Trident-inspired fucoidan-based armor-piercing microcapsule for programmed acute pulmonary embolism treatment

Ning Yang,Weikun Li,Zhicheng Qian,Xin Tan,Zonghao Liu,Feiling Feng,Ling Liu,Liqin Ge

Pulmonary embolism remains the third leading cause of human mortality after malignant tumors and myocardial infarction. Commonly available thrombolytic therapeutic agents suffer from the limitations of very short half-life, inadequate targeting, limited clot penetration, and a propensity for severe bleeding. Inspired by the trident, we developed the armor-piercing microcapsule (MC), fucoidan-urokinase-S-nitrosoglutathione-polydopamine@MC (FUGP@MC), which exhibited a triple combination of photothermal, mechanical and pharmacological thrombolysis for the therapeutic treatment of acute pulmonary embolism (APE). Briefly, the outermost fucoidan layer was utilized for targeting to the APE area. Programmed APE treatment was triggered by near-infrared (NIR) light irradiation. Photothermal thrombolytic therapy was carried out by photothermal conversion of polydopamine. The photothermal conversion broke the S-nitroso bond in S-nitrosoglutathione (GSNO) and produced large amounts of nitric oxide (NO) for mechanical thrombolysis, which subsequently disrupted the interfacial structure of microcapsule to stimulate the release of the urokinase (UK), leading to a triple synergistic thrombolytic effect. The results demonstrated that the embolization residual rate of FUGP@MC (contained ≈ 1452.5 IU/kg UK) group was significantly lower than that of UK (10,000 IU/kg) group (6.35 % VS 16.78 %). Remarkably, FUGP@MC demonstrated a reliable in vivo biosafety proficiency. In summary, trident-inspired armor-piercing microcapsule FUGP@MC reveals a potential avenue for advancing pulmonary embolism therapeutics and promises to be a safer alternative candidate to current drug approaches.

The fucoidan delivery system enhanced the anti-cervical cancer effect of caffeic acid

Xintao Gao,Tao Jiang,Xiaochen Wu,Yantao Li,Jun Xiao,Lin Long,Chuanlong Guo

Cervical cancer remains one of the leading causes of mortality among women, and immunotherapy targeting the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway holds promise for its treatment. This study has developed nanoparticles based on fucoidan (Fu/CA NPs), successfully loading them with caffeic acid (CA) for application in cervical cancer therapy. In vitro experiments revealed that Fu/CA NPs significantly inhibited the proliferation of cervical cancer HeLa cells (by 65.73 ± 4.06 %) and induced apoptosis through the accumulation of reactive oxygen species and mitochondrial damage. Furthermore, treatment with Fu/CA NPs activated the cGAS-STING pathway, attributed to the cytoplasmic release of mitochondrial DNA (mtDNA) and the induction of DNA double-strand breaks (dsDNA) by Fu/CA NPs. In vivo results confirmed that Fu/CA NPs suppressed solid tumor growth (by 67.8 %), with even more pronounced antitumor effects observed when combined with cisplatin (96.5 %), a phenomenon also associated with the activation of the cGAS-STING pathway. Excitingly, the combination of Fu/CA NPs and cisplatin alleviated cisplatin-induced nephrotoxicity, as indicated by a decrease in blood urea nitrogen (BUN) by 53.27 % and serum creatinine (SCr) by 74.93 %. In summary, our research presents a potential therapeutic avenue for cervical cancer treatment, particularly highlighting the synergistic benefits of combining Fu/CA NPs with cisplatin.

Development and characterization of soybean protein isolate and fucoidan nanoparticles for curcumin encapsulation

Lihong Fan,Yuqing Lu,Xiao-kun Ouyang,Junhong Ling

Curcumin (Cur) is a natural polyphenol with beneficial biological and pharmacological activities; however, it has limited applications owing to its low solubility and light sensitivity. The protein-polysaccharide complex can effectively embed lipid-soluble drugs to increase their stability and dispensability in aqueous solutions. Soybean protein isolate (Spi) and fucoidan (Fuc) were used as a polymer matrix, and core-shell nanoparticles were prepared to encapsulate Cur via electrostatic interaction under acidic and neutral conditions. The structure of the Spi-Fuc nanoparticles was studied via Fourier-transform infrared spectroscopy, transmission electron microscopy , and scanning electron microscopy. Concurrently, we evaluated the efficacy of the nanoparticles based on stability, drug loading rate, and simulated release. Our results showed that the Spi-Fuc nanoparticles (size, approximately 236.56 nm) had a spherical, core-shell structure and that they could effectively load Cur with an embedding efficiency of >95%; moreover, the system had long-term dispersion stability. Thus, we provide a simple method for Cur delivery, which can also be potentially used for delivering lipid-soluble active ingredients.

Fucoidan Is Not Completely Dependent on Degradation to Fucose to Relieve Ulcerative Colitis

Qiang Wei,Maochen Xing,Ke Wang,Qiong Yang,Jiarui Zhao,Yuan Wang,Xia Li,Kai Ji,Shuliang Song

Recently, fucoidan has been proposed for use as a potential anti-inflammatory drug. The purpose of this study was to investigate the mechanism of fucoidan in the treatment of ulcerative colitis. We compared the anti-inflammatory effects of fucoidan and fucose induced by dextran sulfate sodium, and the effects of fucoidan and fucose on the gut microbiota of mice. Our results showed that low-dose fucoidan significantly improved weight loss, disease activity index scores, colonic shortening, colonic histopathological damage, intestinal fatty acid binding protein 2 levels, and the expression of Occludin, Claudin-4, and Claudin-1. However, both high-dose fucoidan and fucose did not perform as well as low-dose fucoidan as described above. In addition, 16S rDNA high-throughput sequencing showed that low-dose fucoidan significantly increased the abundance ofAlloprevotella, and fucose significantly increasedRuminococcaceae, but neither significantly reversed the imbalance in the gut microbiota. Therefore, we inferred that the regulation of fucoidan on colitis has a unique and complex mechanism, and it is not completely dependent on degradation to fucose to relieve ulcerative colitis, nor is it achieved only by regulating the gut microbiota. The mechanism by which fucoidan treats colitis may also include reducing inflammatory cell infiltration and increasing intestinal barrier function. Keywords:fucoidan;fucose;ulcerative colitis;gut microbiota;dextran sulfate sodium

Novel ROS-responsive marine biomaterial fucoidan nanocarriers with AIE effect and chemodynamic therapy

Chunjing Guo,Yanguo Su,Ziting Cheng,Qiang Chen,Huimin Guo,Ming Kong,Daquan Chen

Chemodynamic therapy (CDT) has been widely used in the treatment of many kinds of tumors, which can effectively induce tumor cell apoptosis by using produced reactive oxygen species (ROS). In this paper, ROS-sensitive multifunctional marine biomaterial natural polysaccharide nanoparticles were designed. Aggregation-induced emission (AIE) molecules tetraphenylethylene (TPE) labeled and caffeic acid (CA) modified fucoidan (FUC) amphiphilic carrier material (CA-FUC-TK-TPE, CFTT) was fabricated, in which the thioketal bond(TK) was used as the linkage arm between TPE and fucoidan chain, giving the CFTT material ROS sensitivity. In addition, amphiphilic carrier material (FUC-TK-VE, FTVE) composed of thioketal-linked vitamin E and fucoidan was synthesized. The mixed carrier material CFTT and FTVE self-assembled in water to form nanoparticles (CFTT – FTVE@PTX-Fe 3+ ) loaded with PTX and Fe 3+ . The CDT effect was combined with the chemotherapeutic drug PTX to achieve tumor inhibition. In vitro cell studies have proved that CT/PTX nanoparticles have excellent cell permeability and tumor cytotoxicity. In vivo antitumor experiments confirmed effective antitumor activity and reduced side effects.

Bioactivity of fucoidan extracted from Laminaria japonica using a novel procedure with high yield

Dong Zhao,Jian Xu,Xia Xu

Due to antitumor and antioxidant activities, there has been increasing interest in fucoidans, sulfated polysaccharides, from brown algae. Currently, the classic hot water extraction and acid extraction are time-consuming, and can cause the partial cleavage of sulfate esters, leading to bioactivity loss. The present study developed a novel process for extracting fucoidans from Laminaria japonica with 0.5% ethylenediaminetetraacetic acid at 70 °C. Crude fucoidans (CFs) obtained with a yield (∼16%) was purified to different fractions of F1, F2 and F3. The fractions displayed similar anticancer activity against Hela cells. Due to the differences in sulfate content, molar ratio of sulfate to fucose and monosaccharide composition, F1 and F2 exhibited stronger antioxidant ability than CFs and F3. This novel extraction provides an efficient procedure for extracting fucoidans from brown seaweeds with high extraction yield and removing pigments with one single step.

Enhancing the stability of zein/fucoidan composite nanoparticles with calcium ions for quercetin delivery

Hong Zhang,Haozhan Feng,Junhong Ling,Xiao-kun Ouyang,Xiaoyong Song

In this study, zein and fucoidan-based composite nanoparticles were prepared by the antisolvent precipitation method. The effects of different calcium ion (Ca 2+ , 0–3.0 mM) concentrations on the stability of the composite nanosystems loaded with quercetin were studied under different conditions (pH, temperature, salt concentration, and ultraviolet light irradiation), and the composite nanoparticles were characterized. Electrostatic interactions, hydrogen bonding, and hydrophobic interactions are the main forces underlying the formation of composite nanoparticles. The addition of Ca 2+ led to improved release of the active substances from the composite nanoparticles in simulated digestive solutions (especially when the Ca 2+ concentration was 1.5 mM). The composite nanosystems based on alcohol-soluble proteins and anionic polysaccharides with added Ca 2+ can be potentially applied for the delivery of active substances.

Oxidized fucoidan-based nanocomposite hydrogel for cryptotanshinone delivery and prevention of postoperative abdominal adhesions

Tao Zhang,Xianmin Shi,Yanjuan Huang,Yujun Gong,Yuanfeng He,Danni Xiao,Shengzhi Wang,Chunshun Zhao

Postoperative abdominal adhesions (PAA) are common diseases following abdominal surgery and can cause various serious complications. The excessive inflammation and oxidative stress are the main causes of PAA formation. Herein, we developed a nanomicellar hydrogel, OFu/HF@CTS, composed of oxidized fucoidan (OFu) and cryptotanshinone (CTS)-loaded hydrazine-functionalized pluronic F127 nanomicelles (HF127@CTS) for the effective prevention of PAA. The hydrogels exhibited satisfactory viscoelasticity, rapid self-healing ability, and good tissue adhesion properties, and were able to slowly release CTS for one week, with a cumulative release rate of 80 % on the 7th day. Additionally, the hydrogels could effectively reduce fibroblast cells and protein adhesions due to the high negative charge of OFu and have good biocompatibility towards RAW 264.7 and L929 cells. Moreover, CTS released from OFu/HF@CTS could efficiently reduce oxidative stress in macrophages and promote M1 macrophages polarized to M2 phenotype to relieve inflammation. In vivo results showed that OFu/HF@CTS hydrogel had good biodegradability and biosafety, and could effectively reduce PAA formation in a rat cecum-abdominal wall abrasion model through the mechanism of alleviating oxidative stress and inflammation, regulating fibrinolysis, and inhibiting fibrosis. This work highlights the therapeutic potential of drug-loaded nanomicellar hydrogels as a preventive strategy for PAA in clinical applications.

Therapeutic Potential of Fucoidan in Alleviating Histamine-Induced Liver Injury: Insights from Mice Studies

Mengyao Zhang,Huiqian Liu,Linlin Xu,Xizi Zhang,Wei Chen,Chengtao Wang

Histamine, a bioactive component in certain foods such as Huangjiu has been associated with liver injury and disrupted intestinal balance. This study explored the potential therapeutic effects of fucoidan (FCD) in mitigating histamine-induced imbalances in mice. We found that FCD mitigated liver injury, reducing transaminases, oxidative stress, and inflammation. Histological improvements included decreased cell infiltration and necrosis. FCD restored tight junction proteins and suppressed inflammation-related genes. Western blot analysis revealed FCD’s impact on TGF-β1,p-AKT, AKT, CYP2E1, Grp78, NLRP3, Cas-1, and GSDMD. Gut LPS levels decreased with FCD. Gut microbiota analysis showed FCD’s modulation effect, reducing Firmicutes and increasing Bacteroides. FCD demonstrates potential in alleviating histamine-induced liver injury, regulating inflammation, and influencing gut microbiota. Further research exploring higher dosages and additional parameters is warranted.

Soybean protein isolate-oxidized fucoidan nanocomplexes: Structural and interaction characterization, quercetin delivery potential evaluation

Sai Yang,Ziteng Lian,Tingting Zhu,Xiaolei Guo,Qianqian Zhang,Huan Wang,Lianzhou Jiang

A novel nanocomplex was prepared using soybean protein isolate (SPI) and oxidized fucoidan (OFU) to explore the structural and interaction variations and evaluate its potential for quercetin delivery. The optimized SPI to OFU mass ratio of 10:1 (SFU3) resulted in a nanocomplex particle size of 198.1 nm and increased ζ-potential. The incorporation of OFU altered the structure of SPI with the decrease in α-helix and β-sheet, and the redshift and intensity drop in fluorescence spectra. X-ray photoelectron spectroscopy (XPS) confirmed the Schiff base reaction between the two, interacting through covalent imine bonds. Moreover, OFU improved the micromorphology, antioxidant capacity, and stability of Quercetin (Que) nanocomplexes, with SFU3 showing the highest encapsulation efficiency and loading amount (94.80 %, 16.96 μg/mg). The nanocomplexes achieved an effective controlled release of Que. in vitro simulated digestion. This study will provide important insights into the development of SPI-OFU as nutrient carriers.

Inflammatory macrophage reprogramming strategy of fucoidan microneedles-mediated ROS-responsive polymers for rheumatoid arthritis

Xiaowei Liu,Ningning Diao,Shiqing Song,Wenxin Wang,Min Cao,Weili Yang,Chunjing Guo,Daquan Chen

During the pathogenesis of rheumatoid arthritis, inflammatory cells usually infiltrate synovial tissues, notably, M1-type macrophages, whose redox imbalance leads to the degradation of joint structures and deterioration of function. Natural active products play a vital role in immune modulation and antioxidants. In this study, we constructed a ROS-responsive nanoparticle called FTL@SIN, which consists of fucoidan (Fuc) and luteolin (Lut) connected by a ROS-responsive bond, Thioketal (TK), and encapsulated with an anti-rheumatic drug, Sinomenine (SIN), for synergistic anti-inflammatory effects. The FTL@SIN is then dispersed in high molecular weight Fuc-fabricated dissolvable microneedles (FTL@SIN MNs) for local administration. Therapy of FTL@SIN MNs afforded a significant decrease in macrophage inflammation while decreasing key pro-inflammatory cytokines and repolarizing M1 type to M2 type, thereby ameliorating synovial inflammation, and promoting cartilage repair. Additionally, our investigations have revealed that Fucoidan (Fuc) demonstrates synergistic effects, exhibiting superior mechanical strength and enhanced physical stability when compared to microneedles formulated solely with hyaluronic acid. This study combines nanomedicine with traditional Chinese medicine, a novel drug delivery strategy that presents a promising avenue for therapeutic intervention in rheumatoid arthritis.

Poly-allylamine hydrochloride and fucoidan-based self-assembled polyelectrolyte complex nanoparticles for cancer therapeutics

Pei Wang,Ranjith Kumar Kankala,Biaoqi Chen,Ruimin Long,Duanhua Cai,Yuangang Liu,Shibin Wang

Herein, we fabricated the novel drug delivery system based on the self-assembly of two polyelectrolytes, poly-allylamine hydrochloride (PAH) and fucoidan, as the polycation and polyanion, respectively, under mild conditions for cancer therapeutics. Furthermore, the designed polyelectrolyte complex nanoparticles as well as the methotrexate (MTX) disodium salt-loaded composites were systematically characterized using various techniques. The MTX loading in the nanoparticles was confirmed by zeta potential values that changed from −36.2 ± 2.2 to −28.3 ± 3.1 mV at a loading amount of 13.3 ± 1.2%. Furthermore, the obtained eventual particle sizes of nanoparticles were various with different concentrations and ratios of polyelectrolytes. The particle size also has increased from 130 ± 2.6 to 162.9 ± 2.3 nm after loading MTX. The drug release investigations in vitro at a pH value of 6.0 (acid environment) showed that the release of MTX was sustained in the conditions provided. Finally, we investigated the anticancer efficacy of MTX-loaded nanoparticles on MCF-7 cells and HeLa cells and the satisfactory results were obtained. Together, these self-assembled PAH/fucoidan nanoparticles with sustained drug release property will become the promising delivery system for cancer therapeutics. © 2018 Wiley Periodicals, Inc.
