An efficient method for the synthesis of a polymer brush via click chemistry and its ultrasensitive electrochemical detection of AFP

Dongcheng Yang,Jing Wang,Hou Chen,Hui Xu,Wenxiang Wang,Huawei Yang,Donglei Wei,Liangjiu Bai

A sensitive sandwich-type electrochemical immunosensor based on polyacrylonitrile-g-poly(hydroxyethyl methacrylate) (PAN-g-PHEMA)/graphene oxide (GO) composites was prepared for the ultrasensitive detection of tumor markers (alpha fetal protein, AFP as a model). The functional polymer brush was efficiently prepared by the union of cyano click chemistry and controlled/“living” radical polymerization (CRP). The designed immunosensor showed a low limit of detection (LOD, ∼0.285 pg mL−1), with a wide detection range (2.5 × 101 to 2.5 × 104 pg mL−1) (R2 = 0.9876). The electrochemical immunosensor also exhibited high sensitivity, favorable stability, excellent reproducibility and good selectivity towards AFP. Moreover, the immunosensor could be applied to the analysis of AFP in serum samples with satisfactory results. This present work may provide a general model for the detection of tumor markers based on sandwich-type immunosensors.

Fabrication of a Modified Polyethersulfone Membrane with Anti-Fouling and Self-Cleaning Properties from SiO2-g-PHEMA NPs for Application in Oil/Water Separation

Jun Yin

To prepare anti-fouling and self-cleaning membrane material, a physical blending modification combined with surface grafting modification has been carried out; first, poly (2-hydroxyethyl methacrylate) grafted silica nanoparticles (SiO2-g-PHEMA NPs) were synthesized using surface-initiated activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP) and used as a blending modifier to fabricate a polyethersulfone (PES)/SiO2-g-PHEMA organic–inorganic membrane by the phase-inversion method. During the membrane formation process, hydrophobic PES segments coagulated immediately to form a membrane matrix, and the hydrophilic SiO2-g-PHEMA NPs migrated spontaneously to the membrane surface in order to reduce interfacial energy, which enhanced the hydrophilicity and anti-fouling properties of the PES/SiO2-g-PHEMA membrane. Importantly, the membrane surface contained abundant PHEMA segments, which provided active sites for further surface functionalization. Subsequently, the carboxyl-terminated fluorocarbon surfactant (fPEG-COOH) composed of hydrophilic polyethyleneglycol segments and low-surface-energy perfluorinated alkyl segments was synthesized via the esterification of fPEG with succinic anhydride. Lastly, the PES/SiO2-g-PHEMA/fPEG membrane was prepared by grafting fPEG-COOH onto surface of the PES/SiO2-g-PHEMA. Thus, a versatile membrane surface with both fouling-resistant and fouling-release properties was acquired. The PES/SiO2-g-PHEMA/fPEG membrane has a large oil–water flux (239.93 L·m−2·h−1), almost 21 times that of PES blank membrane and 2.8 times of the PES/SiO2-g-PHEMA membrane. Compared with the unmodified PES membrane, the flux recovery ratio increased from 45.75% to 90.52%, while the total flux decline ratio decreased drastically from 82.70% to 13.79%, exhibiting outstanding anti-fouling and self-cleaning properties. Moreover, the grafted fPEG segments on the membrane surface show excellent stability due to the presence of stable chemical bonds. The grafted segments remain at the surface of the membrane even after a long shaking treatment. This suggests that this PES/SiO2-g-PHEMA/fPEG membrane material has potential for application in oil/water separation. Keywords:modified polyethersulfone membrane;oil–water emulsion separation;fouling-resistance;self-cleaning;carboxyl-terminated fluorocarbon surfactant

Long-Lasting Anti-Swelling Sustained-Release Estradiol Hydrogel for Promoting Vaginal Wound Healing

Tianyue Zhang,Hongyi Lv,Yijing Zhang,Lingyun Yu,Yonghong Li,Hechun Yan,Chenyan He,Dongmei Zhao,Lijuan Zhao,Yuedong He,Yi Wang,Zhongyi Zhu

In both clinical and research settings, the rapid healing of vaginal injuries remains a significant challenge globally. Conventional treatment methods often involve extended recovery periods and increased risk of retrograde infections, causing significant pain and discomfort for patients. To address this issue, a mechanically robust and stable poly(hydroxyethyl methacrylate) (PHEMA)/alginate hydrogel loaded with estrogen has been developed to facilitate vaginal wound repair. This hydrogel is formed by an ionically crosslinked alginate network and a covalently crosslinked PHEMA network, which effectively enhances network strength and dissipates energy, leading to excellent fatigue resistance and anti-swelling behavior. Its exceptional mechanical stability and anti-swelling properties prevent local adhesions and mucosal ischemic necrosis. Additionally, the hydrogel sustains the release of estradiol, promoting vaginal epithelial proliferation and collagen deposition, thereby significantly accelerating vaginal injury healing. Furthermore, the hydrogel eliminates the need for frequent dressing changes, reducing the risk of retrograde infections. The hydrogel demonstrates outstanding biocompatibility both in vitro and in vivo. These combined advantages enable the hydrogel to reduce vaginal recovery time, prevent adhesions, and lower the risk of infection. Therefore, the estrogen-loaded PHEMA/alginate hydrogel demonstrates great potential for achieving satisfactory healing in cases of vaginal injury.

Photonic Crystal Hydrogels Fabricated from Nanoparticles of Fe3O4/SiO2 with Programmable Colors through Photopolymerization for Applications as Anticounterfeiting Applications

Jia Deng,Shaoge Fu,Yifan Zhong,Ming Jiang,Tao Chen,Haili Zhao

Photonic crystal (PC) hydrogels with unique micro/nanostructures have excellent prospects in many fields, such as sensing, displays, anticounterfeiting, information encryption, and adaptive camouflage. Herein, magnetic core–shell Fe3O4/SiO2 nanoparticles with a controlled size were synthesized, followed by PC hydrogels of arbitrary structures that were fabricated via the magnetically inducing self-assembly of magnetic nanoparticles in the poly(ethylene glycol) diacrylate (PEGDA) pregel solution and a subsequent digital micromirror device (DMD)-based UV polymerization process. By controlling the size of nanoparticles and magnetic field intensity, various PC hydrogel patterns with tunable structural colors were fabricated. Combining magnetic field control with DMD-based light modulation, high-throughput preparation of structural color PC hydrogels was achieved. Furthermore, the Fe3O4/SiO2 nanoparticles were assembled into poly(2-hydroxyethyl methacrylate) (PHEMA) with solvent responsiveness. Interestingly, the obtained PC hydrogels could swell in ethanol, resulting in a change in the structural color. Meaningfully, a dual-component PC hydrogel consisting of a PEGDA pattern and a PHEMA background was fabricated, and the invisible pattern in the initial state could be displayed and hidden through cyclic exposure to ethanol and ethylene glycol. The presented strategy for constructing the PC hydrogels from Fe3O4/SiO2 nanoparticles could provide valuable insights for the development of intelligent structural color materials, which exhibit significant potential in applications such as information storage, visual sensing, and anticounterfeiting.

Confined Channels Induced Coalescence Demulsification and Slippery Interfaces Constructed Fouling Resist-Release for Long-Lasting Oil/Water Separation

Yajie Ding,Bihan Hu,Liwei Zhuang,Jianqiang Wang,Jindan Wu,Fu Liu,Jiping Wang

Superwetting membranes based on steric exclusion and affinity difference have drawn substantial interest for oil/water separation. However, the state-of-the-art membranes fail to literally sort out fouling and permeability decline and so limit their viability for long-term separation. Inspired by Dayu’s philosophy of “draining rather than blocking water”, herein, we achieve a long-lasting and efficient separation for viscous emulsions by designing poly(hydroxyethyl methylacrylate) (PHEMA)- and polydimethylsiloxane (PDMS)-compensated poly(vinylidene fluoride) membranes based on coalescence demulsification via chemical coordination phase separation. The symmetric and torturous microporous structure facilitated oil spatial confining and coalescence demulsification, while the synergistic compensation of PHEMA and PDMS coordinated the fouling resist and release properties, which was confirmed by multichannel confocal laser scanning microscopy. The developed membrane shows an unprecedented permeability half-life (τ) for viscous emulsions (e.g., decamethylcyclopentasiloxane, soybean oil paraffin, n-hexadecane, and isooctane) under cross-flow operation, far more beyond common superwetting membranes under applied bench-scale dead-end filtration. Our technique for designing “nonfouling” membranes opens up opportunities for advancing next-generation membranes for oil/water separation.

The thickness of poly-phenoxyethyl methacrylate brush interferes with cellular behavior and function of myofibers

Tao Huang,Jiangwei Xiao,Shuhao Wang,Zhaohong Liao,Tao Huang,Ruicai Gu,Junhua Li,Gang Wu,Hua Liao

Introducing or grafting molecules onto biomaterial surfaces to regulate muscle cell destination via biophysical cues is one of the important steps for biomaterial design in muscle tissue engineering. Therefore, it is important to understand the interaction between myoblasts and myofibers with substrates modified by biomimetic layer with different thicknesses. In this study, we used a surface-induced atom transfer radical polymerization method to synthetize and graft poly-phenoxyethyl methacrylate (PHEMA) brushes having different lengths on the glass substrates. C2C12 myoblasts were seeded on the PHEMA brushes and differentiated using horse serum, for analyzing the sensibility of muscle cells to feel environment changing, and further investigating whether the depths of grafting layer on the biomaterial surface are important factors in regulating muscle cell behaviors. Our results demonstrated that on the thicker PHEMA brushes surface (200 and 450 nm), C2C12 myoblasts showed a better survival and proliferation and were favorable for cell fusion and myotube formation. Furthermore, myofibers survived on the thicker brushes were more functional and upregulated cytoskeleton proteins (tubulin, vimentin, and vinculin) and FAK levels, and enhanced the expression levels for mechanical stress molecules (HGF, NOS-1, and c-Met). These results suggest that grafting thickness of PHEMA layer on the substrate led to the myoblasts/myofiber behavior change, which would be valuable for the design and preparation of the modified layer on muscle tissue engineering scaffolds. © 2019 Wiley Periodicals, Inc. Non-Swelling and Anti-Fouling MXene Nanocomposite Hydrogels for Underwater Strain Sensing

Shaoshuai He,Xia Sun,Zhihui Qin,Xiaoru Dong,Haitao Zhang,Mingyue Shi,Fanglian Yao,Hong Zhang,Junjie Li

Flexible wearable sensors are of interest for underwater applications such as aquatic robots and marine exploration. Non-swelling hydrogels would be a preferable candidate for underwater sensing with stable sensing performance. Herein, hydrogels comprised of MXene and polyhydroxyethyl methacrylate (PHEMA) are designed and manufactured. Owing to the synergistic effect of phase separation zones and hydrophilic/hydrophobic interaction, MXene/PHEMA hydrogels exhibit non-swelling performance in various liquid media (e.g., water, seawater) and effectively avoid the adhesion of proteins and bacteria. In addition, MXene/PHEMA hydrogels exhibit a large sensing range (more than 400%), high tensile strength (0.54 MPa), and skin-compliance modulus (120 kPa). Furthermore, the resulted hydrogel-based sensors possess superior sensibility (4.42 at 150–450% strain), fast responsiveness (200 ms), and durable sensing ability (over 1000 cycles). The hydrogel-based sensors can accurately monitor human motions (both large and subtle motions) in air and in various liquid media. Benefiting from the non-swelling and anti-fouling ability, the hydrogels could maintain high mechanical strength and cyclic stability after being immersed in water for 30 days, and precisely detect the movement of human and shark models underwater. It is conceivable that non-swelling and anti-fouling MXene/PHEMA hydrogels provide a feasible material to construct electronic skins for various harsh liquid environments.
