Introduction As a biofilm-associated disease, dental caries benefits from nanoparticle (NP)-based therapies. Streptococcus mutans (S. mutans) is a primary aetiologic agent for dental caries development. We successfully applied a synergistic therapy of Ag/ZnO nanocomposites combined with light-emitting diode (LED) radiation to inhibit S. mutans biofilms. However, the antibiofilm mechanism has not been fully elucidated, and little is known about the biofilm formation ability of bacteria that survive NP-based therapies. Methods This study explored the antibiofilm formation mechanism of this synergistic therapy by an integrated approach based upon proteomics. Results Synergistic therapy killed 99.8% of bacteria, while the biofilm formation ability of 0.2% surviving bacteria was inhibited. The proteomic responses of S. mutans to synergistic therapy were comprehensively characterized to unveil the mechanism of bacterial death and biofilm formation inhibition of the surviving bacteria. In total, 55 differentially expressed proteins (12 upregulated and 43 downregulated) were recorded. The bioinformatic analysis demonstrated that cellular integrity damage and regulated expression of structure-associated proteins were the main reasons for bacterial death. In addition, the proteomic study indicated the potential inhibition of metabolism in surviving bacteria and provided a biofilm-related network consisting of 17 differentially expressed proteins, explaining the multiantibiofilm formation actions. Finally, we reported and verified the inhibitory effects of synergistic therapy on sucrose metabolism and D-alanine metabolism, which disturbed the biofilm formation of surviving bacteria. Conclusion Our findings demonstrated that synergistic therapy killed most bacteria and inhibited the surviving bacteria from forming biofilms. Furthermore, the antibiofilm formation mechanism was revealed by proteomics analysis of S. mutans after synergistic therapy and subsequent metabolic studies. Our success may provide a showcase to explore the antibiofilm formation mechanism of NP-based therapies using proteomic studies.

The purpose of the work is to evaluate the effect of extracellular polymeric substance (EPS) on the surface properties of Streptococcus mutans , and on the interaction between the bacteria and anti-adhesion nanostructured surfaces. Interactions between Streptococcus mutans with or without EPS and nanostructured surfaces were measured with atomic force microscope. Acid-base, Van der Waals, and electrostatic interactions were evaluated according to XDLVO theory. The absorption of glucan was evaluated by quartz crystal microbalance method. Results indicate Streptococcus mutans adhere stronger on surfaces with lower surface energy in both EPS and non-EPS conditions, because acid-base interaction is dominant. It indicates the superhydrophobic surface is lack of durability and reliability in the underwater anti-adhesion. Larger adhesion forces of Streptococcus mutans are shown in EPS than non-EPS conditions. That is due to more hydrogen bonds formed between glucan and substrate surfaces, and the EPS reducing surface energy and affecting acid-base interaction.

Background The dentin exposure always leads to dentin hypersensitivity and/or caries. Given the dentin’s tubular structure and low mineralization degree, reestablishing an effective biobarrier to stably protect dentin remains significantly challenging. This study reports a versatile dentin surface biobarrier consisting of a mesoporous silica-based epigallocatechin-3-gallate (EGCG)/nanohydroxyapatite delivery system and evaluates its stability on the dentinal tubule occlusion and the Streptococcus mutans (S. mutans) biofilm inhibition.Materials and Methods The mesoporous delivery system was fabricated and characterized. Sensitive dentin discs were prepared and randomly allocated to three groups: 1, control group; 2, casein phosphopeptide–amorphous calcium phosphate (CPP–ACP) group; and 3, the mesoporous delivery system group. The dentin permeability, dentinal tubule occlusion, acid and abrasion resistance, and S. mutans biofilm inhibition were determined for 1 week and 1 month. The in vitro release profiles of EGCG, Ca, and P were also monitored. Results The mesoporous delivery system held the ability to sustainably release EGCG, Ca, and P and could persistently occlude dentinal tubules with acid and abrasion resistance, reduce the dentin permeability, and inhibit the S. mutans biofilm formation for up to 1 month compared with the two other groups. The system provided prolonged stability to combat oral adverse challenges and served as an effective surface biobarrier to protect the exposed dentin. Conclusion The establishment of the dentin surface biobarrier consisting of a mesoporous delivery system indicates a promising strategy for the prevention and the management of dentin hypersensitivity and caries after enamel loss.

The integration of nanomaterials with clinical therapeutic instruments is a promising approach to improve the effects of nanomaterials. We reported an efficient synergistic antibacterial strategy formed through the combination of Ag/ZnO nanocomposites with a light-emitting diode (LED) curing light, which is a commonly used small instrument in dental clinics. The as-designed integration depicted a significantly enhanced bactericidal effect on facultative anaerobic oral pathogen Streptococcus mutans (S. mutans) both in planktonic and biofilm phases over a very short irradiation time (≤5 min). Further study showed that the combination of LED and Ag/ZnO nanocomposites induced more ·OH and ·O2– generation, which is responsible for the enhanced antibacterial activity. Moreover, this combination could destroy S. mutans biofilm by killing the bacteria embedded within biofilm, inhibiting exopolysaccharide production and down-regulating the biofilm-related gene expression. Therefore, it is proposed that this combination could be applied in dental clinics to realize dental caries prevention and dental restoration simultaneously.

Stereolithography (SLA) printable denture-tooth resin was modified with magnesium oxide nanoparticles (nMgO) to suppress Streptococcus mutans ( S. mutans ) biofilms while maintaining key material properties. nMgO was characterized prior to incorporation: ζ-potential ≈ +15 mV, cubic phase (XRD), reactive oxygen species generation (EPR), and near-spherical to polyhedral particles of ∼50–100 nm (TEM). Liquid resin containing 0–5 wt% nMgO was printed and post-cured under a standardized protocol. Antibiofilm activity was evaluated after a 24 h S. mutans challenge using viable biofilm counts and live/dead imaging. At 5 wt% nMgO, viable counts decreased by ∼89 % (p < 0.001); microscopy indicated increased bacterial death. Degree of conversion (FTIR), wettability, wear, compressive strength, and microhardness showed no significant differences from the unfilled control (p > 0.05). Flexural properties and water sorption and solubility remained within ISO 10477 limits, and cell-extract tests with human gingival fibroblasts met ISO 10993–5 non-cytotoxicity. These results indicate that embedding nMgO confers antibiofilm activity while retaining properties and standards compliance for denture-tooth materials, with potential utility at plaque-prone tooth–prosthesis interfaces.

Streptococcus mutans is one of the most significant pathogens leading to dental caries by producing highly cariogenic biofilms within the oral cavity. However, the increasing antimicrobial resistance brings great difficulties to the prevention and treatment of dental caries. Therefore, it is very important to develop new and effective antimicrobial agents from natural sources. Clausena lansium (Lour.) Skeels, a plant commonly cultivated in Southern China, is rich in various active constituents. In this study, the main constituents of the essential oils extracted from Clausena lansium leaves (EOL), seeds (EOS), peels (EOP) were precisely analyzed by gas chromatography–mass spectrometry. The antimicrobial and anti-biofilm activities of EOL, EOS, and EOP, along with their main chemical constituents, against Streptococcus mutans were comprehensively evaluated. The primary constituent in EOL was β-caryophyllene, with a content of 112.29 mg/mL. EOS was found to be abundant in sabinene and 4-terpineol, with constituents of 147.21 mg/mL and 54.36 mg/mL, respectively. In EOP, the most predominant constituent was β-phellandrene, with a content of 173.95 mg/mL. The minimum inhibitory concentrations of EOL, EOS, and EOP were 0.12, 2.00, and 1.00 mg/mL, respectively. The minimum concentrations of inhibiting at least 90% biofilm formation of EOL, EOS, and EOP was 0.12 mg/mL, 2.00, and 2.00 mg/mL, respectively. Intriguingly, the antimicrobial and anti-biofilm efficacies of the main constituents were significantly lower than those of the essential oils. By observing the dynamic changes of the biofilm formation of S. mutans treated with essential oils, it was found that the process of biofilm formation was greatly delayed. In some cases, there was even no biofilm formation. The results of real-time quantitative PCR indicated that the expression levels of brpA, gtfB, gtfC, gtfD, and luxS genes were down-regulated after treatment with the essential oils. This study reinforces the potential of Clausena lansium as a promising source for developing new antimicrobial and anti-biofilm agents. They could inhibit the proliferation and virulence of S. mutans, thereby contributing to the control of dental caries.
