Proline-rich protein PRPL1 enhances Panax notoginseng defence against Fusarium solani by regulating reactive oxygen species balance and strengthening the cell wall barrier

Linlin Su,Wenyun Li,Xiaohua Chen,Pengcheng Wang,Diqiu Liu

The root rot mainly caused by Fusarium solani is a bottleneck in the cultivation of Panax notoginseng . In this study, we reported a gene encoding a plant cell wall structural protein, P. notoginseng proline-rich protein (PnPRPL1), whose transcription was upregulated by F. solani and induced by some hormone signals. The PnPRPL1 recombinant protein significantly inhibited the growth and conidial germination of the root rot pathogens. Downregulation of PnPRPL1 by RNA interference (RNAi) in P. notoginseng leaves increased the susceptibility to F. solani , whereas overexpression of PnPRPL1 in tobacco ( Nicotiana tabacum ) enhanced the resistance to F. solani . Compared with wild-type tobacco, the PnPRPL1 -overexpressing transgenic tobacco had higher reactive oxygen species (ROS)-scavenging enzyme activities, lower ROS levels, and more lignin and callose deposition. The opposite results were obtained for the P. notoginseng expressing PnPRPL1 RNAi fragments. Furthermore, the PnPRPL1 promoter transcription activity was induced by several plant hormones and multiple stress stimuli. In addition, the transcription factor PnWRKY27 activated the expression of PnPRPL1 by directly binding to the promoter region. Thus, PnPRPL1 , which is positively regulated by a WRKY transcription factor, encodes an antimicrobial protein that also mediates ROS homoeostasis and callose/lignin deposition during the response to F. solani infection.

Auxin and Root Hair Defective Six-Like 4 Regulate Azospirillum brasilense-Induced Root Hair Development in Arabidopsis

Hongcheng Zhao,Jiarui Xu,Lin Huang,Yizhe Ding,Xianyong Lin,Chengliang Sun

Beneficial microbiomes significantly modulate root hair morphogenesis, yet the underlying signalling pathways remain unclear. Through comprehensive phenotypic analysis, we demonstrated that Azospirillum brasilense , a well-known plant-growth-promoting rhizobacteria (PGPR), promotes root hair development by prolonging the elongation period. The promotion of root hair growth by A. brasilense is attributed to increased auxin accumulation in root apices and enhanced shootward transport from the root tip, which is mediated by the accumulation of the auxin importer auxin transporter protein 1 (AUX1) in the lateral root cap and the overlying epidermis. Chemical or genetic disruption of auxin signalling abolished bacteria-induced root hair responses. Furthermore, A. brasilense upregulated several root hair-related basic helix-loop-helix family transcription factors, including root hair defective 6 (RHD6) and RHD6-LIKE 1 (RSL1), which control root hair development through RSL4. Morphological analysis revealed that while the rsl2rsl4 double mutant remained hairless after inoculation, A. brasilense successfully rescued root hair growth in rhd6rsl1 . These findings suggest that A. brasilense can bypass RHD6 to activate RSL4, which then drives root hair development dependent on ROS. Collectively, our study uncovers the core signalling components modulated by A. brasilense to fine-tune root hair responses.

Cold-Induced Promoter Methylation Attenuates ScMYB7-Mediated Repression of the CBF Pathway: A Proposed Mechanism for Sugarcane Cold Adaptation

Weiwei Liu,Qiuyue Ou,Meichang Feng,Yue Li,Linghan Huang,Xuan Peng,Xiaoqiang Zhong,Guoqiang Huang,Hengbo Wang,Jinlong Guo

Cold stress is a critical environmental factor adversely affecting plant growth and development. As a tropical-origin crop constituting the primary global source of sucrose, sugarcane ( Saccharum spp. hybrid) exhibits particular vulnerability to suboptimal temperature conditions, with chilling injury substantially compromising its yield potential. Despite its agricultural significance, the molecular mechanisms underlying cold acclimation in sugarcane remain poorly characterized. Here, we report a cold-repressed 1R-MYB gene, ScMYB7 , from sugarcane, whose promoter ( pro-ScMYB7 ) contains multiple cis -acting elements, including two cytosine—phosphate diester—guanine (CpG) islands. Bisulfite sequencing PCR (BSP) and qPCR results showed that low-temperature treatment increased the methylation level of the CpG islands in the promoter to reduce the transcription of the ScMYB7 gene. The outcomes of GUS enzyme activity measurement of the promoter also indicated that low-temperature treatment inhibits the promoter's transcriptional activity, and methylation inhibitors could alleviate this inhibition. By generating transgenic Arabidopsis lines overexpressing ScMYB7 , ScMYB7's roles in regulating cold tolerance were investigated. We observed that the transgenic plants reduced cold tolerance, featured by a decreased survival rate after recovery, fluctuated physiological traits, and significantly lower expression levels of the C-repeat binding factor (CBF)-dependent pathway genes ( AtCBFs , AtCOR15 , and AtRD29A ). Yeast one-hybrid assays demonstrated direct binding of ScMYB7 to the AtCBF1 promoter, while repression of sugarcane ScDREB1A occurred indirectly. Furthermore, the dual-luciferase reporter assay indicated that ScMYB7 was able to inhibit the expression of the AtCBF1 or ScDREB1A . Taken together, we propose a model in which ScMYB7 acts as a repressor of cold tolerance via the CBF-dependent pathway. Under low-temperature stress, increased methylation of the pro-ScMYB7 promoter reduces ScMYB7 expression, thereby alleviating its repression of sugarcane DREB/CBF-type transcription factors and enhancing cold adaptation.

Synthetic Metabolites Derived From Host-Exudates Modulate the Bacterial Wilt Occurrence

Mengyuan Yan,Cunpu Qiu,Clement K. M. Tsui,Guilong Li,Kai Liu,Ming Liu,Meng Wu,Zhongpei Li

Root exudate-mediated microbial community assembly is critical for host health and growth. However, disease-induced variations in plant-microbe interactions remain ambiguous. Here, we explored the intrinsic distinctions and interactions between the secretion patterns and microbial community composition of diseased and healthy tomatoes. Our results showed that rhizosphere microbial communities in healthy and diseased tomatoes were dominated by potentially beneficial genera ( Bacillus , Rhodanobacter , Pseudolabrys , Gemmatimonas , Dongia , and Bradyrhizobium ) and putative pathogens ( Ralstonia and Neocosmospora ), respectively, which were correlated with differential metabolites. Further metabolite addition experiments demonstrated the differential regulation mechanism of specific metabolites on host health. Drawing inspiration from synthetic communities, we displayed a way to construct the synthetic metabolites (SynMets), and we found that the SynMets (cortisol, quercetin, pyridoxal, and levodopa), which were enriched in healthy tomatoes could resist diseases by inhibiting pathogen growth and constructing beneficial microbial communities. Conversely, the SynMets (pyridoxine, N-benzylformamide, isoquinoline, and xanthine) enriched in diseased tomatoes could result in microbial imbalances by facilitating pathogen growth, thereby causing disease occurrence and growth limitation. In total, our research indicated the importance of SynMets-mediated pathogen reproduction and microbial community assembly for plant health and lays a foundation for targeted regulation of rhizosphere microecology through synthetic metabolites.

Both Jasmonic Acid- and Abscisic Acid-Mediated Signalling Pathways Regulate the Ovicidal Defence of Plants Against Phloem-Feeding Insects

Qing Gao,Nuo Jin,Zhifan Shen,Jingran Guo,Haiping Lu,Shanjie Han,Wenhan Xiao,Jing Lu,Yonggen Lou

Plants perceive signals associated with herbivore eggs and in response initiate ovicidal defence. However, which phytohormone pathways regulate this defence and which defensive compounds dominate it remains largely unknown. Here, we found that the hatching rate of eggs of white-backed planthopper (WBPH) Sogatella furcifera was significantly lower on a japonica rice variety P81 than an indica rice variety NB44. When infested by WBPH, P81 plants showed higher jasmonic acid (JA) and abscisic acid (ABA) responses than did NB44 plants; moreover, P81 plants produced the ovicidal compound benzyl benzoate and exhibited higher levels of some flavonoids, phenolamides, and volatiles than were found in NB44 plants. Impairing the ABA-signalling pathway, especially the JA-signalling pathway in P81 plants enhanced the survival of WBPH eggs. Decreasing levels of some flavonoids and phenolamides in P81 plants promoted WBPH egg survival. In vitro bioassays revealed that both naringenin and sakuranetin promote the ovicidal effect of benzyl benzoate on WBPH. The results demonstrate that JA- and ABA-signalling pathways jointly regulate the rice ovicidal defence against WBPH, and that benzyl benzoate, as well as some other compounds, such as naringenin and sakuranetin, contribute to the mortality of WBPH eggs.

A structural variation in the promoter of the leucoanthocyanidin reductase gene AaLAR1 enhances freezing tolerance by modulating proanthocyanidin accumulation in kiwifruit (Actinidia arguta)

Shihang Sun,Xiujuan Qi,Zhenzhen Zhang,Leiming Sun,Ran Wang,Yukuo Li,Jinyong Chen,Hong Gu,Jinbao Fang,Miaomiao Lin

Proanthocyanidins (PAs) are important metabolites that enhance freezing tolerance of plants. Actinidia arguta , especially freezing-tolerant germplasms, accumulate abundant PAs in dormant shoots and thereby enhance freezing tolerance, but the underlying mechanism is unknown. In this study, we used two A. arguta with contrasting cold-resistant phenotypes, KL and RB, to explore the mechanisms in response to cold tolerance. We determined that a leucoanthocyanidin reductase gene ( AaLAR1 ) was more highly expressed in freezing-tolerant KL than in freezing-sensitive RB. Moreover, overexpressing AaLAR1 in kiwifruit promoted PAs biosynthesis and enhanced cold tolerance. The AaLAR1 promoters of various A. arguta germplasms differ due to the presence of a 60-bp deletion in cold-tolerant genotypes that forms a functional binding site for MYC-type transcription factor. Yeast one-hybrid and two-hybrid, dual-luciferase reporter, bimolecular fluorescence complementation and coimmunoprecipitation assays indicated that the AaMYC2a binds to the MYC-core cis -element in the AaLAR1 promoter with the assistance of AaMYB5a, thereby promoting PAs accumulation in the shoots of cold-tolerant kiwifruit. We conclude that the variation in the AaLAR1 promoter and the AaMYC2a–AaMYB5a– AaLAR1 module shape freezing tolerance in A. arguta . The identification of a key structural variation in the AaLAR1 promoter offers a new target for resistance breeding of kiwifruit.

Tea Aphid-Induced β-Elemene Biosynthesis by CsELE Enhances JA-Dependent Herbivore Resistance in Tea Plants

Lanxin Luo,Ting Gao,Yanni Deng,Mengyao Chai,Bo Li,Hao Ni,Kai Wang,Mengting Zhang,Yaojia Liu,Hao Jiang,Chuankui Song,Tingting Jing

Herbivore-induced plant volatiles play a crucial role in enhancing plant resistance and adaptability. However, the key volatiles triggering this response and their activation mechanisms remain largely unknown. Although tea aphids ( Toxoptera aurantii ) are common tea plantation pests, the underlying biochemical pathways by which aphid-induced volatiles in Camellia sinensis contribute to aphid resistance are not well understood. In this study, we measured jasmonic acid (JA) accumulation induced by aphid feeding using LC-MS and DESI-IMS analysis. We also identified a diverse array of aphid-induced volatiles through GC-MS. Notably, tea aphids stimulated β-Elemene accumulation, enhancing ‘Shuchazao’ resistance to tea aphids. We also evaluated the insecticidal activity of β-Elemene through contact toxicity and repellent bioassays. For the first time, we identified aphid-induced CsELE as a terpene synthase enzyme catalyzing the final step in β-Elemene biosynthesis from farnesyl pyrophosphate. Transient overexpression and gene silencing of CsELE affected β-Elemene accumulation, thereby influencing tea plant resistance to aphids. Furthermore, Dual-Luciferase and Y1H assays revealed that MYC2a, a core transcription factor in the JA signaling pathway, positively regulates CsELE expression. Overall, this study advances our understanding of the transcriptional regulation of tea volatiles in response to pest attacks, providing new insights into the ecological significance of terpene volatiles under biotic stress.

Microplastics and Nanoplastics Alter the Physicochemical Properties of Willow Trees and Lead to Mortality in Leaf Beetle Larvae

Peipei Zhu,Yanping Zhang,Mengqi Deng,Yuxin Zhang,Jing Luo,Runhua Han,Letian Xu

Polystyrene micro- and nanoplastics (MNPs) are increasingly found in terrestrial environments, posing risks across the food web. However, the potential impacts of MNPs transfer on plant-insect interactions remains largely unknown. In this study, consumption of willow plants ( Salix maizhokunggarensis ) exposed to 10.0 mg/L MNPs for 21 days inhibited survival and reduced body weight in Plagiodera versicolora larvae unlike those exposed to lower concentrations or shorter durations (0.1, 1.0 and 10.0 mg/L MNPs for 7 or 14 days). MNPs exposure increased lignin content and leaf thickness in willows, leading to decreased leaf consumption and increased mouthpart wear in P. versicolora larvae. Transcriptome and gut microbiota analyses revealed significant downregulation of genes related to digestion, intestinal homoeostasis, immunity, and growth/development along with profound alterations in gut microbiota composition. Notably, the abundance of the pathogenic bacterium Pseudomonas increased significantly. The gut barrier was disrupted, allowing gut bacteria to translocate into the haemolymph, accelerating larval mortality. Overall, MNPs altered plant physiology, making willow plants unsuitable for herbivore consumption and indirectly influenced herbivore survival by modulating gut bacteria. These findings offer novel insights into the cascading ecological effects and risks of MNPs, highlighting potential impacts on plant-herbivore interactions, biodiversity, and ecosystem health in terrestrial ecosystems.

Metabolic and Molecular Insights Into Nicotiana benthamiana Trichome Exudates: An Ammunition Depot for Plant Resistance Against Insect Pests

Jun-Xia Wang,Wen-Hao Han,Rui Xie,Feng-Bin Zhang,Zhi-Wei Ge,Shun-Xia Ji,Shu-Sheng Liu,Xiao-Wei Wang

Nicotiana benthamiana , a widely acknowledged laboratory model plant for molecular studies, exhibits lethality to certain insect pests and can serve as a dead-end trap plant for pest control in the field. However, the underlying mechanism of N. benthamiana 's resistance against insects remains unknown. Here, we elucidate that the lethal effect of N. benthamiana on the whitefly Bemisia tabaci arises from the toxic glandular trichome exudates. By comparing the metabolite profiles of trichome exudates, we found that 51 metabolites, including five O -acyl sugars ( O -AS) with medium-chain acyl moieties, were highly accumulated in N. benthamiana . Silencing of two O -AS biosynthesis genes, branched-chain keto acid dehydrogenase ( BCKD ) and Isopropyl malate synthase-C ( IPMS-C ), significantly reduced the O -AS levels in N. benthamiana and its resistance against whiteflies. Additionally, we demonstrated that the higher expression levels of BCKD and IPMS-C in the trichomes of N. benthamiana contribute to O -AS synthesis and consequently enhance whitefly resistance. Furthermore, overexpression of NbBCKD and NbIPMS-C genes in the cultivated tobacco Nicotiana tabacum enhanced its resistance to whiteflies. Our study revealed the metabolic and molecular mechanisms underlying the lethal effect of N. benthamiana on whiteflies and presents a promising avenue for improving whitefly resistance.

Linalool and 1,8-Cineole as Constitutive Disease-Resistant Factors of Norway Spruce Against Necrotrophic Pathogen Heterobasidion Parviporum

Kai Wang,Wenzi Ren,Liang Hong,Qingao Wang,Rajendra Ghimire,Matti Haapanen,Minna Kivimäenpää,Pengfei Wu,Xiangqing Ma,Fred O. Asiegbu

Norway spruce is an important coniferous species in boreal forests. Root and stem rot diseases caused by the necrotrophic pathogen Heterobasidion parviporum threaten the wood production of Norway spruce which necessitates the search for durable control and management strategies. Breeding for resistant traits is considered a viable long-term strategy. However, identification of potential resistant traits and markers remains a major challenge. In this study, short-term disease resistance screening was conducted using 218 Norway spruce clones from 17 families. Disease resistance was evaluated based on the size of necrosis lesion length following infection with the pathogen. A subset of needles/branches from clones with small (partial resistant) or large (susceptible) lesions were used for terpene analysis and transcriptomic profiling. The results revealed that the content of monoterpene linalool and 1,8-cineole and their respective encoded genes were significantly more abundant and highly expressed in the partial resistant group. Furthermore, linalool and 1,8-cineole were demonstrated to have inhibitory effect on the growth of the pathogen H. parviporum , with morphological distortion of the hyphae. RNAseq analysis revealed that transcript of pathogen genes involved in the regulation of carbohydrate metabolism and stress responses were significantly decreased in presence of the terpenes. The results suggest the relevance of monoterpenes together with jasmonic acid precursor and some genes involved in phenylpropanoid biosynthesis, as constitutive tolerance factors for Norway spruce tolerance against necrotrophic pathogen. The high level of necrosis related cell death gene expression might be factors critical for host susceptibility and disease development.

GLR36T807I Mutation of Casuarina equisetifoli Is Associated With a Decreased JA Response to Insect Feeding by Lymantria xylina

Huan Chang,Chengli Li,Tengfei Zhu,Shouping Cai,Jie Chen,Fangfang Zhan,Liqiong Zeng,Yu Fang,Gongfu Ye,Jian Li,Jun Su

Lymantria xylina is the most important defoliator, damaging the effective coastal windbreak tree species Casuarina equisetifolia . However, the underlying genetic mechanisms through which C . equisetifolia responds to L . xylina attacks remain unknown. Here, we compared the transcriptional, phytohormone and metabolic differences between susceptible (S) and resistant (R) C . equisetifolia cultivars in response to L . xylina feeding. The main L . xylina -induced resistance in C . equisetifolia was a jasmonate (JA) response and JA synthesis was highly induced by L . xylina feeding at both the transcriptional and metabolic levels, thus promoting flavonoid accumulation. The JA response was highly activated by L . xylina feeding on the R but not in the S cultivar, although the JA signalling pathway was intact in both cultivars. We found a single amino acid mutation in the homologues of glutamate receptor-like protein 3.6 (CeGLR3.6 T807I ) in the S cultivar. Compared with the GLR3.6 homologues in the R cultivar, phosphorylation of CeGLR3.6 T807I was not induced by insect feeding, leading to a decreased JA response in the S cultivar. Collectively, this study provides new insights into the function of CeGLR3.6 in regulating the JA response of C . equisetifolia to L . xylina feeding.

Low nitrate alleviates iron deficiency by regulating iron homeostasis in apple

Wei-Jian Sun,Jiu-Cheng Zhang,Xing-Long Ji,Zi-Quan Feng,Xun Wang,Wen-Jing Huang,Chun-Xiang You,Xiao-Fei Wang,Yu-Jin Hao

Iron (Fe) is an essential element for plant growth, development and metabolism. Due to its lack of solubility and low bioavailability in soil, Fe levels are usually far below the optimum amount for most plants' growth and development. In apple production, excessive use of nitrogen fertilizer may cause iron chlorosis symptoms in the newly growing leaves, but the regulatory mechanisms underlying this phenomenon are unclear. In this study, low nitrate (NO 3 − , LN) application alleviated the symptoms of Fe deficiency and promoted lower rhizosphere pH, which was beneficial for root Fe acquisition. At the same time, LN treatment increased citrate and abscisic acid accumulation in roots, which promoted Fe transport from root to shoot and maintained Fe homeostasis. Moreover, qRT-PCR analysis showed that nitrate application caused differential expression of genes related to Fe uptake and transport, as well as transcriptional regulators. In summary, our data reveal that low nitrate alleviated Fe deficiency through multiple pathways, demonstrating a new option for minimizing Fe deficiency by regulating the balance between nutrients.
