Curcumin in Pectin-Caseinate Enhances Resveratrol Bioavailability

Yijun Zhou,Yanan Song,Ling Zhu,Hui Zhang

Resveratrol and curcumin, natural plant-derived polyphenols with antioxidant, anti-inflammatory, and anticancer properties, are limited by poor bioavailability and stability. Delivery systems offer an effective strategy to address these limitations. Notably, their co-encapsulation shows synergistic effects, significantly enhancing encapsulation efficiency, stability, and bioactivity. In this study, nanoparticles were prepared from pectin and sodium caseinate to deliver resveratrol and curcumin using a solvent evaporation method combined with electrostatic deposition, and their physicochemical and structural properties were characterized. The nanoparticles (PE-SC-C-RNP) had a narrow size distribution (247.57 nm), low polydispersity index (0.32), and high encapsulation efficiency (95.24 ± 0.76%). Structural analyses indicated that electrostatic interactions between pectin and sodium caseinate formed the nanoparticle outer layer, with resveratrol and curcumin in an amorphous state. In vitro digestion experiments showed that curcumin enhanced the binding of resveratrol to carriers without affecting the release rate of resveratrol or free amino acids, leading to a significant increase in antioxidant activity. Animal model experiments showed that the resveratrol concentrations were greater in the plasma of mice administered PE-SC-C-RNP than those administered nanoparticles encapsulating only resveratrol orally, with a 0.83-1.27-fold increase in the maximal tissue concentration, and better pharmacokinetics. The maximum tissue concentration of resveratrol increased 0.83–1.27-fold in the liver, kidney, and other tissues within 6.0 h of administration. These results demonstrated that pectin sodium caseinate nanoparticles and co-encapsulated curcumin effectively improved the bioavailability of resveratrol for the delivery of bioactive compounds.

Photodynamic inactivation of Shigella flexneri by curcumin

Zuxin Liang,Xuling Liu,Zhiran Qin,Jingshu Li,Jianhai Yu,Xiaoen He,Li Zhu,Hongying Fan,Yushan Hu,Jinxiu Yao,Chenguang Shen,Chengsong Wan,Bao Zhang,Wei Zhao

This study aimed to inactivate Shigella flexneri using curcumin-based photosensitization and determine the sensitivity of S. flexneri to the combination of epsilon (ε)-polylysine (ε-PL) and curcumin-based photosensitization for decontamination. Bacterial numbers were reduced by 4.65 log CFU/mL after incubation with 80 μM curcumin and illumination for 20 min. Photosensitization resulted in cell morphology changes and increased cell permeability and protein degradation. Oxidative stress gene ( oxyR ) and efflux pump genes ( marA , acrA ) were upregulated. Decontamination of S. flexneri after incubation with curcumin and ε-PL for 1 h followed by photoactivation for 60 s has been investigated. It was found that the growth of bacteria was completely inhibited. Additionally, the number of bacteria was reduced by > 6 log CFU/mL. Experiments on the uptake of curcumin molecules by bacteria showed that ε-PL promoted the penetration of bacterial cells by curcumin, thus enhancing the effects of photosensitization.

Improving curcumin solubility and bioavailability by encapsulation in saponin-coated curcumin nanoparticles prepared using a simple pH-driven loading method

Shengfeng Peng,Ziling Li,Liqiang Zou,Wei Liu,Chengmei Liu,David Julian McClements

Curcumin is a bioactive phytochemical that can be utilized as a nutraceutical or pharmaceutical in functional foods, supplements, and medicines. However, the application of curcumin as a nutraceutical in commercial food and beverage products is currently limited by its low water-solubility, chemical instability, and poor oral bioavailability. In this study, all-natural colloidal delivery systems were developed to overcome these challenges, which consisted of saponin-coated curcumin nanoparticles formed using a pH-driven loading method. The physicochemical and structural properties of the curcumin nanoparticles formed using this process were characterized, including particle size distribution, surface potential, morphology, encapsulation efficiency, and loading capacity. Fourier transform infrared spectroscopy and X-ray diffraction indicated that curcumin was present in the nanoparticles in an amorphous form. The curcumin nanoparticles were unstable to aggregation at low pH values (<3) and high NaCl concentrations (>200 mM), which was attributed to a reduction in electrostatic repulsion between them. However, they were stable at higher pH values (3 to 8) and lower NaCl levels (0 to 200 mM), due to a stronger electrostatic repulsion between them. They also exhibited good stability during refrigerated storage (4 °C) or after conversion into a powdered form (lyophilized). A simulated gastrointestinal tract study demonstrated that the in vitro bioaccessibility was around 3.3-fold higher for curcumin nanoparticles than for free curcumin. Furthermore, oral administration to Sprague Dawley rats indicated that the in vivo bioavailability was around 8.9-fold higher for curcumin nanoparticles than for free curcumin. These results have important implications for the development of curcumin-enriched functional foods, supplements, and drugs.

Curcumin-involved self-healing hydrogels as flexible sensors

Xinyu Wei,Zeng Liu,Hongli Fang,Zhiyue Cui,Shu He,Wei Shao

The rapid development of conductive hydrogels shows great potentials in the field of flexible sensors. Accordingly, the research on the hydrogel sensors with great self-healing property for long-term durability is noticeably essential. In the present study, curcumin (Cur)-involved polyacrylic acid (PAA) and polyethyleneimine (PEI) based hydrogels noted as PAA/PEI-Al-Cur were constructed. The effect of curcumin on the mechanical properties of the hydrogels was firstly studied. The introduction of curcumin could effectively improve the mechanical properties that the tensile strength of the prepared PAA/PEI-Al-Cur hydrogel could reach as high as 119.0 kPa. The existence of Al 3+ and the porous network structure of the hydrogel entrust to a high conductivity (σ=1.11 S/m). Thus, the hydrogel could be applied as both strain sensor and pressure sensor to detect different deformations and various human activities including large-scale human movements and subtle facial expressions. It is worth noting that the self-healing ability of the hydrogels could be regulated by the amount of introduced curcumin that the optimal dose exhibited ultrafast self-healing ability and the mechanism was illustrated. More to the point, the healed hydrogel could be applied as the sensor to repeatedly and stably perform sensing behavior as the original one. This work provides an effective construction strategy to prepare novel conductive hydrogels for flexible sensors.

Electrospun curcumin-loaded fibers with potential biomedical applications

Xiao-Zhu Sun,Gareth R. Williams,Xiao-Xiao Hou,Li-Min Zhu

Polyvinyl alcohol (PVA) nanofibers loaded with curcumin or its β-cyclodextrin (CD) inclusion complex were successfully prepared using an electrospinning process. The influence of curcumin or CD–curcumin complex content on fiber formation and quality was investigated. X-ray diffraction and differential scanning calorimetry analyses of the fibers, together with electron microscope evidence, demonstrated that curcumin is likely to be present as crystalline aggregates in the fibers, while its CD complex is more evenly distributed. 1 H NMR analysis indicated that the chemical structure of curcumin was preserved during the electrospinning process. Thermogravimetric analysis demonstrated that inclusion into nanofibers enhanced the thermal stability of curcumin. In vitro dissolution tests showed that the drug release profiles of the PVA/curcumin and PVA/complex fibers were different, with release from the latter occurring more rapidly. Release from both fiber types was found to be largely governed by a diffusion-controlled mechanism; two sequential stages for drug release were observed.

Cellulose–halloysite nanotube composite hydrogels for curcumin delivery

Huang Biao,Liu Mingxian,Zhou Changren

Halloysite nanotubes (HNTs) were added to cellulose NaOH/urea solution to prepare composite hydrogels using epichlorhydrine crosslinking at an elevated temperature. The shear viscosity, mechanical properties, microstructure, swelling properties, cytocompatibility, and drug delivery behavior of the cellulose/HNT composite hydrogels were investigated. The viscosity of the composite solution increases with the addition of HNT. The compressive mechanical properties of composite hydrogels are significantly improved compared with pure cellulose hydrogel. The compressive strength of the composite hydrogels with 66.7% HNTs is 128 kPa, while that of pure cellulose hydrogel is only 29.8 kPa in compressive strength. Rheological measurement suggests the resistance to deformation is improved for composite hydrogels. X-ray diffraction and Fourier transform infrared spectroscopy show that the crystal structure and chemical structure of HNT are not changed in the composite hydrogels. Hydrogen bonding interactions between HNT and cellulose exist in the composites. A porous structure of the composite hydrogels with pore size of 200–400 μm was found by scanning electron microscopy. The addition of HNT leads to decreased swelling ratios in NaCl solution and pure water for the composite hydrogels. Cytotoxicity assays show that the cellulose/HNT composite hydrogels have a good biocompatibility with MC3T3-E1 cells and MCF-7 cells. Curcumin is further loaded into the composite hydrogel via physical adsorption. The curcumin-loaded composite hydrogels show a strong inhibition effect on the cancer cells. All the results illustrate that the cellulose/HNT composite hydrogels have promising applications such as anticancer drug delivery systems and anti-inflammatory wound dressings.

Curcumin-functionalized silk biomaterials for anti-aging utility

Lei Yang,Zhaozhu Zheng,Cheng Qian,Jianbing Wu,Yawen Liu,Shaozhe Guo,Gang Li,Meng Liu,Xiaoqin Wang,David L. Kaplan

Curcumin is a natural antioxidant that is isolated from turmeric ( Curcuma longa ) and exhibits strong free radical scavenging activity, thus functional for anti-aging. However, poor stability and low solubility of curcumin in aqueous conditions limit its biomedical applications. Previous studies have shown that the anti-oxidation activity of curcumin embedded in silk fibroin films could be well preserved, resulting in the promoted adipogenesis from human mesenchymal stem cells (hMSCs) cultured on the surface of the films. In the present study, curcumin was encapsulated in both silk fibroin films (silk/cur films) and nanoparticles (silk/cur NPs), and their anti-aging effects were compared with free curcumin in solution, with an aim to elucidate the mechanism of anti-aging of silk-associated curcumin and to better serve biomedical applications in the future. The morphology and structure of silk/cur film and silk/cur NP were characterized using SEM, FTIR and DSC , indicating characteristic stable beta-sheet structure formation in the materials. Strong binding of curcumin molecules to the beta-sheet domains of silk fibroin resulted in the slow release of curcumin with well-preserved activity from the materials. For cell aging studies, rat bone marrow mesenchymal stem cells (rBMSCs) were cultured in the presence of free curcumin (FC), silk/cur film and silk/cur NP, and cell proliferation and markers of aging (P53, P16, HSP70 gene expression and β-Galactosidase activity) were examined. The results indicated that cell aging was retarded in all FC, silk/cur NP and silk/cur film samples, with the silk-associated curcumin superior to the FC.

Inhaled curcumin mesoporous polydopamine nanoparticles against radiation pneumonitis

Ting Chen,Bo Zhuang,Yueqi Huang,Yan Liu,Bochuan Yuan,Wanmei Wang,Tianyu Yuan,Lina Du,Yiguang Jin

Radiation therapy is an effective method to kill cancer cells and shrink tumors using high-energy X-ray or γ -ray. Radiation pneumonitis (RP) is one of the most serious complications of radiation therapy for thoracic cancers, commonly leading to serious respiratory distress and poor prognosis. Here, we prepared curcumin-loaded mesoporous polydopamine nanoparticles (CMPN) for prevention and treatment of RP by pulmonary delivery. Mesoporous polydopamine nanoparticles (MPDA) were successfully synthesized with an emulsion-induced interface polymerization method and curcumin was loaded in MPDA via π‒π stacking and hydrogen bonding interaction. MPDA owned the uniform spherical morphology with numerous mesopores that disappeared after loading curcumin. More than 80% curcumin released from CMPN in 6 h and mesopores recovered. CMPN remarkably protected BEAS-2B cells from γ -ray radiation injury by inhibiting apoptosis. RP rat models were established after a single dose of 15 Gy 60 Co γ -ray radiation was performed on the chest area. Effective therapy of RP was achieved by intratracheal administration of CMPN due to free radical scavenging and anti-oxidation ability, and reduced proinflammatory cytokines, high superoxide dismutase, decreased malondialdehyde, and alleviated lung tissue damages were observed. Inhaled CMPN paves a new avenue for the treatment of RP.

Curcumin Attenuates Colistin-Induced Peripheral Neurotoxicity in Mice

Chongshan Dai,Xilong Xiao,Yuan Zhang,Biao Xiang,Daniel Hoyer,Jianzhong Shen,Tony Velkov,Shusheng Tang

Peripheral neurotoxicity often occurs in patients receiving parenteral polymyxin therapy (i.e., colistin methanesulfonate or polymyxin B). The present study aimed to investigate the protective effect of curcumin on colistin-induced peripheral neurotoxicity using a murine model. Female C57BL/6 mice (n = 10 in each group) were randomly divided into the following: (1) control group (saline), (2) curcumin only group (200 mg/kg/day; orally), (3) colistin only group (18 mg/kg/day; i.p.), (4) colistin (18 mg/kg/day) plus curcumin 50 mg/kg/day group, (5) colistin (18 mg/kg/day) plus curcumin 100 mg/kg/day group, (6) colistin (18 mg/kg/day) plus curcumin 200 mg/kg/day group; all mice were treated for 7 days. Orally applied curcumin was detected in the brain, cerebellum, and sciatic nerve. Co-administration of oral curcumin markedly improved colistin-induced impaired sensory and motor dysfunctions in a dose-dependent manner. Curcumin supplementation at 100 and 200 mg/kg significantly decreased lipid peroxidation and upregulated catalase (CAT) and superoxide dismutase (SOD) activities, ATP levels, and Na+/K+-ATPase activity in sciatic nerve tissue, compared to the colistin alone group. Curcumin supplementation at 200 mg/kg upregulated the levels of AKT, NGF, mTOR, Nrf2, and HO-1 mRNA and concomitantly downregulated Bax, caspases-3, and -9 mRNA; it also decreased caspase-3 and caspase-9 activity. In summary, for the first time, our study reveals that the protective effect of oral curcumin on colistin induced peripheral neurotoxicity is associated with the activation of NGF/Akt and Nrf2/HO-1 pathways and inhibition of oxidative stress. This study highlights the potential clinical application of curcumin as an oral neuroprotective agent coadministered during colistin therapy.

Curcumin-loaded strontium-based metal-organic framework hydrogel microspheres for bone regeneration: Combined effects of curcumin-mediated immunomodulation and Sr2+-induced osteogenesis

Jianqi Xiong,Hang Zheng,Yijiu Zhang,Zefeng Xie,Zheng Xiang,Tao Sun,Zhigang Zhou,Zhikun Li,Zhuoying Cai,Sailang Shi,Yanming Liu

A multifunctional hydrogel microsphere system with antioxidant, immunomodulatory, and osteogenic functions was developed. • The Sr-MOF enabled efficient curcumin loading for ROS scavenging and macrophage polarization regulation. • Microfluidics encapsulated Cur-loaded Sr-MOF into GelMA microspheres for sustained release of Cur and Sr 2+ . • Cur@Sr-MOF@GelMA microspheres promoted osteogenesis by establishing a favorable immune microenvironment. Bone regeneration in large bone defects is often impeded by an unfavorable microenvironment characterized by excessive reactive oxygen species (ROS), dysregulated macrophage polarization, and poor osteogenic capacity. Therefore, it was hypothesized that a bioactive material targeting these disadvantages is promising in bone defect repair. In the study, we synthesized a hydrogel microsphere system (Cur@Sr-MOF@GelMA), in which the polyphenolic compound curcumin (Cur) was adsorbed onto a strontium-based metal - organic framework (Sr-MOF) and subsequently encapsulated within gelatin methacryloyl (GelMA) microspheres. The Cur@Sr-MOF@GelMA microspheres enabled the sustained release of Cur and Sr 2+ . Cur effectively scavenged ROS and induced M2 polarization of macrophages (RAW264.7 cells), establishing a favorable microenvironment for bone regeneration. Furthermore, Cur@Sr-MOF@GelMA microspheres demonstrated a dual osteogenic effect on MC3T3-E1 cells, directly via Sr 2+ and indirectly through Cur-mediated immunomodulation of RAW264.7 cells. Transcriptomic sequencing analysis indicated that Cur@Sr-MOF@GelMA microspheres might enhance M2 polarization of RAW264.7 cells through suppression of pro-inflammatory pathways (NLR, TLR and TNF signaling), and promote osteogenic differentiation of MC3T3-E1 cells via upregulation of the Wnt signaling pathway. In vivo experiments in rats demonstrated that Cur@Sr-MOF@GelMA microspheres established an M2 macrophage-dominant microenvironment at early stages, accelerated bone formation and eventually resulted in the highest bone volume fraction (BV/TV) in critical-sized calvarial defects at 12 weeks. In conclusion, the Cur@Sr-MOF@GelMA microsphere system possesses antioxidant, immunomodulatory, and osteoinductive properties, and represents a promising material for bone defect repair.

Genipin-enriched chitosan-Zein nanoparticles for improved curcumin encapsulation

Lingling Sha,Husnain Raza,Chengsheng Jia,Imran Mahmood Khan,Huqing Yang,Gang Chen

Polysaccharide-protein nanocomplexes are considered one of the desired systems to encapsulate bioactive compounds. The study prepared chitosan (CS)/genipin (GP)/zein nanoparticles with chemical cross-linkage for encapsulating and releasing curcumin. Fourier transform infrared spectra demonstrated that cross-linkage between molecules was attributed to electrostatic interactions and the formation of amido bonds and hydrogen bonding. Transmission electron microscopy showed that all prepared nanoparticles showed a spherical morphology. Particularly, cinnamaldehyde screening –NH 2 groups in CS significantly reduced the encapsulation efficiency of nanoparticles, demonstrating the significant role of the –NH 2 group in CS-coated zein nanoparticles. Additionally, the encapsulation efficiency was largely increased to 82.3 % at the ratio of CS/GP/zein ( w /w, 20:1:100), compared with 38.4 % using zein as a single encapsulation for curcumin. In vitro digestion, the addition of GP and CS decreased the release of curcumin from 84.8 % to 57.1 % after the initial digestion of 2 h. The GP/CS/zein particles retained 12 % of curcumin after 6- h of digestion, indicating the improvements in encapsulation efficiency and release properties.

Smart Mesoporous Silica Nanoparticles Loading Curcumin Inhibit Liver Cancer

Shuli Man,Wei Liu,Jingxian Bi,Jingjing Bai,Qiong Wu,Bin Hu,Jing Hu,Long Ma

Curcumin (CUR) as one of the natural edible pigments is approved by the World Health Organization due to its nontoxic and anticancer effect. However, the utility of CUR is restricted due to its low oral bioavailability. Nanoparticle drug delivery systems like mesoporous silica nanoparticles (MSNs) have been extensively used due to their high specific surface area, high loading rate, and ease of modification. This study developed lactobionic acid (LA)-modified carboxymethyl chitosan (CMCS)-coated MSNs to deliver CUR specifically targeting hepatocellular carcinoma. Among these nanoparticles, LA targets liver cancer cells. CMCS utilizes pH-responsive release of CUR. The LA-CMCS-MSN@CUR (MSN@CUR) were evaluated using several methods, including Fourier transform infrared spectroscopy, transmission electron microscopy, and zeta potential measurements. Liver cellular uptake of MSN@CUR depends on a specific LA receptor-mediated endocytosis mechanism. Additionally, MSN@CUR performed with a better antitumor effect than Cur in H22 orthotopic transplantation of liver cancer and H22 solid tumor mouse models. Treatment with MSN@CUR significantly reduced the protein of VEGF, p-PI3K, and AKT, increased the protein of caspases 3 and 8, ultimately inhibited tumor migration, and promoted apoptosis. This study provides a new path for delivery of natural active ingredients with excellent bioavailability in the antitumor field.
