Tremella fuciformis Polysaccharides Alleviate Early Brain Injury in Experimental Subarachnoid Hemorrhage by Inhibiting the KDR-Mediated P38 MAPK/NF-κB Pathway

Wan Yu-hui,Cheng Zeng-jing,Fan Ling-xiang,Yang De-hong,Chen Bing-lin,Chen Xiang-xin,Zhu Qing

Subarachnoid hemorrhage (SAH) is associated with high mortality and morbidity rates. In its early stages, a substantial influx of blood into the subarachnoid space triggers excessive activation of microglia, which markedly contributes to early brain injury (EBI), a pivotal determinant of poor prognosis. Tremella fuciformis polysaccharides (TFPSs), as acidic heteropolysaccharides from the fruiting bodies of Tremella, exhibit robust anti-inflammatory characteristics and many biological properties. Nonetheless, the impact of TFPSs on EBI after SAH has yet to be reported, and the molecular mechanisms underlying these effects remain elusive. We used in vivo and in vitro models to study the effects of TFPSs on microglia post-SAH. Network pharmacology analysis was used to predict the targets of TFPSs and the pathways through which it exerts its therapeutic effects. These predictions were subsequently corroborated through flow cytometry, Western blotting, immunofluorescence staining, enzyme-linked immunosorbent assay (ELISA), and quantitative real‑time polymerase chain reaction, both in vivo and in vitro. After 24 h post-SAH, TFPS-treated mice presented improved neurological function and reduced cerebral edema. TFPSs reversed microglial activation, enhanced phagocytic ability, and reduced neuronal apoptosis. Network pharmacology identified KDR as a potential target of TFPSs, with the P38 MAPK pathway as the downstream pathway. TFPSs attenuated KDR expression, inhibited the P38 MAPK/NF-κB pathway, reduced inflammatory cytokine expression, and increased microglial phagocytic capacity post-SAH. This investigation revealed that TFPSs may ameliorate EBI after SAH, potentially via the regulation of the KDR-mediated P38 MAPK/NF-κB pathway and phagocytic function.

Compositional analysis and physicochemical characterization of a purified tremella fuciformis polysaccharide-rich fraction and its intestinal barrier- and microbiota-related effects in DSS-induced colitis

Xinjiu Deng,Haikun Li,Jiamei Hou,Changci Tong,Jinpeng Liu,Yanwen Zhu,Yunen Liu

• Purified Tremella polysaccharide-rich fraction was physicochemically defined. • • TPS alleviated DSS-induced colitis and preserved barrier-related proteins. • • TPS reduced colonic inflammatory cytokines under DSS-induced injury. • • TPS was associated with partial shifts in DSS-disrupted gut microbiota. • • TLR4/MyD88/NF-κB changes were interpreted as downstream readouts. Tremella fuciformis is a widely consumed edible fungus and a source of structurally diverse polysaccharides, but the intestinal biofunctionality of a purified Tremella polysaccharide-rich fraction remains insufficiently defined. In this study, a purified T. fuciformis polysaccharide-rich fraction (TPS) was obtained by hot-water extraction, ethanol precipitation, anion-exchange chromatography, and gel filtration. TPS was characterized using carbohydrate/protein analysis, gel permeation chromatography (GPC), monosaccharide composition analysis, Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), atomic force microscopy (AFM), scanning electron microscopy (SEM), 1 H NMR, dynamic light scattering (DLS), zeta-potential analysis, and thermogravimetric analysis. TPS contained 96.13% total carbohydrate and 5.61% protein and showed features of a mannose-rich acidic heteropolysaccharide, including mainly amorphous organization, aggregated morphology, mixed anomeric proton signals, hydrated colloidal aggregates, negative surface charge, and staged thermal degradation behavior. Its intestinal effects were evaluated in a dextran sulfate sodium (DSS)-induced colitis mouse model. Oral TPS administration alleviated DSS-induced colonic injury, as indicated by improved disease activity and histological features, preservation of ZO-1 and Occludin expression, and reduced colonic TNF-α, IL-1β, and IL-6 levels. TPS was also associated with partial changes in DSS-disrupted gut microbiota profiles and with lower downstream inflammatory protein readouts in the TLR4/MyD88/NF-κB pathway. Molecular docking of TPS-associated monosaccharides was retained only as supplementary exploratory in silico context and was not interpreted as evidence of intact TPS binding, validated receptor engagement, or direct in vivo TLR4 targeting. Overall, this study connects fraction-level compositional and physicochemical characterization of a food-derived Tremella polysaccharide-rich fraction with measurable intestinal barrier-, inflammation-, and microbiota-related responses, providing a basis for future structure-function studies of edible fungal polysaccharides in intestinal health. Biomacromolecular composite microspheres based on sodium alginate and tremella fuciformis polysaccharide for enhanced protection and delivery of camellia oil: A comprehensive study in simulated digestion

Yuanhong Cai,Peipei Xu,Runbing Huang,Jiayi Huang,Junhua Zhan,Junpeng Su,Ruiyun You,Yudong Lu

This study presents the development of a novel polysaccharide biomolecule composite microsphere system based on sodium alginate (SA) and tremella fuciformis polysaccharide (TFP) for the encapsulation and controlled release of camellia oil (CO). The system leverages the synergistic properties of these two bioactive biopolymers to enhance the protection and bioavailability of CO during simulated digestion. The microspheres, characterized by their spherical structure and uniform particle size distribution, were comprehensively evaluated for stability, elemental composition, and thermal stability using techniques such as scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), Fourier transform infrared spectroscopy (FT-IR), and thermogravimetric analysis (TGA). The results confirmed a significant improvement in the elemental composition and surface properties of the microspheres due to the incorporation of CO, which enhanced their stability. In simulated digestion experiments, the microspheres demonstrated excellent gastric acid tolerance and intestinal release, achieving a high encapsulation efficiency of 99.59 % and an oil loading capacity of 80.67 %, effectively protecting and targeting the release of CO. Moreover, the microspheres exhibited significant antibacterial activity against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ). Overall, the SA/TFP/CO microspheres represent an ideal delivery system for CO, demonstrating considerable potential in the fields of food science and biomedicine.

Novel natural microbial preservative nisin/Tremella fuciformis polysaccharide (TFP)/Lactobacillus plantarum (LP) live particle (NTN@LP) and its effect on the accumulation of biogenic amines during sausage fermentation

Yaowen Liu,Yue Wang,Siwei Shen,Md Alomgir Hossen,Dur E. Sameen,Saeed Ahmed,Jianwu Dai,Siyu Xiao,Suqing Li,Wen Qin

In order to increase the stability and the release time of nisin, nisin/ Tremella fuciformis polysaccharide nanoparticles (NTNs) were prepared and evaluated based on their uniform particle size dispersion (168–356 nm) and high loading rate (70–86%). The condensation reaction between 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride and hydroxysuccinimide (EDC/NHS) amide was successfully used to prepare novel live particles (NTN@LP). When the mass ratio of NTN and LP was 2:1, the bacterial viability (85.6 ± 2.11%) and grafting efficiency (68.8 ± 1.96%) of the NTN@LP were both relatively high. The hemolytic activity and in vitro cytotoxicity indicated that NTN@LP is safe to use in food. The NTN@LPs were then used as natural microbial preservatives in sausages. The pH value, water activity, thiobarbituric acid-reactive substances value, and sensory evaluation of the sausages were used to assess the effect of NTN@LPs on the quality of the sausages during fermentation. NTN@LPs played an important role in reducing and decomposing biogenic amines (BAs) throughout the sausage fermentation process and displayed a strong inhibitory effect on histamine (>95%). Compared to that of the control group, the total amount of BAs in the 2:1 group was reduced by 67.90%. Tyramine content in the 2:1 NTN@LP group decreased by 78.16% compared to that in the control group, while tyramine content in the pure LP group was decreased by only 38.25%. These results indicate that NTN@LP can be potentially applied as a natural microbial preservative in the food industry.
