Polysaccharide gels consisting of nanofibers offer considerable promise for biomaterials; however, except for bacterial cellulose, nanofiber gels formed by solely natural polysaccharides are still rare. Here, we present a self-assembled polysaccharide nanofiber gel based on Curdlan, exhibiting high elasticity, toughness, and antineuroinflammatory effects. A gelling method, termed phase separation+annealing (PSA), facilitated the formation of Curdlan hydrogels with a dense network of self-assembling nanofibers (∼20 nm in diameter), structurally analogous to natural β-glucan fibrils in fungi. These hydrogels exhibited exceptional mechanical performance with a tensile strength of up to 2.1 MPa and a fracture strain of 361%. Structural and formation analyses via X-ray diffraction, scanning electron microscopy, differential scanning calorimetry, and molecular dynamics simulations demonstrated that the PSA method enabled the hierarchical organization of Curdlan chains into highly ordered triple helices, forming interconnected nanofibers. Additionally, the Curdlan hydrogels with enhanced nanofiber networks (CD-E, where “CD” denotes Curdlan and “E” refers to the annealing-induced enhanced nanofiber structure) exerted antineuroinflammatory effects in a lipopolysaccharide-induced neuroinflammation model by attenuating nitric oxide production, downregulating pro-inflammatory cytokines, and inhibiting iNOS and COX2 expression, while promoting a neuroprotective microglial phenotype via increased CD206 and decreased CD86 expression. These findings shed light onto Curdlan gels and can expand their applications from food additives to biomaterials.

In this work, carboxymethylated curdlan (CMCD) was utilized as a capping and stabilizing agent for the green synthesis of silver nanoparticles. Subsequently, quaternized curdlan (QCD) was introduced as the second capping layer through electrostatic attraction, leading to the preparation of double-capped silver nanoparticles (AgNPs@CQ). The successful synthesis of silver nanoparticles was characterized using UV–vis, FTIR, XRD, TEM, and DLS. AgNPs@CQ were incorporated into gelatin and a AgNPs@CQ/Gel composite hydrogel was obtained. The incorporation of AgNPs@CQ imparts excellent antibacterial properties to the composite hydrogel, thereby enhancing its antimicrobial efficacy. The presence of double-capping layers significantly retards the release rate of silver, contributing to prolonged antimicrobial activity. The MTT and live/dead fluorescence staining results demonstrate that the gelatin hydrogel incorporating double-capped AgNPs exhibits enhanced cell viability compared to the one incorporating single-capped AgNPs. Additionally, the composite hydrogel exhibits remarkable mechanical strength and adhesive performance. The AgNPs@CQ/Gel composite hydrogel demonstrates a cost-effective and facile preparation, showing significant potential in the field of dressings.

Developing nucleic acid-based tools to control disease-relevant gene expression in human disorders, such as siRNAs, opens up potential opportunities for therapeutics. Because of their high molecular weight and polyanionic nature, synthetic siRNAs fail to cross biological membranes by passive diffusion and therefore, generally require transmembrane siRNA delivery technologies to access the cytoplasm of target cells. To create a biocompatible siRNA delivery agent, we chemically modified natural polysaccharide curdlan derivative 6AC-100 in a regioselective manner to introduce different ratios of imidazole rings in the amino units (denoted as Curimi) and evaluated their siRNA binding ability, cytotoxicity, endosome buffering capacity and siRNA transfection efficiency. The novel curdlan based Curimi polymers formed nanoparticles with siRNA at pH 7.4 in range of 85–105 nm and their size distribution increased along with decreasing pH condition. The zeta potential increased by lowering pH value as well. Curimi polymers showed lower toxicity and higher buffering capacity compared to 6AC-100, and efficiently delivered siRNA against to PLK1 into cancer cells, and subsequently, significantly inhibited target mRNA level. Our result suggested that novel curdlan based Curimi polymers may be used as efficient siRNA carrier for cancer therapy.

Curdlan, a bacteria-derived polysaccharide resource, possesses substantial potential for periodontal antimicrobial delivery. Here, the facile engineering of a functionalized curdlan/polydopamine (PDA) composite hydrogels was reported. The physiochemical evaluations of composite hydrogels proved their tunable properties associated with concentration of PDA including pore size, rheological property and swelling behavior. We have systematically assessed biocompatibility in vitro and found these hydrogels toxicity-free. Moreover, photothermal performance upon near infrared light (NIR) exposure was conducted and eventually indicated the best matches for antibacterial application. The acetate chlorhexidine (CHX) was chosen as a model antimicrobial and the release profiles demonstrated the entrapped CHX could be triggered and nicely controlled by NIR. The optimized bacteriostatic rate reached 99.9 %. Overall, we aimed to provide new curdlan-based hydrogels for periodontal antibacterial treatment by combining photothermal effect and antimicrobial simultaneously.

Curdlan demonstrates significant biological activities and exhibits functional properties such as thickening, water retention, and gelling capabilities. However, the inherent structural simplicity and constrained functionality of pure curdlan-based hydrogels present significant limitations for their utilization in specialized applications requiring multifunctional properties. In this study, curdlan were used as the matrix to prepare multifunctional hydrogels with photothermal conversion, antibacterial and antioxidant capabilities through the addition of tannic acid and iron ions and by employing heating-annealing approach. The molecular interactions between tannic acid and curdlan were elucidated systematically, and the detailed characterization of the hydrogel's functional properties, encompassing its rheological characteristics, thermal stability, swelling kinetics, photothermal conversion capability, and biological activities including antioxidant capacity and antimicrobial efficacy were investigated. The results demonstrated that tannic acid formed stable hydrogen-bonding interactions with curdlan molecules, which significantly affected the structural reorganization of curdlan's triple-helix architecture during renaturation. After introducing the Fe 3 + , the iron-tannic acid chelation complex was served as an effective cross-linking agent for curdlan polymer chains, resulting in enhanced mechanical strength of the composite hydrogel while simultaneously imparting remarkable photothermal conversion efficiency to the material system. And the composite hydrogel demonstrated significant free radical scavenging capacity, potent antibacterial activity, and good cell compatibility. The developed composite hydrogel including superior antioxidant activity, effective antibacterial performance, and efficient photothermal conversion capability, shows the potential as a promising candidate for wound dressing, thereby significantly expanding the potential utilization of curdlan-based biomaterials.

Aminated curdlan derivatives are excellent carriers for delivery of short interfering RNA (siRNA), but their efficiency for mRNA delivery has not been explored. Degradable polymers like poly(β-amino ester) can deliver mRNA due to their biodegradability which is essential for timely release of mRNA cargo upon entering the cell. Herein, we report the synthesis of curdlan derivatives bearing α- or β-amino ester branches for brain targeted mRNA delivery. The β-amino ester functionalized curdlan (denoted by 6AC-MEP) efficiently encapsulated mRNA to form polymeric nanoparticles (PNPs), which readily entered neural cells and initiated the expression of cargo mRNA in vitro . In mouse traumatic brain injury (TBI) model, systemic administration of 6AC-MEP PNPs carrying luciferase mRNA resulted in the strong luminescent signal in the perilesional region of the brain. Two consecutive administrations of 6AC-MEP PNPs loaded with brain-derived neurotrophic factor (BDNF) mRNA resulted in strong expression of BDNF in the ipsilateral hemisphere of TBI mouse brain. The overexpression of BDNF in the brain reduced TBI-induced edema in the brain and improved cognitive function. Taken together, our data suggested 6AC-MEP PNP is a promising carrier for targeted delivery of therapeutic mRNAs.

Background: Patients with melanoma are confronted with postoperative challenges such as tumor relapse, impaired wound healing, infection, and hypertrophic scarring, with chronic wound inflammation potentially increasing the risk of tumor recurrence. Photoresponsive hydrogels have great potential for integrated postsurgical treatment of melanoma by embedding photosensitizers or photothermal agents in porous polymer networks, combining the advantages of hydrogels and phototherapy technologies. Methods: The photoresponsive PCCUR hydrogel was synthesized via a thermal induction method, incorporating polydopamine nanoparticles (PDA-NPs), Chlorella extract (CE), and curdlan (CUR). The physicochemical properties of PCCUR and its effects on skin cells and B16F10 cells were systematically evaluated through in vitro experiments. The antitumor recurrence efficacy of the PCCUR hydrogel was assessed via an incomplete melanoma resection model. Furthermore, the regulatory effects of PCCUR on infected wound healing and its inhibitory effects on hypertrophic scar formation were investigated under temporally controlled laser irradiation. Results: The photoresponsive PCCUR hydrogel possesses temperature sensitivity, injectable properties, optimal mechanical strength, and significant antimicrobial activity and is tailored to mitigate postsurgical issues through healing phase-specific laser irradiation adjustments. During the inflammatory phase, synergistic photodynamic therapy (PDT) and photothermal therapy (PTT) not only eliminate infections but also induce immunogenic cell death (ICD), releasing tumor antigens that bypass tumor heterogeneity and are captured by the in situ hydrogel. The antigen-captured hydrogel acts as an “antigen reservoir” and releases captured antigens to recruit dendritic cells (DCs) to stimulate an antitumor immune response. In the proliferation phase, the inherent properties of the hydrogel inhibit inflammation and facilitate cell proliferation and angiogenesis. The remodeling phase involves low-intensity laser-based phototherapy to modulate myofibroblast activity and collagen remodeling, preventing hypertrophic scarring. Conclusions: The dynamic photoresponsive properties of PCCUR by introducing a 3R paradigm (Remove-Residual tumor cells/bacteria; Remodel-Immune microenvironment; Repair-Skin tissue) provide a safe and efficient therapeutic approach tailored to the three stages of wound healing, making it a promising dressing option for the comprehensive management of postoperative wounds in melanoma patients.

In this study, the effects of grafting chlorogenic acid (CA) on the antioxidant and probiotic activities of curdlan oligosaccharides (CDOS) were investigated. CDOS with degrees of polymerization of 3–6 was first obtained by degradation of curdlan with hydrogen peroxide and then grafted with CA using a free radical-mediated method under an ultrasonication-assisted Fenton system. The thermal stability and antioxidant ability of CDOS were enhanced after grafting with CA. In vitro fermentation, supplementation of CDOS-CA stimulated the proliferation of Prevotella and Faecalibacterium while inhibiting the growth of harmful microbiota. Notably, the concentration of total short-chain fatty acids and the relative abundance of beneficial bacteria markedly increased after fermentation of CDOS-CA, indicating that CA grafting could improve the probiotic activity of CDOS. Overall, the covalent binding of CDOS and CA could enhance the antioxidant and probiotic activities of CDOS, suggesting potential improvements in gastrointestinal and colonic health.

The use of protein-polysaccharide complexes as fat substitutes to reduce fat and maintain fat functionality has received much attention in the food industry. In the present study, complex gels were developed using curdlan (CR) and potato protein (PP) for the potential of development as a solid fat analog. The effect of PP concentrations (1–6%) on the texture, water-holding capacity, freeze-thaw stability, and microstructure of the resultant curdlan-potato protein (CP) gel was investigated. The results showed that the texture properties of the gel with 3% PP were the most similar to those of pork backfat. The addition of PP contents had a pronounced enhancement in the water-holding capacity, and freeze-thaw stability tended to increase and subsequently decrease after 4% PP ( p < 0.05). The low-field NMR confirmed the addition of PP improved the migration and tighter binding of water. The elastic modulus of CP gels increased initially before declining, reaching a maximum at a protein concentration of 3% and a minimum at 5%. The microstructure demonstrated a gradual aggregation and phase separation process of PP and CR, resulting in a strengthened skeleton network. These findings provide a reference for the development of a novel non-animal fat analog in the food industry.

This study presents an engineered approach for sustainable biosynthesis of curdlan by Alcaligenes faecalis using orange peels. To confirm the substrate suitability a four step study was organized. Firstly, drying of substrate was carried within temperature range of 60–120 °C, along with the application of moisture diffusion control model. Secondly, fermentation medium was obtained via saccharification and detoxification, releasing highest sugar at 72.34 g/L with phenolics removal of 95–98%. Thirdly, curdlan fermentation was conducted in detoxified orange peel hydrolysate followed by optimization of batch culture fermentation via kinetic modeling using Logistic and Luedeking-Piret equations. In 5 L bioreactor, highest specific growth rate ( μ m = 0.233/h), highest curdlan production ( P m = 23.24 g/L) and growth associated rate constant ( α = 3.403) were achieved. Moreover, the total sugar consumption and conversion rates were 83.27% and 53.20%. Lastly, characterization techniques such as FTIR, NMR, XRD, TGA, HPGPC and EDS were applied to biosynthesized curdlan for qualitative validation.

Methicillin-resistant Staphylococcus aureus (MRSA) is rapidly spreading worldwide, emerging as a leading cause of bacterial infections in healthcare and community settings. This poses serious risks to human health. The shortage of novel antibiotics and the absence of effective vaccines make MRSA particularly challenging to treat. Existing vaccine development strategies often fail to provide early protection against infections, highlighting the urgent need for solutions. Herein, we propose a novel strategy combining trained immunity with a multi-epitope subunit vaccine to combat MRSA infections. We comprehensively evaluated the trained immune phenotypes induced by β-glucan from barley and curdlan. Macrophages trained with curdlan exhibited a more balanced inflammatory response compared to β-glucan from barley, expressing higher levels of IL-1β , IFN-β , TGF-β , and CCL2 upon secondary stimulation. Furthermore, curdlan-induced trained immunity rapidly provided excellent protection against S. aureus infection in mice. RNA-sequencing analysis revealed that curdlan modulates the Wnt signaling pathway in macrophages, resolves inflammation, and promotes tissue repair. When combined with one or two doses of S. aureus multivalent epitope antigen against MRSA infection, curdlan-induced trained immunity enhanced early protection and promoted recovery. Our study demonstrates the feasibility of combining trained immunity with vaccine protection against MRSA, providing a strategy against multi-drug resistant bacteria.

Curdlan (CUR)-based hydrogels have shown promise for wound healing but suffer from poor flexibility, high brittleness, and limited drug-loading capacity. To overcome these limitations, a curdlan-based complementary hydrogel (CM Gel) was developed by integrating CUR with mangiferin (MF), a polyphenol capable of self-assembly. Non-covalent interactions between CUR and MF led to a stable and flexible network with improved mechanical properties and gelation under mildly acidic pH conditions (pH ∼6.5), which aligns with skin wound conditions. Furthermore, the hydrogel was loaded with dihydromyricetin (DMY), a natural antibacterial agent, using a solvent-free one-pot pH/heating-driven process to fabricate the enhanced comprehensive hydrogel properties D@CM Gel. Multispectral analysis suggested that MF, CUR, and DMY were transformed from the crystalline state to the amorphous state. In vitro studies confirmed its potent antibacterial activity against S. aureus and E. coli , strong antioxidant capacity, and effective biofilm inhibition capacity. In an S. aureus -infected full-thickness wound model, D@CM Gel significantly accelerated wound closure, reduced inflammation (TNF-α, IL-6), promoted collagen deposition and angiogenesis (VEGF, CD31, α-SMA), and outperformed commercial hydrogel dressings with excellent biocompatibility. These findings suggest that D@CM Gel is a promising and easily fabricated wound dressing with synergistic therapeutic functions for treating infected skin wounds.
