Maltol attenuates polystyrene nanoplastic-induced enterotoxicity by promoting AMPK/mTOR/TFEB-mediated autophagy and modulating gut microbiota

Ming-hui Jin,Jun-nan Hu,Ming Zhang,Zhaojie Meng,Guo-ping Shi,Zi Wang,Wei Li

The production and application of nanoplastics has been increased during decades, and the enterotoxicity caused by their bioaccumulation has attracted vast attention. Maltol was proved to exert a protective effect on gut damage induced by carbon tetrachloride and cisplatin, indicating its confrontation with nanoplastics-induced intestinal toxicity. To explore the ameliorative effects of maltol on polystyrene nanoplastics (PS)-mediated enterotoxicity and the underlying mechanism, the mice were exposed to PS (100 mg/kg), combining with or without the treatment of maltol treatment at 50 and 100 mg/kg. We found PS exposure caused intestinal barrier damage and enterocyte apoptosis, while lysosomal dysfunction and autophagic substrate degradation arrest in enterocytes of mice were also observed. In addition, PS exacerbated the disturbance of the intestinal microbial community , affected the abundance of lysosome and apoptosis-related bacterial genes, and decreased the number of known short-chain fatty acid (SCFA) producing bacteria. However, those alterations were improved by the maltol treatment. Maltol also protected the human intestinal Caco-2 cells from PS-induce damages. Mechanistic studies showed maltol promoted TFEB nuclear translocation through the AMPK/mTOR signaling pathway to restore lysosomal function and reduce autophagy dependent apoptosis. The findings in the present work might help to elucidate the potential molecular mechanisms of PS-induced enterotoxicity. For the first time to our knowledge, the protective effect of maltol on PS-induced intestinal injury was studied from multiple perspectives, which provided a potential therapeutic approach for diseases caused by environmental pollution .

A starch-like polysaccharide isolated from Hirsutella sinensis mycelia alleviates DSS-induced colitis in mice by strengthening the intestinal barrier, maintaining immune homeostasis, and modulating the gut microbiota and SCFA metabolism

Yu Li,Hailiang Wang,Hongtao Bi,Jingyue Feng,Tingting Gao,Dai Dong,Guoqiang Li,Boxuan Liu,Hongyan Yuan,Weihua Ni

Hirsutella sinensis ( H. sinensis ), a substitute for a valuable edible and medicinal fungus, Cordyceps sinensis , is well known for its various of beneficial pharmacological properties. However, research on the effects of H. sinensis on colitis in mice is limited. In our previous studies, we carried out a systematic classification and structural identification of the polysaccharides in H. sinensis . Here, we utilized a murine model of colitis induced by a 3% dextran sulfate sodium (DSS) administration to investigate the protective effects of the H. sinensis mycelium-derived starch-like polysaccharide (HSWP-1a) on colitis. Our findings demonstrate that HSWP-1a effectively mitigates DSS-induced weight loss, reduces colon tissue damage, increases tight junction protein expression, and restores intestinal barrier function. Notably, during this process, HSWP-1a reversed the DSS-induced increase in M1/M2 macrophage polarization and the Th17/Treg cell balance, ameliorating the DSS-induced decrease in anti-inflammatory cytokine expression while inhibiting the DSS-induced increase in proinflammatory cytokine levels and mediating immune tolerance to maintain local intestinal immune homeostasis. Furthermore, HSWP-1a is capable of regulating the intestinal microbiota, enhancing the abundance of beneficial bacteria such as Lachnospiraceae_NK4A136_group , and reducing the abundance of harmful bacteria like Proteobacteria and Cyanobacteria. HSWP-1a also restores the levels of short-chain fatty acids (SCFA) to maintain the balanced intestinal microenvironment. Overall, HSWP-1a achieves its colitis-protective effects through its ability to strengthen the intestinal barrier, regulate the immune system, and restore gut microbiota interactions. These findings suggest that HSWP-1a has potential as a valuable food supplement or nutritional health product for alleviating colitis.

Short-chain fatty acid (SCFA) and medium-chain fatty acid (MCFA) concentrations in human milk consumed by infants born at different gestational ages and the variations in concentration during lactation stages

Xinyue Dai,Tinglan Yuan,Xinghe Zhang,Qin Zhou,Huiya Bi,Renqiang Yu,Wei Wei,Xingguo Wang

This study aimed to quantify the short-chain fatty acids (SCFAs) and medium-chain fatty acids (MCFAs) in human milk triacylglycerols (TAGs) and investigate their concentrations in human milk consumed during lactation by infants born at different gestational ages. One hundred and eighty milk samples from the mothers of 30 full-term, 10 early-preterm (≤32 weeks), 10 mild-preterm (32–34 weeks), and 10 near-term (34–37 weeks) infants were collected from the colostrum, transitional, and mature milk. The human milk TAGs were transferred into fatty-acid methyl esters via potassium methoxide in methanol and determined using gas chromatography (GC). The total SCFA (4:0) and MCFA concentrations (6:0 and 8:0) were highest in the mature milk (1.47 ± 0.66 mg g−1 fat from full-term infant milk), approximately 42.18% higher than those in transitional milk. Significantly higher SCFA and MCFA concentrations were found in full-term milk than in preterm milk (p = 0.001). The milk TAGs were analyzed using ultra-high-performance supercritical fluid chromatography with quadrupole-time-of-flight mass spectrometry (UHPSFC-Q-TOF-MS), which showed that the SCFAs and MCFAs were mainly esterified with long-chain fatty-acid groups (16:0, 18:1 n-9, and 18:2 n-6) at the glycerol backbone. The infants’ daily SCFA intake from human milk was estimated; this was highest from mature milk for full-term infants (∼14 mg d−1) which was significantly different from that of preterm infants from colostrum and transitional milk (p < 0.001). The correlation between dietary SCFAs and MCFAs in human milk and nutrition in newborns, especially in the gut microbiotas of preterm infants, requires further study.

Polysaccharides from Fu brick tea ameliorate obesity by modulating gut microbiota and gut microbiota-related short chain fatty acid and amino acid metabolism

Mingzhi Zhu,Jian Ouyang,Fang Zhou,Chenjie Zhao,Wan Zhu,Chunfang Liu,Peifang Huang,Jiafeng Li,Junwei Tang,Zhang Zhang,Jianan Huang,Miaomiao Wu,Kunbo Wang,Zhonghua Liu

Fu brick tea (FBT) is a traditional tea manufactured by solid-state fermentation of tea leaves ( Camellia sinensis ). Although anti-obesity effects have been reported for FBT, the associated role of FBT polysaccharides (PSs) and the underlying mechanisms remain unknown. In this study, we found that FBTPSs inhibited obesity, hyperlipidemia , and inflammation; improved intestinal barrier function; and alleviated gut microbiota dysbiosis in high-fat diet-fed rats. Akkermansia muciniphila , Bacteroides , Parasutterella, Desulfovibrio , and Blautia were the core microbes regulated by FBTPSs. FBTPSs regulated the production of gut microbiota-related metabolites, including short-chain fatty acids (SCFAs), branched-chain amino acids , and aromatic amino acids throughout the development of obesity, and regulated the SCFA-GPR signaling pathway . FBTPS-treated fecal microbiota transplant ameliorated obesity, alleviated gut microbiota dysbiosis, and improved gut microbiota-associated metabolites, suggesting that the anti-obesity effect of FBTPSs was gut microbiota-dependent. FBTPSs may serve as novel prebiotic agents for the treatment of obesity and dysbiosis of gut microbiota.

Maternal resveratrol improves the intestinal health and weight gain of suckling piglets under high summer temperatures: The involvement of exosome-derived microRNAs and immunoglobin in colostrum

Changming Hong,Yujian Huang,Guan Yang,Xiaolu Wen,Li Wang,Xuefen Yang,Kaiguo Gao,Zongyong Jiang,Hao Xiao

Previous studies have shown that maternal resveratrol improved growth performance and altered the microbial composition of suckling piglets under hot summer conditions. However, it remains unclear how maternal resveratrol improves growth performance of suckling piglets under high summer temperatures. A total of 20 sows (landrace × large white; three parity) were randomly assigned to 2 groups (with or without 300 mg/kg resveratrol) from d 75 of gestation to d 21 of lactation in a high temperature environment (from 27 to 30 °C). The results showed that maternal resveratrol supplementation increased total daily weight gain of piglets under hot summer conditions, which is consistent with previous studies. Furthermore, we found that maternal resveratrol improved the intestinal morphology and intestinal epithelial proliferation in suckling piglets. Dietary resveratrol supplementation affected the characteristics of exosome-derived microRNAs (miRNAs) in sow colostrum, as well as the genes targeted by differentially produced miRNAs. MiRNAs are concentrated in the tight junction pathway. As a result, the expression of intestinal tight junction proteins was increased in suckling piglets ( P < 0.05). Notably, maternal resveratrol increased the intestinal secretory immunoglobulin A (sIgA) levels of suckling piglets via colostrum immunoglobins ( P < 0.05), which could increase the abundance of beneficial microbiota to further increase the concentration of short chain fatty acids (SCFA) in suckling piglets' intestine ( P < 0.05). Finally, our correlation analysis further demonstrated the positive associations between significantly differential intestinal microbiota, intestinal sIgA production and SCFA concentrations, as well as the positive relation between total daily weight gain and intestinal health of suckling piglets. Taken together, our findings suggested that maternal resveratrol could promote intestinal health to improve piglet growth under high summer temperatures, which might be associated with the immunoglobin and exosome-derived miRNAs in sows’ colostrum.

Integrated metabolome and microbiome strategy reveals the therapeutic effect of nervonic acid on Alzheimer's disease rats

Ziyi Chen,Shu Liu,Fengrui Song,Zong Hou,Hui Zhou,Yuting Fan,Rongjin Wang,Zhongying Liu

Alzheimer's disease (AD) is a complex neurodegenerative disease. Nervonic acid is a component of breast milk and is also found in fish oil and specific vegetable oils. Studies have shown that nervonic acid is essential for the development of the human nervous system. In this study, Morris water maze (MWM) test and pathological analysis showed that nervonic acid could improve cognitive deficits and brain nerve damage in AD rats. Then, through sequencing, we found that nervonic acid increased the abundance of beneficial bacteria such as Lactobacillus and Bacteroides , and decreased the abundance of Pseudomonadaceae_Pseudomonas . Not only that, nervonic acid also regulates the production of short-chain fatty acids (SCFA) and the levels of 29 fecal metabolites, and affects the metabolism of linoleic acid, α-linolenic acid, arachidonic acid, and sphingolipid. Finally, we verified the regulatory effect of nervonic acid on metabolic enzyme activity.

Bile acids supplementation modulates lipid metabolism, intestinal function, and cecal microbiota in geese

Li Guangquan,Wang Xianze,Liu Yi,Gong Shaoming,Yang Yunzhou,Wang Cui,Wang Huiying,He Daqian

Bile acids(BAs) are important components of bile and play a significant role in fat metabolism. However, there is currently no systematic evaluation of the use of BAs as feed additives for geese. This study aimed to investigate the effects of adding BAs to goose feed on growth performance, lipid metabolism, intestinal morphology, mucosal barrier function, and cecal microbiota. A total of 168 28-day-old geese were randomly assigned to four treatment groups and fed diets supplemented with 0, 75, 150, or 300 mg/kg of BAs for 28 days. The addition of 75 and 150 mg/kg of BAs significantly improved the feed/gain (F/G) (p  < 0.05).The addition of BAs decreased abdominal fat percentage and serum total cholesterol (TC) levels, with 150 mg/kg of BAs significantly reducing serum triglyceride levels and increased expression of Farnesoid X Receptor (FXR) mRNA in the liver(p  < 0.05), 300 mg/kg of BAs significantly increasing the expression level of liver peroxisome proliferator-activated receptor α (PPARα) (p < 0.05). In terms of intestinal morphology and mucosal barrier function, 150 mg/kg of BAs significantly increased villus height (VH) and VH/crypt depth (CD) in the jejunum (p < 0.05). The addition of 150 and 300 mg/kg of BAs significantly reduced the CD in the ileum, while increasing VH and VH/CD (p<0.05). Additionally, the addition of 150 and 300 mg/kg of BAs significantly increased the expression levels of zonula occludens-1 (ZO-1) and occludin in the jejunum. Simultaneously 150mg/kg and 300mg/kg BAs increased the total short-chain fatty acids (SCFA) concentrations in the jejunum and cecum(p < 0.05).Supplementation with BAs resulted in a significant increase in the ɑ-diversity of cecal microbiota and a decrease in the abundance of Proteobacteria in the cecum. The addition of 150 mg/kg of BAs significantly reduced the abundance of Bacteroidetes and increased the abundance of Firmicutes. Moreover,Linear discriminant analysis Effect Size analysis (LEfSe) showed that the abundances of bacteria producing SCFA and bile salt hydrolases (BSH) were increased in the BAs-treated group. Furthermore, Spearman’s analysis showed that the genus Balutia, which is negatively correlated with visceral fat area, was positively correlated with serum high-density lipoprotein cholesterol (HDL-C), while Clostridium was positively correlated with intestinal VH and VH/CD. In conclusion, BAs can be considered an effective feed additive for geese, as they increased SCFA concentration, improve lipid metabolism and intestinal health by enhancing the intestinal mucosal barrier, improving intestinal morphology, and altering the cecal microbiota structure.
