Spray-drying microencapsulation of β-carotene by soy protein isolate and/or OSA-modified starch

Xi-Xiang Deng,Zhong Chen,Qiang Huang,Xiong Fu,Chuan-He Tang

Microencapsulation is an effective strategy to improve the storage stability of β - carotene. This article investigated the potential and effectiveness of soy protein isolate (SPI) and octenylsuccinic anhydride-modified starch (MS), alone or in combination (1:1, w/w), to encapsulate β - carotene by spray drying. The results indicated that the microcapsule with MS exhibited much lower encapsulation efficiency (NE) and poorer dissolution behavior, but much better redispersion behavior, than that with SPI or its blends with MS. The NE was basically unaffected by total solid content (TC) or core/wall ratio; increasing the TC impaired the dissolution and/or redispersion behavior. The dispersion behavior was closely associated with the morphology of the microcapsules. The encapsulated β - carotene suffered a progressive loss upon storage under high humid or temperature environment, but it exhibited extraordinary stability at low temperatures (e.g., 4°C). The β - carotene degradation was independent of sunlight. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2014 , 131 , 40399.

Utilization of polysaccharide-based high internal phase emulsion for nutraceutical encapsulation: Enhancement of carotenoid loading capacity and stability

Chi Yan,Xiaolin Wu,Yi Wang,Shengfeng Peng,Jun Chen,Liqiang Zou,David Julian McClements,Wei Liu

The main goal of the present work was to access the ability of high internal phase emulsions (HIPEs) to encapsulate β -carotene. The carotenoid loading capacity of the HIPEs was around 20-fold higher when OSA-starch/chitosan complexes were used than when only OSA-starch was used. This impact could be mainly assigned to the capacity of the former HIPEs to trap carotenoid caystals in a stable form. The OSA-starch/chitosan complexes were shown to absorb on the oil droplets interface and form a 3D network in the aqueous phase, which helped to prevent droplet coalescence induced by β -carotene crystal. The incorporation of β -carotene within the oil droplets enhanced its resistance to chemical degradation when exposed to heat, ultraviolet radiation, or gastrointestinal conditions. Our results provide information that may aid the design and development of edible soft solids containing high carotenoid levels, which may be applied in food and pharmaceutical industry.

Modulating the multiscale structure and digestibility of OSA starch by phenolic acid substituents and heating process

Ran-Ran Deng,Ran Feng,Han Tao,Bao Zhang

The synergistic effect of methoxy-to-phenolic hydroxyl group ratios in phenolic acid and heat treatment on modified starch structures and underlying mechanism remain unexplored. This study aimed to elucidate the effects of octenyl succinic anhydride (OSA) starch (OR) multiscale structure and digestive properties by constructing complexes between OR and caffeic acid (CA), ferulic acid (FA), and sinapic acid (SA) at 25 °C/90 °C. Results indicated that OR-CA was more ordered among the complexes prepared at 25 °C. Methoxy groups enhanced phenolic acid reactivity, and OR was most stably bound to SA. The increased phenolic acid content at 90 °C promoted the formation of V-type complexes through hydrophobic interactions, where the resistant starch content of the complexes containing SA was 36.90 %. Phenolic acids synergistically reduced digestibility by inhibiting amylase activity or forming densely ordered crystalline structures with OR. The discovery enables targeted design of anti-obesity starchy foods by leveraging phenolic-starch complexes.

Emulsion-templated oleogel based on an OSA starch-gelatin-tannic acid ternary complex for algal oil delivery

Wenbo Miao,Junqing Han,Siew Young Quek,David Julian McClements,Keyu Lu,Zhengyu Jin,Chao Qiu

Electrostatic and hydrogen bonding drove OS-GE-TA ternary complex formation. • The incorporation of GE-TA enhanced the viscoelasticity of OS oleogels. • The ternary oleogels exhibited high oil-holding capacity and oxidative stability. • The ternary oleogels show excellent potential for DHA delivery. In this study, ternary complexes composed of octenyl succinate acetylated starch (OS), gelatin (GE), and tannic acid (TA) were used to fabricate oleogels via an emulsion-template method combined with freeze-drying for algal oil delivery. The results indicated that electrostatic interactions and hydrogen bonding drove the formation of the ternary complex, resulting in a stronger oleogel network and improved viscoelastic properties and storage stability. Under accelerated oxidation, OS-GE-TA oleogels exhibited significantly ( p  < 0.05) lower peroxide values than OS oleogels, with reductions of 9.55–12.87 meq/kg oil, and effectively inhibited fishy odor-related volatiles. In vitro digestion results showed that the free fatty acid (FFA) release of OS-GE-TA 0.4 and OS-GE-TA 0.8 oleogels reached 71.17% and 80.42%, respectively, both significantly ( p  < 0.05) higher than algal oil (64.28%), indicating enhanced lipid digestion and DHA release under simulated gastrointestinal conditions. Overall, the novel oleogels developed in this study have considerable potential for DHA delivery. Download: Download high-res image (407KB) Download: Download full-size image

Sinapic acid-functionalized chitosan and OSA-modified corn starch composite films: Characterization and application in meat packaging

Haijun Chang,Yu Hu,Yu Huang,Xin Wen,Li Wu

Developed OSAS/CH/SA film for active food packaging. • SA enhanced the mechanical strength and UV-barrier of the film. • Films showed high antioxidant activity and biodegradability. • 0.06 g/hg SA composite film extended pork shelf life to 7 days. The study aimed to develop an active packaging material based on octenyl succinic anhydride modified corn starch (OSAS) and chitosan (CH) incorporated with sinapic acid (SA), and to evaluate its application in pork preservation. The results demonstrated that the esterification between OSA and corn starch was achieved while preserving the crystalline structure, with a substitution degree of 0.02. Subsequently, antioxidant-active films were successfully prepared using CH and OSAS as the matrix materials (CO) with varying concentrations of SA. The physicochemical and mechanical properties of the CO/SA films were systematically characterized. SA endowed the films with superior mechanical strength, UV barrier properties, and biodegradability. The composite films exhibited significant antioxidant activity, effectively reducing lipid oxidation and microbial growth in pork, thereby enhancing meat quality. Compared with conventional polyethylene (PE) packaging, the CO/SA0.06 g/hg composite film exhibited excellent preservation capabilities, extending the shelf life of pork to 7 days. These findings suggest that the CO/SA film presents a promising alternative to traditional plastic packaging materials.

Characterization and Drug Delivery Properties of OSA Starch-Based Nanoparticles Prepared in [C3OHmim]Ac-in-Oil Microemulsions System

Liang Qi,Guangyin Ji,Zhigang Luo,Zhigang Xiao,Qingyu Yang

In this work, an enhanced starch-dissolving room-temperature ionic liquid 1-hydroxypropyl-3-methylimidazolium acetate ([C3OHmim]Ac) was tailor-made to serve as the polar phase in the formation of [C3OHmim]Ac/TX-100 + 1-butanol/cyclohexane (IL/O) microemulsion. Its superior solubility to starch was verified by the polarized-light microscope and turbidity test. Furthermore, a pseudoternary phase diagram, conductivity measurement, and dynamic light scattering (DLS) were used to analyze the phase behavior and microregion of IL/O microemulsion. On this basis starch nanoparticles with a mean diameter of 86.69 nm were successfully prepared in this novel IL/O microemulsion with octenyl succinic anhydride (OSA) maize starch as the raw material and epichlorohydrin as a cross-linker through microemulsion cross-linking reaction. For starch nanoparticles, Fourier transform infrared spectroscopy (FTIR) data demonstrated the formation of ester bonds, and cross-linking reaction in starch molecules and X-ray diffraction (XRD) disclosed the destruction of the crystal structure. In addition, scanning electron microscopy (SEM) intuitively revealed that OSA-modified starch nanoparticles possessed good dispersibility, nanoscaled size, and a relatively concentrated size distribution. Finally, drug loading and releasing properties of starch nanoparticles were investigated with indomethacin as a drug model.

Fabrication, characterization and potential application of biodegradable polydopamine-modified scaffolds based on natural macromolecules

Yiyu Wang,Xinyu Wang,Xingxun Liu,Chunqing Niu,Guiting Yu,Yuanjing Hou,Chao Hu,Kai Zhao,Jian Shi

Sodium alginate (SA)-based implantable scaffolds with slow-release drugs have become increasingly important in the fields of biomedical and tissue engineering. However, high-molecular-weight SA is difficult to remove from the body due to the lack of SA-degrading enzymes. The very slow degradation properties of SA-based scaffolds limit their applications. Herein, we designed a series of biodegradable oxidized SA (OSA)-based scaffolds through amide bonds, imine bonds and hydrogen bridges between OSA and silk fibroin (SF). SF/OSA-0.4 with a blend ratio of 4/1 was chosen for further polydopamine (PDA) surface modification studies through the optimization of those parameters such as different OSA oxidation degrees, and blend ratios. PDA modified SF/OSA-0.4 (Dopa/SF/OSA-0.4) showed the excellent stability, better stretchable properties, a uniform interconnective porous structure, high thermal stability, a low hemolysis ratio and cytotoxicity. In vitro degradation experiments showed that the degradation rate of SF/OSA was significantly higher than that of SF/SA, but the degradation slowed again after PDA modification. Interestingly, the degradation of Dopa/SF/OSA-0.4 in vivo was significantly faster than that in vitro . Dopa/SF/OSA-0.4 was also more conducive to new tissue growth and collagen bundle formation. Moreover, Dopa/SF/OSA-0.4 improved the absorbability of RhB (model drug) and reduced the sudden release of RhB during the sustained release.

Encapsulation of astaxanthin in OSA-starch based amorphous solid dispersions with HPMCAS-HF/Soluplus® as effective recrystallization inhibitor

Yinglan Li,Qipeng Wei,Jianshuo Su,Huaizhen Zhang,Zhiping Fan,Zhuang Ding,Min Wen,Min Liu,Yanna Zhao

In this study, the interaction among multifunctional excipients, including polysaccharides, cellulose derivatives, and surfactants, was particularly investigated, together with its impact on the physicochemical properties of astaxanthin amorphous solid dispersions (ASTX ASDs). It was indicated that Span 20 could rapidly form hemimicelles or aggregates in the presence of hypromellose acetate succinate HF (HPMCAS-HF, HF) or Soluplus®, while octenyl succinic anhydride modified starch (OSA-starch) efficiently assisted in the coalescence inhibition of drug-excipients aggregates, which was jointly beneficial to the recrystallization inhibition of amorphous ASTX. ASTX ASDs were further prepared with OSA-starch, HPMCAS-HF/Soluplus®, and Span 20 as the wall materials. DSC, SEM, and XRD confirmed that crystalline ASTX had transformed to amorphous state in the ASDs, while FT-IR spectra provided evidence suggesting the existence of hydrogen bonds and hydrophobic interaction between ASTX and the excipients. The dissolution of ASTX ASDs in different media revealed significant promotion, while the pharmacokinetic results further demonstrated the oral bioavailability of ASTX ASDs enhanced remarkably, exhibiting 2.75-fold (SD 1 ) and 1.87-fold (SD 2 ) increase, respectively, compared to ASTX bulk powder. In summary, the cellulose derivatives-surfactant interaction had great impact on the physicochemical properties of ASTX ASDs, and their combinations exhibited great potential for delivering the hydrophobic bioactive compounds efficiently.

Chitosan films incorporating Pickering emulsions stabilized by OSA-modified yeast dietary fiber for controlled citral release and berry preservation

Yaqiu Kong,Jieling Chen,Haiyan Tang,Zhiyu Yan,Yang Hu,Qilin Huang

OSA-YDF Pickering emulsions regulated film microstructure and interfaces. • Interfacial structure governed swelling–diffusion–controlled citral release. • Droplet dispersion enabled release, while aggregation hindered mass transfer. • The films exhibited enhanced UV-blocking, antioxidant and antibacterial activities. • Shelf life of strawberries and blueberries increased by 50% and 33.3%. Citral-loaded liquid high internal phase Pickering emulsions stabilized by OSA-modified yeast dietary fiber (OSA-YDF) were incorporated into a chitosan matrix to fabricate composite films for berry preservation. Compared with neat chitosan film, incorporation of the citral-loaded Pickering emulsion markedly enhanced the surface hydrophobicity, water vapor barrier properties, antioxidant and antibacterial performance. Specifically, the composite films exhibited inhibition rates exceeding 90% against E. coli , S. aureus , and B. cinerea . Citral in different food simulants showed a sustained release profile over 72 h and was strongly influenced by the interfacial characteristics of OSA-YDF-stabilized emulsion droplets dispersed within the chitosan matrix. Kinetic modeling indicated that citral release was primarily governed by chitosan matrix swelling and concentration-driven diffusion. At relatively low emulsion contents, the emulsion droplets were uniformly distributed within the chitosan matrix, allowing citral to diffuse across the OSA-YDF interfacial layer, resulting in sustained citral release. In contrast, excessive emulsion incorporation induced droplet aggregation and interfacial layer crowding, generating heterogeneous composite structure that hindered citral transport. An optimal balance between droplet dispersion and matrix integrity was achieved at 7.5% emulsion content, leading to superior overall film properties and controlled release performance. Preservation experiments further confirmed that the composite film effectively delayed berry deterioration by reducing weight loss, nutrient loss, and texture softening, extending shelf life from 4 d to 8 d for strawberries and from 12 d to 18 d for blueberries. This work provides valuable insights for active food packaging applications. Download: Download high-res image (193KB) Download: Download full-size image

Injectable oxidized sodium alginate/gelatin (OSA/Gel) hydrogel loaded with MgO/SZS for ion-mediated osteogenesis and macrophage polarization

Haiyu Li,Xianghua Wang,Jiangjie Tang,Ruoyu Ning,Liangjian Chen

Injectable OSA/Gel hydrogel with MgO and Sr–Zn–Si silicate bioceramic is developed. • Dual crosslinking enables rapid gelation, stable mechanics, and controlled degradation. • Sustained release of Mg, Sr, Zn and soluble Si species promotes osteogenic responses. • The hydrogel promoted a more anti-inflammatory, pro-regenerative macrophage profile. • Enhanced bone regeneration is achieved in a rabbit calvarial critical-sized defect model. Bone defect healing remains challenging due to trauma-associated microenvironmental disruption, persistent inflammation, and impaired coupling between bone formation and resorption. Here, we developed an injectable, biodegradable oxidized sodium alginate/gelatin (OSA/Gel) hydrogel with tunable gelation kinetics, favorable mechanical performance, and high shape fidelity for defect filling. By incorporating magnesium oxide (MgO) and Sr₂ZnSi₂O₇ (SZS) at different ratios, the formulation was optimized and its cytocompatibility and osteoimmunomodulatory activity were evaluated in vitro and in vivo. The hydrogel can be delivered minimally invasively to irregular defects and rapidly gels in situ into a porous three-dimensional scaffold, enabling sustained release of bioactive ions (Mg 2+ , Zn 2+ , Si 4+ , and Sr 2+ ). These ionic cues promoted osteogenic responses in MC3T3-E1 cells, as indicated by enhanced osteogenesis-related gene expression, increased alkaline phosphatase activity, and mineral deposition, and were concurrently associated with a shift of RAW264.7 macrophages toward a pro-regenerative M2 phenotype. In a rabbit calvarial defect model, the OSA/Gel–MgO/SZS hydrogel induced early osteoid-like tissue deposition and promoted the formation of low-mineral-density newly formed bone spanning the defect margins and extending toward the central region compared with controls. Overall, the OSA/Gel hydrogel containing 1.0% MgO and 0.5% SZS promoted bone regeneration, accompanied by enhanced osteogenic activity and a pro-regenerative immune response, demonstrating strong potential as a functional biomaterial for bone defect repair.

Salmon protein gel enhancement for dysphagia diets: Konjac glucomannan and composite emulsions as texture modifiers

Siyuan Fei,Yu Li,Kangjing Liu,Haitao Wang,A.M. Abd El-Aty,Mingqian Tan

The growing prevalence of dysphagia among the aging population presents a significant challenge. Many highly nutritious foods, like salmon, are often unsuitable for the elderly due to their firm texture when heated. To address this concern, a combination of salmon myofibrillar protein (SMP), Konjac glucomannan (KGM), and different emulsion fillers—such as oil droplets, octenyl succinic anhydride (OSA)-modified potato starch emulsion, and high methoxylated pectin (HMP) emulsions—was selected to enhance the network of salmon protein gels with the aims to create potential applications as dysphagia-friendly foods. The International Dysphagia Dietary Standardization Initiative (IDDSI) test indicated that all gel samples were classified as level 5. The OSA-SMP-KGM gel exhibited notably higher cohesiveness ( P  < 0.05), reduced adhesion, and enhanced mouthfeel. The OSA-SMP-KGM gel exhibited a smooth surface and excellent water retention (92.4 %), rendering it suitable for individuals with swallowing difficulties, particularly those prone to experiencing dry mouth. The yield stress of OSA-SMP-KGM gel was 594.14 Pa and stable structure was maintained during chewing and swallowing (γ e /γ v  = 62.5). This study serves as a valuable reference for developing salmon-based products that are not only highly nutritious but also fulfill the criteria for a desirable swallowing texture.

Replacing OSA starch: A novel maltodextrin-vitamin E succinate conjugate with superior emulsifying and antioxidant performance in resveratrol-loaded nanoemulsions

Yuanyuan Chen,Chang Zhang,David Julian McClements,Yanyan Zhou,Xuguang Yin,Runpu Shen,Xin Tang,Kewu Zhu,Xiang Luo

This study developed novel maltodextrin-vitamin E succinate (DE19-VES) conjugates were synthesized at different mass ratios of VES to maltodextrin (5 % and 10 %) as multifunctional emulsifiers for resveratrol encapsulation. Compared to nanoemulsions stabilized by octenyl succinic anhydride-modified starch (OSA S), those stabilized by DE19-VES conjugates exhibited significantly smaller droplet sizes, higher encapsulation efficiencies, and superior physical stability. Crucially, the DE19-VES conjugates imparted exceptional antioxidant properties to the nanoemulsions, enhancing radical scavenging activity, oxidative stability, and markedly improved protection of encapsulated resveratrol against degradation. In vitro digestion demonstrated significantly enhanced resveratrol bioavailability from DE19-VES-stabilized nanoemulsions versus OSA S. Cytotoxicity evaluation using human gastric mucosal epithelial cells (GES-1) confirmed that all formulations were non-toxic. These results highlight the dual functionality (emulsification and potent antioxidant activity) of the engineered maltodextrin conjugates, establishing their strong potential as advanced carbohydrate-based emulsifiers for protecting and delivering sensitive bioactives in pharmaceuticals, functional foods, and supplements.
