Biomimetic silicification enhanced PAH degradation by a microalgae–bacteria consortium. • Silica shells mitigated acid-induced oxidative stress and stabilized cellular metabolism. • Biodegradation remained the dominant PAH removal pathway under strong acidity. • Transcriptomics revealed preserved xenobiotic degradation and energy metabolism. • Silicification shifted microbial response from stress defence to metabolism-driven degradation. High-molecular-weight polycyclic aromatic hydrocarbons (HMW PAHs) are persistent pollutants whose biodegradation is strongly inhibited under acidic conditions. Here, a microalgae-bacteria consortium composed of Parachlorella kessleri and Paenibacillus illinoisensis was subjected to an integrated fabrication process comprising PDADMAC-assisted aggregation and nanosilicate deposition. Structural characterization confirmed sequential charge reversal, microbial aggregation, and formation of a silica-associated outer interface. Relative to the unmodified consortium, the fabricated group retained more chlorophyll, accumulated less reactive oxygen species, and maintained higher cytochrome P450 content under acid-PAH co-stress. PAH mass-balance analysis showed that biodegradation-associated transformation remained the dominant removal fraction, including an 84.44% biodegradation-associated fraction for benzo[ a ]pyrene at pH 3. Transcriptomic profiles further revealed reduced stress/repair investment together with preservation of energy and xenobiotic-degradation pathways. These multilevel results demonstrate that the complete PDADMAC-nanosilicate treatment stabilizes consortium function and sustains HMW-PAH transformation in a controlled acidic soil-extract system. Objective Coal miners are exposed to polycyclic aromatic hydrocarbons (PAHs), a group of neurotoxicants formed and released during incomplete combustion of coal. High levels of anxiety and depression have been reported in coal miners, but little is known about neurobehavioral functions in populations that are occupationally exposed to PAHs. We tested neurobehavioral performance in coal miners and correlated it with levels of urinary markers of PAH exposure. Methods Levels of urinary PAH metabolites were measured in 652 male coal miners as an indicator of exposure. Subjects were divided into a high-exposure group and a low-exposure group based on the median level of total PAH metabolites. A neurobehavioral core test battery and a questionnaire were used to assess neurobehavioral performance and collect demographic information, respectively. Results The median level of total PAH metabolites in urine was 4.88 μmol/mol creatinine. Highly exposed workers exhibited more fatigue-inertia than less-exposed workers ( p < 0.05), and had lower scores in forward digit span, digit symbol, and Benton visual retention tests ( p < 0.05). Multiple linear regression indicated that age, education, and shift work were significantly correlated with test scores. Conclusions PAH exposure may be associated with neurobehavioral alterations, which should be monitored in coal miners to prevent neurobehavioral disorders.

Catalytic etherification is a new and developing method for the upgradation of pyrolysis bio-oil into high performance bio-based long-chain ethers. In this work, the application of bio-based long-chain ether oxygenated additives in diesel engines have been checked by focusing on their regulated emissions and PAH emissions. Four bio-based long-chain ethers with similar structures, including: Polyoxymethylene dimethyl ether , diglyme , dipropylene glycol dimethyl ether and tripropylene glycol methyl ether have been blended with diesel fuel and tested in a small-duty diesel engine. The results showed that long-chain ethers were beneficial to the reduction of regulated emissions by comparing to pure diesel. Polyoxymethylene dimethyl ether and tripropylene glycol methyl ether showed best performance among the four tested ethers. Polyoxymethylene dimethyl ether could reduce 56% CO, 23% NO and 93% soot emissions, while Tripropylene glycol methyl ether could reduce 52% CO, 28% NO and 88% soot emissions. Besides, the particle sizes of soot particles from the blended fuels were also reduced. What's more, the addition of bio-based long-chain ethers could reduce particulate PAHs emissions by 39% ~ 67% and reduce gaseous PAHs emissions by 25% ~ 44%, and the PAHs toxicity was also reduced by 32% ~ 55%. This work proved that the structure of oxygen atoms evenly distributed in the chain could efficiently suppress the production of soot precursors and eventually reduce the soot emission

Bacteria play an important role in the biodegradation of polycyclic aromatic hydrocarbons (PAHs), especially the more recalcitrant high-molecular-weight compounds like pyrene. In this study, we isolated and characterized a novel pyrene-degrading bacterium, Glutamicibacter soli ENR6, from petroleum-contaminated soil. Strain ENR6 was able to utilize various PAHs as sole carbon sources and degrade 88.8% of 50 mg L−1 pyrene and 76.8% of 100 mg L−1 pyrene within 15 days. During pyrene degradation, eight metabolites were detected, with 2,2′-diphenic acid emerging as the predominant intermediate. By day 7, 40.6% of the degraded pyrene had accumulated in the form of 2,2′-diphenic acid, which was subsequently metabolized via the phthalate pathway. To our knowledge, this is the first report identifying 2,2′-diphenic acid as an intermediate in pyrene degradation. Integrated genomic and transcriptomic analyses revealed four gene clusters (clusters I–IV) involved in pyrene degradation in strain ENR6. Reverse transcription-quantitative PCR (RT-qPCR) analysis demonstrated that these clusters were specifically induced by pyrene and its intermediates, with genes chr_3400-chr_3404 in cluster I strongly associated with 2,2′-diphenic acid degradation. Based on these findings, we propose a distinct 2,2′-diphenic acid-centered pathway for pyrene degradation in strain ENR6. This study not only broadens the bacterial repertoire available for PAH bioremediation but also provides new insights into the metabolic diversity of PAH-degrading microorganisms.

Resin-bonded restorations, a prevalent treatment modality for dental defects, are prone to complications including nanoleakage, secondary caries, and pulpal pathology over extended service periods. To address these challenges, this study developed a multifunctional biomaterial, HZN@PAH-ACP, specifically designed for application at the dentin bonding interface. The material was fabricated by etching ZIF-8 with tannic acid to produce HZN, which was subsequently loaded with poly(allylamine)-stabilized amorphous calcium phosphate (PAH-ACP). The structure and formation mechanism of this biomaterial were thoroughly investigated using electron microscopy and molecular dynamics simulations. Meanwhile, HZN@PAH-ACP effectively induced intrafibrillar mineralization of collagen fibers and promoted remineralization of demineralized dentin, significantly reducing nanoleakage within the hybrid layer. Furthermore, the material inhibited the activity of Streptococcus mutans and exhibits excellent biocompatibility, while simultaneously enhancing the odontogenic differentiation and mineralization of human dental pulp stem cells. All powered by the simultaneous release of calcium, phosphate, zinc ions, and tannic acid, and the reduction in pH further accelerates this process. The development of this biocompatible, multifunctional repair biomaterial presents considerable clinical potential, as it addresses multiple challenges concurrently through a single application, that can be used in adhesive primers and pulp capping agents, offering a promising solution for dental restoration.

There is increasing concern regarding soils contaminated with polycyclic aromatic hydrocarbons (PAHs).In the present study, the remediation of soil spiked with PAHs was explored by the combination of soil washing with sodium dodecyl sulfate (SDS) and subsequent oxidation through persulfate (PS) activated by Fe2+, nanoscale zero-valent iron (nZVI), and SiO2-coated nZVI (SiO2/nZVI). Results demonstrated that the removal of phenanthrene (PHE), fluoranthene (FLU), and pyrene (PYR) by SDS is an efficient means for soil decontamination. At SDS concentration of 20 g/L, the removal efficiencies of PHE, PYR, and FLU were 37%, 40%, and 44%, respectively. For the degradation of PAHs and SDS in the soil washing effluents, the efficiencies of PS activated with SiO2/nZVI were not significantly different from those of PS activated with nZVI and Fe2+(p> 0.05). In practice, SiO2/nZVI is more preferable due to the improved antioxidation and dispersibility. At the dosage of 2 g/L (in the amount of iron) of SiO2/nZVI, the removal efficiencies of PHE, FLU, PYR, and SDS within 30 min of treatment were 75%, 85%, 87%, and 34%, respectively. The degradation of SDS was much lower than those of PAHs, which facilitated the recycle of SDS. Our findings suggest that PS activated with SiO2/nZVI is a promising method for the treatment of soil washing effluents containing SDS and PAHs.Keywords:soil remediation;PAHs;SDS;SiO2/nZVI;activated persulfate

ObjectivesPolycyclic aromatic hydrocarbons (PAHs) and 3-monochloropropane-1,2-diol ester (3-MCPDE) were studied in camellia oil. It is important to study the changes in the content of PAHs and 3-MCPDE at different refining stages (from crude oil to the final refined oil product) to elucidate the influence of the refining procedures on their change.Materials and methodsThe PAHs and 3-MCPDE in camellia oil from different refining stages (from crude oil to the product) of a plant were analyzed by gas chromatography–mass spectrometry and calculated by the internal standard method.ResultsThe overall PAH content was (79.64±2.43) µg/kg in crude camellia oil. After refining treatment, the PAH content decreased to (18.75±0.55) µg/kg. The 3-MCPDE content increased during the refining process from 0 mg/kg in the crude oil to 4.62 mg/kg in the refined oil product.ConclusionsThis is the first study to simultaneously monitor changes in both PAH and 3-MCPDE contents during the production of camellia oil. These results confirmed the effectiveness of the refining method on PAH removal and the increase in 3-MCPDE at high temperature. It is suggested that novel processing methods or refining parameters need further optimization to decrease the overall concentrations of PAHs and 3-MCPDE in camellia oil.

Background: The relationship between human immunodeficiency virus (HIV) infection and pulmonary arterial hypertension (PAH) has garnered significant scrutiny. Individuals with HIV infection have a higher risk of developing PAH. However, the specific mechanism of HIV-associated PAH remains unclear. Our study aims at investigating the shared biomarkers in HIV infection and PAH and predicting the potential therapeutic target for HIV-associated PAH.Methods: Data for HIV infection and PAH were downloaded from Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) analysis was performed to detect shared genes in HIV infection and PAH. Enrichment analysis was conducted to identify the function of common DEGs. Protein-protein interaction (PPI) analysis was used to detect key genes. These crucial genes were subsequently verified by RT-qPCR. Finally, candidate drugs were identified by using the Drug Signatures Database (DSigDB).Results: Nineteen common DEGs were identified in HIV infection and PAH. Enrichment analysis exhibited that the functions of these genes were mainly enriched in inflammatory responses, mainly including cellular immunity and interaction between viral proteins and cytokines. By constructing PPI networks, we identified the key gene CC-type chemokine ligand 5 (CCL5), and we verified that CCL5 was highly expressed in hypoxia induced human pulmonary artery endothelial cells (hPAECs) and human pulmonary artery smooth muscle cells (hPASMCs). In addition, we predicted 10 potential drugs targeting CCL5 by Autodock Vina. Conclusion: This study revealed that CCL5 might be a common biomarker of HIV infection and PAH and provided a new therapeutic target for HIV-associated PAH. However, further clinical validation is still indispensable.

Although a protective shell based on a metal-organic framework (MOF) can effectively improve the remediation of polycyclic aromatic hydrocarbons (PAHs) by microorganisms, the functional characteristics of the shell are often limited by the surface charge, chemical properties and intrinsic strain characteristics. This study explored the potential of mixed organic acids (MA) as an eco-friendly modifier for zeolite imidazolate framework-8 (ZIF-8), facilitating the formation of a biomimetic mineralized shell around Bacillus subtilis ZL09-26. The addition of MA into the ZIF-8 protective shell was found to stimulate growth and bolster cell viability. Notably, adding MA contribute resulted in a 1.98- fold enhancement of the PHE degradation efficiency. After five cycles of reuse, ZIF-8-MA@ B. subtilis ZL09-26 still maintained almost 90 % of the initial PHE degradation ability. Proteomic analysis revealed a coordinated regulation of multiple metabolic pathways that facilitated PHE uptake and degradation, including central carbon metabolism, direct PHE biodegradation, oxidative phosphorylation, aminoacyl-tRNA biosynthesis, fatty acid biosynthesis, ABC transporters, and the biosynthesis of valine, leucine, isoleucine, and lysine. This work broadens the application potential of biomineralized microorganisms, providing novel strategies for the sustainable bioremediation of xenobiotic pollutants in the environment.

Fundamental knowledge about the production and oxidation chemistry of polycyclic aromatic hydrocarbons (PAHs) is essential for developing predictive soot models. In contrast to the significant progress in understanding the formation/growth kinetics of PAHs, the detailed oxidation kinetics of PAHs remains to be fully established. This study investigates the oxidation kinetics of indene, the simplest PAH, as a foundational step for examining larger PAHs. Experiments were conducted in a flow reactor at temperatures from 850 to 1350 K, under a pressure of 0.04 atm and an equivalence ratio of 0.5. Characteristic products were quantified using synchrotron vacuum ultraviolet photoionization mass spectrometry, including large resonance-stabilized PAH radicals with 3–4 aromatic rings. Results indicate that even under highly oxidative conditions, the mass growth pathways of indene remain competitive, leading to the production of various large PAHs. This phenomenon is unique to indene oxidation and has not been observed in other bicyclic aromatics. A detailed kinetic model was developed to interpret these findings, revealing that PAH formation is predominantly driven by the indenyl radical. This study further calculates the rate coefficients for the dominant mass growth pathway of indenyl, specifically its self-recombination reaction, to enhance predictions of indenyl and bi-indene. Notably, while spiran and bridging mechanisms are comparably important in cyclopentadienyl mass growth, the spiran mechanism is favored for indenyl, suggesting that the addition of a benzenoid ring significantly alters the reactivity of cyclopenta-like radicals. Comparative analysis with indene pyrolysis experiments reveals that under oxidative conditions, the indenyl radical preferentially decomposes into smaller species, e.g., cyclopentadienyl or cyclopentadiene, thereby enhancing the yields of PAHs that depend on C 5 species as precursors. In the early reaction stage (<1220 K), oxidation reactions promote indenyl formation, thus increasing the proportion of mass growth products. However, at higher temperatures (>1220 K), enhanced oxidation reactions of indenyl lead to its decomposition, outcompeting mass growth reactions and sharply reducing the proportion of mass growth products.

Simple SummaryPolycyclic aromatic hydrocarbons (PAHs) are a large group of persistent organic pollutant chemicals that have significant carcinogenic effects on humans and animals. Degradation of PAHs by microbial laccase is a low-cost and high-efficiency pipeline, but the catalytic activity and expression level for most of the native bacterial laccases are low, limiting their wide use in industry. A recombinant enzyme called LacH5 showed high-efficiency PAH degradation activity, and its degradation activity was further improved by fusion expression and the expression level was augmented by a novel strong promoter replacement. The results from the current investigation provide new insights and strategies for PAH degradation and the development of a new candidate laccase for PAH biodegradation.AbstractLaccases catalyze a variety of electron-rich substrates by reducing O2to H2O, with O2playing a vital role as the final electron acceptor in the reaction process. In the present study, a laccase gene,lach5, was identified fromBacillus atrophaeusthrough sequence-based screening. LacH5 was engineered for modification by fusion expression and promoter replacement. Results showed that the purified enzyme LacH5 exhibited strong oxidative activity towards 2,2’-azinobis(3-ehtylbenzothiazolin-6-sulfnic acid) ammonium salt (ABTS) under optimum pH and temperature conditions (pH 5.0, 60 °C) and displayed remarkable thermostability. The activity of the two fusion enzymes was enhanced significantly from 14.2 U/mg (LacH5) to 22.5 U/mg (LacH5-vgb) and 18.6 U/mg (Vgb-lacH5) toward ABTS after LacH5 fusing withVitreoscillahemoglobin (VHb). Three of six tested polycyclic aromatic hydrocarbons (PAHs) were significantly oxidized by two fusion laccases as compared with LacH5. More importantly, the expression level of LacH5 and fusion protein LacH5-vgb was augmented by 3.7-fold and 7.0-fold, respectively, by using a novel strong promoter replacement. The results from the current investigation provide new insights and strategies for improving the activity and expression level of bacterial laccases, and these strategies can be extended to other laccases and multicopper oxidases. Keywords:thermostable laccase;PAH biodegradation;fusion expression;promoter screening;strong promoter replacement

Currently, bifunctional agents with vasodilation and ameliorated vascular remodeling effects provide more advantages for the treatment of pulmonary arterial hypertension (PAH). In this study, we first screened the hit 1 with heat shock protein 110 (Hsp110) inhibition effect from our in-house compound library with soluble guanylate cyclase (sGC) activity. Subsequently, a series of novel bisamide derivatives were designed and synthesized as Hsp110/sGC dual-target regulators based on hit 1. Among them, 17i exhibited optimal Hsp110 and sGC molecular activities as well as remarkable cell malignant phenotypes inhibitory and vasodilatory effects in vitro. Moreover, compared to riociguat, 17i showed superior efficacy in attenuating pulmonary vascular remodeling and right ventricular hypertrophy via Hsp110 suppression in hypoxia-induced PAH rat models (i.g.). Notably, our study successfully demonstrated that the simultaneous regulation of Hsp110 and sGC dual targets was a novel and feasible strategy for PAH therapy, providing a promising lead compound for anti-PAH drug discovery.
