Simple SummaryThe widespread cultivation of genetically engineered crops producing not only toxic proteins from the bacteriumBacillus thringiensis(Bt) but also plant defensive compounds known as allelochemicals, combined with occasional use of insecticides, is the major tactic to manage some economically important pests. Better understanding of the toxicological interactions of the three types of toxins is needed to rationally deploy them to protect crops from pests. The aim of this study is to examine the sequential and simultaneous interactions of the allelochemical flavone, Bt toxin Vip3A, and insecticide emamectin benzoate in the fall armyworm (Spodoptera frugiperda), a worldwide target pest of Bt crops. Bioassays ofS. frugiperdaneonates revealed that all interactions of the three toxins, except for 1-day pre-exposure to a sublethal dose (LC5) of flavone followed by 6-day simultaneous exposure to flavone LC5+ emamectin benzoate LC50, are synergistic or additive. The results suggest that the combined use of the three toxins is basically a great strategy to manageS. frugiperda.AbstractTarget pests of genetically engineered crops producing both defensive allelochemicals andBacillus thuringiensis(Bt) toxins often sequentially or simultaneously uptake allelochemicals, Bt toxins, and/or insecticides. How the three types of toxins interact to kill pests remains underexplored. Here we investigated the interactions of Bt toxin Vip3A, plant allelochemical flavone, and insecticide emamectin benzoate inSpodoptera frugiperda. Simultaneous administration of flavone LC25+ Vip3A LC25, emamectin benzoate LC25+ Vip3A LC25, and flavone LC15+ emamectin benzoate LC15+ Vip3A LC15but not flavone LC25+ emamectin LC25yielded a mortality significantly higher than their expected additive mortality (EAM). One-day pre-exposure to one toxin at LC5followed by six-day exposure to the same toxin at LC5plus another toxin at LC50showed that the mortality of flavone LC5+ Vip3A LC50, emamectin benzoate LC5+ Vip3A LC50, and Vip3A LC5+ emamectin benzoate LC50were significantly higher than their EAM, while that of flavone LC5+ emamectin benzoate LC50was significantly lower than their EAM. No significant difference existed among the mortalities of Vip3A LC5+ flavone LC50, emamectin benzoate LC5+ flavone LC50, and their EAMs. The results suggest that the interactions of the three toxins are largely synergistic (inductive) or additive, depending on their combinations and doses. Keywords:allelochemical;Bt toxin;chemical insecticide;combined toxicity;induced toxicity;synergistic interactions

The fall armyworm,Spodoptera frugiperda(Lepidoptera: Noctuidae) (FAW), is an invasive and destructive polyphagous pest that poses a significant threat to global agricultural production. The FAW mainly damages maize, with a particular preference for V3–V5 (third to fifth leaf collar) plant stages in northern China. How the FAW moth precisely locates maize plants in the V3–V5 stage at night remains unclear. The aims of this study were to evaluate the visual and olfactory cues used by the FAW to identify its host plant, maize, in order to select attractants with better trapping efficacy. Hyperspectral analysis of maize plants at different growth stages using the ASD Fieldspec 4 spectrometer was performed using mimics (moths or maize leaves sealed with transparent plastic sheets) and black cloth-covered plants for single visual and single olfactory attraction experiments. Gas chromatography–mass spectrometry (GC-MS) was used to analyze volatiles emitted from V3–V5 stage maize leaves. Volatile organic chemicals (VOCs) were screened using electroantennography (EAG) and Y-tube. Attractor efficacy was validated using mimics + VOCs. Results showed very little variance in the spectral reflectance curve of the maize at different growth stages. Fifteen VOCs were identified in the V3–V5 stage leaves of three different maize varieties, of whichcis-3-hexenyl acetate and myrcene were found in relatively high concentrations in these maize varieties. The frequency of visits attracted by single visual stimuli was significantly lower than that attracted by single olfactory or olfactory + visual cues. The attractiveness of foliarcis-3-hexenyl acetate increased as its concentration decreased. The combination of mimics +cis-3-hexenyl acetate (1 ng/μL) increased host detection efficiency and stimulated mating behavior. These results indicate that the nocturnal insect FAW primarily uses olfactory cues for host identification, with visual cues serving as a complementary modality. The synergistic effect of olfactory and visual cues increases the efficiency of host recognition. We found thatcis-3-hexenol acetate at a concentration from maize leaves is a reliable olfactory signal for the FAW. When using host plant VOCs as attractants to control adult FAWs, the role of visual cues must be considered.

BACKGROUND The fall armyworm (FAW), Spodoptera frugiperda (J. E. Smith), is an invasive pest. Maize is one of its preferred host plants. The traditional application of granules has been effective for the control of FAW. Nevertheless, the challenge is to develop an application method that is less labor-intensive and also efficient. RESULTS Tiny granules in the size range of 0.38–0.55 mm were prepared by the centrifugal granulation method for drone application. The in-swath distribution patterns of granules deposited on sticky tapes were similar to the distribution of aqueous droplets through drone spraying. In the range of 1.5–3.5 m, a relatively higher aircraft height of flight can provide a more uniform distribution pattern of deposited granules, but resulted in less deposit of tiny granules in the whorl of maize plants. Similarly, the deposit of granules in the whorl of maize plants significantly decreased when the flight speed increased from 4 to 6 m s −1 . Granules gathered in the whorl accounted for 50.8–58.7% of total granules deposited in the maize canopy. The field experiment demonstrates granular formulations (containing 0.25% chlorantraniliprole + 0.15% emamectin benzoate, or higher concentrations) can provide better and longer duration of FAW control than the aqueous spray formulation at the whorl stage of maize plants. CONCLUSION Broadcasting of tiny granules by drone can provide better FAW control than liquid spray. The architecture of the maize plant and the downward airstream of the drone create favorable conditions for the deposition of granules in whorls. © 2021 Society of Chemical Industry.

The fall armyworm, Spodoptera frugiperda, has become a worldwide pest and threatens world food production. A previous study indicated that azadirachtin, the most effective botanical insecticide for S. frugiperda , inhibits larval growth of the insect. The effect of azadirachtin on the tissues of the larvae, however, remains to be determined. In this study, the effects of azadirachtin on the structure of fat bodies were analyzed. Comparative transcriptomic analysis was conducted between controls and samples treated with 0.1 μg/g azadirachtin for 7 days to explore potential relevant mechanisms. The expression of 5356 genes was significantly affected after azadirachtin treatment, with 3020 up-regulated and 2336 down-regulated. Among them, 137 encode detoxification enzymes, including 53 P450s , 20 GSTs , 27 CarEs , 16 UGTs , and 12 ABC transporters . Our results indicated that azadirachtin could destroy fat body structure and change the mRNA levels of detoxification-related genes. The up-regulated genes encoding detoxification enzymes might be related to detoxifying azadirachtin. Our results elucidate a preliminary mechanism of azadirachtin detoxification in the fat bodies of S. frugiperda larvae.

The fall armyworm, Spodoptera frugiperda , is a polyphagous pest worldwide and feeds on many grain and cash crops, which threatens the safety of agriculture and forestry production. Toosendanin (TSN) is a commercial insecticidal active ingredient used to manage various pests in the field and showed adverse effects against S. frugiperda , while the effects of TSN on the larval midguts are not yet known. In this study, the effects of 10 and 20 mg/kg TSN exposures on the larval midguts were analyzed. The structural changes of the larval midgut induced by TSN treatments were also determined by hematoxylin-eosin staining. Besides, TSN treatments also changed the enzyme activities of three digestive enzymes (α-amylase, lipase, and trypsin) and two detoxification enzymes (CarE and GST). A total of 2868 differentially expressed genes (DEGs) were identified by RNA-Seq in the larval midguts with 20 mg/kg TSN treatment, and the DEGs responsible for food digestion and detoxification were further examined. Our findings revealed the preliminary modes of action of TSN on the larval midguts of S. frugiperda , which provide a preliminary rationale for controlling S. frugiperda with TSN in the field.

The fall armyworm, Spodoptera frugiperda (Noctuidae: Lepidoptera), is a wide-reaching notorious insect pest of important cereal crops, which has developed resistance to multiple classes of insecticides. It invaded the Sichuan Province of China in 2019. In this study, we performed resistance monitoring of insecticides for 11 field-collected populations from Sichuan, and all the populations were susceptible to emamectin benzoate and chlorpyrifos. The variations in resistance level to indoxacarb (resistance ratio (RR), 9.23–45.53-fold), spinetoram (RR, 4.32–18.05-fold), and chlorantraniliprole (RR, 2.02–10.39-fold) were observed among these populations. To investigate the resistance mechanism of chlorantraniliprole, synergism tests were performed and showed that piperonyl butoxide had a slight synergistic effect on chlorantraniliprole for the QJ-20 population (1.43-fold) in moderate resistance (RR, 10.39-fold) compared with the treatment group without synergist. Furthermore, the expression scanning for resistance-related genes showed that five P450 genes (CYP6AE43, CYP321A8, CYP305A1, CYP49A1, and CYP306A1) and the ryanodine receptor gene (Ryr, chlorantraniliprole target) were overexpressed in the QJ-20 population. These results indicated that the fall armyworm in Sichuan has exhibited diverse susceptibilities to several classes of insecticides, and the overexpression of Ryr and several P450 genes may contribute to the development of resistance in S. frugiperda to chlorantraniliprole.

The fall armyworm, Spodoptera frugiperda, has become a global invasive pest in recent years, threatening agricultural production. Carvacrol (CAR) is a multifunctional plant-derived compound and exhibited significant bioactivities against many pests. Our previous study found that CAR inhibited growth and development in S. frugiperda . However, the effects of CAR on the midgut of this pest remain unknown. In this study, the actions of CAR on the larval midgut of S. frugiperda were investigated. The results found that 1.0 and 2.0 g/kg CAR treatments destroyed the structure of the larval midgut based on hematoxylin-eosin staining and transmission electron microscope observation. In addition, 2.0 g/kg CAR exposure significantly altered the larval midgut bacterial community composition and structure. The results of the linear discriminant analysis effect size (LEfSe) analysis suggest that six bacterial genera contributed to the different structures of the larval bacterial community. Furthermore, PICRUSt2 analysis found that 16 bacterial function categories were altered by CAR treatment, of which 8 were significantly increased. Our results provided new insight into the toxicological mechanisms of CAR against S. frugiperda larvae and laid the foundation for the field application of S. frugiperda control.

Fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is the main destructive insect pest of grain crops that occurs in all maize growing regions of the Americas. It has rapidly invaded the Southern China since January 2019. However, the current status of insecticide resistance in S. frugiperda has not been reported in China. In this study, we determined the susceptibility of eight populations of FAW to eight insecticides by an artificial diet incorporation method. The results showed that among eight insecticides, emamectin benzoate, spinetoram, chlorantraniliprole, chlorfenapyr, and lufenuron showed higher toxicity to this pest, while lambda-cyhalothrin and azadirachtin exhibited lower toxicity. Susceptibility of S. frugiperda to indoxacarb was significantly different (10.0-fold for LC 50 ) across the various geographic populations. To investigate the biochemical mechanism of FAW to lambda-cyhalothrin, we performed the synergism tests and the results showed that piperonyl butoxide (PBO) and triphenyl phosphate (TPP) produced a high synergism of lambda-cyhalothrin effects in the two field populations. Sequencing of the gene encoding the acetylcholinesterase (AChE) gene in the two field populations identified two amino acid mutations, all of which have been shown previously to confer resistance to organophosphates (OPs) in several arthropod species. The results of this study provided valuable information for choosing alternative insecticides and for insecticide resistance management of S. frugiperda .

CA inhibited the growth and altered the development of S. frugiperda. • CA exposures altered detoxifying and digestive enzyme activities of S. frugiperda. • DEGs enriched in pathways of Toll/Imd signaling, lysosome, and peroxisome. • DEGs also enriched in detoxification, digestion, hormone synthesis, etc. The fall armyworm, Spodoptera frugiperda , has become a prominent global agricultural pest, threatening food production significantly. Caffeic acid (CA) is a phytochemical demonstrating considerable biological effectiveness against several pests. Nonetheless, the impact of this chemical on S. frugiperda is currently unascertained. This study assessed the growth inhibitory effects of CA on S. frugiperda larvae. The impact of 0.50 and 2.0 mg/g CA treatments on the growth and development of S. frugiperda was further examined. The administration of 0.50 and 2.0 mg/g CA could modify the detoxifying and digesting enzyme activity in the larvae. A total of 791 and 1704 differentially expressed genes (DEGs) were identified in the groups administered 0.50 and 2.0 mg/g CA treatments, respectively, in comparison to the control group. The differentially expressed genes were significantly enriched in immune-related pathways, including Toll/Imd signaling, lysosome, and peroxisome. In addition, the DEGs involved in detoxification, hormone synthesis, nutrient digestion and absorption, and fatty acid metabolism were also analyzed. Our findings suggest that CA may regulate the expression of these DEGs, hence influencing process functions and ultimately leading to growth inhibition in Spodoptera frugiperda larvae, underscoring CA's potential for pest management.

The fall armyworm (Spodoptera frugiperda), a lepidopteran pest of highly destructive, has developed resistance to organophosphate and carbamate insecticides through target-site mutations, including A201S and F290V in the ace-1 gene encoding acetylcholinesterase (AChE). Developing rapid and reliable methods to detect these mutations is crucial for monitoring resistance and guiding effective management strategies. Although S. frugiperda populations remain sensitive to the tested insecticides, the A201S and F290V mutations were identified in collected field samples. To facilitate detection, a sensitive and field-deployable loop-mediated isothermal amplification (LAMP) assay was developed, targeting these mutations within the ace-1 gene of S. frugiperda. The specificity and sensitivity of the LAMP assay was optimized for rapid detection, with reactions performed at 68 °C for 80 min. Results were visualized through color change and confirmed by gel electrophoresis. Validation using DNA from laboratory and field samples demonstrated complete agreement between the LAMP assay and DNA sequencing, affirming the accuracy and reliability of the method. The developed LAMP assay provides a simple, rapid, and effective tool for detecting the A201S and F290V mutations in the ace-1 gene of S. frugiperda. The assay’s simplicity, isothermal operation, and clear visual results make it a valuable tool for resistance monitoring and integrated pest management in agricultural settings.

RNA interference (RNAi) has shown excellent potential for pest management worldwide. However, the application of RNAi against insects can be inefficient and unreliable. Finding a better delivery system is a crucial factor for enhancing RNAi efficacy. Here, we present a novel and effective approach of conjugating double-stranded RNA (dsRNA) with a cell-penetrating disulfide polymer (CPD) to improve dsRNA stability and RNAi efficiency. The fall armyworm (FAW), Spodoptera frugiperda , is a globally agricultural insect. Chitin synthase B ( CHSB ) and methoprene-tolerant ( Met ) genes, which are essential for the development and growth of FAW, were selected as target genes. The CPD was synthesized using a two-step method for dsRNA delivery. The synthesized CPD/dsRNA complex protected dsRNA from nuclease degradation. The biological application of CPD in Sf9 cells demonstrated low cytotoxicity and high cell viability. Moreover, the CPD-loaded dsRNA entered the cells within 6 h. Bioassays of FAW showed that the relative expression levels of the CHSB and Met genes were reduced. Specifically, at 72 h, the relative expression levels of the CHSB and Met genes were 48.14 % and 37.60 % of those in the control group, respectively. The weight and body length of the larvae decreased significantly, and the mortality rate of CPD/ds CHSB reached 30 %. This CPD was demonstrated for the first time to have excellent delivery performance in insects and is expected to become a new and effective tool for pest control, representing a significant advancement in pest management.

Harmine , a useful botanical compound, has demonstrated insecticidal activity against some pests. However, harmine's mechanism of action has not been thoroughly elucidated to date. To preliminarily explore harmine's insecticidal mechanisms, the cytotoxicity of harmine against Spodoptera frugiperda Sf9 cells was comprehensively investigated. Our results indicated that harmine induced apoptosis in Sf9 cells, as evidenced by cellular and nuclear morphological changes, DNA laddering and increases in caspase-3-like activities. In addition, activation of the mitochondrial apoptotic pathway by harmine was confirmed by the generation of ROS, opening of mitochondrial permeability transition pores (MPTPs), increase in cytosolic Ca 2+ , changes in mRNA expression levels of genes involved in the mitochondrial apoptotic pathway and increase and release of Cytochrome c. Furthermore, lysosomal membrane permeabilization, release of cathepsin L from the lysosome into the cytosol and cleavage of caspase-3 were also triggered, which indicated that lysosomes were involved in this physiological process . Moreover, the effect of harmine on DNA topoisomerase I activity was investigated by in vivo and molecular docking experiments. These data not only verified that harmine induced apoptosis via comprehensive activation of the mitochondrial and lysosomal pathways and inhibition of DNA topoisomerase I activity in Sf9 cells but also revealed a mechanism of harmine insecticidal functions for pest control.
