Improvement of organoleptic properties of Polygonatum sibiricum-rice wine by Co-fermentation of Saccharomyces cerevisiae, Lacticaseibacillus casei and Limosilactobacillus fermentum

Yu-Tong Han,Yan-Shu Li,Sanabil Yaqoob,Aysha Imtiaz,Chunlai Zeng,Yong-Kun Ma,Feng-Jie Cui,Qing Shen

The growing consumer interest and expanding market for rice wine have intensified the focus on developing high-quality products with the enhanced health-promoting properties. In the present study, the organoleptic and functional properties of Polygonatum sibiricum –rice wine was enhanced by developing a novel fermentation strategy using pre-hydrolyzed P. sibiricum extract as substrate. Results showed that the sequential inoculation of S. cerevisiae followed by L. casei (S C) increased acidity, enhanced antioxidant capacity, promoted the release and transformation of phenolic acids and free amino acids, and improved the volatile aroma profile by increasing esters and acids (e.g., γ-nonalactone and ethyl lactate) with the reduced higher alcohol contents. Multivariate and sensory analyses confirmed that S C group was the optimal strategy since S. cerevisiae provided the alcoholic and ester precursors, while subsequent L. casei contributed to balance the acidity and liberate antioxidant phenolic compounds. The outputs of the present study demonstrated that sequential co-fermentation with S. cerevisiae and L. casei was a promising approach to developing a high-quality, functional P. sibiricum -rice wine with enhanced organoleptic and nutritional properties.

Polygonatum sibiricum polysaccharide delays age-related decline and preserves muscle integrity in Caenorhabditis elegans by modulating the daf-2/IIS pathway

Yuxin Liao,Hongliang Li,Qiong Liu,Jia Zhang,Jiajie Lin,Qiaohui Du,Wanqiao He,Yongyin Li,Zhaoyang Dong,Xuanbin Wang,Lei Gao

Aging is characterized by a progressive decline in muscle strength and mass, posing a significant threat to global quality of life in the absence of effective therapeutic interventions. The edible herb Polygonatum sibiricum and its polysaccharide (PSP) have been shown to delay aging in animal models, although the underlying mechanisms remain poorly understood. In this study, we investigated the anti-aging effects of PSP in Caenorhabditis elegans ( C. elegans ). Our results demonstrate that PSP reduces oxidative stress, stabilizes mitochondrial function, and preserves sarcomere integrity during aging. Notably, the beneficial effects of PSP were abolished upon silencing of daf-2 , confirming their dependence on the insulin/IGF-1 signaling (IIS) pathway. Furthermore, daf-2 silencing eliminated any additional protective effects of PSP, indicating that this pathway is essential for its activity. These findings underscore the potential of PSP-based nutraceuticals in delaying sarcopenia and extending human healthspan.

Screening of the potentially active compounds from Polygonatum sibiricum using RAW264.7 cellular membranes coated magnetic beads fishing followed by HPLC analysis

Qing-ling Qi,Ya-Ming Yu,Cheng Tang

Target biomolecule-immobilized magnetic beads could be used as a powerful tool for screening active compounds present in natural products. Low damage rates of the target proteins, associated with the availability of diverse automated online approaches for analysis, make it a valuable tool for affinity studies. RAW264.7 cells (a kind of murine macrophage cell line) were used in this study. These cellular membranes were immobilized onto the surface of MBs and were used for screening the active compounds of Polygonatum sibiricum. Combining this technique with HPLC led to the identification of an active compound and its biological activity was confirmed. This is the first report establishing the use of RAW264.7 cellular membrane-coated magnetic bead fishing followed by HPLC analysis for screening active compounds from natural products.

Preparation and structure characterization of a natural acetylated fructooligosaccharide from Polygonatum sibiricum and its alleviative effect on colitis by inhibiting NLRP3 pathway

Shengchen Ding,Jun Chen,Guangming Chen,Weiyi Tian,Junping Zheng,Haiming Hu,Lu Cao,Xiaowei Yao,Baifei Hu,Hongtao Liu

This study aimed to isolate oligosaccharides from P. sibiricum , analyze its structure, and elucidate the pharmacological mechanism for colitis improvement. First, a homogeneous oligosaccharide PSO-A was isolated and its structure was characterized. Then, the colitis model was induced with 3 % dextran sulfate sodium (DSS) and intervened with PSO-A. The results show that PSO-A was an agavin-type acetylated fructooligosaccharide with a molecular weight of 2.12 kDa. PSO-A consisted of (2 → 6)-β-D-Fru f residue backbone with an internal α-D-Glc p residue and (2 → 1)-β-D-Fru f residue side chains. Further, PSO-A increased mucin content, protected intestinal barrier integrity of colitis mice, and significantly inhibited the levels of inflammatory factors. Moreover, the protein expressions of NLRP3, ASC, and Caspase-1 in colitis mice was statistically inhibited by PSO-A. In summary, PSO-A could alleviate colitis by inhibiting NLRP3 inflammasome pathway, which provides a basis for the development and application of P. sibiricum oligosaccharides as drug or functional food to relieve colitis.

Enhancing Healing of Infected Wounds with Glycerin-Modified Sodium Alginate/Silk Sericin Composite Film Functionalized with Polygonatum sibiricum Polysaccharide-Capped Silver Nanoparticles

Zicheng Yang,Rongyu Li,Ruonan Wang,Senhe Qian

Graphical An SA/SS/GI/PSP@AgNP composite film was developed, and its physicochemical properties, cytocompatibility, antioxidant activity, and antibacterial properties were investigated. The composite film released PSP@AgNPs in a sustained way, and the film demonstrated favorable mechanical properties, excellent cytocompatibility, and high antioxidant and antibacterial activities in both in vitro and in vivo models. As a result, we conclude that this composite film has potential for application as a medical material. A sodium alginate/silk sericin (SA/SS) film was modified with glycerin (GI) and Polygonatum sibiricum polysaccharide@silver nanoparticles (PSP@AgNPs) to create a SA/SS/GI/PSP@AgNP composite film with antibacterial and antioxidant properties. The physicochemical properties of the SA/SS/GI/PSP@AgNP composite film were analyzed, and its cytocompatibility, antioxidant activity, and antibacterial properties were investigated. The optimum conditions for the preparation of the SA/SS/GI/PSP@AgNP composite film were 2 % SA, 2 % SS, 20 % GI, and 40 μg/mL of PSP@AgNPs. The addition of GI improved the surface smoothness of the composite films, while the presence of PSP@AgNPs did not significantly alter it. Spectroscopic characterization demonstrated that GI and PSP@AgNPs were physically bound to SA/SS. Approximately 53 % of the PSP@AgNP was released from the SA/SS/GI/PSP@AgNP composite film over 24 h of soaking in aqueous solution. The resulting leaching solution did not cause hemolysis or cytotoxicity, indicating its biocompatibility. Additionally, the SA/SS/GI/PSP@AgNP composite film exhibited strong antioxidant and antibacterial properties, and it promoted healing of mouse wounds infected with Staphylococcus aureus .

Optimization of preparation conditions for CTAB-modified Polygonatum sibiricum polysaccharide cubosomes using the response surface methodology and their effects on splenic lymphocytes

Haiyu Ni,Shuwen Xu,Pengfei Gu,Adelijiang wusiman,Yue Zhang,Tianxin Qiu,Zhenguang Liu,Han Ni,Yuanliang Hu,Jiaguo Liu,Yi Wu,Deyun Wang

This study aimed to optimize the preparation conditions for cetyltrimethylammonium bromide-modified Polygonatum sibiricum polysaccharide cubosomes (CTAB-modified PSP-Cubs) by response surface methodology (RSM). Glyceryl-monooleate (GMO) was used as the lipid base for the cubosomes. The optimal preparation conditions of CTAB-modified PSP-Cubs were as follows: the mass percentage of PSP to GMO (X 1 ), Poloxamer 407 (F127) to GMO (X 2 ) and water to GMO (X 3 ) was 1.4%, 9% and 50%, respectively. The encapsulation efficiency (EE) of CTAB-modified PSP-Cubs under the experimental conditions was 62.4 ± 3.6%, which was close to our predicted value. The particle size, polydispersity index (PDI) and zeta potential of CTAB-modified PSP-Cubs were 427.7 ± 8.0  nm , 0.236 ± 0.024 and 19.2 ± 0.4 mV, respectively. Specific modifications were able to slow down in vitro release behaviors and reduce cytotoxicity to some extent. The effects of CTAB-modified PSP-Cubs on splenic lymphocytes were also investigated. When splenic lymphocytes were treated with CTAB-modified PSP-Cubs together with LPS or PHA , the results showed more favorable effects on cellular proliferation than blank cubosomes or free PSP at certain concentrations.

Mechanism elucidation of Polygonatum sibiricum-Prepared Cervus elaphus velvet antler in intervening non-alcoholic fatty liver disease based on network analysis combined with proteomics

Zhao Zihao,Li Minqian,Teng Hongbo,Wang Xv,Shi Mengqi,He Zhongmei,Zong Ying,Chen Weijia,Geng Jianan,Zhou Jia,Zhao Yan

To establish the standardized processing technology of velvet antler stir-fried with Polygonatum sibiricum juice, elucidate the core mechanism underlying the enhanced anti-non-alcoholic fatty liver disease (NAFLD) efficacy of the processed product, and identify the key bioactive components. Taking protein content as the screening index, this study optimized the processing parameters of Polygonatum sibiricum -Prepared Cervus elaphus Velvet Antler (PC-PR) using single-factor experiments combined with response surface methodology (RSM). The in vivo pharmacodynamic efficacy was validated in a high-fat diet-induced NAFLD rat model. Multi-omics technologies were integrated to screen for core differential proteins enriched after processing. The in vitro activity of the target component was verified in an oleic acid-induced inflammatory model of HepG2 cells, and Western blot (WB) analysis was employed to decipher its molecular regulatory mechanism. A standardized processing technology for PC-PR was successfully established, which induced specific alterations in the compositional profile of velvet antler. The processed product significantly improved blood glucose, body weight, liver function, and lipid metabolism indicators in NAFLD model rats, with superior efficacy compared to non-optimally processed velvet antler. Multi-omics screening and in vitro experiments confirmed that Q28237 is a core bioactive component specifically enriched after processing, exhibiting potent lipid-lowering and anti-inflammatory activities. Mechanistic studies revealed that Q28237 alleviates the pathological progression of NAFLD by constructing a “component-pathway-phenotype” regulatory network, which involves inhibiting the overactivation of epidermal growth factor receptor (EGFR), promoting the activation of the phosphatidylinositol 3-kinase-protein kinase B (PI3K-AKT) pathway, and activating estrogen receptor1 (ESR1) to exert synergisticanti-inflammatory effects.

Polygonatum sibiricum polysaccharide-based novel triptolide delivery derived from classic compatibility principles in traditional Chinese medicine: Simultaneous renal protection and toxicity attenuation in diabetic nephropathy

Pei Qiao,Shuang Chen,Mengqing Zhao,Zhiran Ju,Junxiang Chen,Wenhao Xu,Lichan Mao

Triptolide (TP) is a bioactive compound with anti-inflammatory, antitumor, and immunomodulatory effects. Its application, however, is limited due to poor aqueous solubility, low bioavailability, and organ toxicity. Based on the classical compatibility theory of Tripterygium and Polygonatum for toxicity reduction in traditional Chinese medicine, and the super-self-assembly behavior observed in the decoctions of these two herbs, a novel nano-delivery system (TP-PSP) was designed with Polygonatum Sibiricum Polysaccharide (PSP) as the carrier. TP-PSP achieved 6-fold enhancements in solubility and 3-fold enhancements in membrane permeability. The bioavailability of TP-PSP increased by 6.8/5.9-fold in AUC 0-t /AUC 0-∞ , significantly reducing gastrointestinal toxicity by shortening intestinal retention. In the diabetic nephropathy (DN) mouse model, TP-PSP achieved better therapeutic effects by enabling kidney-targeted delivery. TP-PSP reduced serum creatinine and urea nitrogen by 38.7% and 42.5%, respectively. It also alleviated TP-induced renal, cardiac, and hepatic toxicity. Physical characterization and molecular dynamic simulations revealed that TP and PSP form stable supramolecular aggregates via hydrogen bonding. TP-PSP activated the Nrf2/SLC7A11/GPX4 pathway, enhancing antioxidant defenses and suppressing lipid peroxidation/ferroptosis. This study not only reports a clinically viable TP delivery strategy but also highlights the application potential of traditional Chinese medicine principles in developing advanced nanomedicine with better effects but lower toxicity. Significance This work presents a significant advancement beyond classical Chinese medical principles to overcome the major clinical limitations of triptolide (TP), a potent yet therapeutically challenging compound. Guided by the “Sovereign-Minister-Assistant-Envoy” principle and systematic, detailed prescription research, we identified Polygonatum Sibiricum Polysaccharide (PSP) as a promising carrier to enhance the solubility and reduce the toxicity of TP. We developed an engineered nano-delivery system (TP-PSP) with improvements in TP's solubility, dissolution rate, and bioavailability while simultaneously reducing its systemic organ toxicity. The system demonstrates kidney-targeted delivery and a significant reduction in key nephropathy markers, revealing strong therapeutic potential for diabetic kidney disease. Furthermore, our integrated experimental and molecular dynamics approach elucidates the fundamental self-assembly mechanism between TP and PSP, which is driven by hydrogen bonding. Notably, we identified Nrf2 pathway activation and ferroptosis inhibition as a novel protective mechanism against TP-induced nephrotoxicity, effectively expanding its therapeutic window for clinical translation. This study successfully bridges traditional medicine wisdom with modern nanotechnology, addressing critical pharmacological and environmental challenges in drug development. Beyond the specific application of TP, this work validates a scientific principle from Chinese medicine for guiding the design of nanomedicine, offering a generalizable template for reformulating other potent but toxic herbal compounds.

Characterization of the flavor profile of Huangjiu brewed with Polygonatum sibiricum and Broomcorn millet using HS-SPME-GC × GC-TOF-MS, GC-IMS, intelligent sensory and molecular docking approaches

Baoqing Bai,Junnan Li,Ziqing Zhang,Ying Zhang,Tao Bo,Jinhua Zhang,Ligang Yu,Sanhong Fan,Yukun Yang

This study characterized the flavor profile of Huangjiu brewed with Polygonatum sibiricum and Broomcorn millet (HJMHJ) using headspace solid phase microextraction-comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (HS-SPME-GC × GC-TOF-MS), gas chromatography-ion mobility spectrometry (GC-IMS) and intelligent sensory technologies. The results identified six volatile compounds (isobutyric acid, dibutyl phthalate, α-terpineol, β-damascenone, 2-pentyl-furan and γ-nonalactone) as the key aroma compounds. Metabolic network analysis revealed that glycolysis plays a crucial role in Huangjiu metabolism. Ultra-fast gas-phase electronic nose (UF-GC-E-nose) demonstrated a strong association between HJMHJ and acetoin, while electronic-tongue (E-tongue) indicated that HJMHJ exhibited the highest response to freshness, the lowest to sweetness and an intermediate to bitterness. Molecular docking showed that OR1A1 exhibits the strongest binding energy (−8.1 kcal/mol) to β-damascenone through hydrogen bonding and hydrophobic interactions. This study provides theoretical support for the incorporation of herbs into Huangjiu production.
