NIR-II-responsive AuNRs@SiO2–RB@MnO2 nanotheranostic for multimodal imaging-guided CDT/PTT synergistic cancer therapy

Changchun Wen,Xiaolu Guo,Cunji Gao,Zhongkai Zhu,Nianqi Meng,Xing-Can Shen,Hong Liang

Specific tumor-responsive capabilities and efficient synergistic therapeutic performance are the keys to effective tumor treatment. Herein, AuNRs@SiO2–RB@MnO2 was developed as a new type of tumor-responsive nanotheranostic for multimodal imaging and synergistic chemodynamic/photothermal therapy. In AuNRs@SiO2–RB@MnO2, the SiO2 layer wraps the AuNRs, providing light absorption in the second near-infrared (NIR-II) region. The SiO2 layer also adsorbs the MnO2 nanosheets, which have Fenton-like activity, resulting in a fluorescent sensing platform based on the fluorescence quenching properties of MnO2 for rhodamine B dye. The fluorescence can be recovered by the consumption of MnO2 by glutathione, which simultaneously produces Mn2+ in the tumor region. The recovery of fluorescence reflects the consumption of glutathione and the increase in Mn2+, which produces hydroxyl radicals via Fenton-like reaction in the tumor microenvironment to realize chemodynamic therapy. Meanwhile, the AuNRs are a good photothermal reagent that can effectively absorb NIR-II light and convert it into heat energy to kill tumor cells via photothermal therapy. The NIR-II absorption performance of the AuNRs provides good photoacoustic imaging and deep photothermal performance, which is favorable for efficient NIR-II photoacoustic imaging-guided photothermal therapy. As a result, the AuNRs@SiO2–RB@MnO2 nanotheranostic exhibits outstanding imaging and synergistic chemodynamic/photothermal therapeutic performance for tumor imaging and treatment.

The PTT, SDT, and CDT synergistic antitumor effect of Fe3+-coordinated PDA@PLGA-Cur nanoparticles in vitro

Fanfan Zhang,Xuemei Hu,Yue Zhang,Baiyuan Sha,Fengshi Shang,Wenjie Wang,Rui Liu,Hao Wang

Hepatocellular carcinoma (HCC) poses a severe threat to public health, with clinical therapy limited by narrow surgical indications, chemotherapeutic side effects, and targeted drug resistance. Herein, we developed Fe 3+ coordination Polydopamine@Poly (lactic-co-glycolic acid)-Curcumin (Fe 3+ -PDA@PLGA-Cur, FePPC) nanoparticles integrating photothermal therapy (PTT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), and curcumin (Cur)'s intrinsic anticancer activity for synergistic HCC treatment. FePPC was fabricated via emulsion solvent evaporation, in-situ dopamine polymerization, and Fe 3+ coordination chelation, with its structure and physicochemical properties characterized by FT-IR, UV-vis, SEM, XPS, and particle size analysis. FePPC exhibited uniform spherical morphology (198.58 nm), favorable stability, 73.60 ± 0.51% encapsulation efficiency, and NIR-triggered temperature-dependent Cur release. Under 808 nm NIR light (2 W/cm 2 , 4 cm), FePPC achieved a 24.4 °C temperature elevation with robust photothermal stability. Catalytic assays confirmed FePPC generated ·OH via Fe 3+ -mediated Fenton-like reactions, 1 O 2 under NIR irradiation, and ·O₂⁻ upon ultrasound stimulation, realizing synergistic reactive oxygen species (ROS) production. In vitro experiments on HepG2 cells showed time-dependent FePPC internalization; the FePPC + PTT + CDT + SDT group reduced cell viability to 53.45%, induced 47.55% apoptosis, elevated intracellular ROS, disrupted mitochondrial membrane potential, downregulated LDH, modulated Bax/Bcl-2 expression, and inhibited cell migration. This study highlights FePPC's potential as a multifunctional nanoplatform for HCC therapy via coordinated chemical-biological interactions.

Cell membrane camouflaged Cu-doped mesoporous polydopamine for combined CT/PTT/CDT synergistic treatment of breast cancer

Di Meng,Shuoye Yang,Lin Ju,Jinpeng Wang,Yanan Yang,Lu Zhang,Lan Cui

Currently, traditional monotherapy for cancer often results in indiscriminate attacks on the body, leading to the emergence of new health problems. To confront these challenges, multimodal combination therapy has become necessary. However, how to develop new smart nanomaterials through green synthesis methods, delivering drugs while simultaneously synergizing multimodal combination therapies for tumor treatment, remains a topic of great significance. In this study, a biomimetic composite nanomaterial (RM-Cu/P) composed of mesoporous polydopamine (MPDA) as the core and red blood cell membranes (RBCMs) as the shell was synthesized as a drug carrier to deliver doxorubicin (DOX) while achieving synergistic chemotherapy, photothermal and chemodynamic therapy (CT/PTT/CDT). Herein, the nanoparticles were extensively characterized to examine their morphological characteristics, elemental composition, and drug-carrying capacity. Notably, the coating of RBCM reduced the toxicity of the RM-Cu/P@DOX nanoparticles, improved their targeting ability and prolonged their circulation time in vivo . The Cu-doped nanoparticles were capable of initiating a Fenton-like reaction to generate reactive oxygen species (ROS) for CDT, while the photothermal conversion efficiency (η) reached 45.20 % under NIR laser irradiation. Subsequently, the particles were examined by in vivo and in vitro experimental studies in cytotoxicity, cellular uptake, ROS levels, lysosomal escape, and mouse tumor model to evaluate their potential application in antitumor. Compared with monotherapy, the RM-Cu/P@DOX nanoparticles had multiple-stimulation response properties under redox, pH, and NIR, which exhibited the advantage of combined trimodal therapy, resulting in remarkable synergistic antitumor efficacy. In conclusion, this innovative platform exhibited promising applications in smart drug delivery and synergistic treatment of cancer.

NIR-activatable proenzyme DSF/CuS@M for synergistic in situ chemotherapy/CDT/PTT/cuproptosis-based antitumor therapy

Lushuang Xu,Qinyue Chen,Yangwei Qian,Jin Wang,Chenxi Wang,Rui Fan,Jia-Wei Shen,Qiying Shen

Nanozymes provide therapeutic benefits against a diverse range of malignant tumors. However, their antitumor potencies are seriously impaired by their low catalytic efficiencies and the off-target toxicities caused by their “always-on” catalytic activity. Herein, we developed multifunctional nanoparticles co-loaded with ultra-small copper sulfide nanoparticles (CuS NPs) and disulfiram (DSF) (DSF/CuS@M). These nanoparticles had peroxidase-like activity and served as (near-infrared) NIR-activatable proenzymes to achieve synergistic in situ chemotherapy, chemodynamic therapy (CDT), photothermal therapy (PTT), and cuproptosis-based antitumor therapy. DSF/CuS@M not only provided PTT as NIR-activatable proenzymes but also enabled complete DSF and Cu 2+ release. Non-toxic DSF was converted into diethyldithiocarbamic acid (DDC) through a glutathione (GSH)-mediated metabolic reaction, leading to the generation of cytotoxic Cu(DDC) 2 in situ , which enhanced cell apoptosis. Cu 2+ was converted into Cu + by GSH, and this Cu + catalyzed the conversion of H 2 O 2 into hydroxyl radicals (·OH) to induce CDT. Moreover, Cu + could also bind to dihydrolipoyl transacetylase (DLAT) in the tricarboxylic acid (TCA) cycle, thereby inducing cuproptosis. These results demonstrate that the NIR-activatable proenzyme developed in this study is effective and safe for synergistic therapy against malignant tumors.

Mitochondrial Targeted Composite Nanoplatform Activated by a Single NIR Laser for Enhanced Synergistic PTT, PDT, and Chemotherapy

Yu Chen,Zhentan Lu,Dong Wang

The combinations of phototherapeutic modalities with chemotherapy represent an innovative antitumor strategy. Herein, we constructed a NIR laser regulated nanoplatform (PTE@TPP-NM(DOX/IR780)) via the self-assembly process of PEG-b-PCL modified with mitochondrial targeting triphenylphosphine (TPP), reactive oxygen species (ROS)-sensitive polymer (PTE), DOX, and IR780. Under 808 nm laser irradiation, IR780 in PTE@TPP-NM(DOX/IR780) generated abundant reactive oxygen species (ROS) and heat for photothermal therapy (PTT) and photodynamic therapy (PDT), while ROS oxidized PTE to release DOX for chemotherapy. The signal 808 nm laser could effectively regulate PTT, PDT, and chemotherapy in real-time, enhancing the synergistic efficacy. PTE@TPP-NM(DOX/IR780) could result in depolarization of the mitochondrial membrane and inhibition of the expression of heat shock proteins to amplify antitumor effect. Enhanced therapeutic effect was demonstrated via in vivo and in vitro experiment. This study provided a promising strategy for the design of multifunctional nanocarriers to address the limitation of combined therapy.

A tumor-microenvironment-activated nanoplatform of modified SnFe2O4 nanozyme in scaffold for enhanced PTT/PDT tumor therapy

Yang Shuai

Phototherapy has attracted widespread attention for cancer treatment due to its noninvasiveness and high selectivity. However, severe hypoxia, overexpressed glutathione and high levels of hydrogen peroxide (H 2 O 2 ) of tumor microenvironment limit the antitumor efficiency of phototherapy. Herein, inspired by the specific response of nanozymes to the tumor microenvironment, a simple and versatile nanozyme-mediated synergistic dual phototherapy nanoplatform is constructed. In this study, tin ferrite (SnFe 2 O 4 , SFO) nanozyme as a photosensitizer was surface modified with polydopamine (denoted as P-SFO) and incorporated into poly( l -lactide) to fabricate an antitumor scaffold fabricated by selective laser sintering. On one hand, SFO nanozyme could act as a photoabsorber to convert light energy into heat for photothermal therapy (PTT). On the other hand, it played a role of photosensitizer in transferring the photon energy to generate reactive oxygen species (ROS) for photodynamic therapy (PDT). Importantly, its multivalent metal ions redox couples would decompose H 2 O 2 into O 2 for enhancing O 2 -dependent PDT and consume glutathione to relieve antioxidant capability of the tumors. Besides, polydopamine as a photothermal conversion agent further enhanced the photothermal performance of SFO. The results revealed the PLLA/P-SFO scaffold possessed a photothermal conversion efficiency of 43.52% for PTT and a high ROS generation capacity of highly toxic ·O 2 − and ·OH for PDT. Consequently, the scaffold displayed a prominent phototherapeutic effect with antitumor rate of 96.3%. In addition, the PLLA/P-SFO scaffolds possessed good biocompatibility for cell growth. These advantages endow PLLA/P-SFO scaffold with extensive applications in biomedical fields and opened up new avenue towards nanozyme-mediated synergistic phototherapy.

Ruthenium-gallic acid nanozymes encapsulated in 3D bioprintable photosensitive hydrogels for synergistic PTT/CDT therapy of glioblastoma

Jianyu Nie,Zhongyong Wang,Haotian Tian,Yang Qiao,Menghui Liu,Xianwen Wang,Liang Xu,Xingliang Dai

Confronted with the clinical challenges of glioma treatment, including incomplete resection and therapeutic resistance, we developed an integrative bioprinting strategy. At the core of our approach is a 3D-bioprintable, photosensitive hydrogel loaded with ruthenium-gallic acid (Ru-GA) nanoenzymes. This system is designed to seamlessly combine chemodynamic therapy (CDT) with photothermal therapy (PTT) to achieve precise and synergistic treatment of glioblastoma multiforme (GBM). Ru-GA, synthesized via Ru³⁺-gallic acid coordination, has potent peroxidase-like activity to catalyze reactive oxygen species generation from H₂O₂ in the tumor microenvironment. It also efficiently depletes glutathione to disrupt tumor redox homeostasis, and exhibits excellent near-infrared absorption and photothermal conversion efficiency. Based on GelMA and HAMA, the hydrogel is liquid at 37 °C and has the ability to be injected into tumor tissue, ensuring localized retention and sustained action of Ru-GA at tumor sites. In vitro tests verified Ru-GA kills GL261 glioma cells via CDT, and combining it with PTT further inhibits tumor cell proliferation, migration, and clonogenicity while boosting apoptosis. In 3D bioprinted tumor models, the hydrogel suppresses tumor cell aggregation and viability effectively. In vivo experiments showed intratumoral hydrogel injection plus near-infrared irradiation raises tumor temperature to 55 °C, exerting synergistic PTT/CDT effects to significantly inhibit tumor growth with excellent biosafety. Mechanistically, this therapy activates inflammatory pathways and inhibits metastasis-related signaling, offering a safe, efficient GBM treatment and laying a foundation for clinical translation of nanocatalyst-based photosensitive hydrogels.

Synthesis of Silver-Based Plasmas by Plant Extracts for In Vitro PTT/PDT/CT Synergistic Antitumor Therapy

Yiliang Xie,Baowen Dong,Xuan Zhang,Jimin Fan,Jianying Lin,Bing Zhang,Zhihuan Zhao

In this study, biosynthetic Ag@AgCl plasma is used as a photosensitizer for the first time to construct a photothermal/photodynamic/chemotherapy and pH-responsive synergistic antitumor nanomedicine delivery system CS-Ag@AgCl/C. The Ag@AgCl and AgNPs synthesized by the extracts of alpine banana, C itrus reticulata , and Citrus sinensis are characterized. The Ag@AgCl NPs synthesized with banana extract have a regular crystal shape and a granular size of 12 nm, and they are more stable than AgNPs synthesized with the other two extracts. In addition, Ag@AgCl plasma used as a photosensitizer can reduce the dependence of CS-Ag@AgCl/C on oxygen in the photodynamic process. After coating with chitosan, though the photothermal conversion efficiency of CS-Ag@AgCl/C decreases from 44.06% to 29.51%, the pH response character of it increases, and the pH response plays an important role in drug release experiments. Under an acidic environment of pH = 5.0 phosphate-buffered saline buffer, the drug release rate of the samples increases significantly. In the synergistic antitumor experiment, the cell survival rate increases from 36.83% to 78.69% after chitosan-coated Ag@AgCl/C NPs. When CS-Ag@AgCl/C-DOX+808 nm is co-cultured with HeLa cells, the cell survival rate is only 14.89%, which indicates that the constructed plasma drug delivery system has excellent photothermal/photodynamic/chemotherapy combined treatment capability.

Photothermal and Acid-Responsive Fucoidan-CuS Bubble Pump Microneedles for Combined CDT/PTT/CT Treatment of Melanoma

Jiaojiao Tao,Bingjie Wang,Yu Dong,XiangYan Chen,Shuang Li,Tianze Jiang,Xia Zhao

Transdermal cancer therapy faces great challenges in clinical practice due to the low drug transdermal efficiency and the unsatisfactory effect of monotherapy. Herein, we develop a novel bubble pump microneedle system (BPMN-CuS/DOX) by embedding sodium bicarbonate (NaHCO3) into hyaluronic acid microneedles (MNs) loaded with fucoidan-based copper sulfide nanoparticles (Fuc-CuS NPs) and doxorubicin (DOX). BPMN-CuS/DOX can generate CO2 bubbles triggered by an acidic tumor microenvironment for deep and rapid intradermal drug delivery. Fuc-CuS NPs exhibit excellent photothermal effect and Fenton-like catalytic activity, producing more reactive oxygen species (ROS) by photothermal therapy (PTT) and chemodynamic therapy (CDT), which enhances the antitumor efficacy of DOX and reduces the dosage of its chemotherapy (CT). Simultaneously, DOX increases intracellular hydrogen peroxide (H2O2) supplementation and promotes the sustained production of ROS. BPMN-CuS/DOX significantly inhibits melanoma both in vitro and in vivo by the combination of CDT, PTT, and CT. In short, our study significantly enhances the effectiveness of transdermal drug delivery by constructing BPMNs and provides a promising novel strategy for transdermal cancer treatment with multiple therapies.

Self-assembled nano-multiple warheads: A precise targeting platform for combinatorial chemotherapy, PTT and PDT therapy against glioma

Zhihao Chen,Mengru Wu,Qianwen Ren,Wanjun Liang,Xiu Wang

DIPF was fabricated via drug self-assembly, with high encapsulation efficiency and loading capacity of both DOX and ICG. • DIPF exhibits strong ability to penetrate the BBB, to accumulate in gliomas, and enables real-time in vivo imaging. • DIPF integrates chemotherapy, PTT and PDT to inhibit gliomas, overcome drug resistance and reduce DOX-induced cardiotoxicity. • DIPF is a promising delivery platform for multi‑therapy glioma treatment. Combination therapy tactics that incorporate numerous therapies via innovative multi-drug co-loaded nanomedicine delivery systems have shown considerable benefits in crossing the blood-brain barrier (BBB) and slowing glioma growth. However, efficiently developing multi-drug co-loaded delivery systems with high drug loading capacity, the ability to alleviate drug resistance, and minimal in vivo toxicity remains a significant problem. In this study, we created a self-assembling nanomedicine delivery system using a DIPF composite encapsulated with polydopamine and folate. The nanoparticles have an internally coated self-assembled structure that is assembled via electrostatic interactions, π-π stacking, and hydrophobic interactions between doxorubicin (DOX) and indocyanine green (ICG). This achieves high encapsulation efficiency and loading content for the co-delivery of multiple drugs. In this delivery system, the encapsulation efficiencies of DOX and ICG reached 94.08% and 94.01%, respectively, while the inherent cardiotoxicity of DOX was reduced by 64.33%. H&E staining results and anti-tumor experimental outcomes—including expression of TNF-α, Ki67, Caspase-3, and GFAP cytokines—clearly demonstrated that combined DIPF+NIR therapy significantly inhibited tumor growth and substantially extended survival. The highly efficient multi-drug co-loaded delivery system developed here demonstrates significant application potential for active targeting, controlled drug release, treatment visualization, and combined chemotherapy, photothermal (PTT), and photodynamic (PDT) therapy for brain tumors. A highly efficient multi-drug co-loaded drug delivery system that can cross the BBB and delivery combined chemotherapy, photothermal and photodynamic therapy for brain tumors target was established in a self-assembling way via electrostatic interactions, π-π stacking, and hydrophobic interactions between doxorubicin and indocyanine green. NIR-Triggered and ROS-Boosted Nanoplatform for Enhanced Chemo/PDT/PTT Synergistic Therapy of Sorafenib in Hepatocellular Carcinoma

Wang Chonggao,Cheng Xiaolan,Peng Hao,Zhang Yewei

Although being the first-line treatment of advanced hepatocellular carcinoma (HCC), sorafenib (SOR) outcome is limited due to drug resistance and low tumor accumulation. Herein, with MnO 2 as photothermal agent and chlorine6 (Ce6) as photosensitizer, a tumor-targeting and NIR-triggered multifunctional nanoplatform loading sorafenib (MnO 2 -SOR-Ce6@PDA-PEG-FA, MSCPF) was constructed. Owing to oxygen generator MnO 2 , MSCPF could generate excessive ROS, thus can alleviate tumor hypoxia and improve sorafenib accumulation in cancer cells. Besides, ROS production further strengthens Ce6-mediated PDT and PDA-mediated PTT. By exploiting these features, MSCPF exhibited excellent antitumor effects on HCC in the in vitro and in vivo studies, compared to solo sorafenib or PDT/PTT treatment. Further mechanism experiments suggested that MSCPF could inhibit P-gp expression and induce ferroptosis via deactivation of GPX4 and SLC7A11, which ultimately enhanced the antitumor efficacy of SOR. In summary, our work highlights a promising NIR-triggered and ROS-boosted nanoplatform for enhanced chemo/PDT/PTT synergistic therapy of SOR in HCC treatment.

Graphene Oxide Composite for Selective Recognition, Capturing, Photothermal Killing of Bacteria over Mammalian Cells

Gang Ma,Junjie Qi,Qifan Cui,Xueying Bao,Dong Gao,Chengfen Xing

The multifunctional photothermal therapy (PTT) platform with the ability to selectively kill bacteria over mammalian cells has received widespread attention recently. Herein, we prepared graphene oxide-amino(polyethyleneglycol) (GO-PEG-NH2) while using the hydrophobic interaction between heptadecyl end groups of 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[amino(polyethyleneglycol)] (DSPE-PEG-NH2) and graphene oxide (GO). Based on GO-PEG-NH2, the versatile PTT system was constructed with simultaneous selective recognition, capturing, and photothermal killing of bacteria. When the cells undergo bacterial infection, owing to the poly(ethylene glycol) (PEG) chains and positively charged amino groups, GO-PEG-NH2can specifically recognize and capture bacteria in the presence of cells. Meanwhile, the stable photothermal performance of GO-PEG-NH2enables the captured bacteria to be efficiently photothermally ablated upon the irradiation of 808 nm laser. Besides, the GO-PEG-NH2is highly stable in various biological media and it exhibits low cytotoxicity, suggesting that it holds great promise for biological applications. This work provides new insight into graphene-based materials as a PTT agent for the development of new therapeutic platforms. Keywords:photothermal therapy (PTT);graphene oxide (GO);selective recognition and capturing;antibacterial
