Lycorine inhibits lipopolysaccharide-induced iNOS and COX-2 up-regulation in RAW264.7 cells through suppressing P38 and STATs activation and increases the survival rate of mice after LPS challenge

Jingjing Kang,Yushun Zhang,Xiang Cao,Jie Fan,Guilan Li,Qi Wang,Ying Diao,Zhihui Zhao,Lan Luo,Zhimin Yin

As a natural alkaloid extracted from Amaryllidaceae , lycorine shows various biological effects on tumor cells. Here we show that lycorine dose-dependently inhibited the LPS-induced up-regulation of nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein level in RAW264.7 cells. Besides, it also inhibited NO, PGE 2 , TNF-α and IL-6 release from LPS-treated RAW264.7 cells. RT-PCR experiments showed that lycorine suppressed LPS-induced iNOS but not COX-2 gene expression. Moreover, lycorine decreased LPS-induced mortality in mice. Mechanistically, LPS-induced activation of P38 and STATs pathways was suppressed significantly by lycorine. In addition, lycorine did not interfere with the phosphorylation of ERK1/2, JNK1/2 and NF-κB pathways. In conclusion, lycorine inhibits LPS-induced production of pro-inflammatory mediators and increases the survival rate of mice after LPS challenge, suggesting that lycorine could play an anti-inflammatory role in response to LPS.

The Anti-Inflammatory Activities of Sphagnum palustre L. Ethanol Extract to Control Inflammation in RAW264.7 Cells

Zhi Wu,Jiahui Zheng,Yang Xu,Dairong Wang,Qingbin Han,Ping Liu,Xiaorong Liu,Lanyue Zhang

Discomfort caused by inflammation leads to stress and anxiety in patients and seriously decreases the patients' quality of life. People prefer to use natural products instead of anti-inflammatory drugs because of their low toxicity and side effects. Studies have shown that Sphagnum palustre L. ( S. palustre ) can be used as medicinal plant, but few studies have focused on its anti-inflammatory effects. This study explored the mechanism of action of the ethanol extract of the peat moss plant S. palustre on lipopolysaccharide-induced inflammation in macrophage RAW264.7 cells. Components in S. palustre ethanol extracts (SPE) were identified by HPLC-MS, which mainly included 4-methoxybenzaldehyde, 4-methoxycinnamaldehyde and oleanolic acid. The effects of different concentrations (6.25–100 μg/mL) of SPE after 24 h administration were evaluated to establish a cellular inflammation model. Three biological replicates were performed based on each experiment, the MTT assay results showed that a low concentration of SPE promoted cell proliferation marked by Formazan. In a neutral red uptake assay, the SPE group was effectively inhibited the cell phagocytosis rate. With the increase of SPE concentration, intracellular ROS release decreased, which detected by DCFH-DA. Immunofluorescence assay result showed that SPE inhibited the release of reactive oxygen species from macrophages with fluorescent markers and DAPI. SPE inhibited the release of nitric oxide from macrophages as well. What's more, SPE significantly decreased the protein expression of interleukin (IL)-1, IL-6, and nuclear factor (NF)-κB according to enzyme-linked immunosorbent and immunocytochemical assays. SPE reduces inflammation in macrophage RAW264.7 cells and thus is a promising natural anti-inflammatory plant.

Purification and Structural Characterization of Polysaccharides from Polygonum multiflorum Thunb. and Their Immunostimulatory Activity in RAW264.7 Cells

Yan Gou,Donglin Gu,Jing Fan,Jianbo Yang,Ke Zan,Jingjing Liu,Hongyu Jin,Ying Wang,Feng Wei,Shuangcheng Ma

Polygonum multiflorumThunb. (PM) and derived products are broadly utilized in Chinese traditional medicine. According to our previous research, PM mostly contains polysaccharides, which display a wide range of biological activities. Two water-soluble polysaccharides (PMPs-1 and PMPs-2) were obtained from PM by DEAE-Cellulose and Sephadex G-100 column chromatography. Colorimetry, HPGPC-MALLS-RID, HPLC-PDA, methylation, FT-IR, NMR, and SEM were used to characterize these polysaccharides. PMPs-1 and PMPs-2 had average molecular weights of 255.5 and 55.7 kDa, respectively. PMPs-1 consisted of Man, Glc, Gal, and Ara at 0.9:78.6:1.0:1.6 and was a glucan with → 4)-Glcp-(1 → as a backbone. Meanwhile, PMPs-2, an acidic polysaccharide, comprised Rha, GalA, Glc, Gal, and Ara at 3.2:20.3:2.7:1.0:8.3. PMPs-1 and PMPs-2 significantly improved the proliferation of RAW 264.7 cells and induced NO, TNF-α, and IL-6 release. This study reveals that these two polysaccharides can be explored as novel immunomodulators and provide a basis for further development of PM in food and pharmaceutical industries.

Zi Xu,Yong Fang,Yue Chen,Wenjian Yang,Ning Ma,Fei Pei,Benard Muinde Kimatu,Qiuhui Hu,Weifen Qiu

The protective capacity of Se-containing protein hydrolysates with molecular weight below 1   kDa (SPHs-3), against Pb 2+ -induced damage in PC12 and RAW264.7 cells, was investigated in this study. The cell viability, levels of reactive oxygen species (ROS), lactate dehydrogenase (LDH), superoxide dismutase (SOD), nitric oxide (NO), malondialdehyde (MDA) and glutathione (GSH) in cell were analyzed. Results showed that 100   μg/ml of SPHs-3 pretreatment could significantly increase cell viability by 24.9% and 23.0% in Pb 2+ -treated PC12 and RAW264.7 cells, respectively ( P   <   0.01). The levels of ROS, NO, LDH and MDA were reduced by 32.2%, 68.2%, 79.7% and 73.7% in 100   μg/ml SPHs-3 pretreated PC12 cells, respectively ( P   <   0.01). SPHs-3 pretreatment was also associated with increases of SOD activity and GSH content in cells. In conclusion, SPHs-3 could protect cells against Pb 2+ -induced cytotoxicity, suggesting that Se-enriched rice may be a feasible candidate to improve health standard of the Pb 2+ -pollution population.

Ramulus Cinnamomi essential oil exerts an anti-inflammatory effect on RAW264.7 cells through N-acylethanolamine acid amidase inhibition

Wei Jia,Xiwen He,Wenhui Jin,Jinping Gu,Siyu Yu,Jianlin He,Zhiwei Yi,Bing Cai,Huiyuan Gao,Longhe Yang

Ethnopharmacological relevance Ramulus Cinnamomi , the dried twig of Cinnamomum cassia (L.) J.Presl., is a traditional Chinese medicine (TCM) with anti-inflammatory effects. The medicinal functions of Ramulus Cinnamomi essential oil (RCEO) have been confirmed, although the potential mechanisms by which RCEO exerts its anti-inflammatory effects have not been fully elucidated. Aim of the study To investigate whether N -acylethanolamine acid amidase (NAAA) mediates the anti-inflammatory effects of RCEO. Materials and methods RCEO was extracted by steam distillation of Ramulus Cinnamomi , and NAAA activity was detected using HEK293 cells overexpressing NAAA. N -Palmitoylethanolamide (PEA) and N -oleoylethanolamide (OEA), both of which are NAAA endogenous substrates, were detected by liquid chromatography with tandem mass spectrometry (HPLC-MS/MS). The anti-inflammatory effects of RCEO were analyzed in lipopolysaccharide (LPS)-stimulated RAW264.7 cells, and the cell viability was measured with a Cell Counting Kit-8 (CCK-8) kit. The nitric oxide (NO) in the cell supernatant was measured using the Griess method. The level of tumor necrosis factor-α (TNF-α) in the RAW264.7 cell supernatant was determined using an enzyme-linked immunosorbent assay (ELISA) kit. The chemical composition of RCEO was assessed by gas chromatography-mass spectroscopy (GC-MS). The molecular docking study for (E)- cinnamaldehyde and NAAA was performed by using Discovery Studio 2019 software (DS2019). Results We established a cell model for evaluating NAAA activity, and we found that RCEO inhibited the NAAA activity with an IC 50 of 5.64 ± 0.62 μg/mL. RCEO significantly elevated PEA and OEA levels in NAAA-overexpressing HEK293 cells, suggesting that RCEO might prevent the degradation of cellular PEA and OEA by inhibiting the NAAA activity in NAAA-overexpressing HEK293 cells. In addition, RCEO also decreased NO and TNF-α cytokines in lipopolysaccharide (LPS)-stimulated macrophages. Interestingly, the GC-MS assay revealed that more than 93 components were identified in RCEO, of which (E)- cinnamaldehyde accounted for 64.88%. Further experiments showed that (E)- cinnamaldehyde and O -methoxycinnamaldehyde inhibited NAAA activity with an IC 50 of 3.21 ± 0.03 and 9.62 ± 0.30 μg/mL, respectively, which may represent key components of RCEO that inhibit NAAA activity. Meanwhile, docking assays revealed that (E)- cinnamaldehyde occupies the catalytic cavity of NAAA and engages in a hydrogen bond interaction with the TRP181 and hydrophobic-related interactions with LEU152 of human NAAA. Conclusions RCEO showed anti-inflammatory effects by inhibiting NAAA activity and elevating cellular PEA and OEA levels in NAAA-overexpressing HEK293 cells. (E) -cinnamaldehyde and O- methoxycinnamaldehyde, two components in RCEO, were identified as the main contributors of the anti-inflammatory effects of RCEO by modulating cellular PEA levels through NAAA inhibition.

Fractional extraction phenolics from C. oleifera seed kernels exhibited anti-inflammatory effect via PI3K/Akt/NF-κB signaling pathway under Caco-2/RAW264.7 co-culture cell model

Chengyu Jin,Chu Chu,Xianghai Zhu,Yuanchao Lu,Ningxiang Yu,Qin Ye,Yuanxiang Jin,Xianghe Meng

Camellia oleifera Abel ( C. oleifera ) is a multifunctional oilseed, which is rich in many biological active substances with health-promoting properties, especially polyphenols. Previous research revealed that camellia oil phenolics exhibited anti-inflammatory effect, which originated from seed. Thus, we aimed to explore the components of camellia seed phenolics and its potential mechanism of anti-inflammation. Initially, fractional extraction was processed to prepare the phenolics from camellia seed kernels, and we compare four different fractions of phenolics under the LPS-induced Caco-2/RAW264.7 coculturing model. Results showed that free phenolics (FP) had best effect on alleviating pro-inflammatory cytokines (IL-1β, IL-6, IL-8 and TNF-α) compared to esterified-bound phenolics (EP), glycosylated-bound phenolics (GP) and insoluble-bound phenolics (IP). Furthermore, FP reduced inflammation by suppressing the PI3K/Akt/NF-κB signaling pathway and effectively inhibited LPS-induced intestinal permeability increase, tight junction related proteins loss (ZO-1, claudin-1). Same results obtained, as the transepithelial electrical resistance (TEER) and alkaline phosphatase (AKP) activity of high-dose FP treated group was high than model group. Finally, molecular docking was used for evaluating the anti-inflammatory effect for phenolic monomer. KGRG (kaempferol −3-O-(2-O-glucopyranosyl-6-O-rhamnopyranosyl)-glucopyranoside), KXR (kaempferol 3-O-(2′’-xylopyranosyl)-rutinoside) and leucoside (kaempferol 3-O-sambubioside) show lower binding energy docking with NF-κB, PI3K and Akt protein, indicating better interactions, which might be effective constituents against inflammation. Subsequently, five major polyphenols were obtained to validate the docking results, especially, indicating the best anti-inflammatory activities of KGRG. Overall, this research sheds insights on the therapy of phenolics from C. oleifera seed towards LPS-induced intestinal inflammation model in vitro and its related mechanism.

Global immunostimulatory mechanisms of a high-branched glycogen from Glyptocidaris crenularis eggs in RAW264.7 macrophages via BONCAT-based nascent proteomics

Qingchi Wang,Yuhang Luo,Yi Xin,Yuanxiang Jin,Guiling Yang,Quanbin Zhang,Zhengshun Wen

Marine polysaccharides have emerged as promising immunomodulators. However, conventional analytical approaches have failed to elucidate their activation mechanisms from a global perspective. In this study, we isolated a highly branched glycogen (GCEP2a) from eggs of the sea urchin Glyptocidaris crenularis . Structural characterization revealed that GCEP2a possesses a backbone composed of →4)-α-D-Glc p -(1 → residues, with one C6-branch per five residues, and a molecular weight of 8.06 × 10 3  kDa. Employing nascent-proteome bioorthogonal non-canonical amino acid tagging (BONCAT) enrichment followed by DIA-FAIMS, we unraveled the global immunostimulatory mechanisms of GCEP2a in RAW264.7 macrophages. This advanced approach enabled dynamic profiling of nascent protein synthesis, uncovering a multi-receptor and multi-pathway regulatory network. Specifically, GCEP2a activates signaling consistent with engagement of the toll-like receptors (TLRs), C-type lectin receptors (CLRs), and RIG-I-like receptors (RLRs), thereby triggering downstream signaling cascades including the NF-κB, MAPK, and PI3K-AKT pathways. Additionally, GCEP2a downregulates the biosynthesis of lacto- and neolacto-series glycosphingolipids, indicating metabolic reprogramming associated with M1 polarization. These findings not only suggest that GCEP2a exhibits potential as an immune adjuvant, but also provide initial insights into the application of BONCAT-based nascent proteomics for investigating the multi-modal immunostimulatory mechanisms of marine polysaccharides.

Curcumin attenuates oxidative stress in RAW264.7 cells by increasing the activity of antioxidant enzymes and activating the Nrf2-Keap1 pathway

Xinyu Lin,Dingping Bai,Zixi Wei,Ying Zhang,Yifan Huang,Hui Deng,Xiaohong Huang

Large-scale breeding environments often lead to oxidative stress. Macrophages play an important role in the immune system and are vulnerable to reactive oxygen species (ROS), which result in macrophage death. Curcumin is the main active component of turmeric and exerts antioxidant effects. Here, we measured the activity of some antioxidant enzymes and chose the Nrf2-Keap1 signaling pathway to study the protective effects of curcumin on macrophages under oxidative stress in vitro. We used RAW264.7 cells as a research model, and oxidative damage was induced by hydrogen peroxide (H 2 O 2 ). Cell viability was measured by an MTT assay. Flow cytometry was used to measure cellular ROS and apoptosis. The effect of curcumin on Nrf2-Keap1 signaling pathway-related genes was analyzed by qRT-PCR. Furthermore, the translocation of Nrf2 protein was also investigated by Western blot analysis of total and nuclear proteins. All curcumin-treated groups exhibited increased activity of catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX). Low- and middle-dose curcumin decreased malondialdehyde (MDA) and ROS levels, but high-dose curcumin increased MDA and ROS production. We found that low-dose curcumin protected cells from apoptosis, while apoptosis in the middle- and high-dose curcumin-treated groups were stagnant in the early stage. Furthermore, middle-dose curcumin upregulated Nrf2 expression after H 2 O 2 treatment for 4 h. Low- and middle-dose curcumin could activate Nrf2 and promote it to migrate into nuclei. The translocation of Nrf2 to the nucleus to upregulate the expression of haemoxygenase-1 (HO-1) was promoted in the low- and middle-dose curcumin-treated groups. The middle-dose curcumin-treated group also exhibited enhanced expression of glutamate-cysteine ligase, a modifier subunit (GLCM), but inhibited transcription of glutamate-cysteine ligase, a catalytic subunit (GCLC). Curcumin resisted oxidants by increasing the activity of antioxidant enzymes and activating the Nrf2-Keap1 pathway, which could potentially promote cell survival.

Eriocitrin in combination with resveratrol ameliorates LPS-induced inflammation in RAW264.7 cells and relieves TPA-induced mouse ear edema

Junlei Liu,Huarong Huang,Zebin Huang,Yuran Ma,Lanyue Zhang,Yan He,Dongli Li,Wenfeng Liu,Susan Goodin,Kun Zhang,Xi Zheng

Eriocitrin is a flavonoid that is isolated from orange peel. Resveratrol is a polyphenol compound, which is present in various fruits, such as grapes. The aim of the present study was to investigate both in vitro and in vivo anti-inflammatory effects of eriocitrin combined with resveratrol using lipopolysaccharide (LPS)-induced RAW264.7 cells and a mouse model of ear edema. The results showed that eriocitrin combined with resveratrol strongly inhibited LPS-induced secretion of nitric oxide (NO), tumor necrosis factor-α (TNF-α), and Interleukin-1β (IL-1β). Moreover, Eriocitrin combined with resveratrol potently inhibited nuclear factor-κB (NF-κB), phosphor-STAT3, and phosphor-AKT, which was accompanied by inhibition of phosphorylation in mitogen-activated protein kinase (MAPK) signaling pathways . Treatment with eriocitrin combined with resveratrol alleviated edema and subcutaneous tissue inflammation caused by 12-O-tetradecanoylphorbol-13-acetate (TPA) in vivo . This study also demonstrated that treatment with eriocitrin and resveratrol decreased the levels of the pro-inflammatory cytokines TNF-α and IL-1β. The results of the present study indicate that eriocitrin combined with resveratrol effectively inhibits inflammatory responses both in vitro and in vivo .

The Anti-Inflammatory Effect of 6% HES 200/0.5 on RAW264.7 Cells Induced by LPS through HMGB1/NF-κB Signaling Pathway

Jie Zhang,Yongli Wang,Jianzhong Zhang,Shaoyan Huang

We investigated the anti-inflammatory effect of hydroxyethyl starch (HES) 6% 200/0.5 on lipopolysaccharide (LPS)-induced RAW264.7 cells based on the high mobility group protein B1/nuclear transcription factor-κB (HMGB1/NF-κB) signaling pathway. The mouse monocyte macrophage RAW264.7 cells were divided into control group (no additional treatment), LPS group (0.2 pg/ml LPS), LPS+HES group (0.2 pg/ml LPS+6% HES) and LPS+TAK-242 (HMGB1/NF-κB signal pathway blocker) group (0.2 pg/ml LPS The LPS+HES+TAK-242 group (0.2 pg/ml LPS+6% HES+1 μ mol/l TAK-242) was cultured for 48 hours. Griess method and ELISA were used to detect the levels of nitric oxide (NO), tumor necrosis factor α (TNF-α), interleukin (IL)-6 and IL-1β in cell supernatant. The level of intracellular reactive oxygen species (ROS) was detected by flow cytometry. The mitochondrial membrane potential was detected by immunofluorescence. The mRNA expressions of inducible nitric oxide synthase (iNOS), HMGB1, Toll-like receptor 4 (TLR4), nuclear factor-κ b inhibitory protein α (IκBα) and NF-κB were detected by qRT-PCR. The expressions of HMGB1, TLR4, IκBα, phosphorylated IκBα (p-IκBα), NF-κB and phosphorylated NF-κB (p-NF-κB) were detected by western blotting. Compared with the control group, the expression levels of NO, TNF-α, IL-6, IL-1β, ROS, iNOS, HMGB1, TLR4, NF-κB mRNA, HMGB1, TLR4 and p-NF-κB proteins in the LPS group, LPS+HES group, LPS+TAK-242 group and LPS+HES+TAK-242 group increased (P<0.05), the expression levels of mitochondrial membrane potential, IκBα mRNA and p-IκBα protein decreased (P<0.05). Compared with LPS group, the expression levels of NO, TNF-α, IL-6, IL-1β, ROS, iNOS, HMGB1, TLR4, NF-κB mRNA, HMGB1, TLR4 and p-NF-κB proteins in the LPS+HES group, LPS+TAK-242 group and LPS+HES+TAK-242 group increased (P<0.05), the expression levels of mitochondrial membrane potential, IκBα mRNA and p-IκBα protein decreased (P<0.05). The expression levels of mitochondrial membrane potential, IκBα mRNA and p-IκBα protein increased (P<0.05). Six % HES 200/0.5 could down-regulate the expression of molecules related to HMGB1/NF-кB signaling pathway, and reduce the inflammatory response of RAW264.7 cells induced by LPS.

Protection mechanism of Se-containing protein hydrolysates from Se-enriched rice on Pb2+-induced apoptosis in PC12 and RAW264.7 cells

Yong Fang,Zi Xu,Yi Shi,Fei Pei,Wenjian Yang,Ning Ma,Benard Muinde Kimatu,Kunlun Liu,Weifen Qiu,Qiuhui Hu

This study aimed to investigate the protection mechanism of Se-containing protein hydrolysates (SPH) from Se-enriched rice on Pb 2+ -induced apoptosis in PC12 and RAW264.7 cells. Results showed that SPHs could alleviate Pb 2+ -induced morphological changes of apoptosis and the loss of mitochondrial transmembrane potential in both cell types. Besides this, SPHs could significantly reduce the activation of caspase-3, -8, -9 induced by Pb 2+ , reverse the Pb 2+ -induced upregulation of Bax and release of cytochrome C, and downregulate Bcl-2 in cells. HPLC-ICP-MS and SEC-HPLC assays showed that SPHs were low molecular weight peptides (229.4–534.9   Da), and the major Se species found in SPHs was SeMet. Taken together, these findings suggested that SPHs could possibly protect the cells against Pb 2+ -induced apoptosis via a caspase-dependent mitochondrial pathway, and the primary effective constituents in SPHs were SeMet and Se-containing peptides, suggesting that SPHs might be a novel potential candidate to improve the health of people with Se deficiency or in Pb-contaminated areas.

Anti-inflammatory Effects of Astragaloside IV–chitosan Nanoparticles and Associated Anti-inflammatory Mechanisms Based on LC-MS Metabolomic Analysis of RAW264.7 cells

Tingli Qu,Shasha Suo,Liqiu Yan,Liwei Wang,Taigang Liang

Astragaloside IV (ATS) is an active component of Astragalus membranaceus, which has immune regulation and anti-inflammatory effects. However, owing to its large molecular weight and poor solubility in water, the therapeutic effects of ATS are limited. We aimed to prepare astragaloside IV-chitosan (ATS-CS) nanoparticles and determine their anti-inflammatory effect and mechanism of action on RAW264.7 cells using metabolomics. The size of the ATS-CS nanoparticles was 200.3 nm with a zeta potential of 30.5 mV, and the encapsulation rate and drug loading were 69% and 13%, respectively. ATS-CS nanoparticles not only significantly decreased the increase of NO, IL-6 and TNF-α levels induced by LPS, but also exerted an anti-inflammatory effect by acting on arginine and proline, glutathione, sphingolipid, glycerophospholipid, and glycine, serine, and threonine metabolism. Our findings confirmed that ATS-CS nanoparticles have good anti-inflammatory activity and showed that the activity of high molecular weight can be increased by producing nanoparticles. Our study paves the way for exploring the mechanism of nanoparticles through metabolomics.
