Infections caused by microorganisms on the surfaces of medical implants and devices represent a significant challenge in the field of medicine. At present, a considerable number of antimicrobial coatings have been developed with the objective of preventing bacterial contamination. However, the problem of mold contamination is often underestimated. To address this issue, a bi-molecular composite coating was developed using sulfobetaine and borneol unites, which enabled the modification of polydimethylsiloxane (PDMS) substrates. Furthermore, by modifying the ratio of sulfobetaine and borneol groups, the coating can simultaneously demonstrate anti-bacterial, anti-mold adhesion and anti-biofilm properties. Notably, the bi-molecular polymer coating exhibited a synergistic effect in inhibiting bacterial biofilm formation. Subsequent in vivo subcutaneous implantation in mice confirmed its favorable biocompatibility. Therefore, this approach offers novel insights into the development of antimicrobial adhesion coatings for medical device surfaces, particularly those need to prevent biofilm formation.

Infection is a huge obstacle to wound healing. Thus, to enhance the healing of infected wounds, wound dressings that permit the dual delivery of antimicrobials and antioxidants are highly desirable. In this study, a series of gelatin-based nanofiber membranes with different curcumin contents were fabricated via solution electrospinning. The obtained membranes were characterized in terms of their morphologies, in addition to their physical, mechanical, and in vitro properties. The results showed that the membranes maintained an integrated morphology, excellent water absorption capability, satisfactory mechanical properties, and a high dissolution rate of curcumin. The addition of curcumin and borneol conferred the membranes the ability to inhibit Staphylococcus aureus and eliminate free radicals. Furthermore, cytocompatibility testing using the L929 cell line confirmed the excellent biocompatibility of the membranes. These gelatin-based nanofiber membranes loaded with curcumin and borneol can therefore be considered as promising materials for dressing wounds. Moreover, the use of biodegradable polymers and environmentally sustainable production techniques in this system render it suitable for the commercial manufacture of composite membranes.

Nowadays, the development of sustainable nontoxic antifouling coatings is urgent. In this work, the isobornyl as antifouling groups and dodecfluoroheptyl were introduced into the silicone side chains to prepare the environment-friendly silicone antifouling coatings. The synergistic effect of natural borneol and dodecfluoroheptyl improved the static antifouling ability of silicone coating. The coatings exhibited good optical clarity (>92%, 400–800 nm), high adhesion (>2 MPa, steel plates) and excellent fouling release and self-cleaning ability. For example, PDMS-F-PIBO-23 coating had excellent antibacterial and antifouling activities. It’s antibacterial resistance to marine bacteria (Pseudomonas sp.) reached 95.6%, which reduced the coverage area of biofilm by 86.9%, and effectively prevented the adhesion of diatoms. This remarkable antibacterial and antifouling performance was attributed to the complex molecular structure of natural non-toxic borneol and the enrichment of fluoropolymers. Field tests at Yellow Sea showed that the coating had good antifouling ability and could be used for >6 months. Most importantly, compared with traditional coatings containing toxic fungicides, the coating is less harmful to the marine ecological balance. This research provides more possibilities for marine antifouling coatings design and preparation which is expected to be applied in marine, medical and other antipollution fields.

Diabetic peripheral neuropathy (DPN) remains a prevalent and challenging complication of diabetes with limited therapeutic options. Magnolol (Mag), a natural multi-target agent, demonstrates significant potential in managing DPN through synergistic anti-inflammatory, antioxidant, and neurorestorative properties. However, its poor oral bioavailability and limited skin penetration hinder clinical application. To address these challenges, we developed a novel topical nanogel system for enhanced transdermal delivery of Mag. Borneol (Bor), a potent penetration enhancer, was co-assembled with Mag via antisolvent precipitation to form self-assembled nanoparticles (Mag-Bor NPs), which were further encapsulated into a carbomer hydrogel matrix to construct the Mag-Bor nanogel (Mag-Bor NPs Gel). The resulting formulation exhibited markedly improved skin permeability and sustained drug retention. Upon topical administration, Mag-Bor NPs Gel effectively alleviated neuropathic impairments in DPN rats through a “transdermal enhancement – anti-inflammatory – nerve repair” mechanism. This therapeutic strategy comprehensively improved key pathological indicators, including plantar thermal, mechanical pain sensitivity, motor nerve conduction velocity (MNCV), intraepidermal nerve fiber density (IENFD), sciatic nerve myelin structure, and inflammation levels in dorsal root ganglia (DRG). This study is the first to report a nanogel system combining efficient transdermal delivery and neurorestorative functions, offering a novel and promising strategy for topical DPN treatment.

This study focuses on the potential hazards of borneol (BO) to aquatic organisms and human health. BO has antibacterial, anti-inflammatory and antioxidant activities, and is widely used in medicine, cosmetics, and detergents. In this study, zebrafish was used as a model organism to systematically evaluate the effects of BO on the heart, liver, kidney, and nervous system. The effects of BO on metabolites of zebrafish were studied using MALDI-MSI. The results showed that a high concentration of BO (500 μM) could induce morphological abnormalities (swim-bladder loss, spinal curvature, body-length shortening), cardiotoxicity (decreased heart rate, increased SV-BA distance), hepatotoxicity (reduced liver area index), and neurotoxicity (impaired behavioral ability, and dopamine neuron development deficits), but there was no renal toxicity observed in zebrafish. Additionally, MALDI-MSI analysis showed that BO exposure significantly altered the levels of metabolites, including phospholipids, fatty acids, choline, and amino acids. The contents of PC-34:1, PC-34:2, PI-36:4, PE-36:1, LysoPE-22:5, LysoPC-18:1, FA-18:2, phenylalanine, lysine and glutathione were significantly increased, while the contents of PC-38:6 and PC-40:6 were significantly decreased. Notably, BO elicited a significant alteration in the mRNA expression levels of genes associated with phospholipid metabolism, fatty acid metabolism, choline metabolism, and amino acid metabolism (such as elovl5 , chpt1 , chka , setd7 , hgd ). This study revealed that BO exerted toxicity on multiple organs and demonstrated that BO causes metabolic dysregulation in zebrafish. These findings provide a novel insight into the toxicity of BO.

The release of toxic and persistent substances in conventional antifouling paints has elicited substantial environmental concerns, underscoring an urgent requirement for eco-friendly alternatives. This study presents the successful synthesis of an eco-friendly antifouling coating, denoted as PSBZ-X-Y, through the grafting of a hydrolysable borneol-modified zwitterionic precursor, DEA-IBOA, onto a bio- derived rosin resin. The PSBZ-X-Y coating exhibits a controlled self-renewable rate (50–400 μg/cm 2 ·d) tuned by varying DEA-IBOA modified rosin content, which enhances the coating's durability and regulates the release of borneol. Molecular dynamics (MD) simulations and surface structure analyses, including ATR-FTIR and XPS, reveal that hydrolysis of DEA-IBOA leads to the release of the natural antimicrobial agent borneol and thus facilitates the in-situ generation of zwitterionic surfaces. Amphiphilic surfaces and natural antimicrobial borneol as eco-friendly approaches synergistically enhance the antifouling properties of natural rosin, which in turn promotes the sustainability of amphiphilic surfaces due to its self-renewal capability. Consequently, the coating exhibits remarkable resistance to protein adhesion (FITC-BSA adhesion rate: 0.4 %), bacterial biofilm formation (absorbance of biofilm: 1.3), and diatom attachment (150 cells/mm 2 ), as well as remarkable antifouling properties in real marine environments. More importantly, the hydrolyzed products generated during the coating renewal process demonstrate no adverse effects on the marine ecosystem, thus contributing to eco-friendly antifouling systems based on natural products.

Bacterial infection of surface-damaged fruit is one of the troubling issues in daily life. To address this issue, the β-cyclodextrin (CD) grafting N+-borneol ester antibacterial polymer (CD-NBOx), containing borneol with active groups and quaternary ammonium compounds (QACs) positive charges, was successfully synthesized using RAFT and characterized by NMR, Fourier-transform infrared, and gel permeation chromatography. Its antibacterial activity and biofilm inhibition were evaluated against Escherichia coli and Staphylococcus aureus, and the adhesion resistance was measured by crystal violet (CV) staining experiments. It indicated that the antibacterial rate of CD-NBOx reached 99.9 and 87.3% and the biofilm inhibition rates were up to 97.5% against E. coli. Interesting, there was no spoilage of the fruit and only 20% weight loss in the 15-day fruit preservation experiment. CD-NBOx possesses excellent antibacterial and adhesion resistance due to the synergy between cyclodextrin for biocompatibility and N cation for antibacterial and borneol stereochemistry. In summary, CD-NBOx as a constructing “offensive and defensive integrated” is validated as a viable antibacterial material while also expanding potential life applications of N+-borneol ester polymers in fruit preservation in the future.

Natural plant-derived molecules are emerging as promising alternatives to chemical preservatives. Among these, the phytochemical borneol has been shown to inhibit the growth of common foodborne bacteria, but its antifungal activity and underlying mechanisms remain largely unexplored. This study aims to investigate the antifungal properties of borneol and its mechanisms of action against Candida albicans , a foodborne fungus that poses a significant threat to food safety. The results demonstrated that the antifungal efficacy of borneol against Candida albicans was concentration-dependent at the tested time points. Transcriptomic analysis and electron microscopy revealed that borneol primarily targets cell membranes to exert its antifungal effects. Further experiments showed that borneol specifically binds to membrane proteins, particularly chitin synthases 1, 2, and 3, inhibiting chitin biosynthesis and disrupting cellular integrity. Molecular dynamics simulations indicated that borneol diffuses passively from the aqueous phase into the 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphorylcholine bilayer membrane, where it preferentially aggregates in hydrophobic regions, altering the bilayer's thickness and fluidity. Notably, this aggregation likely plays a key role in influencing the structure and function of membrane proteins. Lipidomics analysis further confirmed that borneol disrupts cell membrane integrity, leading to the loss of glycerophosphocholines and glycerophosphoethanolamines lipids. As borneol alters membrane structure and function, it also induces the intracellular accumulation of reactive oxygen species, resulting in membrane lipid peroxidation. In conclusion, these findings provide a theoretical foundation for the potential use of borneol in the food industry to prevent and control pathogenic fungal contamination.

Ulcerative colitis (UC) is a chronic and relapsing inflammatory bowel disease with an increasing global burden. Although various terpenoids have demonstrated significant efficacy against UC, the therapeutic mechanism of bornyl acetate (BA), a monocyclic diterpene derived from pine needle essential oil (PNEO), remains unclear. This study systematically investigated the anti-inflammatory effects and microbiota-modulating mechanisms of PNEO and BA by using an integrated approach that combined in vitro and in vivo models with 16S rRNA sequencing. These results showed that while PNEO significantly inhibited pro-inflammatory mediators like NO and TNF-α, its therapeutic efficacy against UC was modest. In contrast, BA exerted potent anti-inflammatory effects by downregulating the transcriptional activity of p65 in the NF-κB pathway. Furthermore, BA enhanced the transcription and expression of tight junction proteins (ZO-1, claudin-1, and occludin), thereby restoring intestinal barrier integrity in mice with UC. Moreover, BA treatment effectively suppressed the abnormal expansion of opportunistic pathogens (Erysipelotrichaceae, Saccharimonadaceae, Escherichia-Shigella, Turicibacter, Ruminococcus and Candidatus Saccharimonas) while significantly promoting the proliferation of the potential probiotic Akkermansia. Spearman correlation analysis revealed that the abundance of Akkermansia was negatively correlated with p65 transcriptional activity in the NF-κB pathway but positively correlated with anti-inflammatory cytokine IL-10 and the mRNA levels of barrier proteins (ZO-1 and occludin). In conclusion, these findings indicated that BA alleviates UC through a synergistic mechanism encompassing NF-κB pathway inhibition, microbiota homeostasis restoration and intestinal barrier repair. This discovery offers a theoretical basis for novel functional foods leveraging terpenoids to restore gut microecological balance.

Combinations of phytochemical(s) and engineered nanoparticles have attracted immense research interest due to their superior antimicrobial effects against contaminations. Herein, a Pickering emulsion is developed with capsulized phytochemicals including borneol and citral (BC–Cap) stabilized by hydrophilic amine-functionalized silica nanoparticles (SiO 2 ─NH 2 NPs). The droplet sizes of Pickering emulsion were 5.2 ± 1.4 µm under the condition that the concentrations of SiO 2 ─NH 2 NPs ranged from 0.6 to 1.2 wt.%, and the emulsion showed desirable stability during storage at 40°C for 365 days. In addition, the antibacterial and antibiofilm activities of the Pickering emulsion were investigated. The antibacterial effect of BC–Cap increased by two- to fourfold compared with citral or borneol alone. Treatment of BC/BC–Cap for 4 h eliminated the formation of biofilms generated by Listeria monocytogenes (at 5/1.25 mg/ml; 2 × MIC concentration) and Pseudomonas aeruginosa (at 5/2.5 mg/ml; 2 × MIC concentration). Further mechanistic studies revealed that the antibiofilm effects of BC–Cap were attributed to its ability to increase the porosity and lytic effects on the cell membrane of bacteria. Findings from the current study support the antibacterial and antibiofilm effects of BC–Cap Pickering emulsion as a promising food additive. Practical Application The Pickering emulsion has potential applications as bacteriostatic agent in packaging materials and general surface disinfectant. The combination of borneol and citral is stabilized by hydrophilic amine-functionalized silica nanoparticles (SiO 2 ─NH 2 NPs). With the synergistic effects of borneol and citral, the Pickering emulsion shows a promising elimination effect against the formation of biofilms produced by Listeria monocytogenes and Pseudomonas aeruginosa .

Urolithin A (UA), a metabolite of ellagitannins produced by gut microbiota, exhibits a range of beneficial biological activities, particularly its ability to promote mitophagy, indicating its potential for treating ischemic stroke. However, its therapeutic efficacy is limited by poor solubility. In this study, we developed triphenylphosphonium (TPP)-modified UA nanoparticles, utilizing DSPE-PEG as a stabilizer and co-loading borneol (BO) as a permeation enhancer. The resultant UA-BO-TPP-NPs exhibited an average particle size of 172.2 nm, a zeta potential of −2.24 mV, and a substantial drug payload of 39.52%. UA-BO-TPP-NPs demonstrated commendable stability in diverse physiological media and during storage. In vitro experiments showed that UA-BO-TPP-NPs significantly enhanced cellular uptake and achieved high mitochondrial co-localization under both normal conditions and oxygen-glucose deprivation/reoxygenation (OGD/R) conditions. UA-BO-TPP-NPs markedly reduced intracellular reactive oxygen species (ROS) and malondialdehyde (MDA) levels while increasing adenosine triphosphate (ATP) content in human brain microvascular endothelial cells (HBMEC) and SH-SY5Y cells. Furthermore, intravenously injected UA-BO-TPP-NPs effectively accumulated in the brains of rats after cerebral ischemia–reperfusion (I/R) compared to conventional UA-NPs. Consistently, UA-BO-TPP-NPs significantly reduced brain infarction, increased survival rates, preserved blood–brain barrier (BBB) integrity, inhibited oxidative stress, and ameliorated neurological function in cerebral I/R rats. In summary, UA-BO-TPP-NPs effectively delivered encapsulated UA to mitochondria and demonstrated superior therapeutic efficacy in cerebral I/R rats, highlighting the potential for treating ischemic stroke.
