Chemical profiling and arginine kinase inhibitory activity of Angelica dahurica leaves

Aitong Yang,Junyan Zhang,Guangying Lv,Jiabao Chen,Long Guo,Yan Liu,Yuguang Zheng,Lei Wang

Angelica dahurica is a medicinal herb of the Umbelliferae family. The dried root of A. dahurica , also known as Angelicae dahuricae Radix, is widely used in clinical treatment. However, the aboveground part of A. dahurica which accounted for over 70% of the total plant was abandoned in the field. In order to develop the value of the aboveground part of A. dahurica , the chemical constituents and arginine kinase (AK) inhibitory activity of A. dahurica leaves were studied. 85 volatile components were identified from A. dahurica leaves by GC-MS; 39 non-volatile components including sugars, amino acids and organic acids were identified by pre-column derivatization GC-MS analysis; and 7 coumarins were qualitatively and quantitatively analyzed by HPLC. Then, an inhibitory enzyme-linked immunosorbent assay (iEIA) was applied for evaluation of AK inhibitory activity. The extracts of A. dahurica leaves exhibited well inhibitory effects on AK. Further, potential AK inhibitors were screened by grey relational analysis and their inhibitory activities were validated by iEIA. l -aspartic acid exhibited strongest inhibitory effect on AK with its IC 50 value was 0.558 mM, which was much lower than that of chlorpheniramine (6.644 mM). The obtained chemical profiles displayed chemical diversity of A. dahurica leaves and will provide data support for the future development and utilization of A. dahurica leaves. The screened potential AK inhibitors from A. dahurica leaves could be candidates for development of antiallergic substances or insecticides.

Does Arginine Aggregate Formation in Aqueous Solutions Follow Two-Step Mechanism?

Fuge Niu,Xinyu Hu,Christos Ritzoulis,Weiwei Tu,Xurui Zhao,Yuanzhi Xia,Yinghua Lu,Junfeng Yin,Weichun Pan

The formation of aggregates was studied in amino acid aqueous solutions. Regarding to arginine (Arg), the main driving force to form aggregates is hydrogen-bonds between the Arg molecules. The measurement of energy loss during this process, coupled with morphology data indicate that this aggregate is in solid state. The aggregation has two steps, with a liquid intermediate stage. An investigation of the effect of pH and solute concentration upon aggregate formation for other amino acid aqueous solutions verifies that aggregate formation is amino-acid specific, while small size of clusters formed by weak interactions lead to large size aggregation. Homochirality is observed in the aggregates; its existence sheds light on the origin of protein homochirality.

L-Arginine effectively alleviates doxorubicin-induced cardiac dysfunction by inhibiting myocardial fibrosis

Zhu Mo-li,Hao Rui,Zhao Meng-yao,Liu Pei-yao,Zhang Hong-dou,Li Peng,Yin Ya-ling,Liu Hui-jun,Ren Guang-qiu,Zhang Jie,Yang Jing

Background The clinical use of doxorubicin (DOX), a widely used and effective antitumor drug, is limited by its cardiotoxicity. Currently, safe and effective strategies for preventing doxorubicin-induced cardiotoxicity (DIC) remain limited. Therefore, this study aimed to investigate the potential cardioprotective effects and possible underlying mechanisms of L-arginine (L-Arg) against DIC. Methods and Results To investigate the cardioprotective effects and underlying mechanisms of L-Arg, three complementary experimental models were used: male Sprague–Dawley rats ( n  = 6 per group), male AMPKα2 knockout (AMPKα2 KO) mice ( n  = 6 per group), and H9c2 cardiomyocytes. A DIC model was induced in rats by intraperitoneal injection of DOX (2.5 mg/kg/week) for 6 weeks. Serum nitric oxide (NO) and lactate dehydrogenase (LDH) levels were measured to assess oxidative stress and myocardial injury. Cardiac morphology, inflammation, and fibrosis were evaluated by histological staining and protein expression analyses, whereas miR-29b-3p expression was determined by RT-qPCR. AMPKα2-deficient models and miR-29b-3p gain- and loss-of-function models were used to investigate the underlying mechanisms. Compared with the DOX group, L-Arg significantly improved cardiac function and morphology, reduced oxidative stress, myocardial injury, inflammation, and fibrosis, and increased miR-29b-3p expression (all P  < 0.05). Moreover, miR-29b-3p overexpression enhanced, whereas miR-29b-3p inhibition attenuated, the cardioprotective effects of L-Arg ( all P < 0.05 ). These protective effects were also markedly attenuated by AMPKα2 deficiency ( P < 0.05 ). Conclusions L-Arg alleviates DIC by improving cardiac function, reducing oxidative stress, inflammation, and fibrosis, and increasing miR-29b-3p expression. These cardioprotective effects are associated with AMPKα2 activation and enhanced miR-29b-3p expression.

The arginine detection and cytotoxicity of fluorescent probes based on naphthalene derivatives

Xuefang Shang,Congshu Li,Jie Li,Yanmei Chen,Hongli Chen,Tianyun Wang

From a biological point of view, Schiff base probes containing fluorescent groups are chemically modified to provide an effective method for the detection of amino acids. According to this method, three molecular probes containing Schiff base and hydroxyl group have been designed and synthesized. UV-Vis and fluorescent data indicated that compound 2 showed strong sensitivity and high selectivity for arginine (Arg) among normal twenty kinds of amino acids. In addition, compound 2 displayed high combining ability with Arg and low cytotoxicity in MCF-7 cell from 0 to 150 μg/mL. The above results showed the synthesized probes also can be used a biosensor for the Arg detection in vivo.

An effect fluorescence sensor for arginine based on bis-cyanodistyrene Schiff-base

Huan Zhou,Xianyu Huang,Sining Zheng,Hongyu Guo,Fafu Yang

The selective detection of arginine among all kinds of amino acids is of great significance but rarely reported. In this work, a simple fluorescence sensor for sensing arginine in near aqueous media was developed based on bis-cyanodistyrene Schiff-base ( BCS ). BCS was prepared by the Schiff-base condensation of amino-cyanodistyrene derivative and bis-benzaldehyde in yield of 86 %. BCS possessed high fluorescence in near aqueous media and exhibited selective detection for arginine among all kinds of test species including six common metallic ions and twenty amino acids. The detection limitation for arginine was as low as 1.34 × 10 −7 M. BCS showed the good sensing ability in pH = 6–9 and the sensing selectivity kept stable under the interference of other species. The 1:1 binding mechanism was proposed based on the multiple intermolecular hydrogen bonds. The selective sensing ability of BCS for arginine was successfully applied on tested paper and the sample analyses of MinJiang river and adults urine. BCS was further used in the living-cell imaging experiments, suggesting the excellent fluorescence imaging property and the good detecting ability for arginine in living-cell environment. This research supplied a good method for real-time and in-situ sensing arginine in complicated environments and living bodies.

Arginine-assisted Thermal Decomposition for Synthesis of Nanosized Co3O4 with Enhanced Capacitance

Ruyi Zou,Lin Zhu,Guiling Luo,Yanyan Niu,Hui Xie,Ruixia Dong,Hui Cheng,Wei Sun,Laijun Zhang

Arginine-assisted thermal decomposition technique was used to prepare nanosized Co 3 O 4 (A-Co 3 O 4 ) with its supercapacitive performances checked. A-Co 3 O 4 was characterized by different techniques such as X-ray diffraction, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy and scanning electron microscopy. Brunauer-Emmett-Teller analysis revealed that A-Co 3 O 4 had a larger specific surface area than Co 3 O 4 prepared by conventional calcination without arginine (F-Co 3 O 4 ). Electrochemical performances of A-Co 3 O 4 based electrode were better than that of the F-Co 3 O 4 based electrode. Cyclic voltammetric results showed the specific capacitances of A-Co 3 O 4 and F-Co 3 O 4 were found to be 259.2 and 122.2 F/g at a scan rate of 0.01 V/s. Long-term galvanostatic charge-discharge cycling tests indicated the specific capacitance for A-Co 3 O 4 and F-Co 3 O 4 were 128.6 F/g and 106.4 F/g with 104.7% and 103.3% capacitance retention after 1000 cycles at a current density of 5.0 A/g. Electrochemical impedance spectra analysis showed the charge transfer resistance (Rct) of A-Co 3 O 4 was 7.56 Ω and that of F-Co 3 O 4 was 15.56 Ω. All the results indicated the A-Co 3 O 4 exhibited superiority originated from large surface area with porous structure.

Biosynthesis of Putrescine from L-arginine Using Engineered Escherichia coli Whole Cells

Hongjie Hui,Yajun Bai,Tai-Ping Fan,Xiaohui Zheng,Yujie Cai

Putrescine, a biogenic amine, is a highly valued compound in medicine, industry, and agriculture. In this study, we report a whole-cell biocatalytic method inEscherichia colifor the production of putrescine, using L-arginine as the substrate. L-arginine decarboxylase and agmatine ureohydrolase were co-expressed to produce putrescine from L-arginine. Ten plasmids with different copy numbers and ordering of genes were constructed to balance the expression of the two enzymes, and the best strain was pACYCDuet-speB-speA. The optimal concentration of L-arginine was determined to be 20 mM for this strain. The optimum pH of the biotransformation was 9.5, and the optimum temperature was 45 °C; under these conditions, the yield of putrescine was 98%. This whole-cell biocatalytic method appeared to have great potential for the production of putrescine.Keywords:L-arginine;putrescine;Escherichia coli;whole-cell catalysis;co-expression

A Novel Ratiometric Fluorescent Probe for Ag+ Based on Arginine-Naphthalene Imide

Liu Jing,Wang Shan,Wang Xiaofang

A ratio fluorescent probe to detect Ag + using arginine-naphthalene imide (AN) had been elaborately designed and synthesized. The structure of AN was characterized by 1 HNMR and FTIR. The optical properties of AN were investigated by ultraviolet and fluorescence spectroscopy. The results indicated that the fluorescent intensity of AN was enhanced gradually with the increase of Ag + concentration, which was hardly affected by other metal ions such as Bi 3+ , Cd 2+ , Co 2+ , Cr 3+ , Cu 2+ , Fe 3+ , Hg 2+ , Pb 2+ , K + and Na + . A good linear correlation was displayed between the concentration of Ag + (ranging from 4.30 to 11.40 μmol L −1 ) and the fluorescence intensity ( R 2  = 0.9985). The detection limit was 0.1883 μmol L −1 . The mechanism of the sensor may be based on the principle of intramolecular charge transfer. It was expected that this novel ratio sensor could be used for the detection of silver ions in water.

Coordination of l-arginine and iron cation improves stability of hemoglobin concentrates

Zhou Cunliu,Ye Huiqiong,Wang Hui,Qin Hao,Li Junhong

This work evaluated the effects of l -arginine on the stability of porcine hemoglobin concentrates during storage. The addition of l -arginine increased both the oxyhemoglobin content and a* values but decreased both the methemoglobin content and free iron content ( p  < 0.05) during storage. The addition of iron cation decreased both the oxyhemoglobin content and a* values but increased both the methemoglobin content and free iron content ( p  < 0.05). CD spectra disclosed that the free iron induced the transformation of the hemoglobin secondary structure from an α-helix to a β-pleated, β-turn, and random-coil arrangement, while l -arginine hindered the behavior of the free iron. Infrared spectra revealed that the l -arginine coordinated with the free Fe 2+ or Fe 3+ to form the corresponding complex at 25 °C and pH 7.3. These results showed that l -arginine enhanced the stability of hemoglobin concentrates by coordinating with free iron.

Rapid Mass Analysis of Reserpine and Arginine by Ion Trap Mass Spectrometer

Fu-Xing XU,Qian-Kun DANG,Yin-Juan CHEN,Kai YANG,Qiang WANG,Bin CHEN,Yuan-Yuan WANG,Chuan-Fan DING

A fast mass analysis method using digital ion trap technology was reported. The mass analysis was realized by scanning the frequency of digital RF voltage and dipole ion resonance voltage during ion injection period without the conventional ion cooling, ion resonance ejection or ion cleaning process. It can decrease about three fourth of traditional analysis time using ion trap mass spectrometry. The ion signal was optimized through the modification of frequency scanning rate, ion gate voltage and other experimental parameters. For example, an optimal mass spectroscopy of reserpine ( m / z 609) and arginine ( m / z 174) were obtained when the ion gate voltage was 9 V and the digital RF frequency was scanned from 1 MHz to 400 kHz at rate of 2385 Th s −1 . The results indicated that the obtained mass spectra were in very good agreement with conventional analytical method using ion trap mass analyzer.

Lighting up arginine metabolism reveals its functional diversity in physiology and pathology

Rui Li,Yan Li,Kun Jiang,Lijuan Zhang,Ting Li,Aihua Zhao,Zhuo Zhang,Yale Xia,Kun Ge,Yaqiong Chen,Chengnuo Wang,Weitao Tang,Shuning Liu,Xiaoxi Lin,Yuqin Song,Jie Mei,Chun Xiao,Aoxue Wang,Yejun Zou,Xie Li,Xianjun Chen,Zhenyu Ju,Wei Jia,Joseph Loscalzo,Yu Sun,Wei Fang,Yi Yang,Yuzheng Zhao

Arginine is one of the most metabolically versatile amino acids and plays pivotal roles in diverse biological and pathological processes; however, sensitive tracking of arginine dynamics in situ remains technically challenging. Here, we engineer high-performance fluorescent biosensors, denoted sensitive to arginine (STAR), to illuminate arginine metabolism in cells, mice, and clinical samples. Utilizing STAR, we demonstrate the effects of different amino acids in regulating intra- and extracellular arginine levels. STAR enabled live-cell monitoring of arginine fluctuations during macrophage activation, phagocytosis, efferocytosis, and senescence and revealed cellular senescence depending on arginine availability. Moreover, a simple and fast assay based on STAR revealed that serum arginine levels tended to increase with age, and the elevated serum arginine level is a potential indicator for discriminating the progression and severity of vitiligo. Collectively, our study provides important insights into the metabolic and functional roles of arginine, as well as its potential in diagnostic and therapeutic applications.

Arginine alleviates LPS-induced leukocytes inflammation and apoptosis via adjusted NODs signaling

Yuan-Xin Qian,Feng-Feng Zhou,Qiang Chen,Fen Dong,Han-Ying Xu,Yu-Long Sun,Ji-Teng Wang,Tao Han

Arginine plays a key role in regulating the immune function of fish. To evaluate the effect of arginine on the immune response of largemouth bass ( Micropterus salmoides ), the effects of arginine on cell viability, NADPH oxidase activity, respiratory burst activity, and NO production of leukocytes were analyzed both in vitro and in vivo . In this study, we found that arginine could promote the respiratory burst activity of leucocytes both in vivo and in vitro . By incubating leukocytes with the combination of LPS and arginine, we found that arginine supplementation inhibited the expression of inflammatory genes ( tumor necrosis factor-alpha , tnfα ; interleukin ( il ) 8 and il10 ) and apoptotic genes ( caspase 3 , caspase 8 , and caspase 9 ) induced by LPS, as well as promoted the arginine metabolism. Arginine supplementation significantly induced ( cd4-like ) cd4 gene expression after LPS challenge. Further studies showed that LPS could significantly increase nucleotide-binding oligomerization domain containing 1 ( nod1 ) gene expression, but decreased the nod2 gene. The arginine supplementation increased nuclear factor kappa-B (NF-κB) protein level. In conclusion, arginine can alleviate LPS-induced inflammatory response and apoptosis as well as induce cd4 gene expression against LPS challenge via adjusting the expression of NODs signaling.
