Simple SummaryTwo types of diets with broad bean added (additive amounts of 40% and 50%, respectively), and the other two diets further added with the crisping functional package, were tested for the crisping effect on Nile tilapia. They all reached the crisping effect but had no adverse effect on the growth performance, and the supplement of the crisping functional package can further improve the crisping effect (especially the G5 diet). In addition, the crisping functional package also helped to improve the intestinal tissue and microbiota structure.AbstractFour crisping diets were designed to conduct a feeding experiment to investigate the use of broad bean in the formulated feed of Nile tilapia and optimize its use. The growth performance, muscle characteristics, antioxidant capacity, and intestinal health of Nile tilapia with an initial body weight of 617.32 ± 1.64 g were evaluated after being fed with different diets for 90 days. The results showed no adverse effect on the growth performance of Nile tilapia fed with broad bean diets. Contrastingly, some improvements were found in WGR and SGR, but a lower FCR was obtained. The supplementation of broad beans weakened the antioxidant capacity of fish but did not influence liver health and the immune system. Increasing the amount of broad bean addition can increase muscle quality values, and an embrittling functional package being added to the diet can also improve muscle hardness, adhesiveness, and chewiness of Nile tilapia muscle. In addition, the crisping functional package can be applied to tilapia crisping formulated feed, which helps to improve the integrity of the intestinal tissue structure and optimize the intestinal microbiota of Nile tilapia. The final achievement of this study is to provide a theoretical reference for optimizing the breeding technology of crispy Nile tilapia and developing a specialized crisping diet for the species. Keywords:broad bean;crisp;Nile tilapia;intestinal health;muscle

Zn is one of the essential elements to maintain normal fish metabolic process (including growth, development and reproduction), which have different chemical status in nature including inorganic, organic and nano-sized. In this work, the efficacy of gluconic acid zinc (Glu-Zn), nano zinc oxide (Nano ZnO), zinc sulphate (ZnSO 4 ) on the growth performance, tissue mineral elements deposition, antioxidant activity and immune response of the tilapia fish Nile tilapia ( Oreochromis niloticus ) was evaluated. Three experimental diets were prepared by adding different forms of Zn sources (ZnSO 4 as a control group) with same Zn concentration (40 mg/kg) and used to feed fish for 60 days. The biological trial results displayed that the growth performance of Nile tilapia has not obviously affected by different Zn form source supplement. However, Glu-Zn and Nano ZnO supplements have significantly stimulated the Zn and Cu absorption deposition efficiency in the fish tissues as compared to ZnSO 4 group. Besides, there was a significant increase in Cu-Zn superoxide dismutase (35.03 U/mgprot), alkaline phosphatase (79.34 U/mgprot), aspartate aminotransferase (23.42 U/L), alanine aminotransferase (18.45 U/L), glutathione peroxidase (171.24 U/mgprot) and fatty acid synthase (11.55 ng/mL) content in Glu-Zn treatment. Additionally, Glu-Zn supply significantly increased serum lysozyme (9.91 U/mL), immunoglobulin M (0.48 g/L), complement 3 (0.74 g/L) and complement 4 (0.95 g/L) activity of Nile tilapia. Also, compared to ZnSO 4 diet, lysozyme (11.44 U/mL) and complement 4 (0.63 g/L) content were obviously higher in fish fed Nano Zn diet. These findings revealed that supplementation with Glu-Zn as a Zn source in feed is better than ZnSO 4 for improving tissue mineralization, antioxidant stress, alleviate lipid peroxidation and immunity response of Nile tilapia. Meantime, Nano ZnO also played a positive role in the tissue mineralization and immunity response of Nile tilapia.

Src homology 2 domain-containing protein tyrosine phosphatase 1 (SHP1), a kind of protein tyrosine phosphatases (PTPs), is a critical regulator of antigen receptor signal transduction . Signal transduction of BCR is regulated by phosphatases in teleost as in mammals. In this study, SHP1 from Nile tilapia ( Oreochromis niloticus ) (OnSHP1) was identified and characterized, including the expression pattern against bacterial infection and regulation function in BCR signaling pathway. The open reading frame of OnSHP1 contains 1749 bp of nucleotide sequence , encoding a protein of 582 amino acids. The OnSHP1 protein was highly conversed compared to that of other species, including two amino-terminal SH2 domains at the N terminus and a PTP catalytic domain . Transcriptional expression analysis revealed that OnSHP1 was detected in all examined tissues and highly expressed in spleen . The up-regulated OnSHP1 expression was observed in peripheral blood, spleen and anterior kidney following challenge with Streptococcus agalactiae or lipopolysaccharide (LPS) in vivo , as well as that displayed in leukocytes stimulated with S. agalactiae or LPS in vitro . Further, after induction with mouse anti-tilapia IgM monoclonal antibody in vitro , OnSHP1 was significantly up-regulated in leukocytes. When spleen leukocytes treated with PTP Inhibitor II in vitro , the phosphorylation level of OnSHP1 at the phosphorylation sites (Y 535 and Y 557 ) and the cytoplasmic free Ca 2+ concentration were up-regulated significantly. Overall, the findings of this study indicate that SHP1 gets involved in host defense against bacterial infection and BCR signaling pathway in Nile tilapia .

To quantify the optimum requirement of dietary linolenic acid (18:3n-3, LNA) for Nile tilapia ( Oreochromis niloticus ), purified ethyl linolenate was added to the basal diet to provide the dietary LNA level of 0.00, 0.10, 0.32, 0.63, 0.98, 1.56 and 2.04% of dry weight, respectively. All diets contained similar amounts of 18:2n-6 (0.61% of dry weight) and 18:1n-9 from purified ethyl linoleate and oleate , respectively. Basal diet without LNA was considered as a control. After 2-week acclimation with a lipid-free diet, 4 replicate groups of 120 juvenile fish with initial body weight of 2.10 g were fed with experimental diets in a recirculated fresh water rearing system for 10 weeks. Feeding rate was 10% of body weight in the first 2 weeks, 8% in the following 4 weeks, and then gradually reduced to 5% in the next 4 weeks. Fish were fed three times daily at 8:30, 12:00 and 17:00. The results showed that fish fed the diet with 0.63% of LNA had significantly higher weight gain (WG) and specific growth rate (SGR) than fish fed the control diet ( P < 0.05). Feed efficiency (FE) and protein deposition (PD) in fish fed diets with 0.63, 0.98, 1.56 and 2.04% of LNA were significantly higher than those of the control group ( P < 0.05). Fish muscle fatty acid composition reflected dietary fatty acids. Increasing dietary LNA levels resulted in increased contents of LNA and 22:6n-3, but decreased contents of 20:4n-6 and 20:5n-3. When WG and FE were subjected to broken-line regression analysis, the breakpoint was achieved at dietary LNA levels of 0.45 and 0.64% of dry weight, respectively. Based on those results, the recommended optimum requirement of dietary LNA for juvenile tilapia is 0.45–0.64% of dry weight.

Streptococcus agalactiae(SA) is a severe prevalent pathogen, resulting in high morbidity and mortality in the global tilapia industry. With increasing bacterial resistance to antibiotics, alternative strategies are urgently needed. This study aims to investigate the antibacterial activity and the underlying mechanisms of the natural product xanthohumol (XN) against SA infection in tilapia (Oreochromis niloticus). The results showed that XN could significantly reduce the bacterial loads of SA in different tissues (liver, spleen and brain) after treatment with different tested concentrations of XN (12.5, 25.0 and 50.0 mg/kg). Moreover, XN could improve the survival rate of SA-infected tilapia. 16S rRNA gene sequencing demonstrated that the alpha-diversity index (Chao1 and Shannon_e) was significantly increased in the XN-treated group (MX group) compared to the SA-infected group (CG group) (p< 0.05), and the Simpson diversity index significantly decreased. The Bray–Curtis similarity analysis of non-metric multidimensional scaling (NMDS) and principal coordinate analysis (PCA) showed that there were significant differences in microbial composition among groups. At the phylum level, the relative abundance of the phyla Actinobacteria, Proteobacteria and Bacteroidetes decreased in the MX group compared to the CG group, while the relative abundance of the phyla Fusobacteria, Firmicutes and Verrucomicrobia increased. Differences were also observed at the genus level; the relative abundance ofMycobacteriumdecreased in the MX group, but the abundance ofCetobacteriumandClostridium_sensu_stricto_1increased. Metabolomics analysis revealed that XN changed the metabolic profile of the liver and significantly enriched aspartate metabolism, glycine and serine metabolism, phosphatidylcholine biosynthesis, arginine and proline metabolism, glutamate metabolism, urea cycle, purine metabolism, methionine metabolism, betaine metabolism, and carnitine synthesis. Correlation analysis indicated an association between the intestinal microbiota and metabolites. In conclusion, XN may be a potential drug for the prevention and treatment of SA infection in tilapia, and its mechanism of action may be related to the regulation of the intestinal microbiota and liver metabolism.

Tilapia skin collagen was hydrolyzed by five proteases (trypsin, pepsin, neutral protease, alkaline protease and protamex). The results showed that the tilapia skin collagen hydrolysate (TSCH) obtained by 2 h hydrolysis with trypsin exhibited the highest iron chelating rate. The TSCH was then separated by immobilized metal affinity chromatography (IMAC-Fe 2+ ) and obtained the tilapia skin collagen iron-chelating peptides (TSCICP). The iron chelating sites of TSCICP were corresponding to carboxylic groups of Asp/Glu and guanidine nitrogen of Arg/Lys. After chelated with iron ions , TSCICP was folded and aggregated to form spherical particles with increased particle size. The TSCICP-iron complexes could maintain high iron solubility at various pH and in simulated gastrointestinal digestion. The iron bio-accessibility of TSCICP-iron complexes was high than that of ferrous glycinate and ferrous sulfate. Finally, TSCICP was purified by RP-HPLC and identified by LC-MS/MS. Four iron-chelating peptides were identified as GPAGPAGEK (782.39 Da), DGPSGPKGDR (984.46 Da), GLPGPSGEEGKR (1198.59 Da) and DGPSGPKGDRGETGL (1441.68 Da). These results indicating that the iron-chelating peptides derived from tilapia skin collagen could be used as potential dietary iron supplement.

Short-chain fatty acids (SCFAs) can enhance the growth of aquatic animals and improve flesh quality through the gut-muscle axis. However, the effects of valeric acid, one of the SCFAs, on aquatic animals remain unreported. This study sought to explore the effects of valeric acid on Nile tilapia ( Oreochromis niloticus ). A total of 360 Juvenile tilapia (mean weight, 1.820 ± 0.007 g) with 3 tanks for each treatment were randomly assigned to 12 rectangular acrylic tanks and fed with four isolipidic (6 % lipid) and isonitrogenous (37 % protein) diets for 60 days, including a control diet (CK) and three experimental diets supplemented with 500 mg/kg (CK + LV), 1000 mg/kg (CK + MV), and 2000 mg/kg (CK + HV) of sodium valerate. The results discovered that the enhanced flesh hardness ( p < 0.01) of tilapia caused by valerate was attributed to decreased myofiber size ( p < 0.001). Transcriptome analysis revealed significant enrichment of the Forkhead box O (FOXO) signaling pathway in the valerate-treated group ( p < 0.001). Subsequent qPCR, western blot, immunohistochemistry, and immunofluorescence analyses demonstrated that valerate inhibited muscle cell proliferation by activating the FOXO signaling pathway, suppressing FOXO nuclear export, and consequently downregulating cyclins and cyclin-dependent kinases (CDKs) ( p < 0.01). Furthermore, valerate suppressed muscle differentiation by inhibiting the transcription and protein levels expression of myogenic regulatory factors (MRFs) ( p < 0.001). Additional validation in C2C12 myoblasts and myotubes confirmed that valerate indeed inhibited myoblasts proliferation and myotubes differentiation ( p < 0.001). These findings indicated that valerate reduced myofiber size by suppressing myoblasts proliferation and myotubes differentiation. This study elucidated the effects and mechanisms of valerate on skeletal muscle and introduced a novel approach to enhance fish flesh hardness.

An 8-week feeding trial was conducted to evaluate the effect of N -acetyl cysteine (NAC) and glycine supplementation on growth performance, glutathione (GSH) synthesis, anti-oxidative and immune ability of Nile tilapia , Oreochromis niloticus . Four practical diets were formulated, control, control +0.2% NAC, control +0.5% glycine, control +0.2% NAC +0.5% glycine. Each diet was randomly assigned to quadruplicate groups of 30 fish (approximately 9.5 g). The weight gain and specific growth rate were significantly increased with the supplementation of NAC and glycine. While they had no effect on feed efficiency feed intake and survival. Glutathion peroxidase (GPx) was increased by NAC and γ-glutamine cysteine synthase (γ-GCS) in plasma were increased by glycine. After the feeding trail, fish were challenged by Streptococcus iniae , fish fed the diet supplemented with NAC obtained significantly higher survival rate after 72 h challenge test. NAC also decreased malonaldehyde (MDA) in liver, increased glutathione S-transferase (GST) activity in plasma, up-regulated mRNA expression of Superoxide dismutase (SOD) and GPx in liver and headkidney. Dietary supplementation of glycine increased the anti-oxidative ability of tilapia through increase anti-oxidative enzyme activity (SOD, glutathione reductase , myeloperoxidase) and up-regulate anti-oxidative gene expression (SOD). Immune ability only enhanced by the supplementation of NAC through increased interleukin-1β (IL-1β) mRNA expression. These results clearly indicated that the supplementation of NAC and glycine can significantly improve the growth performance of tilapia, and NAC also enhance the anti-oxidative and immune capacity of tilapia, glycine could only enhance the anti-oxidative ability.

Effects of different ionic strengths (0.2, 0.4, and 0.6 mol/L) and different hydrodynamic cavitation (HC) treatment times (0, 1, 2, 3, and 4 min) on the conformation and gel properties of tilapia myofibrillar proteins (TMP) were investigated. The results showed that the solubility of TMP was significantly enhanced (p< 0.05) with the increase in NaCl concentration, and the gel characteristics were significantly improved. After HC treatment of TMP, the average particle size was significantly reduced (p< 0.05) and solubility was significantly enhanced (p< 0.05) with the increase in treatment time, the internal hydrophobic groups and reactive sulfhydryl groups were exposed. The intrinsic fluorescence spectra showed the unfolding of the spatial tertiary structure of proteins, and the circular dichroism spectroscopy showed the significant reduction in the content of α-helix in the secondary structure of the proteins (p< 0.05). In addition, the WHC and gel strength of the TMP heat-induced gels were enhanced, which improved the microstructure of the gels, and scanning electron microscopy showed that the gel network of the TMP gels became denser and more homogeneous. Dynamic rheology results showed that HC treatment resulted in a significant increase in the final G’ and G” values of TMP. In conclusion, HC treatment was able to improve the physicochemical structure and gel properties of TMP at different ionic strengths. This study presents a novel processing technique for the quality maintenance aspect of salt-reduced surimi gel products.

A lipidomic evaluation was performed on the tilapia muscle, head and viscera, including studying the composition, distribution and stereospecific characteristics of fatty acids and lipid species. The head and viscera lipids were significantly richer than the muscle lipids. Triacylglycerols were the predominant fraction (over 80% of total lipid in the muscle and head). Additionally, polyunsaturated fatty acids had higher percentages in phospholipids (30.35–52.05% of total fatty acids) than in triacylglycerols (18.11–25.15%). The C52:2 and C52:3 were the most abundant triacylglycerols, which indicates the potential application in infant food. Moreover, 622, 530 and 513 lipids were identified using ultraperformance liquid chromatography-quadrupole time-of-flight-mass spectrometry in the muscle, head and viscera, respectively. The three tilapia parts were distinguished using multivariate analysis. Five fatty acids and 33 lipid species were considered as the potential biomarkers. This comprehensive analysis will help to evaluate the lipid nutritional values and facilitate exploitation in tilapia consumption and processing.

With the increasing application of tetracycline (TC) in medical treatment, animal husbandry and aquaculture in recent decades, high quantities of TC have been frequently detected in the aquatic environment, and accordingly TC-related toxicity and environmental pollution have become a global concern. The present study was performed to explore the toxicological influences of TC exposure at its environmentally relevant concentrations on the gills of tilapia Oreochromis niloticus , based on the alteration in histopathology, oxidative stress, inflammatory response, cell cycle, mitochondrial function, apoptosis, and transcriptomic analysis. Our findings revealed that TC exposure damaged the structure and function, induced oxidative stress, affected inflammatory responses, and reduced Na + /K + -ATPase (NKA) activity in the gills. TC also caused the inhibition in cell cycle, resulted in mitochondrial dysfunction and activated apoptosis. Further transcriptomic analysis indicated the extensive influences of TC exposure on the gill function, and immune system was the main target to waterborne TC exposure. These results elucidated that environmental TC had more complex toxicological effects on gills of fish than previously assessed, and provided novel insight into molecular toxicology of TC on fish and good basis for assessing the environmental risk of TC.

Immunogenic, methionine copper-induced response had proven to be precedent in providing resistance against certain diseases in fish. This study allocates the fitness strategy for Oreochromis niloticus by introducing and incorporating the well-designed, stabilized, and biocompatible N-carbamoyl-methionine copper (NCM-Cu) as a Cu potent source in diet that enhances the bioavailability and fitness. The synchronized NCM-Cu complex was characterized by directing ultraviolet and visible spectrophotometry (UV–vis), Fourier-transform infrared (FTIR), X-ray diffractometry (XRD), thermogravimetric analysis (TGA), and single-crystal X-ray diffraction. Results revealed blue columnar crystalline, NCM-Cu complex with an empirical formula as C12H30CuN4O10S2. Anonymously, the overall growth performance of the fish remained unaltered with NCM-Cu adjunct feed. NCM-Cu significantly raised the Cu accumulation in the fish muscles, liver, gill, and intestine in contrast to the basic Cu-rich feed. The serum antioxidant enzyme activity elevated up to (ceruloplasmin: 19.38 U/L) and the lowest liver malondialdehyde (MDA) content (8.81 nmol/mg prot.) and triglyceride content (0.39 nmol/g prot.) were observed in the NCM-Cu group as compared to the basic Cu and CuSO4 groups, suggesting that NCM-Cu promoted antioxidative responses and alleviated lipid peroxidation of O. niloticus. Overweening, the synthesized complex, NCM-Cu significantly regulated the expression levels of lysozyme, immunoglobulin M, complement 4, and complement 3 up to 10.93 U/mL, 0.72, 0.77, and 1.18 mg/mL in serum, respectively. Thus, such endorsed results reveal the preeminence of NCM-Cu-supplemented diet for the fitness in O. niloticus.
