Low aqueous dispersibility and poor stability severely restrict the bioaccessibility of curcumin in health-related products. Constructing highly effective delivery carriers is essential to overcome the limitation. In this study, a novel and simple self-assembly process driven by electrostatic interaction was developed to prepare cyclodextrin succinate/chitosan (SACD/CS) nanoparticles. Spherical SACD/CS nanoparticles were obtained by adjusting the ratio of SACD:CS to obtain an average size and ζ-potential of around 500 nm and 20 mV, respectively. By analyzing the intermolecular interactions and crystalline structure of SACD/CS nanoparticles, it is indicated that SACD could act as crosslinking nodes, and contributed to transformation of molecular arrangement in the nanoparticles. The noncytotoxic SACD/CS nanoparticles were further used as curcumin carriers and showed significant improvement on curcumin stability, with the retention index of curcumin under physiological conditions increased from 19% to 62%. Moreover, the Cur-SACD/CS nanoparticles displayed controlled release behavior of curcumin during in vitro digestion, with 9%, 22%, and 44% of curcumin released at the end of the oral, stomach, and small intestine stage, respectively. This study developed a novel and effective strategy for oral delivery of curcumin, showing promising potential for improving the bioaccessibility and bioavailability of bioactive compounds like curcumin.

Conventional chemotherapy is challenged by its inherent limitations. As an emerging drug delivery system, nanoparticles are easily eliminated by the immune system due to their exogenous nature. Moreover, the lack of long-circulation and tumor-targeting greatly limits their application. In our research, to address this problem, the biomimetic nanocarriers combine cell membranes, targeting ligand and nanoparticles into one system. Firstly, an active ester monomer and three terpolymers PMAN containing different proportions of phosphorylcholine, sulfonic acid and active ester group were successfully synthesized. Subsequently, aminated-folate was synthesized and grafted onto the terpolymers to prepare the folate-targeted polymers FA-PMAN. Finally, cell membrane biomimetic and folate-targeted chitosan nanoparticles (FA-PMAN/CS) with average particle size of 100 to 200 nm were prepared through electrostatic interactions. The in vivo blood circulation results showed that the long-circulating properties of PMAN/CS nanoparticles were significantly enhanced, compared with TPP/CS nanoparticle as a control. The half-life extended from 2.71 h to 12.95 h, with five-fold increase. Additionally, the uptake efficiency of FA-PMAN/CS nanoparticles by rat breast cancer cells (MADB-106) was 5–6 times greater than that of nanoparticles without folate. The biological distribution in mice demonstrated that the signal intensity in tumors treated with FA-PMAN/CS nanoparticle was substantially higher than in other organs, indicating that folate on the surface of nanoparticles increased the uptake into cancer cells. This work aims to design more efficient nanocarriers through biomimetic simulation of cell membranes, reduce recognition and clearance by the immune system, prolong blood circulation time and achieve tumor-targeted drug delivery. It is expected to significantly improve efficacy and provide an innovative strategy for clinical treatment.

Wound infections are a growing public health issue, worsened by drug-resistant strains. Quercetin (Que) has shown anti-inflammatory, antioxidant, and antimicrobial properties, but its limited bioavailability hinders therapeutic use. This study introduces a multifunctional self-assembly nanoplatform, QHCNPs, encapsulating quercetin with hordein/chitosan to enhance stability and bioavailability. Transmission electron microscopy and particle size analysis revealed that QHCNPs are spherical structures with a diameter of 435.5 ± 2.9 nm and a Zeta potential of +11.0 mV. QHCNPs demonstrated excellent stability, low cytotoxicity, and MIC values of 512 ppm against Staphylococcus aureus ( S. aureus ) and 256 ppm against methicillin-resistant Staphylococcus aureus (MRSA) . In a bacterial wound model, QHCNPs outperformed quercetin alone by accelerating wound healing, eliminating bacteria, reducing inflammatory markers, scavenging reactive oxygen species (ROS), and promoting collagen and blood vessel regeneration. These results establish QHCNPs as a promising non-antibiotic therapy for treating drug-resistant wound infections, supporting further exploration for clinical applications.

Estrone-modified glycol chitosan nanoparticles (GCNP-ES) based on the mechanisms of ES-mediated endocytosis and intracellular pH-responsive drug release were developed for the treatment of breast cancer. GCNP-ES were prepared by grafting copolymerization of glycol chitosan with 2-(diisopropylamino)ethyl methacrylate to generate GCNP prior to ES conjugation. The particle size, zeta potential , and paclitaxel (PTX) encapsulation efficiency of GCNP-ES were characterized. In particular, GCNP-ES exhibited pH-responsive dissociation properties while maintaining stability under long-term storage and lyophilization. The drug release of PTX-loaded GCNP-ES (PTX/GNCP-ES) was modestly prolonged with considerable pH sensitivity. GCNP-ES promoted internalization in breast cancer MCF-7 cells by approximately 5-fold as compared to GCNP, and the internalized GCNP-ES was mainly localized in the endosomes of MCF-7 cells. PTX/GNCP-ES exhibited higher cytotoxicity and cell apoptosis ratio than GCNP. In mice with MCF-7 breast cancer xenograft, PTX/GCNP-ES showed higher accumulation at the tumor site, which resulted in a higher tumor inhibition ratio (81.4%) than that achieved by PTX/GCNP (69.4%) and PTX solution (48.8%). Furthermore, no histological and hematological toxicity was detected in in vivo studies of PTX/GCNP-ES. Overall, these results suggested the potential applicability of GCNP-ES as a drug delivery system for breast cancer therapy. Statement of Significance Most breast cancers are hormone dependent. Herein, we developed a estrone-modified glycol chitosan nanoparticles (GCNP-ES) as a drug delivery system to overcome the drawbacks of chemotherpeutic drugs, including poor water solubility and lack of specifity. GCNP-ES could provide efficient drug delivery in breast cancer cells. The study demonstrated that GCNP-ES could dissociate under mildly acidic conditions, leading to the timely payload release of the drug in target tumor cells following internalization. The conjugated estrone of the nanoparticles could significantly increase drug accumulation in the tumor site and result in enhanced therapeutic effect. Thus, the potential applicability of GCNP-ES was suggested.

In order to achieve high loading and efficient delivery of curcumin, phenylboronic acid-conjugated chitosan nanoparticles were prepared by a simple desolvation method. These nanoparticles exhibited a regular spherical shape with the average size about 200−230 nm and narrow size distribution, which were kinetically stable under physiological condition. Due to boronate ester formation between curcumin and phenylboronic acid groups in the nanoparticles, and the hydrogen bonding interactions between curcumin and nanocarriers, curcumin was successfully loaded into the nanoparticles with high drug loading content. These curcumin-loaded nanoparticles showed pH and reactive oxygen species (ROS)-triggered drug release behavior. In vitro cell experiments revealed that the blank nanoparticles were completely nontoxic to cultured cells, and the curcumin-loaded nanoparticles exhibited efficient antitumor efficiency against cancer cells. Moreover, the drug-loaded nanoparticles performed an enhanced growth inhibition in three-dimensional multicellular tumor spheroids. Thus, these nanocarriers would be a promising candidate for curcumin delivery in tumor treatment.

As a natural preservative, cinnamaldehyde (CA) has a bright application prospect. However, CA is sensitive to light and can be easily oxidized. What's more, its water solubility is poor. These properties greatly limit its application. In this study, CA/sodium alginate (SA)/chitosan (CS) nanoparticles (CA/SA/CS NPs) were prepared by using SA and CS as wall materials, and CA as core material. The single factor experiment showed that the optimal CA/SA/CS NPs can be obtained when the mass ratio of SA to CS was 5.5/1, SA to CaCl 2 was 5/1, SA to CA was 1/2, and the pH of CS solution was 5.0. Under this condition, the average size of the prepared nanoparticles was 455.2 nm, polymer dispersity index (PDI) value was 0.189, zeta potential was −40.8 mV, and the encapsulation efficiency was 72.92%. The scanning electron microscopy (SEM) and atomic force microscopy (AFM) images confirmed the formation of nanoparticles, which were spherical or ellipsoidal in shape. The fourier transform infrared spectroscopy (FTIR) and multifunctional polycrystalline X-ray diffraction (XRD) spectra indicated that CA was encapsulated in the nanoparticles. Furthermore, in vitro release experiments showed that CA/SA/CS NPs had the function of sustained release. In summary, SA/CS NPs can be used as a promising carrier for CA.

The combination of controlled release technology and targeted drug delivery has become a promising strategy for cancer therapy. In this study, cell-nanoparticle hybrid vector was constructed using mesenchymal stem cells as the targeting cellular carrier and biotinylated chitosan polymer nanoparticles as the drug depot. Drug-loaded nanoparticles (hydrodynamic size =377.0 ± 14.6 nm and zeta potential = 9.6 ± 1.9 mV) were prepared by encapsulating hydrophobic model drug curcumin into biotinylated chitosan polymer. The biotin-modified nanoparticles were anchored on biotinylated mesenchymal stem cells surface by biotin-avidin binding, achieving an upload of 54.73 ± 3.95 pg/cell. The anchorage of nanoparticles on mesenchymal stem cells had no effect on their viability and homing property. Biotin-avidin binding lasted over 48 h, which could be sufficient for cell-directed tumor-tropic delivery. The in vitro and in vivo anti-tumor results advocate that cell-nanoparticle hybrid vector could prove beneficial in pulmonary melanoma metastasis therapy.

Docetaxel (DTX) was effective in the treatment of neoplasm but could only be administered intravenously with the poor oral bioavailability owing to its undesirable solubility, remarkably metabolic conversion, and other factors. Cimetidine (CMD), a classic CYP3A4 isozyme inhibitor, had exhibited a wide range of inhibition on the metabolism of many drugs. The aim of this study was to construct the novel docetaxel-cimetidine (DTX-CMD) complex and the chitosan-deoxycholate nanoparticles based on it to confirm whether this formulation could show advantages in terms of solubility, dissolution rate, small intestinal absorption, and oral bioavailability in comparison with the pure drug. The solid-state characterization was carried out by powder X-ray diffraction (PXRD), Fourier transform infrared spectroscopy (FT-IR), and simultaneous DSC-TGA (SDT). Dissolution rate and kinetic solubility study were determined by evaluating the amount of DTX in distilled water and phosphate buffer solution (pH = 7.4), respectively. And small intestinal absorption and pharmacokinetics study were conducted in rats. The results of this study demonstrated that we successfully constructed DTX-CMD complex and its chitosan-deoxycholate nanoparticles. Furthermore, the DTX-CMD complex increased the solubility of DTX by 2.3-fold and 2.1-fold in distilled water and phosphate buffer solution, respectively. The ultimate accumulative amount of DTX-CMD complex nanoparticles through rat small intestinal in 2 h was approximately 4.9-fold and the oral bioavailability of the novel nanoparticles was enhanced 2.8-fold, compared with the pure DTX. The superior properties of the complex nanoparticles could both improve oral bioavailability and provide much more feasibility for other formulations of DTX.

The extensive development and application of engineered nanoparticles (NPs) in various fields worldwide have been subjected to increasing concern due to their potential hazards to human health and the environment. Therefore, a simple, economical, and effective method for suppressing the toxicity of metal-based nanomaterials is needed. In this study, glutaraldehyde-crosslinked chitosan nanoparticles (CS NPs) were prepared and further modified with lysine (Ly-CS), glutamic acid (Glu-CS), or sodium borohydride reduction (R-CS), and used to suppress cytotoxicity induced by copper oxide NPs (CuO NPs) through chelation with intracellularly released copper ions. All three kinds of CS NPs had similar sizes of ∼100 nm in a dry state and ∼200 nm in cell culture medium, as determined by scanning electron microscopy, transmission electron microscopy, and dynamic light scattering. The chelating efficiency of different CS NPs followed the order Ly-CS > Glu-CS > R-CS. The CS NPs showed minimal or no toxicity to three different cell lines (HepG2, A549, and RAW264.7 cells) at 100 μg mL−1 with similar cell internalization and exocytosis processes. Comparatively, RAW264.7 cells exhibited higher endocytosis and exocytosis rates, as revealed by flow cytometry and confocal laser scanning microscopy. CS NPs were found as agglomerates inside A549 cells and RAW264.7 cells, with the amount of agglomerates inside RAW264.7 cells decreasing significantly with prolonged incubation. All three CS NPs, especially Ly-CS and Glu-CS NPs, efficiently suppressed the cytotoxicity induced by CuO NPs, and reduced the intracellular level of reactive oxygen species.

In this paper, we report a simple approach to fabricate diselenide-crosslinked carboxymethyl chitosan nanoparticles (DSe-CMC NPs) for doxorubicin (DOX) delivery, with disulfide analogs (DS-CMC NPs) as control. DS-CMC NPs and DSe-CMC NPs featured a spherical morphology and narrow size distribution with the average size about 200 nm. Carboxymethyl chitosan (CMC) as the starting material not only improved the biocompatibility of the nanocarriers but also enhanced physiological stability. Due to electrostatic interactions between DOX and CMC, the nanoparticles had high drug encapsulation efficiency (∼25 %). The nanoparticles disintegration and drug release were accelerated by the cleavage of diselenide bonds through oxidation by H 2 O 2 or reduction by GSH. In vitro cell experiments revealed that DOX-loaded DSe-CMC NPs possessed the highest drug accumulation and cytotoxicity in tumor cells. Moreover, DOX-loaded DSe-CMC NPs performed the enhanced growth inhibition in vivo than that of DS-CMC NPs. Thus, the diselenide-crosslinked nanoparticles possess great potentials for DOX delivery.

Natural biopolymer-based nanoparticles are promising candidates for emulsion stabilization and delivery of hydrophobic compounds. In this study, tea protein (TP) and chitosan (CS) were employed to fabricate complex nanoparticles (NPs) using a pH-dependent method. At a mass ratio of 2:1 (TP:CS), the NPs exhibited favorable properties (206.62 nm, −30.86 mV, 85.41° contact angle) owing to electrostatic interactions, hydrogen bonding, and hydrophobic interactions (binding energy of the complex was −5.40 kcal/mol). At a particle concentration of 2 % and an oil-phase volume fraction of 60 %, the Pickering emulsion stabilized by TP-CS 2:1 NPs remained physically stable. Compared with TP alone, TP-CS 2:1 NPs significantly enhanced the encapsulation efficiency and antioxidant capacity (DPPH/ABTS) of curcumin (Cur). In vitro digestion confirmed that the emulsions reduced gastric degradation of Cur, increasing its bioaccessibility by 17.4 %. Accordingly, this study provides a strategy for designing natural protein-polysaccharide NPs for functional emulsions and sustained delivery systems.
