Parkinson's disease (PD), a progressive neurodegenerative disorder, is marked by the degeneration of dopaminergic neurons. Emerging evidence highlights the critical involvement of neuronal ferroptosis and microglial pyroptosis in PD pathogenesis. In this study, we first characterized systemic inflammatory alterations in the peripheral blood of PD patients compared to healthy controls, revealing distinct pro-inflammatory signatures. Based on these findings, we engineered BV2 microglial membrane-camouflaged biomimetic multienzyme gallic acid-selenium nanoparticles (BM@GA-Se nanosomes) and evaluated their neuroprotective potential. In vitro and in vivo experiments demonstrated that BM@GA-Se nanosomes enhanced neuronal survival by simultaneously suppressing ferroptosis of neurons (via GPX4-mediated lipid peroxide reduction) and pyroptosis of microglia (through inhibition of the HSP90/NLRP3/Caspase-1 signaling pathway). These results position BM@GA-Se nanosome as a new dual-pathway targeting therapeutic strategy, offering a promising translational approach to mitigate PD progression by addressing interdependent inflammatory and oxidative mechanisms.

Volumetric muscle loss (VML) is a severe condition in which the loss of skeletal muscle surpasses the body's intrinsic repair capabilities, leading to irreversible functional deficits and potential disability, with persistent inflammation and impaired myogenic differentiation. To address these challenges, a novel zinc-dihydromyricetin (Zn-DHM) nanozyme with superoxide dismutase (SOD)-like activity is developed, designed to neutralize excessive reactive oxygen species (ROS) and restore oxidative balance. Zn-DHM mitigates oxidative stress and promotes polarization of macrophages from the proinflammatory M1 phenotype to the anti-inflammatory M2 phenotype, thereby reducing chronic inflammation and creating a conducive environment for muscle repair. Further, Zn-DHM significantly enhances the myogenic differentiation of C 2 C 12 cells, accelerating wound healing processes. These studies confirm the biosafety and low toxicity of Zn-DHM. As per a murine tibialis anterior VML model, Zn-DHM effectively suppresses inflammation and markedly improves skeletal muscle repair outcomes. Single-cell RNA sequencing reveals that Zn-DHM treatment increases the expression of M2 macrophage markers and enhances the proliferation and differentiation capacity of muscle stem cells (MuSCs). In addition, intercellular communication analysis reveals interactions between MuSCs and macrophages in the Zn-DHM treatment group, suggesting that these interactions may drive tissue regeneration through the activation of the GAS and Notch signaling pathways.

Chronic pancreatitis (CP) is a lifelong progressive fibrotic inflammatory disorder for which no effective cure is currently available. Persistent and recurrent inflammatory stimulation induced by reactive oxygen species (ROS) is a key driver of pancreatic fibrogenesis, making oxidative stress a promising therapeutic target to halt disease progression. In this study, we developed a nanosystem, HC@CeMOF, consisting of a small-sized cerium-based metal–organic framework (CeMOF) core loaded with curcumin and coated with hyaluronic acid (HA), enabling precise targeting of inflamed pancreatic tissue. HC@CeMOF exhibits a small-sized particle size along with favorable cellular and biological safety profiles. Once administered in vivo , the nanosystem exploits the specific binding affinity of HA to CD44 receptors on macrophages to selectively accumulate at inflamed pancreatic sites. Subsequently, the cerium-based nanozyme efficiently scavenges ROS through the reversible redox cycling between Ce 3+ and Ce 4+ , while the slow release of curcumin further suppresses the NF-κB signaling pathway and modulates inflammatory cytokine levels, thereby achieving synergistic anti-inflammatory and antioxidant effects. Collectively, these mechanisms substantially attenuate CP progression. This targeted ROS-scavenging and anti-inflammatory strategy holds promise as an alternative therapeutic approach for chronic pancreatitis.

Excessive oxidative stress and associated inflammation significantly impair bone regeneration around implants. To address this, we developed a novel therapeutic strategy based on metal-polyphenol nanoparticles self-assembled from tannic acid (TA) and manganese ions (Mn 2+ ). These Mn-TA nanoparticles (Mn-TA NPs) were subsequently incorporated into a gelatin methacryloyl (GelMA) hydrogel, which was then applied as a coating onto TiO 2 nanotube (TNT) surfaces by spin coating. The resulting modified titanium was designated as TNT-GelMA@NPs. Surface and interfacial characterization using scanning electron microscopy (SEM), atomic force microscopy (AFM), contact angle measurement, spectroscopic ellipsometry and nanoindentation confirmed the successful fabrication of a uniform, adherent hydrogel coating on the TNT surfaces. The coating reduced surface roughness while maintaining hydrophilicity, and achieved sustained Mn 2+ release, as verified via inductively coupled plasma atomic emission spectroscopy (ICP-AES). TNT-GelMA@NPs exhibited potent ROS scavenging capacity, protected mitochondrial membrane potential, and upregulated antioxidant genes. Moreover, it promoted macrophage polarization from pro-inflammatory M1 to anti-inflammatory M2 phenotype, thereby creating a favorable osteoimmune microenvironment. Additionally, it promoted osteogenic differentiation by increasing ALP activity, mineralization, and the expression of osteogenic genes. In a rat femoral defect model, TNT-GelMA@NPs implants reduced oxidative damage and inflammation, while enhancing bone formation, as evidenced by histological staining and micro-CT analysis. This study demonstrates that the Mn-TA NP-reinforced hydrogel coating acts as a multifunctional biointerface with excellent potential for advanced implant surface engineering.

This study developed a chitosan (CS)‑sodium tripolyphosphate (TPP)‑zinc (Zn) nanogel for treating Helicobacter pylori ( H. pylori ) through synergistic antibacterial, anti-inflammatory, and mucosal repair mechanisms. The nanogel was synthesized via an emulsion method, leveraging electrostatic interactions between CS amino groups and TPP phosphate ions, followed by Zn incorporation. Characterization revealed uniform spherical nanoparticles (~200 nm) with high Zn content (20.18 %) and controlled hydrogen (H 2 ) and Zn 2+ release (15 h) in acidic environments. In vitro assays demonstrated potent antibacterial activity against H. pylori (95.83 % inhibition at pH = 2; 28.54 % inhibition at pH = 7) attributed to Zn 2+ -mediated enzyme disruption. The nanogel exhibited significant antioxidant capacity, scavenging 82.3 % of PTIO and 65.48 % of DPPH radicals. Biocompatibility testing confirmed minimal haemolysis (<5 %) and high cell viability (>95 %). In vivo studies in H. pylori -infected mice demonstrated a bacterial inhibition rate of 97.54 % through upregulation of β-catenin and tight junction proteins. Concurrently, mucosal repair was enhanced, achieving migration rates of 44.2 ± 5.6 % and 72.34 ± 2.1 % at 12 and 24 h respectively. The nanogel also preserved gut microbiota diversity post-treatment, unlike conventional antibiotics. This multifunctional nanogel platform combines biocompatibility, ease of manufacture and dual therapeutic effects. In summary, this study proposes a multifunctional therapeutic strategy capable of simultaneously addressing H. pylori infection, gastric inflammation and mucosal damage. It offers a paradigm shift from traditional antibiotic-centred therapies, providing a promising strategy for eradicating Helicobacter pylori and restoring gastric injury.

As high-efficiency, safe, and low-drug resistant antibacterial agents, silver nanoparticles (AgNPs) have been widely applied in food and biomedicine. AgNPs was prepared using mango peel extract (MPE) as green and cheap reducing agent and stabilizer. In addition, a novel of preservative film material was developed with polylactic acid (PLA) as protective and substrate. AgNPs was characterized by XPS, XRD and TEM, and the size of AgNPs were in the range of 2.5–6.5 nm. The addition of AgNPs improved the mechanical properties of the film and its barrier ability to water vapor and oxygen. The film exhibited excellent antibacterial properties, and the inhibition rate against Escherichia coli and Staphylococcus aureus were above 95%. Furthermore, in terms of safety, the silver migration and cytotoxicity of the film met the relevant standards, and the shelf life of strawberries was significantly extended.

Excessive friction and cellular senescence contribute to osteoarthritis (OA) development. However, synergistic treatments targeting these two causes of OA may have significant implications for OA. In this study, we propose a novel strategy for exosome delivery encapsulated an injectable and lubricated hydrogel to synchronously reduce the excessive friction and the accumulation of aged chondrocytes. A cartilage-targeting peptide HABP-PEG-COLBP was employed to achieve a stable and efficient lubrication. The zwitterions of phosphatidylcholine lipids secreted by the synovial membrane of the joint can adsorb water molecules, forming a hydrated layer around the charge. Moreover, the exosomes derived from bone marrow stem cell (BMSCs) were engineered with a chondrocyte affinity peptide (CAP) in the out membrane and si-STING in the exosome to reshape the senescence microenvironment. Furthemore, the in vivo data showed that the hydrogel/exosome delivery system successfully can alleviate the joint wear and rejuvenate chondrocytes to reverse the development of OA. The HD-T@CAP-Exos-siSTING gels can provide efficient lubrication and potentially alleviate friction-related diseases such as osteoarthritis. Teaser An injectable hydrogel delivering exosomes rejuvenates chondrocytes and reduces mechanical stress, presenting a potential osteoarthritis treatment strategy.

Overactive inflammatory cascade accompanied by oxidative stress in the nucleus pulposus exacerbates intervertebral disc degeneration (IVDD). Hydrogels have been demonstrated to be promising in treating IVDD, yet they remain less efficacious in the case of anti-inflammation associated with antioxidation. In this study, we designed an injectable self-antioxidant hydrogel (HA/CS) with enhanced inflammation inhibitory performance for delivering chondroitin sulfate (CS) with well-documented anti-inflammatory property to treat IVDD. The hydrogel was rapidly formed via dynamic boronate ester bonding between furan/phenylboronic acid and furan/dopamine-modified hyaluronic acid (HA), and mechanically enhanced by Diels–Alder reaction-induced secondary crosslinking, partial dopamine groups of which contribute to grafting phenylboronic acid-modified CS (CS-PBA). This hydrogel exhibits favorable injectability, mechanical property, and pH-responsive delivery behavior. The dopamine moiety endows the hydrogel with efficient antioxidative property. By sustained delivery of CS, the HA/CS hydrogel is well competent to inhibit inflammatory cytokine expression and maintain anabolic/catabolic balance in an inflammation-simulated environment. Most importantly, the HA/CS hydrogel significantly ameliorates degeneration in a puncture-induced IVDD rat model. The self-antioxidant HA/CS hydrogel designed in this work may serve as a novel and promising therapeutic platform for IVDD.

To obtain high-performance tissue-adhesive hydrogel embodying excellent mechanical integrity, a supramolecular hydrogel patch is fabricated through in situ copolymerization of a liquid-liquid phase separation precursor composed of self-complementary 2-2-ureido-4-pyrimidone-based monomer and acrylic acid coupled with subsequent corporation of bioactive epigallocatechin gallate. Remarkably, the prepared supramolecular hydrogel leverages hierarchical multi-strength hydrogen-bonds hinged strategy assisted by alkyl-based hydrophobic pockets, broadening the distribution of binding strength of physical junctions, striking a canonical balance between superb mechanical performance and robust adhesive capacity. Ultimately, the fabricated supramolecular hydrogel patch stands out as a high stretchability (1500 %), an excellent tensile strength (2.6 MPa), a superhigh toughness (12.6 MJ m −3 ), an instant and robust tissue adhesion strength (263.2 kPa for porcine skin), the considerable endurance under cyclic loading and reversible adhesion, a superior burst pressure tolerance (108 kPa) to those of commercially-available tissue sealants, and outstanding anti-swelling behavior. The resultant supramolecular hydrogel patch demonstrates the rapid hemorrhage control within 60 s in liver injury and efficient wound closure and healing effects with alleviated inflammation and reduced scarring in full-thickness skin incision, confirming its medical translation as a promising self-rescue tissue-adhesive patch for hemorrhage prevention and sutureless wound closure.

In order to improve the high-value utilization of mango peel and the performance of the polyvinyl alcohol (PVA) films, mango peel extract (MPE) and PVA were blended to prepare multifunctional composite films in this paper. First, gel permeation chromatography was used to determine the molecular weight distribution of MPE, and the major components of MPE were identified by gas chromatography-mass spectrometry. Subsequently, different ratios of MPE/PVA composite films were prepared. As indicated by the results, the optical and mechanical properties of pure PVA film are significantly affected by the addition of MPE. With addition of 2.4 wt% MPE, the ultimate elongation of this films is significantly increased by 160.24%. The addition of MPE also enhances the water resistibility and thermal stability of the films and significantly improves its antibacterial and antioxidative properties. These results indicate that the prepared composite films is expected to be widely applied in food packaging. Novelty impact statement The mango peel extract (MPE) was prepared, its molecular weight and composition were characterized, and then the MPE/PVA composite films were further prepared. The MPE improved the elongation at break and water resistance of composite films. The MPE gave excellent UV resistance, antibacterial and antioxidant properties to the composite films.

The potential risks of sono-induced nitrosation and nitration side reactions and consequent toxic nitrogenous byproducts were first investigated via sono-degradation of diphenylamine (DPhA) in this study. The kinetic models for overall DPhA degradation and the formation of nitrosation byproduct (N-nitrosodiphenylamine, NDPhA) and nitration byproducts (2-nitro-DPhA and 4-nitro-DPhA) were well established and fitted (R 2 > 0.98). Nitrosation contributed much more than nitration (namely, 43.3 – 47.3 times) to the sono-degradation of DPhA. The contribution of sono-induced nitrosation ranged from 0.4 to 56.6% at different conditions. The maximum NDPhA formation rate and the contribution of sono-induced nitrosation were obtained at 600 and 200 kHz, respectively, as ultrasonic frequencies at 200 to 800 kHz. Both NDPhA formation rate and the contribution of sono-induced nitrosation increased with increasing power density, while decreased with increasing initial pH and DPhA concentration. PO 4 3– , HCO 3 – , NH 4 + and Fe 2+ presented negative impacts on sono-induced nitrosation in order of HCO 3 – >> Fe 2+ > PO 4 3– > NH 4 + , while Br – exhibited a promoting effect. The mechanism of NDPhA formation via sono-induced nitrosation was first proposed.

Quercetin (QCT) has antioxidant, anti-inflammatory, anti-tumor and other important pharmacological activities, but the poor water solubility limits its application. In this work, the amphiphilic dextran (ADEX) was prepared by grafting L-cysteine and octadecylamine onto carboxymethyl dextran with the grafting rate of 21.29 % and 19.35 %. Then, the QCT-loaded nanomicelles (QNMs) were prepared by using ADEX as wall material and the QCT as core material via ultrasonic self-assembly method. The particle size and zeta potential of QNMs were 372 nm and 31.4 mV. Under simulated gastric and simulated intestinal fluids, the cumulative release QNMs were 37.54 % and 52.13 % within 180 min, and the QNMs showed better stability in simulated gastric fluid. The QNMs showed significantly better PTIO , OH and O 2 − scavenging activities than QCT. In addition, QNMs could effectively down-regulate the expression of pro-inflammatory cytokines and promoted the expression of anti-inflammatory cytokine. The cellular uptake results proved that the QNMs were more easily absorbed by cells than free QCT, indicating that the nano-encapsulation procedure effectively improved the uptake efficiency of QCT by cells.
