Management of inflammatory bowel disease (IBD) is a real clinical challenge. Despite intense investigation, the mechanisms of IBD remain substantially unidentified. Some inflammatory conditions, such as matrix metalloproteinases (MMPs) and the nuclear factor-κB (NF-κB) and NOD-like receptor protein 3 (NLRP3) inflammasome signaling pathways, are reported to contribute to the development and maintenance of IBD. Regulation of their common upstream signaling, that is, reactive oxygen species (ROS), may be important to control the progression of IBD. In the present study, we found that procyanidin, a powerful antioxidation flavonoid, has a significant effect on ROS clearance on THP-1 macrophages after lipopolysaccharide (LPS) or LPS-combined adenosine triphosphate stimulation, thus downregulating MMP9 expression, suppressing NF-κB signaling, and interrupting the formation of the NLRP3 inflammasome. Moreover, our in vivo data showed that procyanidin attenuated Dextran sulfate sodium-induced experimental colitis in a dose-dependent fashion by suppressing the expression of MMP9, NF-κB, and NLRP3 inflammasome signaling in colonic tissues in mice. Overall, our results suggested that targeting ROS could be a potential therapeutic choice for colonic inflammation.

Grape seed procyanidin extract was used to green synthesize AuNPs (OPC-AuNPs), in which procyanidin (OPC) accounted for 95% of total content. To improve the dispersion and uniformity of particle size, various conditions were optimized. The final OPC-AuNPs showed excellent stability and were characterized by a series of techniques. Owing to the good quenching properties of OPC-AuNPs, the fluorescence system of OPC-AuNPs and Rhodamine B (RB) mixture was established for prazosin testing. OPC-AuNPs could effectively quench fluorescence based on fluorescence resonance energy transfer (FRET). When prazosin and the testing system of OPC-AuNPs and RB mixture coexisted in the solution, prazosin would replace RB and competitively bond on the surface of OPC-AuNPs, thus RB was dissociated from OPC-AuNPs, resulting in the rapid recovery of fluorescence. This article provided a green, simple one-pot synthesis method for OPC-AuNPs and established a fast, sensitive, visible fluorescence system for prazosin detection which was successfully applied in real sample detection.

Interactions between plant polyphenols and food allergens may be a new way to alleviate food allergies. The non-covalent interactions between the major allergen from peanut (Ara h 2) with procyanidin dimer (PA2) were therefore characterized using spectroscopic, thermodynamic, and molecular simulation analyses. The main interaction between the Ara h 2 and PA2 was hydrogen bonding. PA2 statically quenched the intrinsic fluorescence intensity and altered the conformation of the Ara h 2, leading to a more disordered polypeptide structure with a lower surface hydrophobicity . In addition, the in vitro allergenicity of the Ara h 2-PA2 complex was investigated using enzyme-linked immunosorbent assay (ELISA) kits. The immunoglobulin E (IgE) binding capacity of Ara h 2, as well as the release of allergenic cytokines, decreased after interacting with PA2. When the ratio of Ara h 2-to-PA2 was 1:50, the IgE binding capacity was reduced by around 43 %. This study provides valuable insights into the non-covalent interactions between Ara h 2 and PA2, as well as the potential mechanism of action of the anti-allergic reaction caused by binding of the polyphenols to the allergens.

In this study, we prepared adipocyte-targeted and glutathione-responsive PC nanoparticles by anti-solvent precipitation method, and evaluated the intervention effect of nanoparticles on lipid metabolism disorders. PC@cBSA nanoparticles had spherical morphology with a PC encapsulation efficiency of 79.46 ± 1.82%, which could effectively maintain the antioxidant activity of PC under harsh conditions, and resist the damage of simulated gastrointestinal digestion. The PC release rate from nanoparticles was positively correlated with glutathione concentration, indicating nanoparticles had a glutathione-responsive property. Subsequently, the mature 3T3-L1 adipocyte model was constructed, after intervention with PC@cBSA nanoparticles, the average area of lipid droplets in adipocytes decreased to 17.89 ± 0.91%. The levels of TG, T-CHO and LDL-C were down-regulated, while the level of HDL-C was upregulated, proving that nanoparticles had a regulatory effect on the lipid metabolism disorder of mature adipocytes. The PC nanoparticles developed effectively enhance the stability and achieve favorable targeted delivery efficiency of PC in adipocytes, which provide a novel strategy for the intervention of lipid metabolism-related diseases.

The interactions between bovine α-lactalbumin and procyanidin B2 were fully investigated by spectroscopic methods and molecular docking. This study hypothesized that ALA could spontaneously interact with procyanidin B2 to form protein-based complex delivery carrier. Far UV CD and FTIR data demonstrated ALA’s secondary structures were altered and intrinsic fluorescence quenching suggested ALA conformation was changed with procyanidin B2. Calorimetric technique illustrated ALA-procyanidin B2 complexation was a spontaneous and exothermic process with the number of binding site (n, 3.53) and the binding constant (K b , 2.16 × 10 4 M −1 ). A stable nano-delivery system with ALA can be formed for encapsulating, stabilizing and delivering procyanidin B2. Molecular docking study further elucidated that hydrogen bonds dominated procyanidin B2 binding to ALA in a hydrophobic pocket. This study shows great potential in using ALA as protein-based nanocarriers for oral delivery of hydrophilic nutraceuticals, because procyanidin B2-loaded ALA complex delivery systems can be spontaneously formed.

Inflammatory bowel disease (IBD) is an idiopathic chronic inflammatory bowel disease characterized by inflammation, intestinal barrier injury, and imbalance of gut microbiota. Excess accumulation of reactive oxygen species (ROS) is closely correlated with the development and reoccurrence of IBD. Previous researches demonstrate that procyanidin, as a natural antioxidant, exhibits strong ability of eliminating ROS, thus showing good therapeutic effects in the inflammation-related diseases. Non-etheless, its poor stability and solubility always limits the therapeutic outcomes. Here, we typically designed an antioxidant coordination polymer nanoparticle using the engineering of procyanidin (Pc) and free iron (Fe), named Pc-Fe nanozyme, for effectively scavenging ROS and further inhibiting inflammation while altering the gut microbiome for the treatment of colitis. Furthermore, in vitro experiments uncover that Pc-Fe nanoparticles exert strong multi biomimic activities, including peroxidase, and glutathione peroxidase, for the scavenging of ROS and protecting cells from oxidative injury. In addition, the colon accumulation of Pc-Fe nanozyme effectively protects the intestinal mucosa from oxidative damage while significantly downregulates pro-inflammatory factors, repairs the intestinal barriers and alternates gut microbiome after orally administrated in sodium dextran sulfate (DSS) induced colitis mice. The results collectively illustrate that the multienzyme mimicking Pc-Fe nanozyme owns high potential for treating IBD through scavenging ROS, inhibiting inflammation, repairing gut barriers and alternating gut microbiome, which further promising its clinical translation on IBD treatment and other ROS induced intestinal diseases.

Procyanidin C1 (PCC1) is a biologically relevant oligomeric small molecule and a trimeric epicatechin with the potential to modulate gut microbiota. However, how it reshapes microbiota-linked phosphatidylcholine homeostasis in the context of insulin resistance remains insufficiently defined, limiting the establishment of mechanism-informed design principles for microbiota-directed metabolic modulation. Here, PCC1 improved systemic glucose and insulin sensitivity, promoted hepatic glycogen accumulation, reduced hepatic steatosis, attenuated skeletal muscle fibrosis, and decreased lipid deposition in adipose tissues. Notably, 16S rRNA sequencing revealed that PCC1 treatment enriched Lactobacillus johnsonii and depleted Leptogranulimonas caecicola , which were positively associated with elevated GPCho (16:0/20:5) and circulating eicosapentaenoic acid (EPA) identified by untargeted metabolomics, indicative of enhanced α-linolenic acid metabolism and a gut microbiota-driven contribution to EPA biosynthesis. Meanwhile, lipidomics revealed that PCC1 reduced diacylglycerols, triacylglycerols, ceramides, and lysophosphatidylcholines, while restoring EPA-containing phosphatidylcholines. Transcriptomics further demonstrated that PCC1 induced tissue-specific metabolic reprogramming, activating PPAR signaling in liver, enhancing oxidative phosphorylation in brown adipose tissue (BAT), and stimulating AMPK signaling in skeletal muscle. Importantly, PCC1 reversed HFD-induced phospholipase A2 dysregulation, upregulating Pla2g2d in skeletal muscle and downregulating Pla2g7 in liver and BAT, two genes implicated in EPA release and phospholipid remodeling. Together, integrated multi-omics analyses identify PCC1 as an oligomeric small-molecule modulator associated with improved insulin sensitivity across multiple tissues and implicate an EPA-linked phosphatidylcholine signaling axis that connects microbiota remodeling with host metabolic responses.

Our previous study has revealed that procyanidin A 1 (A 1 ) and its simulated digestive product (D-A 1 ) can alleviate acrylamide (ACR)-induced intestine cell damage. However, the underlying mechanism remains unknown. In this study, we elucidated the molecular mechanism for A 1 and D-A 1 to alleviate ACR-stimulated IPEC-J2 cell damage. ACR slightly activated nuclear factor erythroid 2-related factor 2 (Nrf2) signaling and its target genes, but this activation could not reduce intestine cell damage. A 1 and D-A 1 could alleviate ACR-induced cell damage, but the effect was abrogated in cells transiently transfected with Nrf2 small interfering RNA (siRNA). Further investigation confirmed that A 1 and D-A 1 interacted with Kelch-like ECH-associated protein 1 (Keap1), which boosted the stabilization of Nrf2, subsequently promoted the translocation of Nrf2 into the nucleus, and further increased the expression of antioxidant proteins, thereby inhibiting glutathione (GSH) consumption, maintaining redox balance and eventually alleviating ACR-induced cell damage. Importantly, there was no difference between A 1 and D-A 1 treated groups, indicating that A 1 can tolerate gastrointestinal digestion and may be a potential compound to limit the toxicity of ACR.

Alzheimer's disease (AD) is a chronic neurodegenerative disease with high prevalence, long duration and poor prognosis. The blood-brain barrier (BBB) is a physiologic barrier in the central nervous system, which hinders the entry of most drugs into the brain from the blood, thus affecting the efficacy of drugs for AD. Natural products are recognized as one of the promising and unique therapeutic approaches to treat AD. To improve the efficiency and therapeutic effect of the drug across the BBB, a natural polyphenolic compound, procyanidin C-1 (C1) was encapsulated in glucose-functionalized bovine serum albumin (BSA) nanoparticles to construct Glu-BSA/C1 NPs in our study. Glu-BSA/C1 NPs exhibited good stability, slow release, biocompatibility and antioxidant properties. In addition, Glu-BSA/C1 NPs penetrated the BBB, accumulated in the brain by targeting Glut1, and maintained the BBB integrity both in vitro and in vivo . Moreover, Glu-BSA/C1 NPs alleviated memory impairment of 5 × FAD mice by reducing Aβ deposition and Tau phosphorylation and promoting neurogenesis. Mechanistically, Glu-BSA/C1 NPs significantly activated the PI3K/AKT pathway and inhibited the NLRP3/Caspase-1/IL-1β pathway thereby suppressing neuroinflammation. Taken together, Glu-BSA/C1 NPs could penetrate the BBB and mitigate neuroinflammation in AD, which provides a new therapeutic approach targeting AD.

The metabolite profiles and distributions of procyanidin B2 were qualitatively described using UPLC-DAD-ESI-IT-TOF-MSn without help of reference standards, and a possible metabolic pathway was proposed in the present study. Summarily, 53 metabolites (24 new metabolites) were detected as metabolites of procyanidin B2, and 45 of them were tentatively identified. Twenty seven metabolites were assigned as similar metabolites of (−)-epicatechin by scission of the flavanol interflavanic bond C4–C8, including 16 aromatic metabolites, 5 conjugated metabolites, 3 ring-cleavage metabolites, and 2 phenylvalerolactone metabolites. Additionally, 14 metabolites were conjugates of free procyanidin B2, comprising 9 methylation metabolites, 8 sulfation metabolites, 5 hydration metabolites, 2 hydroxylation metabolites, 1 hydrogenation metabolites, and 1 glucuronidation metabolites. The results of metabolite distributions in organs indicated that the conjugated reaction of free procyanidin B2 mainly occurred in liver and diversified metabolites forms were observed in small intestine. The metabolic components of procyanidin B2 identified in mice provided useful information for further study of the bioactivity and mechanism of its action.

Procyanidins can alleviate small-intestine damage induced by acrylamide (ACR). However, little is known about whether procyanidins, after gastrointestinal digestion, can prevent ACR-induced intestinal barrier damage and the possible mechanism. Here, Caco-2 cells were differentiated into an intestinal epithelial cell monolayer membrane, which was stimulated with or without ACR in the presence or absence of procyanidin A1 (A1) and its digestive products (D–A1). Our findings show that both A1 and D–A1 significantly increased the transepithelial electrical resistance (TEER) value; decreased FITC-dextran 4 kDa (FITC-4 kDa) permeability, apoptosis and lactic dehydrogenase (LDH) release; and enhanced the expression of claudin-1, occludin and zonula occludens-1 (ZO-1) in ACR-induced Caco-2 cell monolayer membrane. In addition, A1 and D–A1 suppressed ACR-induced phosphorylation of mitogen-activated protein kinase (MAPK). Finally, A1 and D–A1 inhibited the myosin light chain kinase (MLCK) signaling pathway, thereby maintaining normal intestinal barrier functions, similar to the MLCK inhibitor in ACR-induced Caco-2 cell monolayer membrane. These findings indicate that A1 can alleviate ACR-induced intestinal barrier dysfunction via inhibiting the MAPK/MLCK signaling pathway, and it still has excellent inhibitory effects after digestion.

A synergistic imprinting strategy of covalent and non-covalent interactions is proposed to prepare magnetic molecularly imprinted polymers (DI-MMIPs) for highly selective separation of procyanidin B2 (PC) from grape seed samples. Dopamine and 3-amino-phenylboronic acid as cooperative functional monomers endow the imprinted sites with synergistic tailoring. Benefiting from the synergistic effect, the DI-MMIPs exhibit enhanced imprinting performance with high adsorption capacity (27.71 mg g −1 ), fast kinetic equilibrium time (within 30 min), outstanding selectivity (IF = 5.8, SC > 3.2), and satisfactory regeneration ability. In addition, the DI-MMIPs possess good magnetism, uniform morphology with typical core-shell structure, and stable crystallization. Furthermore, the established DI-MMIPs coupled with HPLC-UV (~ 280 nm) method has a wide linearity range of 0.05–200 μg mL −1 with correlation coefficient of 0.9997, high recoveries (> 93.1%) with RSDs from 2.9 to 5.5%, and low LOD (0.0008 μg mL −1 ). Consequently, this work provides an effective and easily tailored way to fabricate magnetic imprinted nanomaterials with both rapid recognition rate and high selectivity and thus holds great promise to realize the extraction and detection of PC from real samples.
