A new impedimetric immunosensor for the fast determination of ochratoxin A (OTA) in food samples was developed based on the instant catalyst as enhancer. Initially, the signal tags were prepared via co-immobilization of anti -OTA antibody and amine-terminated dendrimer (PAMAM) on the graphene oxide nanosheets through the covalent interaction, which were utilized as a good platform for combining manganese ion ( anti -OTA-GO-PAMAM-Mn 2+ ). Upon target OTA introduction, a competitive-type immunoreaction was implemented between the analyte and the immobilized OTA-BSA on the electrode for the anti -OTA antibody on the graphene oxide nanosheets labels. After a competitive immunoassay format, the anti -OTA-GO-PAMAM-Mn 2+ were captured onto the electrode surface , which could induce the in situ formation of MnO 2 via classical redox reaction between Mn 2+ and KMnO 4 on the immunesensing platform. Moreover, the generated MnO 2 nanoparticles act as efficient catalyst could catalyze the 4-chloro-1-naphthol (4-CN) oxidation without H 2 O 2 to generate an insoluble precipitation on the platform. Under the optimal conditions, the instant catalyst based impedimetric immunosensor displayed a wide dynamic working range between 0.1 pg mL −1 and 30 ng mL −1 . The detection limit (LOD) of the assay was 0.055 pg mL −1 . The developed method exhibited high selectivity and can be used for the determination of OTA in real red wine samples.

Raman Fingerprinting spectra detection of ochratoxin A (OTA) in food still meets serious challenge because of uncontrolled signal heterogeneity and poor reproducibility at low analyte concentrations. Here, OTA molecularly imprinted three-dimensional (3D) porous SERS substrate was prepared on the surfaces of magnetic inverse photonic crystal microspheres (MIPCMs) @AuNPs, and its Raman analytical enhancement factor can reach to 2.67 × 10 7 . The OTA imprinted polymer was synthesized using 3-aminopropyltriethoxysilane and phenyltriethoxysilane as the dual-functional monomers and Fmoc- d -Phenylalanine as the template molecule on the surfaces of MIPCMs. The functional microspheres can specifically enrich OTA and remarkably enhance the fingerprint Raman spectrum of OTA. The label-free 3D SERS substrate can rapidly quantitatively detect OTA by its fingerprint Raman spectrum, which shows a linear detection range of 10 −1 to 10 3 ng/mL, a limit of detection of 0.078 ng/mL, good recovery rates of 76–97 % and regeneration property. The developed SERS method can integrate enrichment and detection of OTA in one system, which has a great application prospect in simple, rapid and high sensitivity detection of OTA.

A GPE-PET (graphene-polyethylene terephthalate) bipolar electrode-electrochemiluminescence (BPE-ECL) platform was developed for ochratoxin A (OTA) detection. PET served as the electrode sheet substrate, and GPE was drop-coated onto the surface of PET to form a conductive line. On the functional sensing interface, the thiol (-SH) modified OTA aptamer (OTA-Aptamer) are fixed on the surface of the gold-plated cathode through Au S bonds. The efficient electron transfer ability of methylene blue (MB) made the anode ECL signal strong. Due to competition between OTA and MB with OTA-Aptamer, leading to a decrease in ECL intensity of the [Ru(bpy) 3 ] 2+ /TPA system on the BPE anode. Under optimized conditions, the GPE-PET BPE-ECL biosensor displayed superior sensitivity for OTA with a detection limit of 2 ng mL −1 and a wide linear concentration range of 5–100 ng mL −1 . This method could be further applied to detect various toxins and had broad application prospects.

This study describes an aptamer based assay for the mycotoxin ochratoxin A (OTA). The method is based on the use of an OTA-specific aptamer, exonuclease (Exo) III, SYBR Gold as a fluorescent probe, and a complementary strand that specifically combines with the aptamer. In the presence of OTA, the aptamer and OTA hybridize, thereby resulting in the formation of ssDNA, which is not digested by Exo III. Intense fluorescence is observed after addition of SYBR Gold (best measured at excitation/emission wavelengths of 495/540 nm). Fluorescence increases linearly with the log of the OTA concentration in the range from 8 to 1000 ng · mL −1 . The detection limit is 4.7 ng·mL −1 . The assay was applied to the determination of OTA in diluted [2%( v /v)] red wine, and recoveries and RSDs ranged between 93.5% and 113.8%, and between 3.2% and 5.7%, respectively.
In the presence of ochratoxin A (OTA), specific combinations of aptamer and OTA may occur and result in DNA double strands being untied, which avoids being digested by Exo III. Intense fluorescence is observed after SYBR Gold addition.

In the present study, a simple and fast method for simultaneous detection of zearalenone, citrinin, and ochratoxin A utilising capillary zone electrophoresis with an ultraviolet detector was developed. The optimised approach was validated and applied using pepper samples. The proposed method yielded satisfactory linearity between the signal and the mycotoxin concentration in the range of 1.5–150 μg/kg for zearalenone, 4.5–150 μg/kg for citrinin, and 0.8–150 μg/kg for ochratoxin A. The limits of detection for these mycotoxins ranged from 0.3 to 1.5 μg/kg. The corresponding intra- and inter-day precisions were less than 3.5 % and 4.1 %, respectively. Moreover, the matrix effect was also assessed and the result was compared using the capillary zone electrophoresis and high-performance liquid chromatography methods. The developed approach could be used for simultaneous detection of zearalenone, citrinin, and ochratoxin A in pepper.

Ochratoxin A (OTA) is a pervasive and significant mycotoxin that poses serious health risks to humans and animals. The development of efficient biocatalysts for the enzymatic detoxification of OTA is of great importance. In this study, we enhanced the OTA-degrading activity of three amidohydrolases (ADHs) by up to ninefold. This improvement was achieved by reducing steric hindrance in the binding pocket and fine-tuning the hydrophilic interactions between the enzyme and substrate. The most efficient variant, Pw ADH/DM, was immobilized onto magnetic Fe 3 O 4 nanoparticles functionalized by the co -deposition of dopamine and polyethyleneimine. Under optimal conditions, this immobilization process achieved a high immobilization efficiency (85.4%) and activity recovery (76.9%). The immobilized enzyme exhibited enhanced pH stability and thermostability, along with good storage stability, reusability, and recyclability. More importantly, the immobilized enzyme completely degraded 100 ng/mL OTA in contaminated milk without affecting the milk's properties. These findings expand our understanding of the molecular mechanisms governing substrate binding and catalysis in OTA-degrading ADHs. Furthermore, they provide a blueprint for enzyme-based OTA decontamination during food processing.

Ochratoxin A (OTA) as one of the most hazardous mycotoxins has drawn particular concern owing to its widespread occurrence in food and severe threat on human health. Herein, an exonuclease III (Exo III)-amplified electrochemical biosensor is developed for the sensitive detection of OTA by using bimetallic AuPt as signal enhancer. AuPt bimetallic nanoparticles loaded with massive electrochemical species thionine are employed as the excellent signal amplification probes (AuPt@Thi). The specific recognition of OTA triggers the conformational switching of DNA probes and induces the Exo III-assisted nucleic acid cycle amplification, generating a significant amount of DNA segments which are then introduced to the sensing interface to allow the attachment of signal probes AuPt@Thi. Bimetallic AuPt with outstanding electrical conductivity, exceptional loading capacity, and high catalytic activity enables the enhancement of Thi redox signal, resulting in a remarkable increase of signal response. Benefiting from the synergistic effect of Exo III-catalyzed cycle amplification and signal enhancement of electrochemically active AuPt@Thi, the proposed biosensor demonstrates superior analytical performance for OTA detection with the advantages of rapid response, fine robustness, cost-effectiveness and simple operation. Under the optimal conditions, a broad linear detection range of 0.1 pg mL −1 –100 ng mL −1 and the limit of detection down to 0.07 pg mL −1 are achieved. Furthermore, the as-prepared electrochemical biosensor shows good specificity, acceptable stability, satisfactory repeatability, and reliable capability for monitoring the OTA in actual food samples, implying promising practical applicability in the realm of food safety testing.

Ochratoxin A (OTA) is a potentially harmful mycotoxin that is widely found in agricultural products and animal by-products. In order to ensure our physical health, OTA testing is very important. Therefore, we chose to use an optical fiber biosensor based on the principle of local surface plasmon resonance (LSPR), and we designed an asymmetric optical fiber structure using single-mode optical fiber (SMF) to detect different concentrations of OTA. Gold nanoparticles (AuNPs) were applied to the sensing region to stimulate the LSPR effect, and then Nb 2 CTxMXene and CeO 2 -nanorods were sequentially fixed on the probe to change the dielectric environment around the sensing region to improve the performance of the sensor. The sensor has a linear range of 0.5 ng/mL to 60 ng/mL, a sensitivity of 0.0805 nm/(ng/mL), and a limit of detection (LOD) of 3.95 ng/mL. After that, we evaluated it through a series of tests, including repeatability, reproducibility, pH, stability, and selectivity. Finally, we also tested and verified the real samples. According to the experimental results, the sensor shows good performance in OTA detection, indicating that the sensor has good application potential and broad development prospects. This work designed an asymmetric optical fiber biosensor based on LSPR, achieving a broad detection range and a low LOD for OTA detection by coating with AuNPs, Nb 2 CTxMXene, and CeO 2 -nanorods. Its reliability and vast prospects in practical applications were validated through repeatability, stability, and real sample testing.

Ochratoxin A (OTA) is a harmful mycotoxin, which is mainly secreted by Penicillium and Aspergillus species. In this work, an electrochemical aptasensor is presented for OTA detection based on Au nanoparticles (AuNPs) modified zeolite imidazolate frameworks (ZIFs) ZIF-8 platform and duplex-specific nuclease (DSN) triggered hybridization chain reaction (HCR) signal amplification. G-quadruplex-hemin assembled HCR nanowire acted as a nicotinamide adenine dinucleotide (NADH) oxidase and an HRP-mimicking DNAzyme. Besides, thionine (Thi) was enriched as a redox probe for signal amplification in this pseudobienzyme electrocatalytic system. Under the optimal conditions, the analytical response ranged from 1 to 10 7 fg ml −1 with a detection limit of 0.247 fg ml −1 . Furthermore, the aptasensor was proven to be applied in real wheat samples with a recovery between 96.8 and 104.2%, illustrating the potential prospects in practical detection.

The ubiquitous presence of ochratoxin A (OTA), a toxic food contaminant, requires development of fast and sensitive detection tools. However, it is difficult to find a method that can address high sensitivity and wide detection range simultaneously. In this work, a sensing platform composed of praseodymia nanorods (Pr 6 O 11 -NRs) and aptamers for OTA is established. Under the dual-readout modes, this platform provides a highly sensitive detection of OTA with extra-wide range, over three orders of magnitude with limit of detection (LOD) of 0.8 ng/mL. Both fluorometric and colorimetric signals were obtained because of the excellent fluorescent quenching and oxidase-like activity of Pr 6 O 11 -NRs. This probe enables a detection range of OTA which covers the maximum acceptable levels of most foods. Real samples were tested with this method, demonstrating high sensitivity and good recoveries. This sensing strategy has great potential for an expanded application in point-of-care detection of other toxins via portable devices.

Ochratoxin A (OTA) contamination in food is a serious threat to public health. There is an urgent need for development of rapid and sensitive methods for OTA detection, to minimize consumer exposure to OTA. In this study, we constructed two OTA-specific fluonanobodies (FluoNbs), with a nanobody fused at the carboxyl-terminal (SGFP-Nb) or the amino-terminal (Nb-SGFP) of superfolder green fluorescence protein. SGFP-Nb, which displayed better fluorescence performance, was selected as the tracer for OTA, to develop a FluoNb-based nanosensor (FN-Nanosens) via the fluorescence resonance energy transfer, where the SGFP-Nb served as the donor and the chemical conjugates of OTA-quantum dots served as the acceptor. After optimization, FN-Nanosens showed a limit of detection of 5 pg/mL, with a linear detection range of 5–5000 pg/mL. FN-Nanosens was found to be highly selective for OTA and showed good accuracy and repeatability in recovery experiments using cereals with various complex matrix environments. Moreover, the contents of OTA in real samples measured using FN-Nanosens correlated well with those from the liquid chromatography with tandem mass spectrometry . Therefore, this work illustrated that the FluoNb is an ideal immunosensing tool and that FN-Nanosens is reliable for rapid detection of OTA in cereals with ultrahigh sensitivity.

Ochratoxin A (OTA) is a highly toxic potential contaminant of most agricultural products, and OTA is difficult to remove in general food production processes. In this study, we immobilized polyvinyl pyrrolidone-coated carboxypeptidase A (CPA) into zeolitic imidazolate framework material to improve the ability for OTA degradation. In this method, carboxypeptidase A was dispersed well in the material and maintained its stability and ability to degrade OTA, under complex matrix conditions with better repeated use performance than free carboxypeptidase A. The substrate conversion rate of CPA increased 3 times after immobilization. After repeated use of 10 cycles, the composite enzyme still retained 71.37% of its initial activity. The degradation rate of OTA was 30.69% higher than that of free CPA. This method may play an important role in the degradation of mycotoxins.
