Sargassum fusiforme fucoidan modifies gut microbiota and intestinal metabolites during alleviation of hyperglycemia in type 2 diabetic mice

Qifang Wu,Siya Wu,Yang Cheng,Zhongshan Zhang,Genxiang Mao,Shijun Li,Yue Yang,Xu Zhang,Mingjiang Wu,Haibin Tong

Type 2 diabetic mellitus (T2DM) is a complicated metabolic disorder that is now considered as a major global public health problem. Fucoidan exhibits diverse biological activities, especially prevention of metabolic diseases. In this regard, we herein aimed to reveal the beneficial effect of Sargassum fusiforme fucoidan (SFF) on high-fat diet (HFD) and streptozotocin (STZ) induced T2DM mice. We noted that on the one hand, SFF significantly decreased fasting blood glucose, diet and water intake, and hyperlipidemia, while on the other hand, it improved glucose tolerance. Furthermore, SFF reduced epididymal fat deposition, attenuated the pathological changes in heart and liver tissues, and decreased oxidative stress in diabetic mice. To explore the underlying mechanisms of these ameliorative effects, the gut microbiota was analyzed. Notably, SFF highly enriched benign microbes including Bacteroides, Faecalibacterium and Blautia, as well as increased levels of (R)-carnitine and choline in the colon of diabetic mice. This may be a potential mechanism for alleviating T2DM, thus implying the benefits of SFF as an adjuvant agent for T2DM treatment. Taken together, this study demonstrated a promising application of fucoidan as one of the adjuvant agents for the management of T2DM in the future.

The effect of subcritical water treatment on the physicochemical properties and α-glucosidase inhibitory activity of Sargassum fusiforme polysaccharides

Mengqing Zhang,Zhenna Zhang,Lichun Guo,Wei Zhao

Summary In order to make full use of Sargassum fusiforme (S. fusiforme) , subcritical water treatment, an environmentally friendly method, was used to degrade polysaccharides to enhance their functional activity. Results revealed that average molecular weight decreased with increasing subcritical water treatment time or temperature. The depolymerisation kinetic was found to follow a first-order kinetics model fitted to 1/ M t – 1/ M 0 = k • t . Structural characterisation exhibited that subcritical water treatment changed the molar ratio composition of monosaccharides, while the main functional groups of S. fusiforme polysaccharides did not change significantly. Rheological characterisation demonstrated that the three degraded polysaccharides displayed shear-thinning behaviour and viscoelasticity. Except for natural polysaccharides, the α-glucosidase inhibitory activity of the three degraded polysaccharides was superior to that of acarbose. Consequently, this study can provide scientific evidence for subcritical water treatment as a green and efficient method to prepare bioactive polysaccharides with low molecular weight.

In vitro digestibility of Sargassum fusiforme polysaccharide fermented by Lactobacillus acidophilus and its improvement effect on intestinal inflammation in zebrafish

Laijin Su,Ying Yang,Yangli Xu,Lianlian Zhu,Bolun Sun,Jiali Zhou

This study investigated the in vitro digestion characteristics of Sargassum fusiforme polysaccharide fermented using Lactobacillus acidophilus (LA-rSFP) and its effects on dextran sulfate sodium-induced intestinal inflammation in zebrafish. The treatment substantially decreased the molecular weight of LA-rSFP and increased the reducing sugar content during gastrointestinal digestion. LA-rSFP considerably enhanced the activities of superoxide dismutase and catalase and markedly decreased malondialdehyde content, indicating a reduction in the oxidative stress in the intestinal environment. Moreover, LA-rSFP ameliorated colitis-induced intestinal injury and up-regulated the expression of tight junction proteins (occludin, ZO-1, and claudin-1), suggesting strengthened intestinal barrier integrity. LA-rSFP also played an important role in regulating pro-inflammatory cytokines (down-regulated) and anti-inflammatory cytokines (up-regulated) during inflammatory responses in the zebrafish gut. The results of 16S rRNA sequencing showed that LA-rSFP increased the diversity and richness of the intestinal microbiota in zebrafish with inflammatory bowel disease (IBD), enhancing the abundance of beneficial bacteria such as Fusobacteria, Firmicutes, Bacteroidetes, and Lactobacillus, and reducing Proteobacteria levels. These findings suggest that the administration of LA-rSFP alleviates IBD, with the therapeutic mechanisms partially attributed to the regulation of the intestinal microbiota composition and abundance.

Green synthesis of ZnO nanoparticles using enzymatically hydrolyzed Sargassum fusiforme extracts: Characterization and enhanced photocatalytic performance under UV and solar irradiation

Jin Yang,Pengcheng Xue,Lu Zhao,Yajuan Luo,Lianyi Li,Guiying Jiang,Beibei Wang,Shiliang Liu

This study presents an eco-friendly approach for the green synthesis of ZnO nanoparticles (ZnO NPs) using enzymatically hydrolyzed Sargassum fusiforme extracts (SFEE). A comprehensive suite of analytical techniques, including UV-visible absorption spectroscopy, UV-visible diffuse reflectance spectroscopy (UV-Vis DRS), photoluminescence (PL) spectroscopy, Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), field-emission scanning electron microscopy (FESEM), high-resolution transmission electron microscopy (HRTEM), and X-ray photoelectron spectroscopy (XPS), were employed to systematically characterize the structural, morphological, surface chemical, and optical properties of the synthesized ZnO NPs. UV-visible absorption spectroscopy confirmed the formation of nanoparticles through a distinct absorption peak at 375 nm. FTIR and XPS analyses revealed that functional groups derived from SFEE participate in nanoparticle formation and surface stabilization. XRD, FESEM, and HRTEM results demonstrated that SFEE-ZnO nanoparticles possess a well-defined hexagonal wurtzite structure with uniform dispersion, a reduced average particle size of 75.23 nm, and a smaller crystallite size (21.22 nm) compared with chemically synthesized ZnO (C-ZnO) and ZnO prepared using hot aqueous extracts (SFAE-ZnO). UV-Vis DRS analysis revealed a red shift in the absorption edge of SFEE-ZnO, corresponding to a narrowed bandgap of 3.20 eV, while PL analysis indicated suppressed electron-hole recombination. These synergistic structural and electronic features contribute to enhanced photocatalytic activity. The photocatalytic performance of SFEE-ZnO was evaluated by degrading methylene blue (MB, 15 mg L -1 ) using 1 mg mL -1 of catalyst under both UV and solar irradiation. SFEE-ZnO exhibited superior degradation efficiencies of 97.65% under UV light and 99.48% under solar light within 30 min, significantly outperforming C-ZnO and SFAE-ZnO. These results highlight the potential of SFEE-ZnO as an efficient and environmentally sustainable photocatalyst for solar-driven wastewater remediation.

Selenium nanoparticles decorated with polysaccharides from Sargassum fusiforme protects against 6-OHDA-induced neurotoxicity in PC12 cells and rat model of Parkinson's disease

Hongying Zhao,Jiaxin Song,Tian Wang,Xiaodan Fan

Parkinson's disease (PD) is a neurodegenerative disorder and identifying disease-causing pathways and drugs that target them has remained challenging. Herein, selenium nanoparticles decorated with polysaccharides from Sargassum fusiforme (SFPS-SeNPs) were investigated on 6-OHDA-induced neurotoxicity in PC12 cells and rats. 6-OHDA can significantly increase neurotoxicity, oxidative stress and decrease the activity of superoxide dismutase (SOD) and glutathione peroxidase (GPx) both in vitro and vivo. In vitro, treatment with SFPS-SeNPs can significantly decrease 6-OHDA cytotoxicity, reactive oxygen species (ROS) production or malondialdehyde (MDA) levels, and cell apoptosis, significantly increased the activity of SOD and GPx. In vivo, 6-OHDA exposure could also decrease the expression of Nrf2 and OH-1, while treatment with SFPS-SeNPs (1 mg Se/kg) increased. SFPS-SeNPs can protect neurons from 6-OHDA-induced neurotoxicity by regulating apoptosis and Nrf2/ARE pathway. The present study demonstrated that SFPS-SeNPs is a good candidate for developing a new drug against neurodegenerative diseases such as PD.
