PTX-loaded, polysorbate 80-functionalized brain-targeting pullulan nanoparticles for drug delivery

Huabing Yuan,Lu Han,Hao Deng,Yue Tan,Yi Yang,Yi Liu,Jiaxin Ying,Mengqi Fang,Hui Wei,Zhihe Tao

Brain-targeted drug delivery remains a major challenge in pharmaceutical research. In this study, four types of polysorbate 80 and cholesterol-modified pullulan polymers (CHPP) were synthesized and self-assembled into nanoparticles (CHPP NPs) for brain drug delivery. The properties of these NPs, including brain targeting efficiency, were systematically evaluated to investigate the influence of polymer modification on their performance. The hydrophobicity of the polymers increased with the degree of cholesterol substitution, which in turn led to a reduction in nanoparticle size. Furthermore, higher hydrophobicity contributed to an increased drug loading capacity for paclitaxel (PTX) and a more sustained drug release profile. In vivo fluorescence imaging revealed that formulations with higher levels of polysorbate 80 and cholesterol modification exhibited significantly enhanced brain targeting efficiency compared to other variants. The PTX-loaded CHPP NPs (PTX-CHPP NPs) demonstrated potent cytotoxicity and inhibitory effects against SJ-GBM2 glioma cells in vitro, underscoring their potential as a promising platform for brain-targeted drug delivery. These findings provide valuable insights for the rational design of brain-targeting nanocarriers.

Enhancing physical properties of chitosan/pullulan electrospinning nanofibers via green crosslinking strategies

Zeyu Qin,Xiwen Jia,Qian Liu,Baohua Kong,Hao Wang

In this study, electrospun chitosan/pullulan composite nanofiber films were crosslinked by two green methods (heating and cinnamaldehyde). With the increase of chitosan content, the morphology results indicated that the nanostructures of thermal crosslinking chitosan/pullulan (TCP) and cinnamaldehyde crosslinking chitosan/pullulan (CCP) nanofiber films became more stable. The decreases in weight loss (83.65 % to 43.85 % for TCP and 23.42 % to 15.58 % for CCP) and the increases in water contact angle (31.8° to 54.1° for TCP and 83.65 % to 43.85 % for CCP) confirmed the improved water stability. The decreases in water vapor permeability indicated that the crosslinking processes significantly improved the barrier properties of the films. Mechanical and thermal properties of TCP and CCP films were enhanced respectively. Furthermore, the corresponding properties of CCP films were more excellent than TCP films. FTIR results indicated the occurrence of Maillard reaction during the thermal crosslinking process and Schiff base reaction during the cinnamaldehyde crosslinking process.

Superabsorbent polysaccharide hydrogels based on pullulan derivate as antibacterial release wound dressing

Huanan Li,Jing Yang,Xiaona Hu,Jie Liang,Yujiang Fan,Xingdong Zhang

To accomplish ideal wound dressing, hydrogels based on a natural polysaccharide, pullulan were synthesized by chemical cross-linking. The tensile strengths of the hydrogel films (1 mm thick) were determined to range from 0.663 to 1.097 MPa in proportion to cross-linking degrees and water contents. The swelling study of the hydrogels in water showed remarkable water absorption property with swelling ratio up to 4000%, which provided the hydrogel with quick hemostatic ability and prevent the wound bed from accumulation of exudates. The water vapor transmission rate and water retention of the hydrogels were found to be in the range of 2213–3498g/m 2 /day and 34.74–45.81% (after 6 days), indicating that the hydrogel can maintain a moist environment over wound bed, which could prevent the dehydration of the wound bed and prevent the scab formation. Biocompatibility test revealed that the hydrogels were not cytotoxic. The hydrogel could load antimicrobial agents and effectively suppress bacterial proliferation to protect the wound from bacterial invasion. These results suggest that the pullulan hydrogels prepared in this study may have high potential as new ideal wound-dressing materials. © 2011 Wiley Periodicals, Inc. Self-crosslinking and injectable chondroitin sulfate/pullulan hydrogel for cartilage tissue engineering

Tao Li,Xiongbo Song,Changmei Weng,Xin Wang,Li Sun,Xiaoyuan Gong,Liu Yang,Cheng Chen

Injectable hydrogels derived from natural polysaccharides have great application prospects in cartilage tissue engineering (CTE). In this study, chondroitin sulfate was functionalized with adipic dihydrazide (CS-ADH), and pullulan was oxidized (oxPL). We fabricated a self-crosslink, biodegradable, in situ forming, injectable hydrogel by covalent hydrazone crosslinking of polysaccharides, without using extra cross-linking agents and releasing harmful byproducts under physiological conditions. The gelation time, equilibrium swelling, degradation behavior, network morphology and mechanical properties of CS-ADH/oxPL hydrogels were well manipulated by varying the weight ratio and concentration of CS-ADH and oxPL. In addition, encapsulation of rabbit articular chondrocytes in CS-ADH/oxPL hydrogels was observed after 14 days culture. Live/Dead staining assay demonstrated that the hydrogel system possessed good cytocompatibility. Sulfated glycosaminoglycan (sGAG) contents indicated that the hydrogel system favored cartilaginous extracellular matrix (ECM) deposition. Furthermore, the hydrogel system could maintain the chondrocyte phenotype and enhance chondrogenesis, which was evaluated in terms of their protein and gene expression. These features support that CS-ADH/oxPL hydrogels have a potential as an injectable cell delivery carrier scaffold in CTE.

Polydopamine/montmorillonite-embedded pullulan hydrogels as efficient adsorbents for removing crystal violet

Xiaoliang Qi,Qiankun Zeng,Xianqin Tong,Ting Su,Lei Xie,Kai Yuan,Jianxiong Xu,Jianliang Shen

Both secondary pollution and the low mechanical strength of adsorbents have severely impeded the practical application of adsorption methods in the dye wastewater treatment. In this work, we innovatively synthesized a composite hydrogel adsorbent by incorporating polydopamine (PDA) and montmorillonite (MMT) into the pullulan hydrogel matrix for dye adsorption. First, the successful formation of the resultant adsorbents was verified by Fourier-transform infrared spectroscopy and scanning electron microscope elemental mapping analysis. Then, several physicochemical properties (such as thermal and swelling properties, microarchitecture, and mechanical strength) of the five prepared adsorbents (PM1–PM5) were investigated. These results demonstrated the adsorbents had tunable properties, which can be achieved by adjusting the PDA/MMT mass ratio. Next, the dye adsorption performance was systematically explored. The resultant adsorbent (PM3) had a maximum adsorption capacity of 112.45 mg/g and its adsorption data was best described by a Langmuir isotherm and pseudo-second-order kinetic. Finally, the adsorption mechanisms and potential commercial practicability of the adsorbent were studied. Altogether, the designed adsorbent could effectively avoid generating the secondary pollution typical of adsorbents and it displayed excellent mechanical strength, thus opening up a new horizon in mitigating environmental pollution from textile industries.

Synthesis of oxidized pullulan coated mesoporous silica for pH-sensitive drug delivery

Shangji Li,Wei Dai,Zheng-Zhi Yin,Jun Gao,Datong Wu,Yong Kong

The past decades have witnessed the development of mesoporous silica as an attractive carrier for controlled drug delivery due to its outstanding mesoporous feature, facile surface functionalization and good biocompatibility. In this work, NH 2 grafted mesoporous silica nanoparticles (NH 2 -MSN) were first synthesized for the loading of 5-fluorouracil (5-FU), and then the drug loaded NH 2 -MSN were coated with oxidized pullulan (oxPL) via Schiff base reaction for the encapsulation of 5-FU. The resultant drug carrier, 5-FU/MSN/oxPL, was fully characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), Dynamic light scattering (DLS), 1 H NMR, Fourier transform infrared (FT-IR) spectroscopy and nitrogen adsorption/desorption isotherms, respectively. Finally, controlled delivery of 5-FU from the carrier was investigated. Since the acylhydrazone bond between MSN and oxPL is prone to be hydrolyzed under acidic conditions, the encapsulated 5-FU is easily released from the MSN/oxPL carrier in acidic environment due to the separation of oxPL coating from MSN. Cytotoxicity of the developed carrier was also evaluated using LO2 and HepG2 cell lines by conventional CCK-8 assay.

A novel photocurable pullulan-based bioink for digital light processing 3D printing

Feng Zhaoxuan,Li Jinsong,Zhou Dasen,Song Hui,Lv Jiaqi,Bai Wenqin

Digital light processing has significant advantages, such as high repeatability, low failure rate, and no extrusion shear force. To make it possible to print complex structures with high resolution, the consecutive development of photocurable ink materials is indispensable. In this work, photo-functionalized pullulan (Pul-NB) was prepared by introducing norbornene groups into pullulan chains, and an ink material suitable for photocurable printing was prepared by thiol-ene click reaction. The rheology, water absorption, and mechanical properties of the Pul-NB precursor solution and photocurable hydrogel were investigated. The optimal composition of Pul-NB ink for three-dimensional (3D) printing was obtained by adjusting the degree of substitution, Pul-NB concentration, and thiol crosslinking agent. This novel bioink for digital light processing 3D printing showed good printability and high shape fidelity. This ink material provides an excellent alternative for printing biomimetic soft tissue organs, high-throughput tissue models, soft robots, etc.

Dual functional pullulan-based spray-dried microparticles for controlled pulmonary drug delivery

Hao-Ying Li,En-Yu Xu

Two main challenges are associated with current spray-dried microparticles for inhalation, including the enhancement of aerosolization performance of microparticles and the creation of sustained drug release for continuous treatment on-site. For achieving these purposes, pullulan was explored as a novel excipient to prepare spray-dried inhalable microparticles (with salbutamol sulphate, SS, as a model drug), which were further modified by additives of leucine (Leu), ammonium bicarbonate (AB), ethanol and acetone. It was demonstrated that all pullulan-based spray-dried microparticles had improved flowability and enhanced aerosolization behavior, with the fine particle (<4.46µm) fraction of 42.0-68.7% w/w, much higher than 11.4% w/w of lactose-SS. Moreover, all modified microparticles showed augmented emitted fractions of 88.0-96.9% w/w, over 86.5% w/w of pullulan-SS. The pullulan-Leu-SS and pullulan-(AB)-SS microparticles demonstrated further increased fine particle (<1.66µm) doses of 54.7µg and 53.3µg respectively, surpassing that (49.6µg) of pullulan-SS, suggesting an additionally increased drug deposition in the deep lungs. Furthermore, pullulan-based microparticles revealed sustained drug release profiles with elongated time (60mins) over the control (2mins). Clearly, pullulan has a great potential to construct dual functional microparticles for inhalation with improved pulmonary delivery efficiency and sustained drug release on-site.

Regulation of whey protein emulsion gel's structure with pullulan to enhance astaxanthin bioaccessibility

Qianxi Zhao,Yankai Liu,Jiale Feng,Jiahua Zhang,Xinzhu Wang,Xun Li,Wantang Ji,Yu Wang,Bo Nan,Jingsheng Liu,Xia Li,Yuhua Wang

In this study, the potential of using an emulsion gel based on whey protein concentrate (WPC) and pullulan (PUL) to encapsulate and deliver astaxanthin (AST) was investigated. PUL concentration was observed to affect the microstructure of WPC/PUL/AST emulsion gels, and the performance of emulsion gels was evaluated by encapsulation efficiency, simulated gastrointestinal digestion, storage stability, hardness, and water holding capacity tests. The WPC/PUL/AST emulsion gels had the highest encapsulation efficiency, gastrointestinal digestion retention, and bioaccessibility of (91.70 ± 1.47) %, (80.96 ± 5.02) %, and (40.69 ± 1.16) % at 2.0 % PUL concentration, respectively. Furthermore, adding PUL to the emulsion gels improved their hardness, water holding capacity, and storage stability. The findings suggested a potential method to optimize the bioaccessibility of AST in emulsion gels. It provided the possibility for the efficient application of AST as a functional food.

Electrospun gelatin/tea polyphenol@pullulan nanofibers for fast-dissolving antibacterial and antioxidant applications

Jianfeng Zhou,Weiqiang Wang,Xingjian Yang,Deng-Guang Yu,Ping Liu

Bio-based active food packaging materials have a high market demand. We use coaxial electrospinning technology to prepare core–shell structured nanofibers with sustained antibacterial and antioxidant properties. The fiber core layer was composed of gelatin and tea polyphenols, whereas tea polyphenols provide antibacterial and antioxidant properties; the fiber sheath was composed of pullulan polysaccharides with antioxidant properties. By using a scanning electron microscope, it can be seen that the diameter distribution of the prepared nanofibers was uniform and the surface is smooth; using a transmission electron microscope, it can be clearly seen that the nanofibers have a core–shell structure; Fourier Transform Infrared and X-ray diffraction analysis indicate that the nanofibers have an amorphous structure; the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging shows that nanofibers have higher antioxidant properties with the addition of tea polyphenols; antibacterial test showed that nanofibers had obvious inhibitory effect on the growth of Staphylococcus aureus and Escherichia coli ; and the nanofiber film dissolution test shows that nanofibers can be used as fast soluble active packaging. Finally, core–sheath-structured nanofibers can serve as active packaging for instant food, possessing both rapid water solubility and excellent antibacterial and antioxidant activity, making water-soluble nanofibers interesting applications in the field of food packaging.

Polydopamine Nanoparticle-Loaded Antifreezing and Photothermal Organohydrogel Based on Pullulan for Frostbite Therapy

Wenzhuang Cui,Ying Yue,Yujie Liu,Jun Wang,Zhizhou Yang,Yin Qiang,Chu Gong,Jun-Li Yang

Rapid rewarming is the standard and most common strategy for treating frostbite. Due to freezing susceptibility and lack of thermal effects, traditional therapeutic hydrogels are not suitable for being directly applied to frostbite therapy in cold conditions. Contrastively, antifreezing and photothermal hydrogels that are not apt to freeze and capable of rewarming frostbite wounds are deemed to hold great application potential in such therapy. Nevertheless, these hydrogels have rarely been researched. Herein, using glycerol as the cryoprotectant and polydopamine nanoparticles (PDA NPs) as the photothermal agent, a novel pullulan-based antifreezing and photothermal organohydrogel (CPG-PDA organohydrogel) was successfully developed to treat frostbite for the first time. The CPG-PDA organohydrogel formed through 1-ethyl-3-(3-(dimethylamino)propyl)carbodiimide hydrochloride (EDC·HCl)-mediated esterification reactions was found to possess certain mechanical stability, shear-thinning behaviors (injectability), excellent antifreezing properties, superb in vitro and in vivo photothermal performances, and outstanding cytocompatibility and hemocompatibility. Most noticeably, the photothermal rewarming and coating therapy using the CPG-PDA organohydrogel was observed to significantly accelerate the frostbite healing of rats. The CPG-PDA organohydrogel was opined to be a promising platform for the direct treatment of frostbite in a cold environment and would open a new avenue for the design of therapeutic strategies for frostbite.

Novel antibacterial pullulan derivatives modified with quaternary phosphonium salts for infected wound treatments

Wenzhuang Cui,Yujie Liu,Ying Yue,Jun Wang,Xianhua Meng,Zhizhou Yang,Chu Gong,Jun-Li Yang

The development of novel antibacterial agents is urgently needed to tackle bacterial infection, the major global issue menacing human health. Among them, polymeric quaternary phosphonium salts are worth noticing owing to their strong antibacterial activities and other merits including low bacterial drug resistance. Herein, pullulan modified with quaternary phosphonium salts (PQP) was synthesized using esterification reactions of pullulan and (5-carboxypentyl)triphenylphosphonium bromide (CPTPPB) mediated by N,N′‑carbonyldiimidazole (CDI), and was evaluated as novel antibacterial agents for treating wound infection for the first time. The chemical structures, chemical bonding, elemental compositions, crystalline properties and thermostability of PQP were systematically investigated. PQP exhibited in vitro antibacterial activities against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ), which were unveiled by the spread plate method and possibly resulted from the damage of bacterial cell walls/membranes mediated by electrostatic and hydrophobic interactions according to preliminary mechanism studies. Weak cytotoxicity and excellent hemocompatibility of PQP at the effective bactericidal concentration were observed. Moreover, in the infected wound model of mice, PQP was capable of disinfecting the wound and accelerating the healing. We opine PQP in this work is promising for antibacterial applications and will inspire the synthesis of novel antibacterial agents derived from natural polymers.
