringenin (Nar), a natural flavonoid compound, is known for its antibacterial and anti-antioxidant properties. However, the protective effects of Nar against ETEC-induced diarrhea have not been reported yet. This study investigated the protective mechanisms of Nar against ETEC infection in porcine intestinal epithelial cells (IPEC-J2). ETEC infection induced oxidative stress and ferroptosis in IPEC-J2 cells by elevating intracellular iron content and ROS accumulation, increasing MDA levels, downregulating SOD activity and GPX4 expression, and upregulating the transcription of CHAC1 and SLC7A11. In contrast, Nar suppressed ETEC-induced ferroptosis of IPEC-J2 cells by inhibiting the SLC7A11/GPX4 pathway. Specifically, Nar mitigated mitochondrial damage, reduced intracellular iron levels and ROS accumulation, and ultimately reversed the oxidative stress. Network pharmacology and molecular docking identified heat-shock protein 90 (HSP90) as a potential target of Nar. Overexpression and knockdown experiments revealed that ETEC-induced ferroptosis was mediated by upregulation of HSP90, while the protective effects of Nar against ETEC-induced ferroptosis were dependent on the downregulation of HSP90. In conclusion, Nar targets host HSP90 to protect IPEC-J2 cells from ferroptosis caused by ETEC infection. This study demonstrates that Nar is a potent antioxidant natural compound with potential for preventing ETEC-induced intestinal damage.

Hyperuricemia (HUA) is one of main risk factors for liver injury, and xanthine oxidase (XOD) is an important target for HUA-induced liver injury. As a typical natural active ingredient, naringenin (NAR) has been confirmed the good therapeutic effect on variety of diseases. However, studies of NAR ameliorating HUA-induced liver injury have not been reported. Therefore, we evaluated the bioactivity of NAR in ameliorating HUA-induced liver injury and investigated the related molecular mechanisms. The inhibitory activity and type of NAR on XOD was investigated by enzymatic reactions and kinetic analyses, and molecular docking showed that NAR was able to bind tightly to XOD. In vivo activity studies showed that NAR ameliorated liver function while being able to inhibit XOD activity. NAR alleviated oxidative stress in the liver caused by excess reactive oxygen species through antioxidant activity. At the same time, NAR exerted anti-inflammatory activity by regulating the levels of inflammatory factors. The molecular docking results suggested that NAR was able to interact with Keap1 and AMPK to exhibit antioxidant and anti-inflammatory effects. This work demonstrated the therapeutic effect of NAR on HUA-induced liver injury, which was valuable for the further development of related functional foods.
