In porous media, non-wetting phase droplets snapping off in a constricted microchannel are one of the most common phenomena in two-phase flow processes. In this paper, the application range of the classic quasi-static criterion in rectangular cross section microchannels is obtained. For three different droplet breakup phenomena—total breakup, partial breakup, and non-breakup—observed in experiments when a non-wetting phase droplet passes through a microchannel constriction, the breakup is caused by the droplet neck snapping off in a channel constriction. A critical criterion for the dynamic snap-off event in a two-phase flow is proposed considering the effect of viscous dissipation by mechanical analysis, energy dissipation analysis, and many microfluidic experiments. When the droplet front flows out of the constriction, snap-off will occur if the surface energy release exceeds the required energy for viscous dissipation and kinetic energy conversion. The unique partial breakup phenomenon is affected by droplet surfactant distribution and the acceleration effect in the constriction center. This partial breakup phenomenon in experiments is an essential evidence for the non-uniform distribution of surfactants in the droplet surface. The results of this study contribute to understanding pore-scale mass transfer and flow pattern changes within porous media.

Background Chronic brain hypoperfusion (CBH) is closely related to Alzheimer’s disease (AD) and vascular dementia (VaD). Meanwhile, synaptic pathology plays a prominent role in the initial stage of AD and VaD. However, whether and how CBH impairs presynaptic plasticity is currently unclear. Methods In the present study, we performed a battery of techniques, including primary neuronal culture, patch clamp, stereotaxic injection of the lentiviral vectors, morris water maze (MWM), dual luciferase reporter assay, FM1–43 fluorescence dye evaluation, qRT-PCR and western blot, to investigate the regulatory effect of miR-153 on hippocampal synaptic vesicle release both in vivo and in vitro. The CBH rat model was generated by bilateral common carotid artery ligation (2VO).Results Compared to sham rats, 2VO rats presented decreased field excitatory postsynaptic potential (fEPSP) amplitude and increased paired-pulse ratios (PPRs) in the CA3-CA1 pathway, as well as significantly decreased expression of multiple vesicle fusion-related proteins, including SNAP-25, VAMP-2, syntaxin-1A and synaptotagmin-1, in the hippocampi. The levels of microRNA-153 (miR-153) were upregulated in the hippocampi of rats following 2VO surgery, and in the plasma of dementia patients. The expression of the vesicle fusion-related proteins affected by 2VO was inhibited by miR-153, elevated by miR-153 inhibition, and unchanged by binding-site mutation or miR masks. FM1–43 fluorescence images showed that miR-153 blunted vesicle exocytosis, but this effect was prevented by either 2′-O-methyl antisense oligoribonucleotides to miR-153 (AMO-153) and miR-masking of the miR-153 binding site in the 3′ untranslated region (3’UTR) of the Snap25, Vamp2, Stx1a and Syt1 genes. Overexpression of miR-153 by lentiviral vector-mediated miR-153 mimics (lenti-pre-miR-153) decreased the fEPSP amplitude and elevated the PPR in the rat hippocampus, whereas overexpression of the antisense molecule (lenti-AMO-153) reversed these changes triggered by 2VO. Furthermore, lenti-AMO-153 attenuated the cognitive decline of 2VO rats. Conclusion sOverexpression of miR-153 controls CBH-induced presynaptic vesicle release impairment by posttranscriptionally regulating the expression of four vesicle release-related proteins by targeting the 3’UTRs of the Stx1a, Snap25, Vamp2 and Syt1 genes. These findings identify a novel mechanism of presynaptic plasticity impairment during CBH, which may be a new drug target for prevention or treatment of AD and VaD.

Snap beans ( Phaseolus vulgaris L.) have high nutritional value but are susceptible to senescence and spoilage after harvest. Previously, a carboxymethyl cellulose/starch-based photodynamic film (CCPF) was shown to delay the senescence process and maintain the storage quality of snap bean pods (SBPs). However, the mechanism of the CCPF effect on the preservation of snap beans remains unclear. This study first confirmed the beneficial effects of CCPF packaging on the appearance and nutritional quality of SBPs and microbial growth inhibition. Then, the regulatory mechanism by which CCPF packaging impacts the physiological metabolism of postharvest SBPs was investigated via widely targeted metabolomics. The results revealed 1802 metabolites in total SBP samples, among which 464 metabolites were significantly different between SBPs packaged with the CCPF and those packaged with a pure carboxymethyl cellulose/starch-based film (CCF) in the dark. It was found that CCPF packaging could delay the deterioration of the quality of SBPs by regulating the synthesis and decomposition of phenolic acids, flavonoids, lipids, amino acids and derivatives in postharvest SBPs. Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that compared with CCF packaging, CCPF packaging delayed the postharvest senescence of SBPs by affecting three potential metabolic pathways (isoflavone biosynthesis, flavonoid biosynthesis and nicotinate and nicotinamide metabolism), maintaining the storage quality of snap beans. This study revealed the possible mechanism by which CCPF packaging maintains the postharvest storage quality of snap beans.

Established a universal screening platform for CD147-targeted therapeutics. • Integrated the CMC-HPLC system with mass spectrometry (MS) for the identification of unknown compounds. • Two bioactive constituents from Magnolia officinalis, directly interact with CD147. Hepatocellular carcinoma (HCC) is one of the most prevalent digestive malignancies, with CD147 emerging as a promising therapeutic target for HCC. To date, no natural bioactive components targeting CD147 for anti-HCC therapy have been explicitly reported. Given the low toxicity and favorable efficacy of natural small-molecule drugs, identifying and investigating CD147-targeted natural agents is imperative. In this study, based on cell membrane chromatography (CMC) and SNAP-tag protein labeling technology, we constructed a CD147-SNAP-tag-CMC/HPLC-MS two-dimensional coupled system to screen CD147-binding components from Magnolia officinalis , followed by ligand-receptor affinity verification, in vitro pharmacological evaluation, and downstream molecular mechanism validation. Two bioactive compounds, honokiol and magnolol, were successfully identified: both exhibit strong CD147 binding affinity, effectively suppress HCC cell migration, and exert their functions by regulating the CD147/integrin-FAK-paxillin pathway as confirmed by Western blotting. Collectively, the established two-dimensional system serves as a universal platform for CD147-targeted component screening. This study is the first to reveal the strong CD147 affinity of honokiol and magnolol and their anti-migratory effect on HCC cells via modulating the aforementioned pathway.

Exosomes play crucial roles in local and distant cellular communication and are involved in various physiological and pathological processes. Tumour-derived exosomes are pivotal to tumorigenesis, but the precise mechanisms underlying their secretion remain elusive. In particular, the SNARE proteins that mediate the fusion of multivesicular bodies (MVBs) with the plasma membrane (PM) in tumour cells are subject to debate. In this study, we identified syntaxin-4, SNAP-23, and VAMP-7 as the SNAREs responsible for exosome secretion in MCF-7 breast cancer cells and found that a SNARE complex consisting of these SNAREs can drive membrane fusion in vitro. Deletion of any of these SNAREs in MCF-7 cells did not affect MVB biogenesis and transportation, indicating their specific involvement in MVB–PM fusion. In addition, syntaxin-4, SNAP-23, and VAMP-7 play equivalent roles in exosome secretion in both HeLa cervical cancer cells and A375 melanoma cells, suggesting their conserved function in exosome secretion. Furthermore, deletion of VAMP-7 in 4T1 mammary carcinoma cells efficiently inhibited exosome secretion and led to significant attenuation of tumour growth and lung metastasis in mouse models, implying that VAMP-7 may hold promise as a novel therapeutic target for breast cancer.

The characterization of protein complex is vital for unraveling biological mechanisms in various life processes. Despite advancements in biophysical tools, the capture of non-covalent complexes and deciphering of their biochemical composition continue to present challenges for low-input samples. Here we introduce SNAP-MS, a Stationary-phase-dissolvable Native Affinity Purification and Mass Spectrometric characterization strategy. It allows for highly efficient purification and characterization from inputs at the pico-mole level. SNAP-MS replaces traditional elution with matrix dissolving during the recovery of captured targets, enabling the use of high-affinity bait-target pairs and eliminates interstitial voids. The purified intact protein complexes are compatible with native MS, which provides structural information including stoichiometry, topology, and distribution of proteoforms, size variants and interaction states. An algorithm utilizes the bait as a charge remover and mass corrector significantly enhances the accuracy of analyzing heterogeneously glycosylated complexes. With a sample-to-data time as brief as 2 hours, SNAP-MS demonstrates considerable versatility in characterizing native complexes from biological samples, including blood samples.

Cancer recurrence and metastasis are two major challenges in the current clinical therapy. Here, we synthesized a novel diketopyrrolopyrrole-based photothermal reagent (DCN) with unique J-aggregation-induced redshift to achieve efficient tumor thermal ablation under safe power (0.33 W/cm 2 ). Meanwhile, S-nitroso-N-acetylpenicillamine (SNAP) was co-loaded with near-infrared (NIR)-absorbing DCN in amphiphilic polymers to realize heat-induced massive release of nitric oxide (NO), which could form oxidant peroxynitrite (ONOO – ) to active MMPs, thereby degrading the compact tumor extracellular matrix (ECM) to improve the ablation depth and infiltration of immune cells. Through a facile supramolecular assembly method, the DCN/SNAP nanoparticles were anchored to liquid nitrogen-frozen cancer cells (LNF), achieving enhanced anti-tumor immune responses and effective inhibition of distant tumor and pulmonary metastases after only one treatment. The safety and effectiveness of this supramolecular cell-conjugation platform were verified by 2D/3D cellular experiments and bilateral tumor model, confirming the thermal ablation-gas permeation-antigen presentation therapeutic mode had promising anticancer prospects.
