Microplastics (MPs) can adsorb and desorb organic pollutants, which may alter their biotoxicities. Although the toxicity of perfluorooctane sulfonate (PFOS) and its alternative 6:2 chlorinated polyfluorinated ether sulfonate (F-53B) to organisms has been reported, the comparative study of their combined toxic effects with MPs on aquatic organisms is limited. In this study, adult female zebrafish were exposed to 10 μg/L PFOS/F-53B and 50 μg/L MPs alone or in combination for 14 days to investigate their single and combined toxicities. The results showed that the presence of MPs reduced the concentration of freely dissolved PFOS and F-53B in the exposure solution but did not affect their bioaccumulation in the zebrafish liver and gut. The combined exposure to PFOS and MPs had the greatest impact on liver oxidative stress, immunoinflammatory, and energy metabolism disorders. 16S rRNA gene sequencing analysis revealed that the combined exposure to F-53B and MPs had the greatest impact on gut microbiota. Functional enrichment analysis predicted that the alternations in the gut microbiome could interfere with signaling pathways related to immune and energy metabolic processes. Moreover, significant correlations were observed between changes in gut microbiota and immune and energy metabolism indicators, highlighting the role of gut microbiota in host health. Together, our findings demonstrate that combined exposure to PFOS/F-53B and MPs exacerbates liver immunotoxicity and disturbances in energy metabolism in adult zebrafish compared to single exposure, potentially through dysregulation of gut microbiota.

As emerging alternatives to perfluorooctane sulfonate (PFOS), 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (6:2 Cl-PFESA) and sodium p -perfluorous nonenox-benzenesulfonate (OBS) were frequently detected in the four freshwater fish species collected from Poyang Lake. Median concentrations of 6:2 Cl-PFESA and OBS in fish tissues were 0.046–6.0 and 0.46–5.1 ng/g wet weight, respectively. The highest concentrations of 6:2 Cl-PFESA was found in fish livers, whereas OBS was found in the pancreas, brain, gonads, and skin. The tissue distribution pattern of 6:2 Cl-PFESA is similar to that of PFOS. The tissue/liver ratios of OBS were higher than those of PFOS, suggesting that OBS has a greater tendency to transfer from the liver to other tissues. The logarithmic bioaccumulation factors (log BAFs) of 6:2 Cl-PFESA in three carnivorous fish species were greater than 3.7, whereas those of OBS were less than 3.7, indicating that 6:2 Cl-PFESA had a strong bioaccumulation potential. Notably, sex- and tissue-specific bioaccumulation of OBS has also been observed in catfish. Most tissues (except the gonads) exhibited higher OBS concentrations in males than in females. However, no differences were found for 6:2 Cl-PFESA and PFOS. Maternal transfer efficiency of OBS was higher than that of 6:2 Cl-PFESA and PFOS in catfish ( p < 0.05), indicating that OBS presents a higher risk of exposure to males and offspring through maternal offloading.

Perfluorooctane sulfonic acid (PFOS), a persistent environmental pollutant, has been associated with decreased birth weight. The dysregulation of long non-coding RNA (lncRNA) H1 9 has been implicated in pregnancy complications such as intra-uterine growth retardation (IUGR), preeclampsia (PE), however, the expression and function of H19 in PFOS-exerted detrimental effects in the placenta remains to be unveiled. Here, we explored the role of H19 in PFOS-induced placental toxicity. Results showed that PFOS caused decreased cell growth in human HTR-8/SVneo cells. Expression of H19 was increased, while miR-19a and miR-19b expression were decreased in mice placenta tissues and in HTR-8/SVneo cells exposed to PFOS. A significant hypomethylation was observed at the H19 promoter in the placentas of mice that were gestational exposed to high dose of PFOS. H19 was confirmed to bind with miR-19a and miR-19b, targeting SMAD4 . Furthermore, H19 appeared to partially improve the cell growth of HTR-8/SVneo cells exposed to PFOS via upregulation of miR-19a and miR-19b. In summary, our findings revealed that H1 9/miR-19a and miR-19b/SMAD4 axis exerted important functions in PFOS-induced placenta cell toxicity.

Purpose Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) in soils pose a severe threat to the soil ecosystem and human health. Soil aggregates play a critical role in the migration and transformation of pollutants. Materials and methods In this study, soil with different properties (Fluvisol and Planosol) spiked with PFOA/PFOS were incubated for 90 days, and then divided into five particle size fractions by wet sieving with low loss. The concentrations of PFOA and PFOS in soils and fractions were extracted using ultrasound-assisted extraction method with methanol as an extractant. The PFOA and PFOS were analyzed using high-performance liquid chromatography-tandem mass spectrometry (HPLC–MS/MS). Results and discussion The results showed that the ability of the soil aggregates to carry PFOA and PFOS initially decreased and then increased with decreasing particle sizes. Moreover, organic matter was the main factor influencing the PFOA and PFOS distribution in the soil aggregates, whereas soil types did not affect the accumulation capacity of PFOA and PFOS on soil particles. The macroaggregates (2000–250 µm) showed the highest accumulation capacity of PFOA and PFOS, suggesting high load of PFOA and PFOS. Additionally, the microaggregates (< 15 µm) played a dominant role in the PFOA and PFOS distribution due to the high proportion of this fraction in soil samples (51.73 ~ 62.47%). Conclusions Significantly bipolar immigrations of PFOA and PFOS into macroaggregates and microaggregates in soils were observed. This phenomenon can be explained by the fact that the particulate organic carbon (POM) can sorb the PFOA and PFOS in the macroaggregates, while the presence of SOC (highly humified) may increase the PFOA and PFOS contents in the microaggregates. A Chinese perfluorooctane sulfonate (PFOS) substitute frequently detected in the environment, 6:2 chlorinated polyfluorinated ether sulfonate (F-53B), has a similar structure to PFOS and it is proposed to cause thyroid dysfunction. To further confirm this hypothesis, the effects of F-53B on the thyroid endocrine system and underlying mechanisms were investigated in vitro and in vivo using rat pituitary GH3 cells and developing zebrafish, respectively. In GH3 cells, F-53B enhanced cell proliferation in a dose-dependent manner, indicative of thyroid receptor agonistic activity. In zebrafish larvae, F-53B exposure induced significant developmental inhibition and increased thyroxine (T 4 ) but not 3,5,3′-triiodothyronine (T 3 ) levels accompanied by a decrease in thyroglobulin (TG) protein and transcript levels of most genes involved in the hypothalamic-pituitary-thyroid (HPT) axis. Interestingly, T 4 levels remained significantly increased while TG protein and gene transcription levels were markedly upregulated after depuration. Molecular docking studies revealed that F-53B binds to transthyretin (TTR) by forming hydrogen bonds with Lys123 and Lys115, thereby interfering with thyroid hormone homeostasis. Our collective in vitro , in vivo and in silico studies provide novel evidence that F-53B disrupts the thyroid endocrine system at environmentally relevant concentrations, which cannot be recovered after depuration. Given the persistence of F-53B in the environment, the long-term consequences of thyroid hormone disruption by this chemical warrant further investigation.

6:2 Chlorinated polyfluorinated ether sulfonate (F-53B) and sodium p -perfluorous nonenoxybenzene sulfonate (OBS) are widely used as perfluorooctane sulfonate (PFOS) alternatives in the Chinese market. Here, adult zebrafish were chronically exposed to 1 μM PFOS, F-53B, and OBS for 21 days to investigate the comparative immunotoxicity of these three per- and polyfluoroalkyl substances (PFAS). PFOS induced more severe oxidative stress in the liver than F-53B and OBS, and these three PFAS induced similar anti-inflammatory effects by repressing the expression of pro-inflammatory cytokines. The intestinal microbiota analysis showed that the relative abundance of Plesiomonas , Aeromonas , Cetobacterium , Shewanella , and Vibrio changed with the same trend in the three PFAS treatment groups. Furthermore, the PFAS increased the expression of hepcidin , muc , the immune-related genes mpo and saa , and decreased the expression of the tight junction-related gene occ in the intestine; moreover, villus height of the intestine was reduced after PFAS exposure, which indicated the functional disruption of the intestine. In particular, the significant correlation between the changed intestinal microbiota and liver and intestinal indicators also suggested the interaction between the immune system and intestinal microbiota. Taken together, our results indicate that exposure to PFOS and its alternatives F-53B and OBS can induce hepatic immunotoxicity associated with intestinal microbiota dysbiosis in adult zebrafish.

Perfluoroalkyl substances (PFASs) are persistent chemicals in the environment. So far, little is known about their uptake potential in wetland plants. Here, we investigated the uptake and translocation of perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS) in eight common wetland plants, namely, Canna indica (Ci), Thalia dealbata (Td), Cyperus alternifolius (Ca), Phragmites australis (Pa), Arundo donax (Ad), Pontederia cordata (Pc), Cyperus papyrus (Cp), and Alisma orientale (Ao) by hydroponic experiments and visualized their tissue- and cell-level distribution with desorption electrospray ionization mass spectrometry (DESI-MS) and transmission electron microscopy equipped with energy-dispersive spectroscopy (TEM-EDS). The results showed that the PFASs accumulated in plants accounted for 1.67–16.7% of the total mass spiked into the hydroponic systems, and PFOS accumulated largely in roots (48.8–95.8%), while PFOA was stored mostly in the aboveground part (29.3–77.4%). DESI-MS and TEM-EDS analysis showed that PFASs in Ci, Td, Pa, and Ca were transported from the hydroponic solution to the root cortex via both apoplastic (e.g., across cell walls and/or intercellular spaces) and symplastic routes (e.g., across plasma membranes or via plasmodesmata) and further to the vascular bundle via symplastic route in Td and Pa and via both routes in Ci and Ca. These two chemicals were transported from roots to stems mainly through the cortex in Td and through both the cortex and vascular bundles in Ci and Ca.

Perfluorooctanoic acid (PFOA) and Perfluorooctane sulfonate (PFOS) are two typical perfluorinated compounds (PFCs) that poss potential ecological toxicity . In this work, a fluorescence and resonance light scattering (RLS) dual-readout strategy for the detection of PFCs at picomole level based on the water-soluble CdTe quantum dots (CdTe QDs) has been proposed. It is found that the CdTe QDs exhibit a quenching in the presence of PFCs and thus serve as useful probes for PFCs. The linear ranges are 0.032 – 10.0 nM with a limit of detection(LOD) of 32.02 pM for PFOA and 0.044 – 15.0 nM with a LOD of 43.96 pM for PFOS, respectively. Meanwhile, PFCs can form complexes with CdTe QDs in acid medium , resulting in remarkable RLS signals. The enhanced RLS intensities are in proportion to the concentrations of PFOA and PFOS, respectively. And the linear ranges are 0.048 – 5.0 nM with a LOD of 47.78 pM for PFOA, and 0.057 – 5.0 nM with a LOD of 56.72 pM for PFOS, respectively. This dual-mode detection increases the reliability of the measurement. The proposed method is simple, sensitive and cost-effective, with potential applications in environmental monitoring and assessment.

Widespread detection in soils and sediments underscores the potential threats posed by persistent, bioaccumulative and toxic perfluorooctane sulfonate (PFOS) to ecosystems and organisms. Nevertheless, the formidable energy of the C-F bond imparts stability and hampers degradation. This study investigates the potential of boron carbide (B 4 C), a hard-ceramic material often utilized in armor and abrasion contexts, for degrading solid-phase PFOS through ball milling. Following a 60-minute reaction period, PFOS degradation reached 100 %, while desulfurization stabilized at 13.9 %. Defluorination initially rose to 23.7 % but subsequently displayed a gradual decline. Fluoride ions, once released, were secured and the stable reservoir surface of B 4 C during the ball-milling process, leading to the formation of B-C-B-F chemical bonds, which was confirmed through characterization by XPS, FTIR, and solid-state NMR analyses. Furthermore, intermediate byproducts formed during the degradation of PFOS, encompassing perfluoroalkyl sulfonic acids (PFSAs) and perfluoro carboxylic acids (PFCAs), were identified. Eventually, the generation of free electrons, contributing to the facilitation of degradation for both PFOS and its intermediates, was confirmed through the DPPH experiment. This identification underscores the intricacy of the PFOS degradation pathway, which encompasses the generation and subsequent breakdown of these intermediates. These discoveries hold significance for pioneering sustainable and inventive methods to combat PFOS pollution across diverse applications. Furthermore, they enrich the comprehension of how mechanical forces can be leveraged for the eco-friendly removal of PFOS contaminants.

The F and Sb co-doped Ti/SnO 2 electrode containing Sn-Sb interlayer was innovatively synthesized for electro-oxidation (EO) removal of perfluorooctane sulfonate (PFOS) in water. Stronger oxidation capability and remarkably longer lifetime were performed for Ti/Sn-Sb/SnO 2 -F-Sb electrode than these similar electrodes in this study without Sn-Sb interlayer and F/Sb doping, which could removed more than 99% of PFOS ( C 0 = 100 mg L −1 ) after 120-min electrolysis. The attribute of Ti/Sn-Sb/SnO 2 -F-Sb anode and its EO mechanism were surveyed by XRD, XPS, SEM, and LSV. F and Sb co-doping could improve the stability and EO capability of the electrode, which resulted from that the smooth surface was formed conveniently to produce physically adsorbed hydroxyl radical and lead to relatively high OEP. The operating parameters of EO process were also investigated including current density, pH value, and stirring speed, which revealed the effect of these parameters on PFOS decomposition. Additionally, the mineralization parameters, TOC and F − concentration, as well as short-chain perfluorocarboxylic acids were analyzed in solution during the electrolysis. Consequently, the EO pathway of PFOS was proposed through the detection of intermediates. These results manifest that Ti/Sn-Sb/SnO 2 -F-Sb anode is prospective to effectively decompose PFOS in wastewater.

In this study, a detailed analysis of 20 per- and polyfluoroalkyl substances (PFAS), including 13 perfluorocarboxylic acids (PFCAs) (C4–C18), four perfluorosulfonic acids (PFSAs) (C4, C6, C8, C10), and three polyfluoroalkyl substances, in different environmental media of Fu River, which was the main sewage storage body, and located near the Tianhe Airport in Wuhan, China. In the surface water samples, short-chain PFAS, perfluorobutanesulfonic acid (PFBS) and perfluorobutanoic acid (PFBA) were the most prevalent, and highest concentrations of 168 ng/L and 49.7 ng/L, respectively. However, in the sediment samples, PFBS and perfluorooctane sulfonate (PFOS) were the most prevalent, and highest concentrations of 840 ng/kg and 1510 ng/kg, respectively. Meanwhile, PFOS was the most prevalent substance, with the highest concentrations measured at 23.3 μg/kg in egg yolk and 28.1 μg/kg in fish, accounting for proportions of 82% and 88%, respectively. Two new short-chain substitutes of PFOS, 6:2 clorinated polyfluoroalykyl ether sulfonate (6:2 Cl-PFESA) and 8:2 clorinated polyfluoroalykyl ether sulfonate (8:2 Cl-PFESA), emerged in sediment and fish. The PFAS in eggs, fish, and vegetables of Fu River Basin poses a potential health risk to residents according to the suggested value of the European Food Safety Agency.

Background: Perfluorooctane sulfonate (PFOS) is a persistent, widely present environmental pollutant, and its toxicity to male reproduction has gradually attracted attention. Flaxseed oil (FO) is a dietary oil abundant in α-linolenic acid and has been demonstrated to possess multiple health benefits. However, whether FO protects against PFOS-induced testicular injury and its mechanism remain unclear. Methods: C57/BL6 mice were gavaged with different concentrations of FO or PFOS (10 mg kg−1) for 28 days. Blood and testicular tissues were collected for histopathology, proteomics, and biochemical and molecular analyses. Results: Our results showed that FO supplementation significantly attenuated PFOS-induced testicular injury, as indicated by histopathological changes, decreased oxidative stress level, increased sperm count, decreased rate of sperm malformation, and improved functional markers of spermatogenesis. Proteomic analysis showed that differentially expressed proteins were notably enriched in spliceosome pathways. Machine learning algorithms were used to screen the hub gene, and PRPF3 and PUF60 proteins were found to be important for FO to exert protective benefits to testicular injury. Western blot results confirmed that FO supplementation could increase the protein expression of PRPF3 and decrease the protein expression of PUF60 in PFOS-exposed mice. Conclusions: This study revealed that FO can alleviate PFOS-induced testicular dysfunction by regulating RNA alternative splicing. The spliceosome-related proteins PRPF3 and PUF60 may be the potential targets for FO to alleviate PFOS-induced testicular injury. FO supplementation may be an effective dietary intervention to prevent adverse effects of PFOS on testes.
