Preparation of pH-sensitive nanoparticles of poly (methacrylic acid) (PMAA)/poly (vinyl pyrrolidone) (PVP) by ATRP-template miniemulsion polymerization in the aqueous solution

Chen Guoxia,Liu Junxiu,Yang Yadong,Zhang Lijuan,Wu Min,Ni Henmei

Employing polymethacrylic acid (PMAA) as the template and N -vinyl pyrrolidone (N-VP) as monomer, the ATRP-template miniemulsion polymerization was carried out in the aqueous medium by using MBP/CuBr/bpy as initiator. The results were characterized by dynamic light scattering (DLS), transmission electron microscope (TEM), and gel permeation chromatography (GPC). It was observed that the stable particles exhibited amphoteric pH sensitivity, namely that in the range of pH 3.0 to 5.0, the particles precipitated, whereas beyond the range the particles were stable and swollen as pH varied. Moreover, the pH range was variable according to the molecular weight of PVP. The results of GPC indicated that the molecular weight of template polymer PMAA was duplicated by the daughter polymer PVP. Being noncross-linked, unlike the common microgels, the hydrodynamic diameter dramatically increased in a very narrow pH range, e.g., pH 5.5 –6 and 2.0–2.5. Finally, the nanoparticles of PMAA/PVP were applied for the controlled release of rifampicin (RFP) and doxorubicin (DOX).

Polydopamine-Based Composite Nanoparticles with Redox-Labile Polymer Shells for Controlled Drug Release and Enhanced Chemo-Photothermal Therapy

Tian Yefei,Lei Miao

Photothermal therapy (PTT) that utilizes phSUPPotothermal conversion agents (PTC) to ablate tumor under NIR light irradiation has attracted increasing attention due to its excellent therapeutic efficacy and improved target selectivity. Herein, a novel core-shell nanoparticle based on disulfide-crosslinked poly(methacrylic acid) (PMAA) layer coated polydopamine (PDA) particle has been successfully synthesized by precipitation polymerization. For these PDA@PMAA composite nanoparticles, PDA core exhibits high photothermal efficacy, meanwhile, the redox-labile PMAA shell serves as carriers to encapsulate anticancer drugs and selectively release them. Due to the characteristic of the disulfide bond, PMAA shell occurs at selective degradation as well as controlled drug release upon entering cancer cells. Moreover, the DOX-loaded PDA@PMAA nanoparticles demonstrated a synergistic effect, which shows a significantly improved inhibition effect against cancer cells by the combination of photothermal therapy and traditional chemotherapy with low drug dosage and short laser irradiation in an in vitro study.

Vapor-based hybrid grafting of heparin-like coating for dialysis membranes with enhanced hemocompatibility

Ruhao Zhang,Xiao Shi,Xiaocheng Huang,Jiaqi Zhao,Peng Lu,Yu He,Fu Liu,Wenna Liu,Yumin Ye

Heparin is commonly used to improve the hemocompatibility of biomaterials, but high doses of heparin may cause significant side effect in clinical applications. Thus, heparin-mimicking surfaces have been urgently explored to reduce the use of heparin. In this work, heparin-like coating was deposited on polylactide (PLA) dialysis membranes using one-step hybrid grafting of a carboxyl-enriched poly(methacrylic acid) (pMAA) coating via initiated chemical vapor deposition (iCVD). The hybrid grafting was conducted by depositing a prime layer of poly(methacrylic acid- co -ethylene glycol dimethacrylate) (p(MAA- co -EGDMA)) copolymer followed by immediate in situ grafting of a pMAA homopolymer layer. The grafting parameters were systematically studied to obtain the maximum grafting density of pMAA. The resulting membrane with maximized surface functionality shows strong hydrophilicity with a water contact angle of 33° without affecting its permeability, excellent suppression of platelet adhesion and deformation, and significantly lengthened clotting time (APTT, TT, and PT prolonged to 308.4 s, 31.0 s, and 31.9 s, respectively). The substantially enhanced hemocompatibility is attributed to the abundant carboxyl groups achieved by hybrid grafting, which effectively disrupts the coagulation cascade. This facile iCVD grafting method can be a promising candidate for improving the hemocompatibility of biomaterials, such as dialysis membranes, catheters, and implants.

Bioinspired Janus Mesh with Mechanical Support and Side-specific Biofunctions for Hernia Repair

Xiaoli Han,Zhenliang Liu,Liwei Sun,Zexiang Li,Yanhong Dong,Lu Zhou,Lingwan Hao,Jie Zhao,Rujian Jiang

Postoperative adhesion (PA) caused by the combination of proteins, inflammatory response and bacterial infection poses substantial challenges for polypropylene meshes (PPMs) based hernioplasty. Herein, inspired by the peritoneum, a Janus PPMs with side-specific functions was developed via a surface-initiated photoiniferter-mediated polymerization technology. A physical barrier composed of zwitterionic polymer brushes (PS) was firstly constructed on the one side of the PPMs, while the polymethacrylic acid (PMAA) brushes acting as the linker for bioactive nanoparticles (HAP) were precisely situated on the opposite surface subsequently. Our findings reveal that the mesh surface modified with PS demonstrated significant antifouling property that more than 99% of protein adhesion could be inhibited even after the co-incubation for 72 h in the crucial test. Meanwhile, on the other surface of the PPMs modified with HAP achieved satisfactory ROS-scavenging, inflammation-inhibiting and cell adhesion-promoting properties as well as good bactericidal performance (killing rate > 99.9%). Furthermore, the Janus PPMs could maintain comparable mechanical property with pristine meshes. Equipped with the above multiple merits and asymmetric property, the constructed Janus PPMs demonstrated effective treatment for abdominal hernia defects in vivo without any PA formation. Overall, this study duplicates the unique characteristics of peritoneum onto PPMs to successfully address postoperative complications of the hernioplasty and also offers a versatile and innovative idea to construct asymmetrical functions on the one implant. Statement of Significance The implantation of surgical meshes in the hernia defect provides additional firm support to reinforce the abdomen fascia in tension-free way. However, multiple post-surgery complications induced tissue adhesions is of great challenges for commercial mesh-based hernioplasty. Although material designing and surface modification might circumvent these limitations partly, the construction of side-specific biofunctions on the commercial mesh surfaces is very challenging because of its porous structure. Herein, we demonstrate a feasible and promising approach to construct asymmetric biofunctions on the opposite sides of network structured polypropylene mesh, which is rarely achieved previously. The fabricated Janus PPMs maintains the inherent long-term mechanical support; meanwhile, the opposite sides of PPMs could perform multiple biofunctions independently.

Responsive antibacterial surface based on looped poly(methacrylic acid)

Xiaoyu Xia,Xue Jia,Ziqiang Yu,Guangyu Zhang,Zhaohui Su

The adhesion of bacteria to surfaces poses a significant challenge to public health and many industries, making the development of antibacterial surfaces an urgent issue. In this work, α,ω-bisthiol poly(methacrylic acid) (HS-PMAA-SH) was synthesized and grafted in loop conformation onto a substrate primed with polydopamine, in conjunction with Ag NPs prepared in-situ to create a pH responsive composite surface with synergistic fouling-resistant and bacterial killing and releasing capabilities. A monofunctional analogue of half the chain length, PMAA-SH, was also synthesized and used to construct the composite surface with the PMAA in linear conformation for comparison. Quartz crystal microbalance with dissipation experiments revealed that PMAA loops allowed much less protein to adsorb than their linear analogues. Confocal laser scanning microscopy results showed that both composite surfaces were effective in killing the bacteria attached due to the presence of Ag NPs, and exhibited good initial resistance to bacterial adhesion and ability to release the killed bacteria under mild conditions due to the PMAA component; the composite surface conjugated with PMAA-loop was superior to the one with PMAA-linear, presenting lower initial bacterial adhesion and higher release rates, maintaining a low bacterial coverage even after five resist/kill/release cycles. The research opens a new avenue for antibacterial surfaces that integrate fouling-resistance, bacterial killing and release functions.

On-demand storage and release of antimicrobial peptides using Pandora's box-like nanotubes gated with a bacterial infection-responsive polymer

Junjian Chen,Xuetao Shi,Ye Zhu,Yunhua Chen,Meng Gao,Huichang Gao,Lei Liu,Lin Wang,Chuanbin Mao,Yingjun Wang

Background : Localized delivery of antimicrobial agents such as antimicrobial peptides (AMPs) by a biomaterial should be on-demand. Namely, AMPs should be latent and biocompatible in the absence of bacterial infection, but released in an amount enough to kill bacteria immediately in response to bacterial infection. Methods : To achieve the unmet goal of such on-demand delivery, here we turned a titanium implant with titania nanotubes (Ti-NTs) into a Pandora's box. The box was loaded with AMPs (HHC36 peptides, with a sequence of KRWWKWWRR) inside the nanotubes and “closed” (surface-modified) with a pH-responsive molecular gate, poly(methacrylic acid) (PMAA), which swelled under normal physiological conditions (pH 7.4) but collapsed under bacterial infection (pH ≤ 6.0). Thus, the PMAA-gated Ti-NTs behaved just like a Pandora's box. The box retarded the burst release of AMPs under physiological conditions because the gate swelled to block the nanotubes opening. However, it was opened to release AMPs to kill bacteria immediately when bacterial infection occurred to lowering the pH (and thus made the gate collapse). Results : We demonstrated such smart excellent bactericidal activity against a panel of four clinically important bacteria, including Staphylococcus aureus , Escherichia coli , Pseudomonas aeruginosa , methicillin-resistant Staphylococcus aureus . In addition, this box was biocompatible and could promote the osteogenic differentiation of human mesenchymal stem cells. Both in vitro and in vivo studies confirmed the smart “on-demand” bactericidal activity of the Pandora's box. The molecularly gated Pandora's box design represents a new strategy in smart drug delivery.
