Melanin-mediated accumulation of polycyclic aromatic hydrocarbons in human hair: Insights from biomonitoring and cell exposure studies

Shiyi Zhang,Weikeng Luo,Fang Zhao,Lulu Huang,Ruixin Qin,Xiao Yan,Bin Tang,Xiaojun Luo,Bixian Mai,Yunjiang Yu,Jing Zheng

While human hair is widely used to monitor micro-organic contaminants (MOCs), their incorporation mechanisms are poorly understood. Melanin, known to facilitate the accumulation of drugs in hair, hasn’t been studied in the field of MOCs. Here, polycyclic aromatic hydrocarbons (PAHs), a class of priority MOCs, were investigated through hair biomonitoring as well as cell exposure experiments. PAH concentrations and melanin contents were measured in black and white hairs from the same individual. The results showed that five dominant PAHs (phenanthrene, fluoranthene, pyrene, benzo[ a ]anthracene and chrysene) in black hair (0.66 ng/g – 35.1 ng/g) were significantly higher than those in white hair (0.52 ng/g – 29.6 ng/g). Melanin contents in black hair (14.9 – 48.9 ng/g) were markedly higher than in white hair (0.35 – 2.15 ng/g) and were correlated to PAH concentrations, hinting melanin-mediated accumulation of PAHs in hair. The in vitro experiment using murine melanoma cells demonstrates that PAH levels in cells were affected by melanin, suggesting the affinity of melanin to PAHs. Both biomonitoring and cell exposure experiment implicate the pivotal role of melanin in PAH accumulation in hair. Therefore, to ensure the accuracy of hair biomonitoring for MOCs, attention must be paid to the melanin content uniformity.

Quantification of amino-PAHs in human urine using UHPLC- hybrid quadrupole orbitrap high resolution MS (Q-orbitrap-HRMS): Assessing potential carcinogenic risks of Nitro-PAHs

Haixia Jin,Yong Wang,Junning Liu,Qun Gao,Jing Ji,Geyu Liang,Xiaoxia Gu,Kai Luo

Considering only parent polycyclic aromatic hydrocarbons (PAHs), such as benzo(a)pyrene as a representative compound, may underestimate the carcinogenic risk of polycyclic aromatic compounds (PACs). Nitrated polycyclic aromatic hydrocarbons (nitro-PAHs), known for their higher toxicity relative to parent PAHs, are widely present in the environment. In this study, a novel, highly sensitive, and accurate method utilizing ultra-performance liquid chromatography coupled with hybrid quadrupole Orbitrap high resolution MS (Q-Orbitrap-HRMS) was developed to quantify 15 amino-PAHs, which are metabolites of nitro-PAHs, in human urine samples. Our analysis revealed the presence of 10 out of the 15 targeted amino-PAHs in urine samples obtained from adults residing in Nanjing, China. The cumulative concentrations of the 10 amino-PAHs (∑ 10 amino-PAHs) in each urine sample ranged from not detected (ND) to 19.12 ng/mL, with mean and median concentrations of 2.85 and 2.09 ng/mL, respectively. Risk assessment using the potency equivalency factor (PEF) indicated that the mean estimated cancer risks for 1,6-dinitropyrene and 1,8-dinitropyrene, two dinitro-PAHs classified by the International Agency for Research on Cancer (IARC) as possibly carcinogenic to humans (Group 2B), were 2.96 × 10 −4 and 4.93 × 10 −5 , respectively, exceeding the commonly accepted risk threshold of 1 × 10 −6 , and indicating a potential public health concern. Overall, this study established a robust method for the identification and quantification of amino-PAHs in urine samples, highlighting the presence of nitro-PAHs, particularly dinitro-PAHs, in the human body and their significant yet often overlooked carcinogenic potential, necessitating further investigation and attention.

Lactobacillus murinus alleviated lung inflammation induced by PAHs in mice

Yuqi Zhu,Xuna Tao,Tongtong Yan,Shuyuan Cao,Ping Jiang,Zhan Zhang,Lei Li,Qian Wu

Objective This study aimed to investigate the mechanism that Lactobacillus murinu s ( L. murinu s) alleviated lung inflammation induced by polycyclic aromatic hydrocarbons (PAHs) exposure based on metabolomics. Methods Female mice were administrated with PAHs mix, L. murinus and indoleacrylic acid (IA) or indolealdehyde (IAId). Microbial diversity in feces was detected by 16 S rRNA gene sequencing. Non-targeted metabolomics analysis in urine samples and targeted analysis of tryptophan metabolites in serum by UPLC-Orbitrap-MS and short-chain fatty acids (SCFA) in feces by GC-MS were performed, respectively. Flow cytometry was used to determine T helper immune cell differentiation in gut and lung tissues. The levels of IgE, IL-4 and IL-17A in the bronchoalveolar lavage fluid (BALF) or serum were detected by ELISA. The expressions of aryl hydrocarbon receptor ( Ahr ), cyto chrome P45 0 1A1 ( Cyp1a1 ) and forkheadbox protein 3 ( Foxp3 ) genes and the histone deacetylation activity were detected by qPCR and by ELISA in lung tissues, respectively. Results PAHs exposure induced lung inflammation and microbial composition shifts and tryptophan metabolism disturbance in mice. L. murinus alleviated PAHs-induced lung inflammation and inhibited T helper cell 17 (Th17) cell differentiation and promoted regulatory T cells (Treg) cell differentiation. L. murinus increased the levels of IA and IAId in the serum and regulated Th17/Treg imbalance by activating AhR. Additionally, L. murinus restored PAHs-induced decrease of butyric acid and valeric acid which can reduce the histone deacetylase (HDAC) level in the lung tissues, enhancing the expression of the Foxp3 gene and promoting Treg cell differentiation. Conclusion our study illustrated that L. murinus alleviated PAHs-induced lung inflammation and regulated Th17/Treg cell differentiation by regulating host tryptophan metabolism and SCFA levels. The study provided new insights into the reciprocal influence between gut microbiota, host metabolism and the immune system, suggesting that L. murinus might have the potential as a novel therapeutic strategy for lung diseases caused by environmental pollution in the future.

Ce(Ⅲ) activates peroxymonosulfate for the degradation of substituted PAHs

Xuwen Chen,Peixin Wang,Fei Peng,Zhou Zhou,Michael Gatheru Waigi,Wanting Ling

Substituted polycyclic aromatic hydrocarbons (SPAHs) are being intensively investigated, considering their high toxicity. Additionally, the mechanism of the effect of substituents on the removal of SPAHs and the activation of Ce(III) ions on peroxymonosulfate (PMS) have not been explored. Here we evaluated the removal efficiency of SPAHs in the oxidation system constructed by Ce(Ⅲ) ions and PMS, with emphasized the effect of substituents on SPAHs degradation. Ce(Ⅲ) has high catalytic performance for PMS, and the degradation percentage of all pollutants was higher than 92%. The significantly negative correlation between the reaction rate constants of SPAHs and the highest occupied molecular orbital-the lowest unoccupied molecular orbital gap, confirms that substituents lead to the differences in the degradation of SPAHs. The generation of reactive oxygen species (SO 4 •− , • OH, and 1 O 2 ) is based on the electron transfer between Ce(Ⅲ) and PMS, and the contribution of ROS to substituted naphthalene varies due to the role of substituents. The Ce(Ⅳ)/Ce(Ⅲ) cycle accelerates the activation of PMS. Based on the transformation products and condensed Fukui function, the possible degradation pathways are inferred. In addition, inorganic anions and organic matter have little effect on the Ce(Ⅲ)/PMS system, which is a prerequisite for applying this system to real-world waste-water for SPAHs removal. This work demonstrates a new model of the degradation mechanism of SPAHs in the Ce(Ⅲ)/PMS system.

Occurrence, source modeling, influencing factors and exposure assessment of polycyclic aromatic hydrocarbons in water sources: A mega-study from mainland China

Kunfeng Zhang,Sheng Chang,Qi Zhang,Yunsong Bai,Enrui Wang,Yueting Fan,Xiang Tu,Qing Fu,Liangliang Wei,Yanling Yu

This study investigates the composition and distribution of 16 priority polycyclic aromatic hydrocarbons (PAHs) in 70 water samples collected from centralized drinking water sources (CDWSs) across 30 provinces (cities) in mainland China. Additionally, water quality and socio-economic data were collected to determine the influencing factors. The study discovered that the total concentrations of 16 priority PAHs (Σ 16 PAHs) varied from 0.17 to 176.71 ng/L (mean: 56.49 ± 38.58 ng/L). Lakes and reservoirs had significantly higher concentrations of PAHs than groundwater. The primary pollutants were low-molecular-weight (LMW) PAHs, such as phenanthrene, naphthalene, and fluorene. There was a strong correlation between LMW PAHs and Σ 16 PAHs ( p < 0.05). The mean Σ 16 PAHs in the seven geo-environmental zones were ranked as follows, under the comprehensive influence of emission density, natural, and social environmental factors: Northwest China (78.42 ± 66.96 ng/L) > Northeast China (73.70 ± 33.23 ng/L) > East China (64.35 ± 45.66 ng/L) > North China (50.95 ± 31.87 ng/L) > Southwest China (44.05 ± 32.56 ng/L) > Central China (41.68 ± 19.44 ng/L) > Southern China (40.99 ± 16.54 ng/L). The source modeling analysis revealed that the majority of PAHs came from chemical production and metal smelting (42.36%), as well as from the combustion of coal, wood, and biomass (34.17%), and other mixed sources. In addition, the study found a strong correlation between water quality and socio-economic factors with concentrations of PAHs, indicating that these factors may contribute to the re-enrichment, transport, and transformation of PAHs in overlying water to varying degrees. The mean incremental lifetime cancer risk (ILCR) was lower than the risk threshold. These findings provide scientific references for managing hazardous substances in the aquatic environment.

Microplastic-water partitioning of two states halogenated PAHs: Solute and sol

Qing Sun,Feng-Hui Lu,Hong-Gang Ni

The complex interactions of contaminants with microplastics significantly affect ecological risk assessments . Studies of the sorption behavior of freely dissolved hydrophobic organic contaminants (HOCs) on microplastics are common. However, concentrations of HOCs in the actual aquatic environment sometimes exceed their water solubility. A possible explanation is that a microplastic-sol-water three-phase medium (TPM) is formed in the actual water environment. Both states HOCs (in solute and sol) have the potential to migrate to particles suspended in water. To confirm this view, four kinds of microplastics and eight halogenated polycyclic aromatic hydrocarbons (HPAHs) were selected to examine the partitioning of HPAHs between microplastics and water (sol and solute). Both monolayer and multilayer coverage of HPAHs onto microplastics occurred, and chemical sorption dominated the pseudo−sorption mechanism. The microplastic−water partition ratios of HPAHs (0.12–0.74) were approximately four to five orders of magnitude lower than their corresponding K OW values, suggesting that mechanisms other than sorption were involved. Apparently, the sol HPAHs contributed almost identically large increments to both microplastics and water, and closed the gap. For microplastic−supported HPAHs, the contribution of the sol fraction was more than triple that of the dissolved fraction; the key influencing factor was the water solubility of HPAHs.

Intensive removal of PAHs in constructed wetland filled with copper biochar

Xiaotong Shen,Jian Zhang,Huijun Xie,Zhen Hu,Shuang Liang,Huu Hao Ngo,Wenshan Guo,Xinhan Chen,Jinlin Fan,Congcong Zhao

In this study, biochar-loading copper ions (Cu-BC), a novel composite for removing phenanthrene very efficiently from water, was prepared using the impregnation method. The performance of constructed wetlands (CWs) with these modified and original biochar as substrates was analyzed. CW with Cu-BC removed a large amount of phenanthrene (94.09 ± 3.02%). According to the surface characteristics analysis, Cu-BC can promote the removal of pollutants via complex absorption, hydrophobic adsorption, increasing the Lewis Pair and electrostatic attraction. Furthermore the higher nitrate removal rate in the treated system (91.11 ± 1.17%) was observed to have higher levels of bacterial metabolic diversity and denitrifier types. The phenanthrene accumulated in plants with this treatment system was enhanced by the role of copper in photosynthesis. It is able to boost the plant extraction of organic matter.

Enhanced phytoremediation of PAHs and cadmium contaminated soils by a Mycobacterium

Na Li,Rui Liu,Jianjun Chen,Jian Wang,Liqun Hou,Yuemei Zhou

This study investigated Fire Phoenix ( Festuca L.) and Echinacea purpurea (L.) Moench inoculated with a Mycobacterium strain N12 in remediation of soils contaminated with both polycyclic aromatic hydrocarbons (PAHs) and cadmium (Cd). Plant growth and PAH and Cd removal were monitored in 60, 120, and 150 days after transplanting. Results showed that Fire Phoenix plants grown in soil containing 200 mg/kg PAHs and 15 mg/kg Cd inoculated with N12 were able to remove 76.3% PAHs compared to removal of 68.3% of PAHs by the plants without N12 inoculation. On day 150, the underground biomass of Fire Phoenix plants grown in soil inoculated with N12 increased 59.40% compared to that without N12 inoculation. The enhanced removal of PAH by Fire Phoenix and N12 was related to the improved rhizosphere microbial activities . However, inoculation of N12 to E. purpurea grown soil did not significantly improve the removal of PAHs and Cd. Our results showed that phytoremediation of PAHs and Cd can be enhanced by a Mycobacterium strain N12, especially when PLFA concentrations of bacteria and fungi exceeded 60% of the initial concentrations, but the enhancement is plant species dependent.

Particle-bound PAHs induced glucose metabolism disorders through HIF-1 pathway

Siqi Wu,Zhongli Chen,Li Yang,Yulin Zhang,Xiaohe Luo,Jinsong Guo,Ying Shao

Vehicle exhaust, as one of the most important compositions of air pollution, induced various adverse health effects, especially diabetes, on human beings. Even though monitoring and epidemiological data indicates that particle-bound polycyclic aromatic hydrocarbons (PAHs) is an inducing factor of diabetes, the specific causative mechanisms are still unclear. In the current study, the concentration of particulate matters (PMs, including PM 1.0 , PM 2.5 and PM 10.0 ) and PAHs was investigated at rush hour of weekday in three urban underground parking garages (UPGs). To evaluate the impacts of particle-bound PAHs on human beings, analysis of non-target metabolomics and unmetabolized PAHs were conducted for UPG and non-UPG worker urine samples. The results showed that the highest concentrations of PMs and total PAHs were found at the UPG entrance. The concentrations of unmetabolized 5-6 rings PAHs in the UPG worker urine were significantly higher than that in non-UPG worker urine samples, which induced glucose metabolism disorders through hypoxia-inducible factor 1 (HIF-1) signaling pathway. This could be a reason for particle-bound PAHs induced-diabetes on road workers, drivers and garage staff. These findings can serve as a step towards air pollution management and the pathological mechanism analysis of environmental factor induced-diseases.

Fluoranthene degradation and binding mechanism study based on the active-site structure of ring-hydroxylating dioxygenase in Microbacterium paraoxydans JPM1

Jin Jingnan,Yao Jun,Liu Wenjuan,Zhang Qingye,Liu Jianli

In this study, a gram-positive fluoranthene-degrading bacterial strain was isolated from crude oil in Dagang Oilfield and identified as Microbacterium paraoxydans JPM1 by the analysis of 16S rDNA sequence. After 25 days of incubation, the strain JPM1 could degrade 91.78 % of the initial amount of fluoranthene. Moreover, four metabolites 9-fluorenone-1-carboxylic acid, 9-fluorenone, phthalic acid, and benzoic acid were detected in the culture solution. The gene sequence encoding the aromatic-ring-hydroxylating dioxygenase was amplified in the strain JPM1 by PCR. Based on the translated protein sequence, a homology modeling method was applied to build the crystal structure of dioxygenase. Subsequently, the interaction mechanism between fluoranthene and the active site of dioxygenase was simulated and analyzed by molecular docking. Consequently, a feasible degrading pathway of fluoranthene in the strain JPM1 was proposed based on the metabolites and the interaction analyses. Additionally, the thermodynamic analysis showed that the strain JPM1 had high tolerance for fluoranthene, and the influence of fluoranthene for the bacterial growth activity was negligible under 100 to 400 mg L −1 concentrations. Taken together, this study indicates that the strain JPM1 has high potential for further study in bioremediation of polycyclic aromatic hydrocarbon (PAH)-contaminated sites.

Construction and analysis of an engineered Escherichia coli-Pseudomonas aeruginosa co-culture consortium for phenanthrene bioremoval

Xiaoqiang Jia,Yun He,Dawei Jiang,Chang Liu,Wenyu Lu

Polycyclic aromatic hydrocarbons (PAHs) like phenanthrene are harmful to human health and are difficult to eliminate from environments. In this study, a phenanthrene degrading strain, Pseudomonas aeruginosa PH1, was isolated from oil-contaminated soil. An engineered co-culture consortium was subsequently constructed comprising two engineered bacterial strains, Escherichia coli HY1 and P. aeruginosa PH2, for use in phenanthrene bioremoval. E. coli HY1 harbored two modules of terminal dioxygenases and an electron transfer chain, while P. aeruginosa PH2 contained a module of catechol 1,2-dioxygenase. Phenanthrene removal rates by the wild-type degrader, P. aeruginosa PH1, were 45%, 43%, and 81%, in mineral salt medium supplemented with 100 mg/L phenanthrene, 100 mg/L phenanthrene and 2% (wt/vol) oil, and 100 mg/L phenanthrene and 2% (vol/vol) n -hexadecane, respectively. In contrast, the engineered co-culture consortium removed 71%, 80%, and 88% of the phenanthrene in the corresponding treatments, thus achieving much higher removal efficiencies. Moreover, gas chromatography-mass spectrometry confirmed that the engineered co-culture consortium first oxidized phenanthrene to 9,10-dihydroxy phenanthrene and 1,2-dihydroxy phenanthrene and then metabolized these intermediates via catechol to carbon dioxide and water. During this process, E. coli HY1 dominated the initial oxidation steps, while P. aeruginosa PH2 was primarily responsible for ring cleavage. These results indicate that the engineered co-culture consortium described here represents a potential alternative and attractive means to remove PAHs from natural environments.

Involvement of the Gut–Lung Axis in LMW-PAHs-Induced Pulmonary Inflammation

Jiali Qin,Shiyao Jiang,Zhengyi Zhang,Jianding Wang,Yuanjie Li,Yunting Li,Haojun Zhang,Chengyun Li,Haitao Ma,Junling Wang

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous environmental pollutants recognized for their toxicological significance. Increasing evidence suggests that chronic exposure to low-molecular-weight PAHs (LMW-PAHs) contributes to heightened disease vulnerability and immune dysregulation, particularly among rural female populations. Recent studies have further linked a significant association between PAH exposure and gut microbiome (GM) modifications. Considering the common embryonic origin of the intestinal and respiratory systems, cross-organ communication under conditions of PAH exposure warrants deeper exploration. Although current gut–lung axis research largely emphasizes microbial metabolites such as short-chain fatty acids and bile acids, the contribution of arachidonic acid (AA) metabolites in LMW-PAH-induced pulmonary inflammation via this axis remains poorly defined. To address this knowledge gap, we developed an animal model employing integrated 16S rRNA sequencing and metabolomics approaches to systematically examine phenanthrene (Phe) and fluorene (Flu) induced GM compositional shifts and associated metabolic reprogramming. Through comprehensive profiling, we identified candidate microorganisms and metabolites potentially involved in dysbiosis-mediated pulmonary inflammation, thereby elucidating the mechanistic basis of Phe and Flu-associated health risks.
