Marine open traumatic brain injuries (oTBI) face high infection risks due to seawater-borne pathogens, demanding dura scaffolds with dual antibacterial and osteogenic functions under seawater immersion. Conventional scaffolds lack this dual capacity. Here, a biomimetic triple-layer polycaprolactone (PCL)-based scaffold is developed. The cerebral-side electrospun layer incorporates gentamicin sulfate (GS) for antibacterial action, while the cranial-side electrohydrodynamically printed rhombic PCL layer contains nano-hydroxyapatite (nHA) to enhance osteogenesis. A middle GS/PCL electrospun layer reinforces mechanical strength. The PCL/GS/nHA scaffold demonstrates suitable mechanical properties, sustained GS release, and low hemolysis. After seawater immersion or direct S. aureus exposure, it maintains potent antibacterial efficacy and biocompatibility. Additionally, nHA promotes osteogenic differentiation of human bone marrow mesenchymal stem cells in vitro. This multi-functional scaffold combines seawater-resistant antimicrobial performance with osteogenic capacity, offering a promising solution for dura repair in marine environments.

Polymethyl methacrylate (PMMA) bone cement is widely used in orthopedic surgery, and enhancing its antibacterial properties is crucial for preventing perioperative infections. In this study, unique “berry-shaped” micro/nano composite particles were prepared via coaxial electrospraying for the development of antibacterial PMMA bone cement. These particles consist of PMMA microspheres uniformly embedded with nanosilica (SiO 2 ) drug carriers coated with quaternary ammonium chitosan (HACC) and loaded with gentamicin sulfate (GS). Using a polylactic acid (PLA) nanofiber mat as the collection substrate, uniform assembly of the micro/nano composite particles and their strong anchoring to the matrix were achieved by tailoring the core-sheath solution properties and leveraging capillary-driven interactions. This hierarchical design not only improved interfacial adhesion to the PMMA bone cement matrix and improved flexural toughness, but also reduced the curing temperature. The corresponding antibacterial PMMA composite bone cement released GS sustainably for over five days, demonstrating potent antibacterial activity against both Escherichia coli ( E. coli ) and methicillin-resistant Staphylococcus aureus ( MRSA ), as well as good fibroblast biocompatibility. This study proposed an integrated “material-structure-assembly” strategy for the one-step fabrication of functional micro/nano composite particles, providing a new approach for the development of advanced biomedical materials.

Background Gentamicin is a commonly used aminoglycoside antibiotic, with ototoxicity as a significant side effect. Ferroptosis, an iron-dependent form of cell death, has been implicated in a variety of disorders. Whether ferroptosis impacts gentamicin ototoxicity is not yet known. The current work used an in-vitro model to examine the influence of gentamicin-induced ferroptosis on cochlear hair cell damage and probable molecular biological pathways. Methods House Ear Institute-Organ of Corti 1 (HEI-OC1) cells were treated with different concentrations of gentamicin for 24 hours, with or without ferrostatin-1 pretreatment, to observe gentamicin-induced ferroptosis. The role of p53/solute carrier family 7 member 11 (SLC7A11)/glutathione peroxidase 4 (GPX4) signaling in gentamicin-induced ferroptosis was explored by pretreating cells with the p53 inhibitor pifithrin-α (PFT-α). We investigated the effect of gentamicin on cells by assessing cell viability. Cellular proteins were isolated and Western blots were performed to detect changes in the expression of p53, SLC7A11, and GPX4. Fluorescence staining was used to assess levels of reactive oxygen species. An enzymatic detection kit was used to detect glutathione, Fe, and malondialdehyde markers. Results Gentamicin reduced cell viability, glutathione content, and SLC7A11 and GPX4 protein levels, and increased levels of p53 protein, reactive oxygen species, malondialdehyde, and Fe. These effects were largely blocked by pretreatment with ferrostatin-1. Pretreatment with the p53 inhibitor PFT-α prevented the gentamicin-induced reduction in SLC7A11 and GPX4, which alleviated several features of ferroptosis including glutathione depletion, iron overload, and lipid peroxidation build-up. Conclusion Gentamicin induces ferroptosis in the HEI-OC1 cell line, and the mechanism may be related to the p53/SLC7A11/GPX4 signaling pathway.

Applications of chitosan (CS) are hindered by its weak antimicrobial ability, poor solubility in basic and neutral solutions, structural modification is needed to improve its functional properties. Herein, a new chitosan-gentamicin derivative (CS-GT) was successfully synthesized via protection, regioselective bromination, nucleophile substitution reaction, and deprotection. Characterization techniques confirmed the successful introduction of GT onto the C-6 position of CS with a grafting rate of 76.19%. The additional amino groups provided by GT decreased the crystallinity and thermal stability of CS-GT, while enhancing its solubility in neutral and alkaline solutions. Compared to CS, CS-GT exhibited 2.50 and 2.41 times higher antibacterial activities against Escherichia coli and Staphylococcus aureus , respectively. Besides, an active packaging coating was develop by incorporating glycerol into CS-GT. The hydrogen bond networks formed within coatings endowed them good thermal stability and tightly uniform morphology. Furthermore, increasing CS-GT from 1% to 3% improved the coating's ultraviolet resistance and flexibility, but reduced its waterproof performance, visual appearance, and water barrier properties. Preservation experiments with papaya and litchi showed that CS-GT coatings effectively inhibited microbial growth and maintained quality and appearance up to 16 and 8 days of storage, respectively. In conclusion, CS-GT show promising potential for applications in food packaging.

Constructing bio-based antibacterial composite multilayer coatings with sustained drug release is important for reducing the infection risk of bone tissue repair. In this paper, a layer-by-layer aqueous electrodeposition method was applied to prepare a bio-based composite coating of chitosan, hyaluronic acid, and chitosan quaternary ammonium salt on the titanium surface. By pretreating with poly (acrylic acid), controlling the electrochemical deposition process, and post-treating with glutaraldehyde, a multi-layer composite coating with a smooth surface, good interface bonding with the titanium substrate, and a thickness more than ten microns was obtained. The composite coating was used as a drug-carrying system for gentamicin sulfate, which showed obvious antibacterial effects against Escherichia coli and Staphylococcus aureus . The bio-based composite coating with a sandwich structure loaded with antibiotics effectively inhibited the burst release of antibiotics, and the 14-day release rate is only 68.4 ± 2.1 %, achieving sustained release of the drug. The hydrophilic chitosan quaternary ammonium salt on the surface of the composite coating can effectively inhibit the early adhesion of bacteria, while the slow release of gentamicin sulfate in the interlayer can exert a long-lasting antibacterial effect. The preparation of bio-based antibacterial composite coatings using a layer-by-layer electrodeposition strategy has promising applications in titanium orthopedic implants.

Adding gentamicin to silk fibroin enhances both the antibacterial performance and degradation rate of silk-based materials. The increased material degradation rate can affect the strength of early internal fixation, resulting in internal fixation failure. This study sought to adjust the gentamicin concentration to control the material degradation rate, thereby better meeting clinical application requirements. The in vitro degradation, water absorption rate, and expansion rate of silk-based materials containing different gentamicin concentrations were studied. A gentamicin-loaded silk-based screw was implanted into the femurs of New Zealand rabbits. Micro-computed tomography was used to measure the screw diameter, which was then used to calculate the degradation rate. The specimens were stained with hematoxylin and eosin and Masson’s trichrome. The in vitro results revealed increasing material degradation rates with increasing gentamicin concentration but no significant differences in water absorption rates with different gentamicin concentrations. The degradation rates of gentamicin-loaded (4 mg/g) silk-based rod-like materials were approximately 11.08% at three months in vitro and 9.4% in the animal experiment. The time for complete degradation was predicted from the fitting curve to be approximately 16 months. No inflammatory hyperplasia was observed in bone or soft tissue. The degradation and biocompatibility of the material containing 4 mg/g gentamicin meet clinical application requirements, and previous experimental results demonstrate good antibacterial performance of materials containing this gentamicin concentration.

Carbonated hydroxyapatite coatings (CHACs) on titanium alloy substrates have been widely used as bone substitute materials. However, implant-associated infection becomes a serious problem in orthopedic surgery, leading to prolonged antimicrobial treatment, extensive bone debridement and failure of implants. Herein, gentamicin-loaded CHACs (Gent-CHACs) were fabricated according to the following steps: (i) the conversion of CHACs with hierarchically porous structures from calcium carbonate coatings (CCCs) by treatment with phosphate buffer solutions, and (ii) loading gentamicin into the CHACs. The main constituents in the CHACs were plate-like carbonated hydroxyapatite particles with a low crystallinity. Mesopores with a pore size of ∼3.8 nm and macropores (or apertures) with an aperture size of ∼1 μm existed within and among the plates, respectively. The hierarchically porous structures made the CHACs possess good drug loading–release properties. The sustained release of gentamicin from the Gent-CHACs inhibited significant bacterial adhesion and prevented biofilm formation against S. epidermidis (ATCC35984). In vitro cell tests indicated that human bone marrow stromal cells (hBMSCs) had good cell adhesion, spreading and proliferation on the Gent-CHACs. Moreover, the released gentamicin from the Gent-CHACs had no toxic effects on the hBMSCs. The good biocompatibility, drug delivery properties and bactericidal properties of the Gent-CHACs suggest that they have great potential as bone substitute materials.

Biofilms associated bacterial infections on material surfaces have become a tremendous biomedical challenge. Developing antimicrobial coatings on biomaterial surfaces and endowing them with decent mechanical stability and biofilm inhibition capabilities is an efficient way to resist bacterial attachment and biofilm formation. Herein, we integrated 2-hydroxyethyl methacrylate (HEMA) and D-amino acid mixtures based antibiofilm combinations with sulfobetaine methacrylate (SBMA) hydrogel coatings. The obtained hydrogel coatings demonstrated high stability in various transport and service environments. The proper incorporation of the HEMA achieves only ∼3% weight loss of SBMA hydrogel coatings after swelling, flushing and abrasion damages. In addition, both biofilm formation inhibiting D-amino acid mixtures and bacteria-killing gentamicin components were loaded in the coatings. The synergistic action of these two components was able to significantly reduce the bacterial numbers with up to 2.3 log reduction. The bacteria exposed to D-amino acid mixtures was difficult to form biofilm, which was more susceptive to the harm of gentamicin. This work provides an effective paradigm to integrate mechanically stable SBMA-HEMA hydrogel with natural D-amino acid mixtures based antibiofilm agents to generate biomedical surfaces to combat biofilms associated bacterial infections.

Antibiotic-loaded calcium phosphate cement (CPC) has emerged as a promising biomaterial for drug delivery in orthopedics. However, there are problems such as the burst release of antibiotics, low cumulative release ratio, inappropriate release cycle, inferior mechanical strength, and poor anti-collapse properties. In this research, montmorillonite-gentamicin (MMT-GS) was fabricated by solution intercalation method and served as the drug release pathways in CPC to avoid burst release of GS, achieving promoted cumulative release ratios and a release cycle matched the time of inflammatory response. The results indicated that the highest cumulative release ratio and release concentration of GS in CPC/MMT-GS was 94.1 ± 2.8 % and 1183.05 μg/mL, and the release cycle was up to 504 h. In addition, the hierarchical GS delivery system was divided into three stages, and the kinetics followed the Korsmeyer-Peppas model, the zero-order model, and the diffusion-dissolution model, respectively. Meanwhile, the compressive strength of CPC/MMT-GS was up to 51.33 ± 3.62 MPa. Antibacterial results demonstrated that CPC/MMT-GS exhibited excellent in vitro long-lasting antibacterial properties to E. coli and S. aureus . Furthermore, CPC/MMT-GS promoted osteoblast proliferation and exhibited excellent in vivo histocompatibility. Therefore, CPC/MMT-GS has favorable application prospects in the treatment of bone defects with bacterial infections and inflammatory reactions.

A series of gentamicin-modified anion exchange membranes (AEMs) immobilized by mussel-inspired dopamine coating is prepared in this work. The purpose is to improve antibacterial activity and fouling resistance of the commercial AEMs (AEM Type I, Fujifilm Co.). As a result of the applied modification, the hydrophilicity of membrane surface improved significantly. Thus, the as-prepared modified AEMs exhibit the good antifouling property, in electrodialysis (ED) experiments by using the sodium dodecylbenzenesulfonate and albumin from bovine serum as model foulants. Furthermore, taking into account the results of the antibacterial test, the resulting modified AEMs exhibit improved antibacterial property for Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). After a brief contact with bacteria, the antibacterial rates of modified AEMs coating with gentamicin-based polymer are 80.11% for E. coli and 90.66% for S. aureus , which are much higher than pristine AEM (6.28% and 9.07%), respectively. In addition, the modified membrane has good operational stability during ED process and recovery ability after contamination, and also maintains satisfactory electrochemical properties. This investigation highlights a design of simple strategy for the preparation of antimicrobial and antifouling AEM, which can be used for ED.

In this paper, Bi 2 S 3 nanospheres with size of 212 nm were prepared by a simple hydrothermal process. The selectively enhanced antibacterial effects of Bi 2 S 3 nanospheres with three classes of ineffective antibiotics, β-lactam (cefuroxime, CXM; cefotaxime , CTX and piperacillin , PIP), quinolone (ciprofloxacin, CIP) and aminoglycoside (gentamicin, GEN) against clinical isolated methicillin-resistant Staphylococcus aureus (MRSA) were investigated for the first time. GEN shows significantly synergistic growth inhibition against MRSA when combined with Bi 2 S 3 nanospheres, while CXM, CTX, PIP and CIP do not. Raman spectroscopy and Z potential studies reveal that Bi 2 S 3 could interact with GEN and the combination showed small electronegativity , which probably induced the increase of GEN content in cytoplasm of bacteria. Furthermore, the combination of Bi 2 S 3 nanospheres and GEN can destroy the bacterial membrane function and induce more bactericidal reactive oxygen generation than that of Bi 2 S 3 or GEN alone. The cytotoxicity test indicates that the combination of Bi 2 S 3 and GEN presented low toxicity to human normal hepatocyte L02. This work shows that Bi 2 S 3 nanospheres can be used to enhance the action of ineffective antibiotic GEN against MRSA, thus strengthening the antibiotic capacity for fighting MRSA infections.

Biofilms contribute to persistent bacterial infections as well as formidable resistances to conventional antibiotics. However, it is still a major challenge to establish an advanced antibacterial nanoplatform that can efficiently eradicate biofilms while overcoming bacterial resistances. Taking advantage of the stimuli-responsive technique and the magnetic guidance strategy, here we present a highly efficient nanoplatform for planktonic inactivation and biofilm disruption. The multilayer films consisting of antibiotic gentamicin (Gen), tannic acid, and silver nanoparticles (AgNPs) were fabricated and coated on magnetic nanoparticles via electrostatic interactions. To achieve controlled drug release and improved biocompatibility, biodegradable hyaluronic acid was capped on the outer surface as a responsive shell. In vitro release profiles suggested that the nanocomposites showed both enzyme and pH-responsive release properties. The nanoplatform could be employed as a powerful nanocarrier for small molecular Gen and AgNPs delivery and on-demand release in response to bacterial infection microenvironment. The nanocomposites also showed satisfying antibacterial capacities against planktonic Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli. Intriguingly, with magnetic field navigation (NdFeB, 2000 gauss), the nanocomposites could be guided to handily penetrate into S. aureus biofilm and performed dual-responsive release, showing significantly enhanced biofilm disruption. Moreover, excess reactive oxygen species production resulting from the nanocomposites contributed to the decomposition of biofilm matrix and ultimate biofilm eradication. As a consequence, the ingenious antibacterial nanoplatform could be promising for combating biofilm infections while overcoming bacterial resistances with extra environmental factors such as magnetic field.
