Background Aging is a physiological phenomenon in the process of life, and skin aging has a significant impact on human appearance. Therefore, the search for methods to delay skin aging is of great significance for improving the quality of human life. Materials and Methods This study investigated the anti-photoaging effect of Tricholoma matsutake (T) extract composition combined with bakuchiol (B) and ergothioneine (E), and explored its potential mechanism through transcriptome, metabolomics, and network pharmacology. Results 57 main chemical components are identified from the ethanol extract of T. matsutake (T), including D-carnitine (24.55%), α,α-trehalose (15.56%), DL malic acid (8.99%), D-(−)-quinic acid (7.46%), erucamide (7.04%) and so on. After TBE treatment, inflammation of the mice dorsal skin is significantly minimized. Hematoxylin and eosin (H&E) staining and toluidine blue staining reveal that TBE has an anti-inflammatory effect on the back skin tissue of mice. Masson staining shows that TBE has a repair effect on mice dorsal skin tissue. In addition, the inflammatory factors (IL-1β, IL-6, TNF-α) in the mice dorsal skin tissues are significantly reduced but collagen (COL-1) is significantly increased. By cellular immunofluorescence assay, TBE is shown to promote PPAR-α expression in cells. Transcriptomics, metabolomics, and network pharmacology have revealed that TBE can regulate exogenous stimuli and cancer-related signaling pathways to prevent skin aging. Conclusion The results suggest that TBE can be a beneficial supplement to natural anti-aging.

Ergothioneine (ERG) is a potent antioxidant with applications in nutrition, pharmaceuticals, dermatology, and related fields. However, its large-scale production is constrained by low yields and high costs linked to extraction and chemical synthesis. Microbial fermentation offers a promising alternative. In this study, a truncated NcEgt1–1 from Neurospora crassa and CtEgtB from Chloracidobacterium thermophilum were fused to convert L-histidine and L-cysteine into hercynylcysteine sulfoxide, subsequently catalyzed by MsEgtE from Mycobacterium smegmatis to produce ERG. Initial expression in Escherichia coli yielded 87.02 mg/L of ERG. With the use of a rigid linker-EAAAK, the ERG titer increased to 144 mg/L. Further amino acid modifications in NcEgt1–1 resulted in the mutant strain M3–02, producing 325 mg/L of ERG. Fed-batch fermentation in a 5-L bioreactor achieved an ERG yield of 790 mg/L with a productivity of 9 mg/L/h. Iterative mutations of CtEgtB further increased ERG production to 478 mg/L in shake flasks and 790 mg/L in the bioreactor, with productivity rising to 14.9 mg/L/h. This study presents an engineered strain with significant ERG production potential, suitable for industrial application.

This study aims to develop a multifunctional polymer nanovesicle system, AG86-β-glucan-poly(methionine-b-carboxybetaine) (AGPMC), which integrates AG86 peptide (a targeting ligand for the α6β4 integrin on basal keratinocytes) and reactive oxygen species (ROS) responsiveness to enhance the transdermal delivery of ergothioneine (EGT). The objective is to overcome the limitations of hydrophilic drug delivery and provide a targeted, ROS-triggered strategy for treating skin lipofuscin. AGPMC was synthesized via RAFT and ROP polymerization and self-assembles into stable nanovesicles (ANVs). Physicochemical characterization confirmed that the unloaded ANVs have an average size of 106.17 ± 3.26 nm and a polydispersity index (PDI) of 0.261 ± 0.027, with excellent stability and clear ROS-triggered structural responsiveness. After EGT loading, the nanovesicles exhibited a size of 140.10 ± 3.33 nm and a PDI of 0.217 ± 0.008. In vitro studies using HaCaT cells demonstrated a significant enhancement in cellular uptake, with an increase of up to 2.2-fold compared to free FSS ( p < 0.0001), as well as effective ROS scavenging and minimal cytotoxicity. The endocytosis mechanism study revealed that ANVs are primarily taken up via energy-dependent active transport, with caveolae-mediated endocytosis being the main pathway. Moreover, transdermal delivery studies confirmed that EGT-loaded nanovesicles significantly improved drug penetration through the stratum corneum. This work represents the first application of the AG86-conjugated nanovesicles for keratinocyte-targeted delivery, combined with ROS responsiveness. The “Loading-Targeting-Responding “ strategy holds significant potential for enhancing hydrophilic drug delivery and offers a promising approach for treating age-related skin lipofuscin accumulation. Graphical Targeted delivery of EGT via the “Loading-Targeting-Responding” strategy.

Acute kidney injury (AKI) is widely recognized as an important risk factor for chronic kidney disease (CKD), and preventing or delaying the AKI-CKD transition represents a critical therapeutic priority. Ergothioneine (EGT), a natural antioxidant, has demonstrated renal protective potential, but its role in attenuating the AKI-CKD transition remains unclear. To investigate this, an AKI-CKD mouse model was constructed by administering multiple small-dose injections of cisplatin. Mice were divided into control, AKI-CKD, and AKI-CKD + EGT groups. The protective function of EGT was assessed using biochemical indexes and histopathology. Proteomic analysis and network pharmacology were applied to identify potential EGT targets, which were further verified by molecular docking and Western blot. Results showed that EGT significantly mitigated renal damage in AKI-CKD mice. Proteomic analysis identified 35 differentially expressed proteins (DEPs) potentially associated with EGT treatment, with GO and KEGG enrichment implicating complement activation as a key pathway. Protein-protein interaction (PPI) network analysis, topology analyses, and molecular docking identified C4B and PON1 as potential core EGT targets. In conclusion, EGT attenuates AKI-CKD progression by targeting C4B and PON1, modulating complement activation and oxidative stress. These findings highlight the therapeutic potential of EGT and provide novel insights for drug development targeting the AKI-CKD transition.

In this study, a novel protein-polysaccharide based edible film was developed by sodium alginate (SA) incorporating with lactoferrin (LF) and ergothioneine (EGT). A small quantity of LF (0.15% w/v) equipped edible films with excellent moisture protection performance caused by enhanced hydrophobicity, evidenced by the descending water vapor permeability and ascending water contact angle (rising from 38.1° to 74.1°). Meanwhile, the films exhibited strengthened tensile strength and declining moisture content because of the filling of LF. EGT was employed to improve the properties of films as well, which endowed the films with homogeneous microstructure and ultraviolet shielding ability. Notably, the films loaded with EGT witnessed a dramatic growth of DPPH (2,2-diphenyl-1-picrylhydrazyl) inhibition from 8.50% to 64.90% and remained antioxidant active during storage. In this context, the samples containing the EGT-LF complex showed superior physical properties and multiple functional activities against ultraviolet light, bacterium, and oxidation. The analysis aimed at the film-forming solutions and spectrum of films provided a better understanding of the structure of samples: The adhesion of LF on the SA network driven by electric interaction improved the crystallinity of films; whereas EGT reduced the surface charge of LF according to molecular docking model. Overall, these results indicated that LF-assisted SA might be utilized as a promising material for active packaging, while EGT could be adopted to modify the protein/polysaccharide compounds for improved functional features.

This study investigated the feasibility of substituting SO 2 with ergothioneine- and glutathione-producing yeasts in cider fermentation. The results indicated that the Optiwhite+E91-8 group reduced dissolved oxygen by 61.36% and enhanced total flavonoids by 5.54%, achieving antioxidant capacity comparable to the SO 2 -treated controls. Furthermore, organic acid content increased by 30.50% compared to apple juice, accompanied by a richer ester profile, enhanced fruity aroma, and improved flavor quality without dimethylamine formation. Ergothioneine is more stable than glutathione in acidic environments and may synergize with glutathione via redox cycling, regenerating post-oxidation to inhibit phenolic oxidation and sustain fermentation stability through sustained oxygen consumption. These findings confirm that inoculation with antioxidant-producing strains can effectively replace SO 2 , providing both a theoretical and technical foundation for the development of sulfur-free cider that boasts enhanced antioxidant activity and superior sensory qualities. This approach not only advances sustainable winemaking practices but also addresses consumer preferences for natural preservative alternatives.

Ergothioneine (EGT) has attracted great attention due to its extremely potent antioxidant properties, universally acknowledged as ‘longevity vitamin’. In order to comprehensive understanding of its pharmacodynamics and pharmacokinetics, the binding mechanism of EGT with human serum albumin (HSA) was clarified by cutting-edged multi-spectroscopic approaches and in silico molecular docking coupled with molecular dynamic simulation. Our fluorescence quenching results revealed that the binding of EGT to HSA was in a static quenching mode validated by the descending Stern–Volmer constant ( K sv ) values (2.82, 2.36, 1.48 × 10 4 L mol −1 ) and biomolecular quenching rate constant ( K q ) values (2.82, 2.36, 1.48 × 10 12 L mol −1 ) at 298 K, 305 K, and 310 K, respectively. van’t Hoff criterion revealed the combination of EGT with HSA was a spontaneous exothermic process (Δ G = −24.16 kJ mol −1 ) via hydrogen bonding and van der Waals force interactions (Δ H = −60.25 kJ mol −1 , Δ S = −129.44 J mol −1 K −1 ) at 310 K. The analysis of UV–vis absorption spectrum, synchronous fluorescence spectrum, three-dimensional fluorescence spectrum and circular dichroism indicated the addition of EGT affected the microenvironment of Trp214 and rearranged the structure of HSA. The binding replacement assay interpreted their binding site was near the subdomain IIA of HSA (Sudlow’s site I), which was intuitively exhibited by molecular docking. In addition of obvious van der Wall forces, attractive charge and Pi-alkyl interactions, the chiral betaine group (N + (CH 3 + ) 3 ) in the side chain of EGT was inclined to form hydrogen bonds with Lys199, Ser287 and Arg257 in the hydrophobic cavity of albumin. Moreover, the dynamic simulation reinforced the equilibrium and stability of formed docking complex by four indicators (RMSD, RMSF, Rg, SASA) within 100 ns.

This study systematically investigated the interaction mechanism between ergothioneine (EGT) and soy protein isolate (SPI) and its effects on protein structure and functionality by integrating multi-spectroscopic techniques with computational simulation. The results demonstrated that EGT quenches the intrinsic fluorescence of SPI through a static quenching mechanism, with a binding constant (K A ) of 2.242 × 10 5 L/mol and approximately one binding site per molecule at 298 K. Thermodynamic parameters (ΔH = −35.01 kJ/mol, ΔS = −41.48 J/(mol·K)) indicated that the spontaneous interaction was primarily driven by hydrogen bonding and van der Waals forces. Molecular docking confirmed these findings, revealing that EGT binds specifically within the hydrophobic grooves of SPI subunits (7S and 11S) via hydrogen bonds and hydrophobic interactions. The binding induced significant conformational changes in SPI, evidenced by redshifted synchronous fluorescence peaks (Tyr: 282 → 284 nm; Trp: 289 → 296 nm) and alterations in FTIR amide bands, which collectively enhanced molecular flexibility and surface hydrophilicity. These structural modifications directly improved the functional properties of SPI, including significant reductions in particle size and turbidity, along with marked enhancements in solubility, emulsifying activity index (EAI), and emulsion stability index (ESI). Furthermore, the antioxidant capacity of the complexes-assessed by DPPH and ABTS radical scavenging assays and reducing power analysis-exhibited a concentration-dependent increase. This work provides a comprehensive elucidation of the EGT-SPI interaction mechanism and offers a solid theoretical foundation for developing SPI-based delivery systems to enhance the stability and functionality of bioactive compounds in functional foods.

Adjuvant skincare formulations possessing anti-inflammatory and antibacterial properties can alleviate treatment-related side effects and enhance patient adherence in acne management. Paeonol (PAE), totarol (TOT), and ergothioneine (ERG) demonstrate promising anti-acne activity. However, their clinical translation is hindered by poor solubility, limited stability, and insufficient skin permeation. To address these limitations, we co-encapsulated PAE, TOT, and ERG into hyaluronic acid (HA)-modified nanoliposomes (NLPs). Compared to the NLPs modified with HA with high molecular weights, those formulated with miniHA (3-10 kD a ) exhibited superior skin penetration capability and cellular uptake efficiency. The miniHA modified NLPs co-encapsulated PAE, TOT, and ERG (PTE-NLPs) exhibited excellent biocompatibility in both in vitro cytotoxicity assay and chicken embryo chorionic allantoic membrane irritation assays. Compared to free functional ingredients, PTE-NLPs demonstrated significantly enhanced anti-inflammatory and antibacterial efficacy in cellular assays. Furthermore, in 3D skin models, PTE-NLPs suppressed Transient Receptor Potential Vanilloid 1 (TRPV1) expression, indicating a potential mechanism for attenuating neuronal hyperreactivity. The anti-inflammatory activity was subsequently validated in vivo. These findings collectively demonstrate that miniHA-modified NLPs provide a safe and efficient platform for co-delivering synergistic anti-acne actives, highlighting their strong potential as an adjuvant therapy for acne management.

α-Ketoglutarate (AKG), ergothioneine (EGT), ascorbic acid (VC), and pyrroloquinoline quinone (PQQ) are increasingly co-formulated to combat age-related mitochondrial dysfunction and oxidative stress, driving an urgent need for reliable quality control. However, existing analytical approaches remain fragmented, targeting only one or two compounds, relying on time-consuming derivatization, or employing costly mass spectrometry, and critically failing to address chemical incompatibilities among analytes. This study presents the first validated HPLC-UV method for simultaneous quantification of all four bioactives. A segmented extraction strategy was developed to reconcile their physicochemical conflicts: VC, AKG, and EGT were extracted with 0.1% metaphosphoric acid, while acid-labile PQQ was separately extracted with neutral phosphate buffer. Chromatographic separation was achieved on an AQ-C18 column with a gradient of 50 mM Na₂HPO₄ and acetonitrile. The method demonstrated excellent linearity (R² > 0.99), high sensitivity, good precision (RSD < 2%), and satisfactory accuracy (recoveries: 97.2–103.6%). The Mosaic-WAC score of 82.6 further validates the proposed method. Analysis of commercial products indicated that only VC exhibited degradation, with more pronounced losses in liquid matrices, attributable to its inherent oxidizability. This underscores the need for comprehensive multi-component quality control. The proposed method provides a practical platform for routine analysis and regulatory evaluation of complex nutraceutical products.

This study developed a rapid and eco-friendly HPLC method for simultaneous determination of adenosine and ergothioneine in Cordyceps spp. samples. The extracted protocol involved dispersing samples in diatomaceous earth followed by vortex extraction with 80 % methanol. Chromatographic separation was achieved within 5 min using a Shim-pack Velox HILIC column with a gradient mobile phase of 0.1 % formic acid aqueous solution and acetonitrile solution containing 0.1 % formic acid and detection at 260 nm. Both analytes demonstrated excellent linearity ( r > 0.9999) with average recoveries of 102.82 % (adenosine) and 100.10 % (ergothioneine), accompanied by RSD s below 2 %. The contents of adenosine and ergothioneine in 46 batches of Cordyceps spp. samples were 0.000–2.700 mg/g dry weight (DW) and 0.000–0.703 mg/g DW, respectively. The Ophiocordyceps sinensis and its related species were successfully classified by hierarchical cluster analysis. This study determined the content of ergothioneine in Cordyceps spp. samples for the first time, and identifies ergothioneine as a critical marker for differentiating authenticated Ophiocordyceps sinensis from its adulterants. Results of BAGI assessment (score=87.5) confirmed the superior practicality and environmental friendliness of the developed method. This rapid, accurate, and reliable method provided a scientific foundation for enhancing quality evaluation technology of Cordyceps spp. species samples.
