Antifungal activity and mechanism of citral, limonene and eugenol against Zygosaccharomyces rouxii

Rui Cai,Miaomiao Hu,Yijun Zhang,Chen Niu,Tianli Yue,Yahong Yuan,Zhouli Wang

The aim of this study was to investigate the antifungal activity and mechanisms of citral, limonene and eugenol against Zygosaccharomyces rouxii . All the three essential oil ingredients presented strong antifungal activities against Z. rouxii . Minimum inhibitory concentrations of citral, limonene and eugenol were 0.188, 0.75 and 0.4 μL/mL, and the minimum fungicidal concentrations were 0.375, 3 and 0.8 μL/mL, respectively. Scanning electron microscopy showed that the tested essential oil compounds had destructive effects on the yeast cell surface. After treated by citral, limonene and eugenol, the relative electric conductivity and leakage of nucleic acid were increased significantly. SDS-PAGE analysis indicated that the essential oil components could result in the loss of soluble proteins and might also destroy the yeast protein or inhibit their expression. These results demonstrated that citral, limonene and eugenol exerted their antifungal action against Z. rouxii through cell membrane damage and destruction of cellular proteins. This study provided a theoretical possibility that citral, limonene and eugenol could be used as natural food preservatives for the control of Z. rouxii -related spoilage in the fruit juice industry.

Antioxidant and antibacterial starch-based edible films composed of eugenol/gelatin microspheres

Ying Chen,Feng Wei,Wei Mu,Xiaojun Han

The food safety issues caused by microorganisms are a great threat to human life and society. However, most edible packaging films are insufficient to meet antimicrobial applications. Thus, recent research has been focused on the combination of various antimicrobial agents with edible films. Herein, antioxidant and antibacterial starch-based edible films with different concentrations of eugenol/gelatin microspheres were developed. Gelatin microspheres were introduced as the carriers for eugenol to improve its compatibility in the composite films. The optimum composite films presented a compact and uniform structure with low water vapor permeability and low solubility. The tensile strength of the composite film reached 31.87 MPa with 2.14% elongation at break. The eugenol conferred edible films with effective antioxidant and antibacterial activities against E. coli and S. aureus. The apple cubes wrapped by the prepared films can be stored at room temperature at 50% humidity for 7 days. The fabricated composite films show promising practical applications in food packaging.

Release kinetics of pectin/eugenol composite film and application in pork preservation

Wei Wang,Qing Wang,Xiaoju Chen,Yaqiong Kong,Mengqing Wu,Shuangshuang Zhu,Minmin Chen,Linlin Li

Biodegradable active packaging materials have received growing attention for extending the shelf life of foodstuffs. A Pectin/eugenol (Pe/Eu) composite film is prepared by casting from pectin and eugenol. The morphology and wettability of the composite film are characterized. The antioxidant activity of the composite film is determined with DPPH method. Special attention is given to the release kinetics of Eu from Pe/Eu composite film and its application in pork preservation. The results display that Eu increased the surface roughness, hydrophobicity, and antioxidant activity of the composite film. In addition, the release mechanism of Eu from Pe/Eu composite film is controlled by diffusion. The release behavior of Eu in Pe/Eu composite film can prolong the shelf life of pork. Therefore, our results suggest that Pe/Eu composite films can provide new ways for active packaging materials to extend the shelf life of animal-based protein foods.

Eugenol alleviates renal ischemia-reperfusion injury induced-endoplasmic reticulum stress via activating Sestrin2

Jingwei Liu,Xujie Sun,Junfeng Liang,Shiqiang Song

Introduction Renal Ischemia-Reperfusion Injury (RIRI) often arises due to heightened oxidative stress, rendering it a central focus of research. Sestrin2 plays a pivotal role in regulating oxidative stress; nevertheless, its impact on the renoprotective properties of Eugenol (EU) during RIRI warrants further investigation. Methods Mice and TCMK-1 cells were categorically assigned into six groups: Sham/Control, Ischemia-Reperfusion (IR)/HR (Hypoxia-Reoxygenation), IR/HR+EU, Sham/Control+Sestrin2-KO, IR/HR+Sestrin2-KO, and IR/HR+EU+Sestrin2-KO. The effects of EU and the involvement of Sestrin2 in RIRI/HR were evaluated using Urea Nitrogen (BUN), Creatinine (Scr), Superoxide Dismutase (SOD), Glutathione (GSH), Catalase (CAT), and Malondialdehyde (MDA) assay kits; western blotting; cell viability assays; HE-staining; and Reactive Oxygen Species (ROS) detection. Results Following RIRI/HR, a marked deterioration in kidney function and a significant surge in oxidative stress levels were observed. However, EU treatment ameliorated renal injury and inhibited oxidative stress. Additionally, EU upregulated Sestrin2 expression, and the renoprotective effects of EU were reversed upon Sestrin2 knockdown. Conclusion The present study posits that EU effectively mitigates RIRI/HRI (Hypoxia-Reoxygenation Injury), and its mechanism of renal protection potentially involves the upregulation of Sestrin2, coupled with the inhibition of oxidative and Endoplasmic Reticulum Stress (ERS).

Response of xenobiotic biodegradation and metabolic genes in Tribolium castaneum following eugenol exposure

Zhang Yonglei,Gao Shanshan,Zhang Ping,Sun Haidi,Lu Ruixue,Yu Runnan,Li Yanxiao,Zhang Kunpeng,Li Bin

Eugenol, a plant-derived component possessing small side effects, has an insecticidal activity to Tribolium castaneum ; however, the underlying molecular mechanisms of eugenol acting on T. castaneum are currently unclear. Here, a nerve conduction carboxylesterase and a detoxifying glutathione S-transferase were significantly inhibited after eugenol exposure, resulting in the paralysis or death of beetles. Then, RNA-sequencing of eugenol-exposed and control samples identified 362 differentially expressed genes (DEGs), containing 206 up-regulated and 156 down-regulated genes. RNA-seq data were validated further by qRT-PCR. GO analysis revealed that DEGs were associated with 1308 GO terms of which the most enriched GO terms were catalytic activity, and integral component of membrane; KEGG pathway analysis showed that these DEGs were distributed in 151 different pathways, of which some pathways associated with metabolism of xenobiotics or drug were significantly enriched, which indicated that eugenol most likely disturbed the processes of metabolism, and detoxication. Moreover, several DEGs including Hexokinase type 2 , Isocitrate dehydrogenase , and Cytochrome b-related protein , might participate in the respiratory metabolism of eugenol-exposed beetles. Some DEGs encoding CYP , UGT , GST , OBP , CSP , and ABC transporter were involved in the xenobiotic or drug metabolism pathway, which suggested that these genes of T. castaneum participated in the response to eugenol exposure. Additionally, TcOBPC11/ TcGSTs7 , detected by qRT-PCR and RNA-interference against these genes, significantly increased the mortality of eugenol-treated T. castaneum , providing further evidence for the involvement of OBP/GST in eugenol metabolic detoxification in T. castaneum . These results aid eugenol insecticidal mechanisms and provide the basis of insect control.

Synthesis of Eugenol-Derived Furanylbenzoxazine and Properties of Its Copolymer With Epoxy Resin

Jinhe Gao,Shuangle Xue,Caizhao Liu,Caiyu Song,Mehdi Derradji,Mianyuan Wu,Tianyi Xie,Zhigang Yuan,Mingming Sun,Xugang Zhang,Jianhui Li,Bin Zhang

To increase the toughness of epoxy resin, an allyl-terminated furanylbenzoxazine (AFB) derived from eugenol was copolymerized with epoxy resin (E51). Fourier transform infrared (FTIR) spectroscopy and differential scanning calorimetry (DSC) analyses were used to examine the curing behavior of the blended resins. With increasing AFB content, the crosslinking density of the EP-AFB resins decreases, while the residual carbon yield ( Y c ) of the epoxy resins after high-temperature combustion in a nitrogen atmosphere increases. The Y c of EP-4AFB increased from the initial 14.92% to 20.21%. In addition, appropriate concentration of AFB could be conducive to improving the mechanical properties. The flexural and impact strengths of EP-1AFB resins were 3.5% and 81.3% higher than EP resins, respectively. The elongation at break and tensile strength of EP-3AFB resins were 91.2% and 68.4% higher than EP resins, respectively. Furthermore, the addition of AFB significantly modified both adhesive (lap shear strength and peel strength) and dielectric properties of the epoxy resins.

Preparation, characterization, oral bioavailability, and pharmacodynamic study of eugenol-porous silica solidified powder

Yao ZhongWei,Zhang Wei,Hu Yehong,An Zhentao,Fang Zhijun,Wang Jing,Zhang Zhenhai

Eugenol possesses anti-inflammatory and antioxidant properties, and may serve as a potential therapeutic agent for hepatic fibrosis. However, the development of solid eugenol formulations is challenging due to its volatility. To address this issue, this study employed porous silica to adsorb solidified eugenol. The solidified powder was characterized using Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM). In addition, the differences in in vitro release and oral bioavailability between eugenol and solidified eugenol powder were investigated. The effectiveness of eugenol and eugenol powder in treating liver fibrosis was investigated using enzyme-linked immunosorbent assay (ELISA), polymerase chain reaction (PCR), and histopathological observations. Our results indicate that porous silica can effectively solidify eugenol into powder at a lower dosage. Furthermore, we observed that porous silica accelerates eugenol release in vitro and in vivo. The pharmacodynamic results indicated that eugenol has a positive therapeutic effect against hepatic fibrosis and that porous silica does not affect its efficacy. In conclusion, porous silica was able to solidify eugenol, which may facilitate the preparation and storage of solid formulations.

Mechanical and Dielectric Property Enhancement of Epoxy Resins With Eugenol Derived Allyl Benzoxazine

Bo Liu,Zhigang Yuan,Caizhao Liu,Mehdi Derradji,Mingming Sun,Xugang Zhang,Bin Zhang,Lei Wang,Jianhui Li,Tianyi Xie,Jinzi Li,Gang Xue,Ming Zhao,Xue Zhang,Caiyu Song,Shuangle Xue

A bio-based allyl-terminated benzoxazine (AMP) was copolymerized with epoxy resins (E51) to enhance the toughness of epoxy resins. The curing behavior of the blended resins was investigated using differential scanning calorimetry (DSC) analysis and Fourier transform infrared (FTIR) spectroscopy. The crosslinking density and gel content of EP-AMP resins were decreased with the increasing AMP content. AMP significantly improved the residual carbon yield ( Y c ) of epoxy resins following high-temperature combustion in N 2 atmosphere, with enhancements correlating positively with AMP concentration. Compared with the value of EP resins (14.92%), the value of EP-4AMP resins (19.41%) was well increased. The mechanical properties (including flexural, impact, and tensile tests) were evaluated, revealing substantial improvements. The flexural strength, tensile strength, and elongation at break of EP-3AMP resins were 11.7%, 104.8%, and 67.6% higher than EP resins, respectively. The impact strength of EP-2AMP resins was 66.9% higher than EP resins. Furthermore, the incorporation of AMP markedly modified both adhesive (lap shear strength and peel strength) and dielectric properties of the epoxy resins.

Performance of eugenol emulsion/chitosan edible coating and application in fresh meat preservation

Runan Zhao,Yu Zhang,Huanle Chen,Rong Song,Yan Li

A green chitosan-based coating was prepared by incorporating chitosan-stabilized eugenol emulsions into chitosan solution. The physical, mechanical, rheological, and stability property of chitosan coatings were characterized and then the effect of chitosan coatings on the shelf-life of fresh meat was investigated. Emulsion incorporation greatly improved the performance of chitosan coating, including stronger anti-corrosion against pH and UV-light resistance and lower water vapor permeability. Emulsion incorporation greatly increased the inhibition zone for Escherichia coli and Staphylococcus aureus by 6.9 and 8.7 mm, respectively. After the application of coating, meat kept a bright red appearance within 14 days and its spoilage was prevented. After 14-day storage, pH value, the total number of colonies, and TVB-N of meat still met the quality standard, demonstrating that the shelf-life of meat was prolonged from 6 to 14 days. The knowledge provides a green coating to extend the shelf-life of fresh meat. Novelty impact statement Chitosan-stabilized emulsions were added into chitosan-coating materials. Eugenol-loaded chitosan coating had a good protection effect on the color of meat. Shelf-life of fresh meat was prolonged to 14 days by eugenol/chitosan coating.

Eco-friendly eugenol curing agent-derived biobased epoxy resins for multi-functional integration

Zhiyong Li,Xiangyan Ji,Xiaoge Li,Da Gao,Lei Sheng,Wei He,Jie Sun,Dingqiang Lu,Jingjing Meng,Kai Guo

The investigation of bio-based epoxy resin has become increasingly significant, particularly for functional resins. However, research on bio-based resins that exhibit comprehensive performance advantages has seldom proceeded. To overcome this dilemma, the liquid amine curing agent AEESN was synthesized from bio-based eugenol and cysteamine, and then furnished the polymerization with three epoxy monomers, viz. DGEBA, TGAP, and TGDDM, respectively. The AEESN-based epoxy resin polymer, specifically DGEBA/AEESN, demonstrated an impact strength of 53.6 ± 7.78 kJ m −2 by a 1060 % increase, after storage at −20 °C, wherein less impact occurred on tensile and bending strength. Among them, the temperature control microphase separation process is responsible for such an impact enhancement. In addition, DGEBA/AEESN is also highly resistant to acidic and basic aqueous solutions. In contrast, the TGDDM/AEESN thermoset demonstrated good resistance to both organic solvents and aqueous solutions. Moreover, among different substrates, it was revealed that wood allowed the strongest adhesion for TGDDM/AEESN, reaching 4.14–7.48 MPa. Delightedly, all thermosets showed superior shape memory ranging from 100 °C to 150 °C and behaved with good sustainability, processability, and design freedom. Besides, superior optical shielding against UV and blue light may promote the use of functional coatings or lenses. Overall, the aforementioned bio-based AEESN epoxy resin exhibits good performance and outstanding functionality for applications.

Antiviral Activity of Eugenol Against Chinese Rice-Field Eel Rhabdovirus in Monopterus albus

Jingwen Jiang,Mingyang Xue,Wenzhi Liu,Yong Zhou,Yiqun Li,Yuding Fan

Chinese rice-field eel rhabdovirus (CrERV) is a serious epidemic pathogen of Chinese rice-field eel and causes severe economic losses to aquaculture. However, there are no commercial drugs presently available to control CrERV infection. Eugenol is a bioactive compound extracted from clove plants and exhibits potential antiviral activity. In the study, the antiviral activity of eugenol against CrERV was investigated in Chinese rice-field eel (Monopterus albus). Eugenol reached the highest inhibition rate of 96.6% at 40 mg/L in Chinese rice-field eel kidney cells (CrEK). Notably, eugenol exhibits antiviral activity by directly targeting CrERV and additionally confers prophylactic effects against infection via its action on CrEK cells. The results of exploring the viral invasion cycle demonstrated that eugenol primarily exerted its antiviral effect during the middle stage and late stage (12 h and 24 h) of viral infection. In addition, eugenol inhibited CrERV-induced apoptosis of CrEK cells, maintained mitochondrial membrane potential levels, maintained physiological cellular morphology and structure, and protected cells from loss of cellular morphology, formation of apoptotic vesicles, and cell fragmentation. For the in vivo study, eugenol increased the survival rate of CrERV-infected rice-field eel by 56% and 48%, in prevention experiments and treatment experiments, respectively. Concurrently, eugenol significantly reduced viral loads and induced the upregulation of anti-inflammatory and antioxidant genes, indicating its potential for immunoregulation. In summary, eugenol holds potential for both preventing and treating CrERV infections in the aquaculture context.
